Studies on the Development of Food Resources from Waste Seeds V. Chemical Composition of Water-melon Seed (폐기종실(廢棄種實)의 식량자원화(食糧資源化)에 관(關)하여 제(第)5보(報) : 수박씨의 화학적(化學的) 조성(組成))

  • An attempt was made to find out the possibility of utilizing water-melon seed as resources of food fats and protein. The water-melon seed contained 40.40% of crude fat and 28.36% of crude protein. The lipid fraction obtained by silicic acid column chromatography was composed of about 97.35% neutral lipid, and the main components of neutral lipid by thin layer chromatography were triglyceride(50.40%), diglyceride(21.84%) and sterol(11.48%). The predominant fatty acids of total and major lipid classes were linoleic acid(55.30-67.85%), palmitic acid(12.07-28.12%) and oleic acid(9.06-16.40%), whereas stearic acid and linolenic acid were detected as small amounts. The salt soluble protein of watermelon seed was highly dispersible in 0.02M sodium phosphate buffer containing about 0.7M $MgSO_4$, and the extractability of seed protein was about 27%. Glutamic acid and arginine were major amino acids, and the essential amino acids such as lysine, threonine, valine, methionine, isoleucine, leucine and phenylalanine were also detected. The electrophoretic analysis showed 6 bands in water-melon seed protein, and the collection rate of the main protein fraction purified by sephadex G-100 and G-200 was 52.4%. The amino acids of the main fraction protein were also mainly composed of glutamic acid and arginine. The molecular weight for the main protein of the water-melon seed was estimated to be 120,000.

Enhanced Anaerobic Digestion Efficiency of Food Waste by Seaweed Addition (해조류 첨가를 통한 음식물쓰레기의 혐기성소화 효율 증대)

  • In this study, we investigated the effect of seaweed (SW) addition on the anaerobic digestion of food waste (FW). Anaerobic batch experiments were carried out at various substrate concentrations (2.5 to 10.0 g VS/L) and mixing ratios (FW:SW=100:0, 75:25, 50:50, 25:75 and 0:100 on VS basis) of FW and SW. The methane yield of FW alone was 394, 377, 276, $49mL\;CH_4/g\;VS_{added}$ at each substrate concentration (2.5 to 10.0 g VS/L). In cases of co-digestion, methane yield decreased (up to 15 %) with increasing mixing ratio of SW at low substrate concentration (2.5 to 5.0 g VS/L), while it increased (up to 240 %) at high substrate concentration (7.5 to 10.0 g VS/L). The synergistic effect was calculated based on the amount of methane generated from the single-feedstock digestion of FW and SW. The synergistic effect was not found at 2.5 and 5.0 g VS/L. However, the synergistic effect increased (up to 25% = synergistic increment/total methane production at 10.0 g VS/L, FW:SW=50:50) with increasing the ratio of seaweed at 7.5 and 10.0 g VS/L. At 10.0 g VS/L of FW alone, the accumulated amount of organic acids was 7,426 mg COD/L, which was decreased to 2,346 mg COD/L by seaweed (FW:SW=50:50) addition. The reason for the synergistic effect was to control the production rate of the organic acids by adding SW that has a relatively lower biodegradability compared to FW.

Recovery of high-purity phosphoric acid from the waste acids in semiconductor manufacturing process (반도체(半導體) 제조공정(製造工程)에서 발생하는 혼산폐액(混酸廢液)으로부터 고순도(高純度) 인산회수(燐酸回收))

  • The waste solution discharged from the LCD manufacturing process contains acids like nitric, acetic and phosphoric acid and metal ions such as Al, Mo and other impurities. It is important to remove impurities less than 1 ppm in phosphoric acid to reuse as an etchant because the residual impurities even in sub-ppm concentration in semiconductor materials play a major role on the electronic properties. In this study, a mixed system of solvent extraction, diffusion dialysis and ion-exchange was developed to commercialize in an efficient system fur recovering the high-purity phosphoric acid. By vacuum evaporation, almost 99% of nitric and acetic acid was removed. And by solvent extraction method with tri-octyl phosphate (TOP) as an extractant, the removal of acetic and nitric acid from the acid mixture was achieved effectively at the ratio A/O=1/3 with 4th stage of extraction stage. About 97.5% of Al and 36.7% of Mo were removed by diffusion dialysis. Essentially almost complete removal of metal ions and purification of high-purity phosphoric acid could be obtained by using ion exchange.

Extraction of Yttrium and Europium with Fatty Acids as Extractants (지방산계 추출제에 의한 이트륨과 유로피움의 분리)

  • Recently, an amount of waste electronic devices such as LED and trichromatic fluorescent lamp has increased with the development of electronic industry. Reportedly, rare earth metals such as yttrium and europium have been discovered in the waste electronic devices. In order to improve the selectivity of yttrium and europium, the effects of the following factors on recovery experiment have been considered : i) fatty acids with various alkyl chain lengths, ii) the concentration of extractant, and iii) pH. The results show that the extraction efficiencies decrease at the same pH with decreasing the concentration of extractant and so $pH_{0.5}$ (That value of pH in an aqueous phase at which the distribution ratio is unity at equilibrium: 50% of the solute is extracted (E = 0.5) only when the phase ratio is unity.) moves into higher pH. The highest selectivity of yttrium and europium was obtained with tetradecanoic acid as extractant. The extraction mechanism of yttrium and europium was varied with the change of concentration of the tetradecanoic acid. $MR_3$single-species was formed from the yttrium and europium ion in the extractant concentration less than 0.1 M. On the other hand, the yttrium or europium ion is solvated with three molecules of tetradecanoic acid monomer like $MR_3{\cdot}$ 3RH in the extractant concentration more than 0.1 M.

Preparation of Yeast Extract from Waste Brewer's Yeast using Various Enzymes (각종 효소를 이용한 맥주 폐효모로부터 효모추출물 제조)

  • This study was performed to investigate the optimum process conditions for manufacturing yeast extract from waste brewer's yeast using various enzymes. Contents of IMP, GMP, free amino acids, and crude protein of yeast extracts were measured by enzymes treatment. Crude protein contents of yeast extracts subjected to cell wall digestion enzyme treatment were 21.1, 33.6, and 28.0% for the control grouup, glucanase (0.5%, 12 h), and tunicase (1%, 18 h), respectively. Crude protein contents of yeast extracts subjected to protease treatment were 22.0, 30.8, and 29.8% for control group, bromelin (1%, 3 h), and protamex (1%, 3 h), respectively. Crude protein content of yeast extract subjected to glucanase and protamex mixed treatment was 34.4%. The total contents of IMP and GMP of yeast extracts subjected to G+P+A (glucanase+phosphodiesterase+adenyldeminase) and G+Pro+P+A (glucanase+protamex+phosphodiesterase+adenyldeaminase) treatments were 1,066 and 1,047 mg/100 g, respectively. The content of free amino acids of yeast extract was the highest (2,302 mg/100 g) in G+Pro+P+A treatment. Optimum concentration and process condition of enzyme treatment to obtain yeast extract with high IMP, GMP, and free amino acid content were in the order of glucanase (0.5%, 12 h), protamex (1%, 3h), phosphodiesterase (0.1%, 3 h) and adenyldeaminase (1%, 1.5 h) treatments.

In this study, we have been clearly established that a mixed system of solvent extraction, diffusion dialysis and ion-exchange technique, which made individually the most of characteristics is developed to commercialize in an efficient system for recovering the high-purity phosphoric acid. By applying vacuum evaporation, the yield of the process are almost 99% removal of nitric acid and acetic acid was achieved. And by applying the solvent extraction method with tri-octyl phosphate(TOP) as an extractant, the removal of acetic and nitric acid from the acid mixture was achieved effectively at the ratio O/A=1/3 with four stages and the stripping of nitric acid from organic phase is attained at a ration of O/A=1 with six stages by distilled water. About 97% and 76% removal of Al and Mo were achieved by diffusion dialysis. Essentially complete less than 1ppm removal of Al, Mo by using ion exchange ion resin and purification of the phosphoric acid was obtain.

Growth characteristics of Lentinula edodes treated with bean sprout waste (콩나물 부산물 첨가량에 따른 표고의 생장특성)

  • In this study, the growth characteristics of Lentinula edodes were confirmed by bean sprout waste(BW) as an alternative raw material for rice bran. The mycelium growth of Sanjo701, a major cultivation variety of L. edodes, was compared between a medium mixed with 8:2(v/v) of oak sawdust and a medium mixed with BW 50% and BW 100%. The mycelium growth in BW 50% was 13.5 cm. Compared to the control, BW 50% increased the diameter of the pileus by 1.6 cm. Additionally, the length of the pileus decreased by 0.4 cm when comparing the growth of the fruit body. In contrast, at BW 50%, the diameter of the pileus decreased by 9.6 cm and the length of the stipe decreased by 1.4 cm. According to analysis of the constituent amino acids, BW 50% showed a lower overall nutritional content than the control, whereas BW 100% had a lower amino acid content than the control. However, glutamic acid and aspartic acid, which are flavor-enhancing ingredients, were observed at levels of 3.954 mg/g and 1.436 mg/g, respectively, in BW 100%. Therefore, if bean sprout by-products are efficiently processed and utilized, it is believed that they will be beneficial to farmers as a substitute for rice bran and reduce the cost of manufacturing substrate

Comparison of Pretreatment Methods for Determination of 55Fe and 63Ni Activity in Nuclear Wastes Sample (원자력 시설 해체 폐기물 내 55Fe 와 63Ni 방사능 분석을 위한 전처리 방법 비교 연구)

  • 55Fe and 63Ni are key factors in deciding the proper handling of the decommissioning of radioactive waste from nuclear facilities. For determining beta emitting radionuclides, the dismantled waste samples should be completely decomposed and separated from the sample matrix. This study reports the comparison results of the recovering efficiencies of Iron and Nickel with wet digestion methods that use various acids and alkali-fusion methods. Various matrices of NIST SRMs (1646a, 1944, 8704, 2709a, and 1633c), the recovering efficiencies of using alkali-fusion methods ranged from 95.3 to 98.3% for Iron, and from 86.6 to 88.1% for Nickel within about 2% of relative standard deviation. On the other hand, those using one of the three wet digestion methods ranged from 77.9 to 105.3% for Iron and from 40.1 to 78.5% for Nickel with over 10% of relative standard deviation. Therefore, one may draw the conclusion that the analytical results derived from Iron and Nickel using alkali-fusion methods are fairly reliable due to the recovering efficiencies observed.

The Preparation and Practical Deinking Property of Liquid Deinking Agent for Recycling of Waste Paper (폐지 재활용용 액상 탈묵제의 제조 및 현장적용 탈묵특성)

  • It is necessary to recycle and reuse waste paper for preservation of the global environment. Instead of a solid deinking agent, liquid deinking agents have been made using various fatty acids with EO/PO added for higher deinking effect. The removal efficiency of ink on waste paper was evaluated by the ERIC and brightness properties after processes of recycling pulping (sample composition ONP: OMG = 5:5, concentration 15%, temperature $45^{\circ}C$, pulping time 20 min.) and flotation (concentration 1%, temperature $45^{\circ}C$, air flow 7 L/min, time 8 min.). Also, the ink reattachment of paper was evaluated by hyperwashing. The results show that it is effective for mass-preparation to saponify oleic acid with KOH at $80^{\circ}C$ and to add EO/PO. This deinking agent was used in a paper plant, and we evaluated the brightness, color, and ERIC. The deinking agent showed excellent performance.

The Methane Production from Organic Waste on Single Anaerobic Digester Equipped with MET (Microbial Electrochemical Technology) (미생물 전기화학 기술이 설치된 단일 혐기성소화조에서 유기성폐기물로부터 메탄생성)

  • Theoretical maximum methane yield of glucose at STP (1 atm, $0^{\circ}C$) is 0.35 L $CH_4/g$ COD. However, most researched actual methane yields of anaerobic digester (AD) on lab scale is lower than theoretical ones. A wide range of them have been reported according to experiments methods and types of organic matters. Recent year, a MET (Microbial electrochemical technology) is a promising technology for producing sustainable bio energies from AD via rapid degradation of high concentration organic wastes, VFAs (Volatile Fatty Acids), toxic materials and non-degradable organic matters with electrochemical reactions. In this study, methane yields of food waste leachate and sewage waste sludge were evaluated by using BMP (Biochemical Methane Potential) and continuous AD tests. As the results, methane production volume from the anaerobic digester equipped with MET (AD + MET) was higher than conventional AD in the ratio of 2 to 3 times. The actual methane yields from all experiments were lower than those of theoretical value of glucose. The methane yield, however, from the AD + MET occurred similar to the theoretical one. Moreover, biogas compositions of AD and AD + MET were similar. Consequently, methane production from anaerobic digester with MET increased from the result of higher organic removal efficiency, while, further researches should be required for investigating methane production mechanisms in the anaerobic digester with MET.
