To develop natural dyeing materials from the unused plant materials, we had several dyeing experiments to investigate the optimum conditions of dyeing fabrics with the dyebath extracted from wisteria. Dyeing experiments were done under different dyeing conditions of dyeing temperature, pH of dyebath, dyeing time, concentration of dyebath and mordant treatment which were treated after dyeing. Experimental fabrics were silk, wool, cotton, ramie and hemp. Color difference(ΔE) and Munsell's HV/C of the dyed fabrics and color fastness of silk and cotton fabrics to dry cleaning, washing, rubbing, perspiration and light were measured. In silk and wool fabrics(non mordant, dyeing time-20min), color differences increased in case of dyeing temperature of $100^{\circ}C$, but in cotton, ramie and hemp fabrics, color differences did not increase. Without mordant treatment, color differences of all experimental fabrics were about 10-20, so in wisteria dyeing mordant treatment was necessary. Color differences were increased by the mordant treatment; the color of dyed fabrics with Al mordant is dull yellow, Cu mordant is gold and Fe mordant is olive tone. Color difference was not affected by pH of dyebath in all experimental fabrics. On the whole, the color fastness of dyed silk and cotton fabrics were good, but color fastness to washing and perspiration in Fe mordant was weak and improved in Cu mordant.
To develope a high value-added fiber products which is useful in the human body physiology, the Macban stone was deposited on the PET fabric by sputtering and its effects were investigated. Then, a Macban stone target was prepared for sputtering treatment and treated on the PET fabric by RF sputtering process. After treatment, surface observation by SEM, far-infrared emissivity and emissive power, the fastness to washing and light, bacteriostatic rate, electrostatic, drape stiffness, and breaking strength of PET fabric were investigated. From these investigation, the following conclusions were obtained. 1) The Macban stone was able to deposit on the PET fabric, by the RF sputtering treatment which is eco-friendly dry textile finishing. 2) The far-infrared emissivity and emissive power of sputtered PET fabrics were increased. 3) When PET fabric was treated by sputtering with Macban stone, the amount of deposited Macban stone increased with increasing treatment time and it was deposited on the fabric surface firmly. 4) The bacteriostatic rate of sputtered PET fabrics was about 20%. 5) The electrostatic charge of fiber surface was reduced by sputtering. 6) The fastness washing to light of dyed fabric were improved by the deposition of Macban stone, but the breaking strength was not changed by sputtering. 7) The drape stiffness of sputtered PET fabrics increased with increasing treatment time.
The purpose of this study was investigate the dyeing property on chitosan crosslinked cotton fabric with cochineal at variable conditions. Chitosan crosslinked cotton fabrics were manufactured by crosslinking agent epichlorohydrin in the presence of chitosan. Chitosan crosslinked cotton fabrics dyed using cochineal were post-mordanted using Al, Fe and Cu. The dyeability(K/S) of chitosan crosslinked cotton fabrics were measured by computer color matching. Additionally the fastness to washing and light were also investigated. The dye-uptake of chitosan crosslinked cotton fabrics increased with the dyeing time. The saturated dyeing time was about 20minutes at $60^{\circ}C$. The dyeability(K/S) was remarkably increased with increasing content of crosslinked chitosan because of having a amine group of chitosan. Chitosan crosslinked cotton fabrics were dyed yellowish red by non and Fe mordanting, blueish red by Al and Cu mordanting, respectively. The washing and light fastness were increased by mordanting, especially Cu and Fe mordanting.

The main reaction for soil washing with using sodium hydroxide(NaOH) and hydrogen peroxide(H$_2$O$_2$) was desorption and flotation of petrochemical contaminant by means of oxygen bubble. We found the rate of decomposition by rate constant according to various temperature. For the purpose of optimizing the operation factor, we examined the effect of concentration of NaOH and H$_2$O$_2$, washing time, and soil:water ratio. The rate of decomposition for H$_2$O$_2$ in liquid phase is the first order reaction by its concentration. The rate constant of k$_1$ was 0.9439 $\times$ exp(-1376.82/RT) when concentration of NaOH was lower than 0.1 M, and the rate constant of k$_2$ was 17.3588 $\times$ exp(-2320.06/RT) when it was higher than NaOH of 0.1 M. It found that NaOH was facilitated at the beyond of specific concentration. We confirmed the optimum concentration of NaOH/H$_2$O$_2$ by means of rate constants during soil washing. Also, the optimum conditions during soil washing were washing time of 15 min, soil : water ratio of 1 : 3, and NaOH/H$_2$O$_2$ concentration of 0.25 M/0.1 M.

The storage attributes of green juice prepared by washing Angelica keiskei with various washing solutions using air bubble method were investigated. The washing solutions were distilled water (DW), sodium hypochlorite electrolyzed water (SHEW), and slightly acidic electrolyzed water (SAEW). During storage at $4^{\circ}C$, the number of bacteria after 1 day was $>10^5$ CFU/mL when DW was used, whereas bacterial growth was $<10^5$ CFU/mL after 4 days when SHEW or SAEW was used. The pH and color were not changed, and the polyphenol content, electron donating ability and total antioxidant ability were decreased slowly with the increase of storage time. No significant difference in any of the measured properties was found among washing methods (p<0.05). Consequently, these results suggest the possible use of electrolyzed water for washing to enhance the shelf life of green juice with A. keiskei because SHEW and SAEW decreased bacterial growth without affecting other properties of the green juice.

The sterilization efficacies of various washing solutions on the surfaces of vegetables such as sesame leaves, lettuce, and mini-cabbage were investigated. The washing solutions were tap water (TW), microbubble water (MB), electrolyzed water (EW), and microbubble electrolyzed water (MB+EW). After Escherichia coli and Bacillus cereus were artificially inoculated onto the surfaces of vegetables, each vegetable was washed for 1, 3, and 5 min with TW, MB, EW 100 (100 mg/L of available chlorine), EW 200 (200 mg/L of available chlorine), MB+EW 100, and MB+EW 200. The washing efficacy of MB was slightly higher than that of TW, and EW was more effective than MB (p<0.05). In all instances, the sterilization efficacies of MB+EW 100 and MB+EW 200 were higher than those of EW 100 and EW 200 (p<0.05). Thus, MB+EW offers an effective means of reducing the studied microorganisms in a short time period. The MB+EW washing method provides microbial reduction on the surfaces of various vegetables and enhances the microbiological safety of the vegetables.

We have studied the physico-chemical change and pharmacological transformation of traditional herbal medicines with processing. Evodia fruit (EF) has processed for the purpose of detoxification and reduction of its bitter taste. There are various methods of processing EF in Oriental medicinal reference. Among them, we processed EF according to the method of Sang-han theory, the most famous medicinal reference. We processed EF with washing in hot water, and then dried. Processed EFs(PEFs) were prepared according to the above procedure through repetition of 1,3,5 and 7 times respectively. The contents of weight loss, water extract, diluted ethanol extract, ether extract, total ash, acid insoluble ash, alkaloids and limonin in non-processed EF (NPEF) and PEFs were examined. The weight loss, and contents of water extract and dilute ethanol extract in PEF showed decrease in proportion to increase of washing time, while the contents of ether extract, total ash and acid insoluble ash showed little change as compared with those of NPEF. And the content of evodiamine and rutaecarpine was not changed distinctly. However, the content of limonin decreased in the final processed material in proportion to increase of washing times with hot water. And the intense of bitter taste in PEF was also reduced. These results were ascribed to the flowing-out of the water-soluble portion. The biological activities of NPEF and PEF were also investigated. In the test of DPPH scavenging effect, xanthine oxidase inhibition effect and TBA-Rs effect, PEF was more effective than NPEF in vitro. Also, both NPEF and PEF showed potent analgesic and anti-inflammatory effects against in mice. Especially, PEF by 3 times washing with hot water was more effective than other PEFs. From these results, it is considered that PEF should be prepared by 3 times washing with hot water.

In this study, nine strains were isolated from heavy metal-contaminated soil in a mine. The high efficiency bacteria, JH1, to be able removal cadmium and copper, was selected by the screen test. JH1 was identified as Ralstonia eutropha by 16S rDNA analysis, fatty acid analysis, and its morphological and biochemical characteristics. After the cadmium-contaminated soil was washed with citric acid solution(pH 6, 10 mM), Ralstonia eutropha JH1 was inoculated in the soil washing water. In order to determine the optimal cell concentration for inoculation, cell concentrations were considered in 0.5, 1.0, 2.0, 4.0 g/L, respectively. The removal efficiencies for cadmium in each cell concentration of Ralstonia eutropha JH1 were 49.9, 84.4, 89.7% and 89.9% of 110 mg/L(Cd), after 5 days culture in soil washing water. When Ralstonia eutropha JH1 was inoculated in soil washing water containing each cadmium(110 mg/L) and copper(100 mg/L), each of them was removed completely during 6 days culture. The completely removing time for cadmium and copper in each low concentration, 10, 30 and 60 mg/L were 12, 18 and 48 hrs, respectively.
Background: With the introduction of Xpert MTB/RIF assay (Xpert), its incorporation into tuberculosis (TB) diagnostic algorithm has become an important issue. The aim of this study was to evaluate the performance of the Xpert assay in comparison with a commercial polymerase chain reaction (PCR) assay. Methods: Medical records of patients having results of both Xpert and AdvanSure TB/NTM real-time PCR (AdvanSure) assays using the same bronchial washing specimens were retrospectively reviewed. Results: Of the 1,297 patients included in this study, 205 (15.8%) were diagnosed with pulmonary TB. Using mycobacterial culture as the reference method, sensitivity of the Xpert assay using smear-positive specimens was 97.5%, which was comparable to that of the AdvanSure assay (96.3%, p=0.193). However, the sensitivity of the Xpert assay using smear-negative specimens was 70.6%, which was significantly higher than that of the AdvanSure assay (52.9%, p=0.018). Usng phenotypic drug susceptibility testing as the reference method, sensitivity and specificity for detecting rifampicin resistance were 100% and 99.1%, respectively. Moreover, a median turnaround time of the Xpert assay was 1 day, which was significantly shorter than 3 days of the AdvanSure assay (p<0.001). Conclusion: In comparison with the AdvanSure assay, the Xpert assay had a higher sensitivity using smear-negative specimens, a shorter turnaround time, and could reliably predict rifampin resistance. Therefore, the Xpert assay might be preferentially recommended over TB-PCR in Korean TB diagnostic algorithm.
