Prevalence and Identification of Cryptosporidium spp. from Swine Slurry

  • Cryptosporidium spp. were detected in 17 of 135 swine lagoon samples from five farms by 18S ribosomal DNA locus and PCR. Seventeen positive samples identified were included two distinctive genotypes C. suis and Cryptosporidium sp. based on a small-subunit rRNA gene-based PCR-restriction fragment length polymorphism analysis. Cryptosporidium spp. were detected out of farrowing, farrowing and nursery (mix), and finishing. Prevalence rate was 12.6% with infection rates between 3.7 and 18.5%. We concluded that Cryptosporidium oocysts can persist in treated lagoon and potentially contaminate surface water through improper discharge. This study was undertaken for the evaluation of the infection status of the genotypes of Cryptosporidium spp. in swine lagoon.

  • The restriction of the use of hazardous substances in electrical and electronic equipment legislation mandates the substitution of lead and other hazardous substances in electronics products by July 2006. Due to this legislative pressure, the electronics industry is moving to adoption of lead free solders. In this paper, we investigated a flux to restrict generating electrochemical migration in lead-free solder. The lead-free solders used in this study were Sn-0.7Cu-0.01P and Sn-3.0Ag-0.5Cu. To measure the resistance of electrochemical migration, the dew-cycle test and water drop test were adopted. As the result, now flux having high durable of electrochemical migration was developed.

A Study on the Liquid Crystal Orientation Characteristics of the Inorganic NiOx Film with Aligned Nanopattern Using Imprinting Process (무기막 NiOx의 정렬 패턴 전사를 이용한 액정의 배향 특성 연구)

  • We demonstrate an alignment technology using an imprinting process on an inorganic NiOx film. The aligned nanopattern was fabricated on a silicon wafer by laser interference lithography. The aligned nano pattern was then imprinted onto the sol-gel driven NiOx film using an imprinting process at an annealing temperature of $150^{\circ}C$. After the imprinting process, parallel grooves had been formed on the NiOx film. Atomic force microscopy and water contact angle measurements were performed to confirm the parallel groove on the NiOx film. The grooves caused liquid crystal alignment through geometric restriction, similar to grooves formed by the rubbing process on polyimide. The liquid crystal cell exhibited a pretilt angle of $0.2^{\circ}$, which demonstrated homogeneous alignment.

  • The low and unstable yield of soybean has been a major problem in Japan. Excess soil moisture conditions are one of the major factors to restrict soybean productivity. More than 80 % of soybean crops are cultivated in converted paddy fields which often have poor drainage. In central and eastern regions of Japan, the early vegetative growth of soybean tends to be restricted by the flooding damage because the early growth period is overlapped with the rainy season. Field observation shows that induced excess water stress in early vegetative stage reduces dry matter production by decreasing intercepted radiation by leaf and radiation use efficiency (RUE) (Bajgain et al., 2015). Therefore, it is necessary to evaluate the responses of soybean growth for excess water conditions to assess these effects on soybean productions. In this study, we aim to modify the soybean crop model (Sinclair et al., 2003) by adding the components of the restriction of leaf area development and RUE for adaptable to excess water conditions. This model was consist of five components, phenological model, leaf area development model, dry matter production model, plant nitrogen model and soil water balance model. The model structures and parameters were estimated from the data obtained from the field experiment in Tsukuba. The excess water effects on the leaf area development were modeled with consideration of decrease of blanch emergence and individual leaf expansion as a function of temperature and ground water level from pot experiments. The nitrogen fixation and nitrogen absorption from soil were assumed to be inhibited by excess water stress and the RUE was assumed to be decreasing according to the decline of leaf nitrogen concentration. The results of the modified model were better agreement with the field observations of the induced excess water stress in paddy field. By coupling the crop model and the ground water level model, it may be possible to assess the impact of excess water conditions for soybean production quantitatively.

Derivation of Ecological Protective Concentration using the Probabilistic Ecological Risk Assessment applicable for Korean Water Environment: (I) Cadmium

  • Probabilistic ecological risk assessment (PERA) for deriving ecological protective concentration (EPC) was previously suggested in USA, Australia, New Zealand, Canada, and Netherland. This study suggested the EPC of cadmium (Cd) based on the PERA to be suitable to Korean aquatic ecosystem. First, we collected reliable ecotoxicity data from reliable data without restriction and reliable data with restrictions. Next, we sorted the ecotoxicity data based on the site-specific locations, exposure duration, and water hardness. To correct toxicity by the water hardness, EU's hardness corrected algorithm was used with slope factor 0.89 and a benchmark of water hardness 100. EPC was calculated according to statistical extrapolation method (SEM), statistical extrapolation $method_{Acute\;to\;chronic\;ratio}$ ($SEM_{ACR}$), and assessment factor method (AFM). As a result, aquatic toxicity data of Cd were collected from 43 acute toxicity data (4 Actinopterygill, 29 Branchiopoda, 1 Polychaeta, 2 Bryozoa, 6 Chlorophyceae, 1 Chanophyceae) and 40 chronic toxicity data (2 Actinopterygill, 23 Branchiopoda, 9 Chlorophyceae, 6 Macrophytes). Because toxicity data of Cd belongs to 4 classes in taxonomical classification, acute and chronic EPC (11.07 ${\mu}g/l$ and 0.034 ${\mu}g/l$, respectively) was calculated according to SEM technique. These values were included in the range of international EPCs. This study would be useful to establish the ecological standard for the protection of aquatic ecosystem in Korea.

  • Environmental-friendly nontocxic flame retardant paint which can overcomes the restriction of harmful materials for human body and environments such as Pb, Hg, Cd, $C^{+6}$, PBB/PBDE by EU and domestic Ministry of Environment was developed. Developed paint is the water-soluble organic inorganic hybrid material that VOC(volatility organic compound) discharge is low, and that human riskiness and environmental pollution is minimized not using the kinds of halogen materials. $Mg(OH)_2$, $Sb_{2}O_{3}$, and Zinc borate were used as flame retardant materials, 2% Micell and water were used as binder and solvent, respectively. Results showed the optimum activity was obtained when the ratio of those frame retardant agents($Mg(OH)_2$, $Sb_{2}O_{3}$, Zinc borate made by 1: 2: 2), binder(2% Micell) and water was 1: 0.5: 0.5.

Bioremediation Potential of a Tropical Soil Contaminated with a Mixture of Crude Oil and Production Water

  • A typical tropical soil from the northeast of Brazil, where an important terrestrial oil field is located, was accidentally contaminated with a mixture of oil and saline production water. To study the bioremediation potential in this area, molecular methods based on PCR-DGGE were used to determine the diversity of the bacterial communities in bulk and in contaminated soils. Bacterial fingerprints revealed that the bacterial communities were affected by the presence of the mixture of oil and production water, and different profiles were observed when the contaminated soils were compared with the control. Halotolerant strains capable of degrading crude oil were also isolated from enrichment cultures obtained from the contaminated soil samples. Twenty-two strains showing these features were characterized genetically by amplified ribosomal DNA restriction analysis (ARDRA) and phenotypically by their colonial morphology and tolerance to high NaCl concentrations. Fifteen ARDRA groups were formed. Selected strains were analyzed by 16S rDNA sequencing, and Actinobacteria was identified as the main group found. Strains were also tested for their growth capability in the presence of different oil derivatives (hexane, dodecane, hexadecane, diesel, gasoline, toluene, naphthalene, o-xylene, and p-xylene) and different degradation profiles were observed. PCR products were obtained from 12 of the 15 ARDRA representatives when they were screened for the presence of the alkane hydroxylase gene (alkB). Members of the genera Rhodococcus and Gordonia were identified as predominant in the soil studied. These genera are usually implicated in oil degradation processes and, as such, the potential for bioremediation in this area can be considered as feasible.

  • In this study, a study on how to use a fire hydrant as a heat wave reduction facility through hydraulic analysis of the water supply pipe network was conducted. Assuming that the fire hydrant installation point is open for heat wave reduction, the water pressure at each point was derived. And the reduction rate of the temperature according to the hydrant watering was compared with the watering area according to the operation of the watering truck. The watering area according to the opening of the fire hydrant was calculated by deriving the pressure value at the node where the fire hydrant was installed through hydraulic analysis of the water pipe network, and then using the watering radius relational expression according to the pressure value. As a result of applying the proposed methodology to two real city areas, the temperature reduction effect of the watering method by a fire hydrant can be derived lower than the watering method by a watering truck according to the difference in the absolute watering area. However, unlike a watering truck, a fire hydrant does not have a relative restriction on the amount of water supply and is expected to allows continuous divided spraying of the same area.

  • Mitochondrial DNAs of two Mdh allelotypes of the dark chub, Z. temmincki inhabiting in Korean fresh water, were analysed. Samples of each type were collected from four populations, and the fragment patterns for mtdNA of each type were explained from 7 of the eleven restriction enzymes with hexanucleotide recognition site. Genome size was approximately 16.7 kilobases. The highly typical mtdNA fragments of each type were discovered in digestion profiles produced by Eco RI and Pst I enzyrnes. The comparisons of restriction fragment patterns and relative digestion maps permitted the estimation of fragment homology (F) and nucleotide sequence divergence(p). Between the two identical types, sequence divergence(p) was 0.128(MS), and 0.045(MM), ; between the two different types, 0.195 (range 0.177-0.226). These result may provide a distinct difference more than the value derived from allozyrne analysis, and a powerful new molecular approach for assessing genetic-evolutionary relationship among fishes.

Numerical Study on Designing Truncated Mooring Lines for FPSO Stability Analysis (FPSO 안정성 평가를 위한 절단계류선 모델링 수치 연구)

  • In this paper, a numerical analysis for an internal turret moored vessel located at a 400-m water depth is conducted. The target vessel has an internal turret that is located at the 0.2 Lpp position from the fore-side, with $3{\times}4$ complex mooring lines installed around the turret circumference. To investigate the motion response of the vessel and the structural reliability of the lines, model tests were conducted. The KRISO ocean basin has a water depth of 3.2 m, which represents 192m using a scaling of 1:60. In order to precisely represent the real-scale condition, equivalent mooring lines needed to be designed. Truncated mooring lines were designed to supplement the restriction of the flume's water depth and increase the reliability of the model testing. These truncated mooring lines were composed of two different chains in order to match the pre-tension, simultaneously restoring the curve and variation in the effective line tension. The static similarities were compared using a static pull-out test and free decaying test, and the dynamic similarities were matched via a regular wave test and combined environments test. Consequently, the designed truncated mooring system could represent the prototype mooring system relatively well in the aspects of kinematics and dynamics.
