Simultaneous qualitative and quantitative analysis of morroniside and hederacoside D in extract mixture of Cornus officinalis and Stauntonia hexaphylla leaves to improve benign prostatic hyperplasia by HPLC-UV

  • With the improvement in the standard of living and extension of life expectancy, the incidence of prostate diseases has increased yearly, thus becoming a serious disease affecting the health of men. The extract mixture of Cornus officinalis and Stauntonia hexaphylla leaves is a developed functional food formula to improve prostate health. This study developed a simultaneous analytical method of bioactive compounds for quantifying the mixture of Cornus officinalis and S. hexaphylla leaves using high-pressure liquid chromatography-ultraviolet (HPLC-UV). HPLC analytical condition was performed on a Hector C18 column with a mobile phase of 0.1 % formic acid in water (A) and 0.1 % formic acid in acetonitrile (B) under the following gradient conditions: 0-50 min, 12 %-40 % (B) at a flow rate of 1.0 mL/min. Meanwhile, this method was validated properly and successfully used to quantify the bioactive components of morroniside and hederacoside D in 20 sample batches and assess the quality of different ages and seasons of S. hexaphylla leaves. The result showed that the content of morroniside in the extract mixture of Cornus officinalis and S. hexaphylla leaves ranged from 1.38-1.62 mg/g, and the hederacoside D ranged from 28.42-32.02 mg/g, suggesting that this novel analytical method will be suitable for the quality control of the extract mixture to improve benign prostatic hyperplasia.

Visitor Behavior and Satisfaction in Suburban Recreational Forests - Focused on the suburbs of Seoul - (도시근교형 자연휴양림 이용 행태와 만족도 분석 - 서울 근교를 중심으로 -)

  • This study researches attributes , behaviors , satisfaction factors and degrees of curiosity of visitors to recreational forests in the suburbs of a large city. It aims to present suggestions for urban forest development and management policy by offering basic data which help to plan, design and manage recreational forests to increase the quality of these environments. The results are as follows First, in attribute, the visiting rate of males is higher than that of females, and the main users are in their thirties and forties. Sixty percent of visitors graduated from university and their rate of employment is evenly distributed. 95 percent of visitors are residents of the metropolitan area. In terms of behavior, major visitors are family units visiting during summer seasons and for overnight stays. 75% are re-visitors. 85% of visitors came to escape the city with families and friends, keep in good health and experience nature. Second, to extract the factors affecting visitor satisfaction in recreational forests, the natural environment, facilities, and management/use systems were identified as independent variables, while subordinate satisfactions were dependent variables. so regression analysis was used. Thus, the variables affecting the natural environment are quality of water, stream use, biodiversity, fresh air and landscape factors. The variables affecting facilities are puking, convenience, play facilities, sanitary arrangement and camping. Most important among the variables affecting management/use systems are educational facilities and access condition. On the basis of generalizing the study in the existing individual site, we must verify the visiting characteristics in recreational forests in the suburbs of a large city. Since development of recreational forests is understood as a sequence considering a site and a given condition, and since management and improvement must unfold according to these characteristics, a strategy is needed to reveal visitors' opinions about the site. Depending on the facilities and service, satisfaction of recreational forests is generally influenced by social and economic qualities. Also, this study can look into the effect according to use pattern motive and season. As suburban recreational forests have many overnight-users and younger men, programs suitable for these groups are needed. On the basis of variables affecting satisfaction according to natural environment, facilities, and use system, policies which can manage the natural environment and introduce educational programs are needed.

The Effect of Pyroligneous Acid on Turfgrass Growth - The Case of Yong-Pyong Golf Course Green - (목초액의 잔디 생육효과 - 용평 골프 코스 그린을 대상으로 -)

  • This study was carried out to investigate the effects of pyroligneous acid on turfgrass growth and to propose usage for sustainable management on Korean golf course green. Each plot was treated with 0, 300, 500, and 800 acid to water solution. On all the plots, turfgrass growth increment and visual quality were measured at fried times. The data were subjected to paired samples t-test and corelation analysis. The summarized results are as follows; 1) Leaf growth increment, density, and root spread depth of turfgrass treated with 1: 500 and 1: 800 diluted pyroligneous acid were significantly superior to the control. Particularly, 1: 500 diluted solution was superior to the others. 2) Color, texture, and uniformity of turfgrass treated with 1: 500 and 1: 800 diluted pyroligneous acid were significantly superior to the control. Especially, 1: 500 diluted solution tended to be superior to the others. 3) There were no symptoms of disease in all plots treated with the diluted pyroligneous acid. 1 500 and 1: 800 diluted pyroligneous acid were not thought to trigger or promote disease. 4) Before and after investigating the effect of pyroligneous acid on turfgrass growth ,the soils were analyzed. In all treatments, the chemical properties of the soils did not change noticeably. The chemical properties of the soils might be not changed by the diluted pyroligneous acid treatments. 5) Considering the results mentioned above, 1: 500 diluted pyroligneous acid was the most effective. Though pyroligneous acid is an organic fertilizer and contains only a little nitrogen and phosphorus, 1: 500 diluted pyroligneous acid promoted turfgrass growth effectively. Thus this use might lead to a reduction in the amount of fertilizers used and result in ecologically responsive management of Korean golf courses.

Validation and Determination of Glycyrrhizic Acid as a Marker Substance in Bu-Zhong-Yi-Qi-Tang by HPLC/DAD (HPLC/DAD에 의한 보중익기탕 중의 글리시리진산 정량분석법의 확립)

  • A high performance liquid chromatographic (HPLC) method for the determination of glycyrrhizic acid was developed for the quality control of traditional herbal medicinal preparation Bu-Zhong-Yi-Qi-Tang (BZYQT), which is well-known herbal medicine used as tonic. RP-HPLC analysis was carried out using Capcell pak $C_{18}$ MG column $(5\;{\mu},\;150{\times}4.6\;mm)$ and a mobile phase consisting of acetonitrile and water containing 0.03% phosphoric acid (pH 2.46) at a flow rate of 1.0 ml/min. The optimum wavelength for the detection of the glycyrrhizic acid was found at 250 nm using diode-array UV/VIS detector. The glycyrrhizic acid in BZYQT shows good linearity $(r^2>0.999)$ in the range of $15\;{\mu}g/ml$ to 500 ${\mu}g/ml$. The limit of detection (LOD) was less than 5 ng and R.S.D for intra-day and inter-day reproducibility was less than 7%. The mean recovery of the glycyrrhizic acid was $97.3{\sim}113.0%$. These results suggest that the developed HPLC method is simple and efficient, and could be contributed for the quality control of commercial BZYQT products.

A convergence study on dental infection management awareness and experience of dental users (치과이용자의 치과 감염관리 인식과 경험에 관한 융복합 연구)

  • The study investigated the perception and experience of infection control targeting dental users. During July-August 2020, 198 adults over the age of 20 were surveyed on general characteristics, infection control awareness and experience, and improvement. Analysis was performed using PASW Statistics ver 18.0. The research results, 91% of dental users recognized that infection control was important. In the recognition of infection control were highly investigated oral treatment equipment sterilization, hand hygiene and glove replacement before and after treatment by dental staff. And dental users was relatively low the replacement of disposable gowns and safety glasses for each patient by medical staff. The dental staff are doing well in personal protection and instrument sterilization. Surface disinfection and water quality management needed improvement. It was meaningful to suggest improvement in infection control based on the perception and experience from the perspective of dental users. It is expected to be used as basic data necessary for high-quality medical services through infection control in dental medical institutions.

Quality Characteristics of Modified Doenjang and Traditional Doenjang (개량된장과 전통된장의 품질 특성)

  • This study was conducted to investigate the relationship between quality factors and palatability of modified Doenjang and traditional Doenjang. Fourteen types of Doenjang, including three modified Doenjang and 11 traditional Doenjang samples, were analyzed to evaluate their physicochemical and sensorial properties. There were differences in e-tongue, which indicates overall acceptability. Water contents, minerals, total sugar contents, salt contents, pH, titratable acidity, and free amino acids of Doenjang did not show significant differences. From the correlation and regression analysis, palatability was closely related to the e-tongue sensor such as $X_1$ (sourness), $X_3$ (saltiness), and $X_4$ (umami) to -0.772, -0.642, and 0.678, respectively. The regression equation for sensorial palatability (Y) was Y=$45.356-0.008X_1-0.010X_3$ with a coefficient of 0.882.

Research on Definition and Economics of Organic Farming Methods (유기농업의 정의와 경제성에 관한 연구)

  • The definitions of organic farming method has rather philosophical meaning than application of agricultural practices. The meaning of organic farming method has certainly basic differences in comparison with the conventional farming methods. The main definitions of organec farming method have certainly to reduce or not to apply any kinds of farm inputs materials by increasing quality of agricultural prod-ucts. However, the quality of produced food by organic farming method should be improved in compari-son with cinvethional farming method. By applying organic method, the qualith of food, envir-onment as well as spiritual attitude of human being should be improved in future. Actually, there are very wide fields of research on organic farming methods such as production tech-niques, soil and water conservation, plant and animal production, marketing and consumers behavior on organic products. In this article the present situation of organic farming methods in the United States of Americal is investigated in cimparison with conventional rice production. The production cost of rice per acre in the United States of America mde 58, 788 Won in 1986, while it was 121, 699 Won in Korea having about 107 percent higher in Korea than tham the U.S.A There is a larger rice farm cost analysis between conventional and organic rice farm in California. The cash cost pre 1 lbs of conventional rice made 4.86 $, while the organic rice was 6.96$ showing about 43.2%higher level of cost in organic products(1 lbs=0.45359kg) At present, there is less econmic advantage of organic rice production in California because of lower yields as well as price levels, Therefore, the total net return over cash cost peracer/yeat was a little lower in organic rice production than conventional rice prouction.

Effects of Pectin and Potato Starch on the Quality Characteristics of Low-Fat Pork Patties (펙틴 및 감자전분 첨가가 저지방 돈육 패티의 품질에 미치는 영향)

  • This study was conducted to investigate the quality characteristics of low-fat pork patties containing fat replacers. Pectin(PE) and potato starch(PO) were added as fat replacers and physicochemical properties of the patties were evaluated after oven-roasting or pan-frying. Uncooked and cooked pork patties formulated with the fat replacers were higher in moisture and ash content and lower in fat content than those of the control. The cooking yield and reduction in diameter of the patties were significantly improved by the addition of PE or PO and oven-roasting exhibited a higher cooking yield and lower reduction in diameter than pan-frying (p<0.05). Water holding capacity(WHC) and fat retention were highest in the patties formulated with PE, while the control had the lowest WHC when cooked by oven-roasting. The Hunter L (lightness) values were not different among the patties but the Hunter a (redness) and b (yellowness) values of the low fat pork patties were higher than those of the control and oven-roasting produced higher redness values than pan frying. Hardness and brittleness were lowest in the patties with PE, while cohesiveness and springiness showed no difference among the patties (p<0.05). The sensory analysis results showed that the use of PE or PO improved the overall acceptability of the low-fat pork patties. After 30 days of storage at $-20^{\circ}C$, pH and cooking yield decreased slightly, while hardness increased in all the patties.

Characterizing a Full Spectrum of Physico-Chemical Properties of Ginsenosides Rb1 and Rg1 to Be Proposed as Standard Reference Materials

  • Good manufacturing practice (GMP)-based quality control is an integral component of the common technical document, a formal documentation process for applying a marketing authorization holder to those countries where ginseng is classified as a medicine. In addition, authentication of the physico-chemical properties of ginsenoside reference materials, and qualitative and quantitative batch analytical data based on validated analytical procedures are prerequisites for certifying GMP. Therefore, the aim of this study was to propose an authentication process for isolated ginsenosides $Rb_1$ and $Rg_1$ as reference materials (RM) and for these compounds to be designated as RMs for ginseng preparations throughout the world. Ginsenoside $Rb_1$ and $Rg_1$ were isolated by Diaion HP-20 adsorption chromatography, silica gel flash chromatography, recrystallization, and preparative HPLC. HPLC fractions corresponding to those two ginsenosides were recrystallized in appropriate solvents for the analysis of physico-chemical properties. Documentation of the isolated ginsenosides was made according to the method proposed by Gaedcke and Steinhoff. The ginsenosides were subjected to analyses of their general characteristics, identification, purity, content quantitation, and mass balance tests. The isolated ginsenosides were proven to be a single compound when analyzed by three different HPLC systems. Also, the water content was found to be 0.940% for $Rb_1$ and 0.485% for $Rg_1$, meaning that the net mass balance for ginsenoside $Rb_1$ and $Rg_1$ were 99.060% and 99.515%, respectively. From these results, we could assess and propose a full spectrum of physicochemical properties for the ginsenosides $Rb_1$ and $Rg_1$ as standard reference materials for GMP-based quality control.

Influence of Milk Co-precipitates on the Quality of Restructured Buffalo Meat Blocks

  • Restructuring had made it possible to utilize lower value cuts and meat trimmings from spent animals by providing convenience in product preparation besides enhancing tenderness, palatability and value. Milk co-precipitates (MCP) have been reported to improve the nutritional and functional properties of certain meat products. This study was undertaken to evaluate the influence of incorporation of milk co-precipitates at four different levels viz. 0, 10, 15 and 20% on the quality of restructured buffalo meat blocks. Low-calcium milk co-precipitates were prepared from skim milk by heat and salt coagulation of milk proteins. Meat chunks were mixed with the curing ingredients and chilled water in a Hobart mixer for 5 minutes, followed by addition of milk co-precipitates along with condiments and spice mix and again mixed for 5 minutes. Treated chunks were stuffed in aluminium moulds and cooked in steam without pressure for 1.5 h. After cooking, treated meat blocks were compared for different physico-chemical and sensory attributes. Meat blocks incorporated with 10% MCP were significantly better (p<0.05) than those incorporated with 0, 15 and 20% MCP in cooking yield, percent shrinkage and moisture retention. Sensory scores were also marginally higher for meat blocks incorporated with 10% MCP than product incorporated with 15 and 20% MCP, besides being significantly higher than control. On the basis of above results 10% MCP was considered optimum for the preparation of restructured buffalo meat blocks. Instrumental texture profile analysis revealed that meat blocks incorporated with 10% MCP were significantly better (p<0.05) in hardness/ firmness than control although, no significant (p>0.05) differences were observed in cohesiveness, springiness, gumminess and chewiness of both type of samples.
