Characterization of the Production of Biogenic Amines and Gamma-Aminobutyric Acid in the Soybean Pastes Fermented by Aspergillus oryzae and Lactobacillus brevis

  • The production of gamma-aminobutyric acid (GABA) using GABA-producing lactic acid bacteria (LAB) has been considered to be an attractive strategy. However, some LAB may produce biogenic amines (BA), which may be of concern from the safety viewpoint. The aim of the present study was to characterize the production of GABA and BA in the soybean pastes fermented by Aspergillus oryzae (A. oryzae) FMB S46471 and GABA-producing Lactobacillus brevis (L. brevis) GABA 100. After a ripening period of 90 days, the levels of BA (putrescine, cadaverine, histamine, and tyramine) and GABA in the fermented soybean were assessed by highperformance liquid chromatography. The soybean pastes fermented by A. oryzae and L. brevis showed a range of 7,130-11,592 mg/kg for GABA, 178-305 mg/kg for tyramine, 139-163 mg/kg for putrescine, 7.4-10.8 mg/kg for histamine, and 7.1-7.9 mg/kg for cadaverine, whereas the soybean pastes fermented by A. oryzae only showed a range of 30-1,671 mg/kg for GABA, 0.8-189 mg/kg for tyramine, 1.3-85 mg/kg for putrescine, up to 3.6 mg/kg for histamine, and 0.2-2.4 mg/kg for cadaverine. The results showed that the production of GABA was accompanied by the increase in the production of BA, even though the production levels of histamine and cadaverine were very low. This is the first study to simultaneously characterize the production of BA and GABA in GABA-enriched fermented soybean pastes, and warrants further study to minimize the production of BA while optimizing the production of GABA.

  • Low sensitivity to anticancer drugs such as 5-fluorouracil (5-FU) has been associated with decreased expression of genes involved in cell proliferation, apoptosis and metastasis. Recently, it has been shown that the expression levels of some of these genes are reduced by transcription inhibition due to epigenetic silencing on CpG islands. Therefore, epigenetic therapy has been proposed, where epigenetic silencing is repressed with DNA methyltransferase (DNMT) inhibitors and histone deacetylase (HDAC) inhibitors alone or in combination with other chemotherapeutic agents. The aim of our study was to evaluate the combination effect of 5-FU and its association with the status of epigenetic silencing using methylation-specific PCR of $p14^{ARF}$ when given with S-aza-2'-deoxycytidine (5-aza-dC), a DNMT inhibitor and depsipeptide, an HDAC inhibitor in DLD-1 human colorectal cancer cells. The combination of 5-aza-dC with depsipeptide showed a synergism and induced unmethylation of $p14^{ARF}$. However, triplet combination of 5-aza-dc/depsipeptide and 5-FU resulted in antagonistic effects and abrogated unmethylation of $p14^{ARF}$. These results suggest that unfavorable interaction of 5-aza-dC/depsipeptide with 5-FU in DLD-1 cells may be related with the failure in repression of epigenetic silencing, which warrants further investigation.

수상안전을 위한 Sculling 동작의 전산유체역학적 연구 (A Computational Fluid Dynamic Study on the Sculling Motion for Water Safety)

  • This study analyses the effects of various angles in sculling on human body lift and drag by means of computational fluid dynamics, discusses the importance of sculling and provides a basis for the development of future water safety education programmes. Study subjects were based on the mean data collected from males in the age of 20s from a survey on the anthropometric dimensions of the Koreans. Moreover, lift, drag as well as coefficient values, all of which were governed by the angle of the palm, were calculated using 3-dimentional modelling produced by computational fluid dynamics programmes i.e. CFD. Interpretations were performed via general k-${\varepsilon}$ turbulence modelling in order to determine lift, drag and coefficient values. Turbulence intensity was set to one per cent as per the figures from preceding research papers and 3-dimentional simulations were performed for a total of five different angles $0^{\circ}$, $15^{\circ}$, $30^{\circ}$, $45^{\circ}$ and $60^{\circ}$. The drag and lift values for the differing angles of the hands during sculling movement are as follows. The lift and drag values gradually increased with the increasing angle of the palm, however, the magnitude of increase for drag started to predominate lift from $45^{\circ}$ and lift gradually decreased from $60^{\circ}$. Overall, it is concluded that the optimal efficiency of sculling can be achieved at the angles $15^{\circ}$ and $30^{\circ}$, and it is anticipated that greater safety and informative education can be ensured for Life saving trainees if the results were to be applied to practical settings. However, as the study was conducted using simulation programmes which performed analyses on the collected anthropometric dimension, the obtained results cannot be made universal, which warrants furthers studies involving varied study subjects with actual measurements taken in water.

자가 보고형 질문지 측정 속성에 대한 평가: 국내 간호학술지에 게재된 논문을 중심으로 (Measurement Properties of Self-report Questionnaires Published in Korean Nursing Journals)

  • Purpose: The purpose of this study was to evaluate measurement properties of self-report questionnaires for studies published in Korean nursing journals. Methods: Of 424 Korean nursing articles initially identified, 168 articles met the inclusion criteria. The methodological quality of the measurements used in the studies and interpretability were assessed using the COnsensus-based Standards for the selection of health Measurement INstruments (COSMIN) checklist. It consists of items on internal consistency, reliability, measurement error, content validity, construct validity including structural validity, hypothesis testing, cross-cultural validity, and criterion validity, and responsiveness. For each item of the COSMIN checklist, measurement properties are rated on a four-point scale: excellent, good, fair, and poor. Each measurement property is scored with worst score counts. Results: All articles used the classical test theory for measurement properties. Internal consistency (72.6%), construct validity (56.5%), and content validity (38.2%) were most frequently reported properties being rated as 'excellent' by COSMIN checklist, whereas other measurement properties were rarely reported. Conclusion: A systematic review of measurement properties including interpretability of most instruments warrants further research and nursing-focused checklists assessing measurement properties should be developed to facilitate intervention outcomes across Korean studies.

Transferrin-Conjugated Liposome/IL-12 pDNA Complexes for Cancer Gene Therapy in Mice

  • Transferrin ($T_{f}$) has been used as a targeting ligand for delivering liposome/interleukin-12 (IL-12) pDNA complexes to cancer cells mostly due to the greater number of transferrin receptors ($T_{f}R$) found on tumor cells than on normal cells. $T_{f}$ was conjugated to liposomes via the reaction of MPB-PE with thiol groups of $T_{f}$ introduced by a heterobifunctional cross-linking agent, N-succinimidyl-3-(2-pyridyldithio)propionate (SPDP). Four days after C26 inoculation when the tumor volume reached ${\sim}100mm^{3}$, tumor-bearing Balb/c mice were injected intravenously with $T_{f}-liposome/IL-12 pDNA$complexes twice a week for 3 weeks. Significant suppression of tumor growth was achieved in the group treated with the $T_{f}-liposome/IL-12 pDNA$ complexes, with a dose of $10{\mu}g$ of IL-12 pDNA showing the highest suppression effect among the tested doses. Similar results were obtained when the therapy was initiated one day after tumor inoculation, although in this case $30{\mu}g$ IL-12 pDNA/$T_{f}-liposome$ complexes showed a significant suppression of tumor growth between 19 and 23 days after tumor inoculation. This result indicates that the transferrin receptor-targeted liposomal system is an efficient delivery agent of therapeutic genes, such as IL-12, in mice and that its potential clinical use warrants further research investigation.

하악편차 교정을 이용한 통증완화 효과에 대한 보고 (A Study on Pain Control Using Balancing Therapy of Occlusal Plane : Case Series)

  • Objectives : This study was performed to evaluate the effect of occulsal plane balancing therapy on pain control without another therapy. Methods : This clinical study was carried out in 6 cases of patients which had neck and shoulder pain. In this study we treated the patients by balancing therapy of occulsal plane using paper. We measured Visual Analogue Scale(V.A.S.) for pain intensity, Range of Motion(R.O.M.) and difference of thermographic temperature between pain site and the opposite before and 15 minutes after treatment and analyzed the change of variables. Also, we analyzed the correlation among hypomovable site of neck rotaion(HS), pain site(PS) and thick site of paper(TS). Results : 1. The average of skin temperature was decreased from $0.45{\pm}0.11$ to $0.24{\pm}0.13$ significantly(p<0.05). 2. The average of V.A.S was decreased from $10.00{\pm}0.00$ to $3.83{\pm}0.99$ significantly(p<0.05). 3. In the correlation analysis among HS, PS and TS, there is negative correlation between HS and TS, but there was no statistical significance. Conclusions : In the study, the balancing therapy of occulsal plane warrants further investigation in the change of skin temperature and R.O.M. of the joint, pain control.

인삼(Panax ginseng) 항암 효과에 관한 문헌고찰 - 실험연구와 역학연구 결과를 중심으로 - (The Cancer-Preventive Potential of Panax ginseng - A Review of Human and Experimental Evidence -)

  • Objective : We have reviewed the potential cancer preventive and other relevant properties of Panax ginseng C. A. Meyer, which has been traditionally used as a natural tonic in oriental countries. Data identification and study selection: Publications on Panax ginseng and its relation to cancer were obtained from the Medline database (1983-2000) and by checking reference lists to find earlier reports. The reports cover experimental models and human studies on cancer-preventive activity, carcinogenicity and other beneficial or adverse effects. In addition, possible mechanisms of chemoprevention by ginseng were also considered. Results : Published results from a cohort and two case-control studies in Korea suggest that the intake of ginseng may reduce the risk of several types of cancer. When ginseng was tested in animal models, a reduction in cancer incidence and multiplicity at various sites was noted. Panax ginseng and its chemical constituents have been tested for their inhibiting effect on putative carcinogenesis mechanisms (e.g., cell proliferation and apoptosis, immunosurveillance, angiogenesis); in most experiments inhibitory effects were found. Conclusion : While Panax ginseng C. A. Meyer has shown cancer preventive effects both in experimental models and in epidemiological studies, the evidence is currently not conclusive as to its cancer-preventive activity in humans. The available evidence warrants further research into the possible role of ginseng in the prevention of human cancer and carcinogenesis.

Effects of a low-FODMAP enteral formula on diarrhea on patients in the intensive care unit

  • BACKGROUND/OBJECTIVES: A dietary restriction on the intake of fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAPs) has been reported to be effective in the treatment of gastrointestinal (GI) tract complications. Enteral nutrition (EN) is widely used for patients who cannot obtain their nutritional requirements orally, but many studies have reported EN complications, especially diarrhea, in up to 50% of patients. SUBJECTS/METHODS: We performed a single-center, non-randomized, controlled trial to determine the effects of a low-FODMAP enteral formula on GI complications in patients in intensive care units (ICUs). Patients in the ICU who needed EN (n = 66) were alternately assigned to the low-FODMAP group (n = 33) or the high-FODMAP group (n = 33). RESULTS: Anthropometric and biochemical parameters were measured, and stool assessment was performed using King's Stool Chart. We excluded patients who received laxatives, GI motility agents, proton pump inhibitors, antifungal agents, and antibiotics other than β-lactams. There were no differences in GI symptoms during 7 days of intervention, including bowel sound, abdominal distension, and vomiting between the 2 groups. However, diarrhea was more frequent in the high-FODMAP group (7/33 patients) than the low-FODMAP group (1/33 patients) (P = 0.044). CONCLUSIONS: Our results suggest that a low-FODMAP enteral formula may be a practical therapeutic approach for patients who exhibit enteral formula complications. Our study warrants further randomized clinical trials and multicenter trials.

Facial reanimation using the hypoglossal nerve and ansa cervicalis: a short-term retrospective analysis of surgical outcomes

  • Background: Transferring the hypoglossal nerve to the facial nerve using an end-to-end method is very effective for improving facial motor function. However, this technique may result in hemitongue atrophy. The ansa cervicalis, which arises from the cervical plexus, is also used for facial reanimation. We retrospectively reviewed cases where facial reanimation was performed using the ansa cervicalis to overcome the shortcomings of existing techniques of hypoglossal nerve transfer. Methods: The records of 15 patients who underwent hypoglossal nerve transfer were retrospectively reviewed. Three methods were used: facial reanimation with hypoglossal nerve transfer (group 1), facial nerve reanimation using the ansa cervicalis (group 2), and sural nerve interposition grafting between the hypoglossal nerve and facial nerve (group 3). In group 1, the ansa cervicalis was coapted to neurotize the distal stump of the hypoglossal nerve in a subset of patients. Clinical outcomes were evaluated using the House-Brackmann (H-B) grading system and Emotrics software. Results: All patients in group 1 (n= 4) achieved H-B grade IV facial function and showed improvements in the oral commissure angle at rest (preoperative vs. postoperative difference, 6.48° ± 0.77°) and while smiling (13.88° ± 2.00°). In groups 2 and 3, the oral commissure angle slightly improved at rest (group 2: 0.95° ± 0.53°, group 3: 1.35° ± 1.02°) and while smiling (group 2: 2.06° ± 0.67°, group 3: 1.23° ± 0.56°). In group 1, reduced tongue morbidity was found in patients who underwent ansa cervicalis transfer. Conclusion: Facial reanimation with hypoglossal nerve transfer, in combination with hypoglossal nerve neurotization using the ansa cervicalis for complete facial palsy patients, might enable favorable facial reanimation outcomes and reduce tongue morbidity. Facial reanimation using the ansa cervicalis or sural nerve for incomplete facial palsy patients did not lead to remarkable improvements, but it warrants further investigation.

Moderating effect of self-esteem and depression on the correlation between the frequency of community volunteer activities and job consciousness in dental hygiene students

  • Objectives: This study examined dental hygiene students' frequency of participation in community volunteer activities, job consciousness, depression, and self-esteem, and then identified the correlation among them. We also analyzed the moderating effect of depression and self-esteem on the correlation between participation in community volunteer activities and job consciousness. Methods: We surveyed 312 dental hygiene students attending four universities in South Chungcheong Province, Korea, from May 2 to June 15, 2016, through direct visits. They were given structured questionnaires containing four items on community volunteer activities, three on job consciousness, ten on self-esteem, and 13 on depression. To examine the moderating effect of depression and self-esteem, we performed analysis of variance, correlation analysis, and structural equation modeling analysis (based on the path analysis model and by inserting interaction terms) using SPSS and AMOS. Results: We observed a negative correlation between self-esteem and depression (r=-0.062) but a positive correlation between self-esteem and job consciousness (r=0.125). Depression and job consciousness had a negative correlation (r=-0.176). Depression had a statistically significant impact on job consciousness (r=-0.519, p<0.01). The interaction term between depression and frequency of community volunteer activities also had a statistically significant influence on job consciousness (r=0.090, p<0.05). These findings indicate that depression moderates the correlation between frequency of community volunteer activities and job consciousness at a statistically significant level. Conclusions: More frequent participation in volunteer activities enhances dental hygiene students' self-esteem, reduces depression, and raises job consciousness. The positive impact of volunteering on self-esteem, depression, and job consciousness warrants encouraging students' participation in community volunteer activities, creating supportive structures, and developing various volunteer programs relevant to the students' area of study.
