Soy Oligosaccharides and Soluble Non-starch Polysaccharides: A Review of Digestion, Nutritive and Anti-nutritive Effects in Pigs and Poultry

  • Soybean contains a high concentration of carbohydrates that consist mainly of non-starch polysaccharides (NSP) and oligosaccharides. The NSP can be divided into insoluble NSP (mainly cellulose) and soluble NSP (composed mainly of pectic polymers, which are partially soluble in water). Monogastric animals do not have the enzymes to hydrolyze these carbohydrates, and thus their digestion occurs by means of bacterial fermentation. The fermentation of soybean carbohydrates produces short chain fatty acids that can be used as an energy source by animals. The utilization efficiency of the carbohydrates is related to the chemical structure, the level of inclusion in the diet, species and age of the animal. In poultry, soluble NSP can increase digesta viscosity, reduce the digestibility of nutrients and depress growth performance. In growing pigs, these effects, in particular the effect on gut viscosity, are often not so obvious. However, in weaning piglets, it is reported that soy oligosaccharides and soluble NSP can cause detrimental effects on intestinal health. In monogastrics, consideration must be given to the anti-nutritive effect of the NSP on nutrient digestion and absorption on one hand, as well as the potential benefits or detriments of intestinal fermentation products to the host. This mirrors the needs for i) increasing efficiency of utilization of fibrous materials in monogastrics, and ii) the maintenance and improvement of animal health in antibiotic-free production systems, on the other hand. For example, ethanol/water extraction removes the low molecular weight carbohydrate fractions, such as the oligosaccharides and part of the soluble pectins, leaving behind the insoluble fraction of the NSP, which is devoid of anti-nutritive activities. The resultant product is a high quality soy protein concentrate. This paper presents the composition and chemical structures of carbohydrates present in soybeans and discusses their nutritive and anti-nutritive effects on digestion and absorption of nutrients in pigs and poultry.

Water-soluble ginseng oligosaccharides protect against scopolamine-induced cognitive impairment by functioning as an antineuroinflammatory agent

  • Background: Panax ginseng root is used in traditional oriental medicine for human health. Its main active components such as saponins and polysaccharides have been widely evaluated for treating diseases, but secondary active components such as oligosaccharides have been rarely studied. This study aimed to assess the impact of water-soluble ginseng oligosaccharides (WGOS), which were isolated from the warm-water extract of Panax ginseng root, on scopolamine-induced cognitive impairment in mice and its antineuroinflammatory mechanisms. Methods: We investigated the impact of WGOS on scopolamine-induced cognitive impairment in mice by using Morris water maze and novel object recognition task. We also analyzed the impact of WGOS on scopolamine-induced inflammatory response (e.g., the hyperexpression of proinflammatory cytokines IL-$1{\beta}$ and IL-6 and astrocyte activation) by quantitative real-time polymerase chain reaction and glial fibrillary acid protein (GFAP) immunohistochemical staining. Results: WGOS pretreatment protected against scopolamine-induced learning and memory deficits in the Morris water maze and in the novel object recognition task. Furthermore, WGOS pretreatment downregulated scopolamine-induced hyperexpression of proinflammatory cytokines interleukin (IL)-$1{\beta}$ and IL-6 mRNA and astrocyte activation in the hippocampus. These results indicate that WGOS can protect against scopolamine-induced alterations in learning and memory and inflammatory response. Conclusion: Our data suggest that WGOS may be beneficial as a medicine or functional food supplement to treat disorders with cognitive deficits and increased inflammation.

Quantification of Inulo-oligosaccharides Using High pH Anion Exchange Chromatography with Pulsed Amperometric Detector (HPAEC-PAD)

  • Inulo-oligosaccharides (IOS, $F_n$, n=2-6) were purified from enzymatic hydrolysates of water-soluble extract of Jerusalem artichoke tubers. Quantification of inulo-oligosaccharides was done using high pH anion exchange chromatography with pulsed amperometric detector (HPAEC-PAD) at the concentration range of 10-100 mg/L, which was compared with that of fructo-oligosaccharides (FOS, $GF_n$, n=1-7). Peak areas per mg IOS were higher than FOS at the same degree of polymerization (DP). Specific peak areas of IOS increased proportionally as DP increased up to six, in contrast to FOS showing no linearity.

  • Chitin, a poly $\beta$-(1longrightarrow14)-N-acetyl-D-glucosamine, is best known as a cell wall component of fungi and as a skeletal materials of invertebrates. Chitosan is derived from chitin by deacetylation in the presence of alkali. Chitosan has been developed as new physiological materials since it possesses antibacterial activity, hypocholesterolemic activity and antihypertensive action. However, the actions of chitosan in vivo still remain ambiguous as the physiological functional properties because most animal intestines, especially the human gastrointestinal tract, do not possess enzyme such as chitosanase which directly degrade the $\beta$-glucosidic linkage in chitosan, and consequently the unbroken polymers may be poorly absorbed into the human intestine. Therefore, recent studies as chitosan have attracted interest for chitosan oligosaccharides, because the oligosaccharides process not only water-soluble property but also versatile functional properties such as antitumor activity, immune-enhancing effects, enhancement of protective effects against infection with some pathogens in mice and antimicrobial activity (Kingsnorth et al., 1983, Mori et al., 1997).   • To obtain low molecular weight water soluble chitosan (LMWSC) with various molecular weights, chitosan oligosaccharides (COS) with lactic acid was separated by using ultrafilteration technique and LMWSC with a free amine group was prepared by the novel salts-removal method. The characterization of LMWSC removed the lactic acid and degree of deacetylation (DDA) were identified by FT-IR and $^1H-NMR$ spectra. Polydispersity index (PDI) was $1.278{\sim}1.499$, which indicates a relatively molecular weight distribution. To identify the potential as a gene carrier, we confirmed the transfection efficiency of COS fractioned according to molecular weight successfully and the salt-removed LMWSC using 293T cell. Also, LMWSC derivatives prepared for improvement transfection efficiency were evaluated using Balb/C mice.

  • Chitosan is a natural biopolymer of N-acetylglucosamine (-1,4 linked 2-acetamido-D-glucose), which is obtained after alkaline deacetylation of the chitin derived from the cell walls of lower plants and in the skeletal tissues of lower animals including arthropods and mollusks. In the recent studies, chitin and chitosan have attracted interest for converting them to oligosaccharides because the oligosaccharides are not only water-soluble but also possess versatile functional properties such as antimicobial activity (Jeon and Kim, 2000; Jeon et al., 2001).   • To develop water-soluble chitosan as an effient gene delivery carrier, chitosan oligosaccharides (COSs) with various molecular weights (MW) were studied for gene transfection agents. MWs of COSs fractionated by ultrafiltration techniques were identified as narrow MW distributions with the average MW ranging from 1 to 10 kDa through gel permeation chromatography (GPC) measurement depending on the applied ultrafiltration membranes. Their structural characterizations were analyzed by FTIR spectrophotometer and $^1H$ NMR. The degree of deacetylation was determined by UV spectroscopy showing the degree of deacetylation above 90%. The relative cell viabilities were maintained over 100% (10 mg/mL), independent of the MW of the fractionated COSs. The fractionated COSs of 10 mg/mL concentration with narrow MW distributions showed non-cytotoxicity in Caco-2 cells.

  • To enjoy a healthy life, it is important to have a well-balanced diet. However, in today's society, there is an increase in the consumption of preprocessed foods and frequency of eating out. Also the western diet, which is becoming move popular worldwide, contains relatively high levels of protein and fat, and a low amount of fiber, Furthermore, the increased availability of favorite foods has created a condition were the individual diet is less variable. With these conditions, it is difficult to maintain a diet that is nutritionally balanced. With these unbalanced diets, which are difficult to change, there has been an increase in adult disease and health problems, such as colon and breast cancer, It is speculated that metabolites for carcinogens are produced from diet components and that intestinal bacteria contribute to the production of these metabolites. Therefore, it is necessary to evaluate the relationships between health, diet, and intestinal microflora. Soybean oligosaccharide is composed of water-soluble saccharides that have been extracted from soybean whey, a by-product from the production of soy protein. This is mainly a mixture of mono-, di-, tri-, and tetrasac-charides, with the principle components being the oligosaccharide raffinose and stachyose. When consumed by humans, the oligosaccharides cannot be digested in the human duodenal and small intestinal mucosa, and these are selectively utilized by beneficial bifidobacteria in intestines. The results of acute and subacute toxicity tests, soy-bean oligosaccharides were nonpoisonous. Soybean oligosaccharides promote the growth of indigenous bifido-bacteria in the colon which by their antagonistic effects, suppress the activity of putrefactive bacteria. Also, they reduce toxic metabolites, detrimental enzymes and plasma lipid, and increase in the frequency of bowel evacuation and fecal quantities. Consequently, soybean oligosaccharides as functional foods components have potential roles in the prevention and medical treatment of chronic adult diseases. The study of processing property and physiological function of soybean oligosacchavides and development of high oligosaccharide variety allow the creation of new and exciting foodstuffs that aye functional healthy.

Antioxidative Effects of Water-Soluble Chitinous Compounds on Oxidation of Low Density Lipoprotein in Macrophages (대식세포에서 지단백 산화에 대한 수용성 Chitinous Compounds의 항산화 효과에 대한 연구)

  • It has been proposed that oxidative modification of LDL (oxLDL) plays a significant role in the pathogenicity of atherogenesis. We tested the hypothesis that chitin and chitosan may function as antioxidants with respect to 0.1 mg cholesterol/ml LDL incubated with 5 $\mu$ M Cu$^2$$^{+}$alone or in the P338Dl mouse macrophage system using L-ascorbic acid as a standard classical antioxidant. The degree of oxLDL formation was ascertained by the relative electrophoretic mobility (rEM) in the combination of thiobarbituric acid reactive substances (TBARS) levels, and the cytotoxicity of oxLDL was detected by macrophage viability. The oxLDL uptake and foam cell formation of macrophages were measured by Oil Red O staining. Incubation with Cu$^2$$^{+}$and macrophages increased rEM of LDL and stimulated TBARS formation. Culture of macrophages with LDL in the presence 5 $\mu$ M Cu$^2$$^{+}$induced macrophage death. In cell-free system 200 $\mu$g/ml water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation. Water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation near-completely relative to L-ascorbic acid, whereas water-soluble chitin and chitin-oligosaccharide had no measurable antioxidant effect. In macrophage system water-soluble chitosan and chitosan-oligosaccharide blocked oxidation of LDL with a significant increase in cell viability, and decreased TBARS in medium. As for the inhibitory effect on macrophage foam cell formation, chitosan and its oligosaccharide, but not watersoluble chitin, revealed the effectiveness. The endothelial expression of lectin-like oxLDL receptor-1 (LOX-1) was tested by Western blot analysis, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide blocked LOX-1 expression. These results indicate that water-soluble chitosan and its oligosaccharide showed the inhibitory effect on Cu$^2$$^{+}$-induced LDL oxidation of macrophages, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide had blocking effect on oxLDL receptor expression in the human umbilical vein endothelial system. Thus, water-soluble chitosan and its oligosaccharides possess anti-atherogenic potentials possibly through the inhibition of macrophage LDL oxidation or endothelial oxLDL receptor expression depending on chemical types.l types.

Dietary Fiber and Large Bowel Cancer

  • Large bowel cancer correlates tightly to dietary factors such as dietary fiber and fat. Dietary fiber prevents the large bowel cancer in different modes of action which depend upon physicochemical and fermentable properties. Water-soluble fiber is fermented easily by intestinal microbes producing short chain fatty acids ; in contrast, water-insoluble fiber occurs effectively more rapid transit time due to greater bulk of gut content, though it is unfermentable. Not only short chain fatty acid is utilized in the proximal and distal colon as primary energy source, but also it lowers pH in the colon to normalize cellular differentiation and helps to stimulate peri staltic movement by acting as an osmotic laxative. In particular, butyric acid may also regulate gene expression and cell growth, though it is an important respiratory fuel for the colonocyte. Since dietary fiber and non-digestible oligosaccharides are the major source of butyric acid, this provides a possible link between dietary fiber and oligosaccharide and prevention of large bowel cancer. But, as with many links between dietary fiber and large bowel cancer, a direct casual association has not been established. In addition, RDA of dietary fiber which is 20~25g/day for adult Japanese, appears to be reasonable for the defecation once daily and the prevention of large bowel cancer.
