본 연구는 댐파괴나 급격한 방류로부터 발생되는 홍수파가 하도내에서 사류의 흐름으로 나타날 때, 이를 해석하기 위한 수치모형을 개발한 것이다. 기본방정식으로서는 dynamic wave equation을 사용하였고 이식을 general Preissmann sceme에 의하여 유한차분화 시켰으며 fore-sweep algorithm에 의하여 해석하였다. 본 모형을 Buffalo-Creek flood의 경우에 적용하고 실측치 및 DAMBRK에 의한 계산 결과치와 비교검토하여 모형의 적용성을 입증하였다. 국내 효기댐의 경우에 대하여서는 파괴당시의 실측자료를 기초로한 유출수문곡선을 유도하였고 댐 하류부의 홍수파 전달양상을 해석하였다.
Trichomonad species inhabit a variety of vertebrate hosts; however, their potential zoonotic transmission has not been clearly addressed, especially with regard to human infection. Twenty-one strains of trichomonads isolated from humans (5 isolates), pigs (6 isolates), rodents (6 isolates), a water buffalo (1 isolate), a cow (1 isolate), a goat (1 isolate), and a dog (1 isolate) were collected in Indonesia and molecularly characterized. The DNA sequences of the partial 18S small subunit ribosomal RNA (rRNA) gene or 5.8S rRNA gene locus with its flanking regions (internal transcribed spacer region, ITS1 and ITS2) were identified in various trichomonads; Simplicimonas sp., Hexamastix mitis, and Hypotrichomonas sp. from rodents, and Tetratrichomonas sp. and Trichomonas sp. from pigs. All of these species were not detected in humans, whereas Pentatrichomonas hominis was identified in humans, pigs, the dog, the water buffalo, the cow, and the goat. Even when using the high-resolution gene locus of the ITS regions, all P. hominis strains were genetically identical; thus zoonotic transmission between humans and these closely related mammals may be occurring in the area investigated. The detection of Simplicimonas sp. in rodents (Rattus exulans) and P. hominis in water buffalo in this study revealed newly recognized host adaptations and suggested the existence of remaining unrevealed ranges of hosts in the trichomonad species.
Present investigation was conducted to determine the post-thaw sperm motility and acrosomal damage of filtered and non-filtered frozen semen of Murrah buffalo bulls. Twenty semen ejaculates (from four Murrah buffalo bulls collected at weekly interval) were diluted in Tris egg yolk glycerol extender and divided into two parts. One was filtered through sephadex G-100 column and the other portion was kept as such (non-filtered). Both fractions were frozen in liquid nitrogen ($-196^{\circ}C$) by the standard method developed in the laboratory. After 24 h of freezing, non-filtered and filtered semen samples were thawed at $37^{\circ}C$ for 1 min. These samples were incubated at $37^{\circ}C$ in a water both. The different seminal characteristics i.e. percent progressive sperm motility, live and abnormal spermatozoa and spermatozoa with damaged acrosome were assessed at hourly interval till they remained motile. The filtered frozen and thawed semen showed significantly (p<0.05) high sperm viability and acrosomal integrity as compared to non-filtered semen.

Twenty crossbred steers ($400{\pm}40.1kg$ of initial body weight) were used to assess the effects of a dietary supplementation with crude glycerin (CG) as a substitute for corn grain on performance, carcass traits, and meat quality. Four isocaloric and isonitrogenous diets were offered to the experimental animals (5 steers per treatment) for 121 days using randomized complete block design. The steers individually received dietary treatments containing 0%, 7%, 14%, and 21% of CG (88.91% pure) on a dry matter (DM) basis. The diets were offered ad libitum as total mixed rations twice daily. Weight gain and carcass traits were determined. At the end of the experimental period, the harvest data and carcass characteristics of the steers were recorded, and meat quality was determined. No significant effect of CG inclusion was observed in any of the growth performance and carcass characteristics traits studied. Also, there were no apparent effects of diets (p>0.05) on meat quality (pH, water holding capacity, drip losses, and cooking losses). The study concluded that CG could be used as a substitute for corn grain up to the level of approximately 21% of DM in the diets of finishing steers.
Restructuring had made it possible to utilize lower value cuts and meat trimmings from spent animals by providing convenience in product preparation besides enhancing tenderness, palatability and value. Milk co-precipitates (MCP) have been reported to improve the nutritional and functional properties of certain meat products. This study was undertaken to evaluate the influence of incorporation of milk co-precipitates at four different levels viz. 0, 10, 15 and 20% on the quality of restructured buffalo meat blocks. Low-calcium milk co-precipitates were prepared from skim milk by heat and salt coagulation of milk proteins. Meat chunks were mixed with the curing ingredients and chilled water in a Hobart mixer for 5 minutes, followed by addition of milk co-precipitates along with condiments and spice mix and again mixed for 5 minutes. Treated chunks were stuffed in aluminium moulds and cooked in steam without pressure for 1.5 h. After cooking, treated meat blocks were compared for different physico-chemical and sensory attributes. Meat blocks incorporated with 10% MCP were significantly better (p<0.05) than those incorporated with 0, 15 and 20% MCP in cooking yield, percent shrinkage and moisture retention. Sensory scores were also marginally higher for meat blocks incorporated with 10% MCP than product incorporated with 15 and 20% MCP, besides being significantly higher than control. On the basis of above results 10% MCP was considered optimum for the preparation of restructured buffalo meat blocks. Instrumental texture profile analysis revealed that meat blocks incorporated with 10% MCP were significantly better (p<0.05) in hardness/ firmness than control although, no significant (p>0.05) differences were observed in cohesiveness, springiness, gumminess and chewiness of both type of samples.
To understand the caused for poor response to superovulation in water buffalo compared to crossbred cows, follicular events, before start of superovulation, during superovulation and after superovulation were compared. Follicular development was monitored a day before start of superovulation, daily upto superestrus and on the day of flushing. A real time B mode diagnostic instrument equipped with a linear array, 5 MHz transducer was used in five crossbred cows and five Murrah buffaloes. Crossbred cows yielded significantly (p < 0.01) higher number of corpora lutea than buffaloes (21 vs 10). The mean number of small size (2 to 5 mm); medium size (6 to 9 mm) and large size $({\geq}10mm)$ follicles, a day before start of superovulation were almost similar or even slightly higher in buffalo. Though initial shift in the mean number of follicles was higher in buffalo than cow, yet, from Day 2 to Day 3 of the treatment, the average increase in medium (3.2 vs 1.2) and large size (5.0 vs 2.0) follicles was higher in cows than buffaloes. The mean number of medium and large size follicles was 9.8 and 14.4 in cows and 6.4 and 7.6 in buffaloes. On the day of flushing, the number of large size follicle was more in buffaloes than cows, indicating the ovulation problem in this species. The major conclusion from this investigation was that, a day before start of superovulatory treatment, the number of small and medium size follicles was slightly higher in buffaloes, even then superovulatory response was better in cows, due to shift, recruitment and passage of follicles from smaller size to larger size from Day 2 of treatment. Ovulation problem in buffaloes was also responsible for lower superovulatory responses as revealed by the presence of higher number of large size follicles on the day of flushing.
Six adult female Murrah buffaloes of about 12 years were exposed to solar radiation during summer when minimum and maximum ambient temperatures were 27.1 and $44.1^{\circ}C$, respectively. The skin surface temperature at forehead, middle pinna, neck, rump, foreleg, hind legs were recorded using non-contact temperature measuring instrument and respiration rate and rectal temperature were measured throughout the 24 hours starting from 6:30 AM. The diurnal fluctuations and temperature gradients have been reported for buffaloes. During summer when ambient temperature and solar radiation was maximum, adult buffaloes were not able to maintain their thermal balance even after increasing the pulmonary frequency 5 - 6 times. The changes in skin temperature at various sites indicate that the temperature of skin surface not only varies in relation to exposure but also due to water diffusion and evaporation.
An experiment was conducted to study the effect of feeding ammoniated wheat straw treated with and without HCl on meat quality and various sensory attributes of growing male buffalo (Bubalus bubalis) calves. Due to urea-ammoniation, the CP content of wheat straw increased from 2.90 to 6.96%. The addition of HCl along with urea during urea-ammoniation further increased the CP content to 10.09%. The proximate composition (% fresh basis) of psoas major, longissimus dorsi and semitendinosus were comparable among the groups. However, comparatively higher cumulative muscle mean protein and ash percentage and lower moisture percentage in groups II and III in comparison to group I indicated the desirable effect of feeding AWS and HCl-AWS. The cumulative muscle mean pH in group III (5.54) was comparatively lower than group I (5.65) and II (5.62). The cumulative group mean pH and water holding capacity (% water retained) in semitendinosus was comparatively lower than psoas major and longissimus dorsi. The cumulative muscle mean SSP (g%) was significantly (p<0.05) lower in group III (41.78) than group I (42.45). The cumulative group mean cooking loss (%) was significantly (p<0.01) lower in psoas major (31.61) and longissimus dorsi (29.78) than semitendinosus (35.10). The cumulative group mean SFV (kg/$cm^{2}$) of psoas major, longissimus dorsi and semitendinosus were 6.38, 6.32 and 6.56, respectively, being lowest in psoas major and highest in semitendinosus. However, the cumulative muscle mean SFV (kg/$cm^{2}$) in group I, II and III were 6.53, 6.56 and 6.17, respectively, being lowest in group III and highest in group II. The scores of the cooked (2% common salt) buffen for various sensory attributes viz. appearance, flavour, juiciness, texture, mouth coating and overall palatability were comparable among the groups. Results suggested that feeding of ammoniated wheat straw treated with and without HCl to growing male buffalo calves for 180 days had no adverse effect on the meat quality and various sensory attributes.
Eighteen water buffalo calves of Nili-Ravi breed (about 15 months age and of $147{\pm}12$ kg average body weight and mixed sex) were used with six animals on each treatment. All the animals were fed long wheat straw for ad libitum intake as the basal ration. Animals in group-I were supplemented with ad libitum amount of urea molasses block having cotton seed meal (CSMB) while the animals in group-II were supplemented with ad libitum urea molasses block containing sunflower seed meal (SFMB) and group III animals were supplemented with a fixed amount of commercial concentrate feed (CCF). The experiment lasted for a period of 80 days (April to June). Results revealed a significantly decreased total feed intake (wheat straw+supplements) in group-III ($1,666{\pm}52$ g/h/d) as compared to group I ($2,299{\pm}194$ g/h/d) and group-II ($2,193{\pm}230$ g/h/d). Average daily supplement intakes were $891{\pm}87$; $666{\pm}104$ and $593{\pm}0$ grams per head in group I, II and III, respectively. Supplement intakes among groups were different (p<0.05). Average daily body weight gains (g/h) were $214{\pm}25$, $174{\pm}23 $and $183{\pm}24$ for group I, II and III, respectively. Feed conversion ratio (FCR) was found to be $10.74{\pm}1.12$, $12.60{\pm}0.88$ and $9.90{\pm}1.33$ grams for group No. I, II and III, respectively. The economic net benefit of live weight gain of calves were 7.63, 6.11 and 7.33 rupees/h/d for group No. I, II and III, respectively. Thus SFM can replace CSM and urea molasses blocks can replace commercial concentrates as supplement to basal ration of wheat straw.
Ovarian follicular fluid contains both stimulatory and inhibitory agents that influence the growth and maturation of oocyte. In the present study, an attempt was made to isolate and study the biological properties of ovarian follicular fluid peptide(s) in buffaloes. Bubaline ovarian follicular was made steroid- and cell-free. A protein fraction was obtained by saturation (30-35% level) of the follicular fluid with ammonium sulfate. The protein fraction was purified with Sephadex-G 50 gel filtration chromatography and a single peak was obtained in the eluant volume, which was lyophilized. SDS-PAGE of the lyophilized fraction revealed a single band and the molecular weight of the peptide was 26.6 kDa. The peptide stimulated the cumulus cell expansion and in vitro maturation rate of oocytes in buffaloes in a dose dependent manner when it was incorporated at different dose levels (0, 10, 25, 50, 100 and 1,000 ng $ml^{-1}$ of maturation medium). The basic culture medium consisted of TCM 199 with Bovine serum albumin (0.3%). The in vitro maturation rates were comparable to those obtained with a positive control medium (TCM 199+20 ng EGF $ml^{-1}$+steer serum (20%)). Further purification and biological assays may throw more light on the nature and functions of this peptide.
