양돈 슬러리의 암모니아 발생 특성에 관한 연구 (Study on Ammonia Emission Characteristic of Pig Slurry)

  • 본 연구에서는 육성 비육돈 슬러리를 이용하여 슬러리의 환경조건에 따라 슬러리로 부터의 암모니아 발생 특성을 분석코자 하였다. 슬러리의 온도와 pH를 조절하여 슬러리로부터 발생하는 암모니아 농도를 측정 분석한 결과 슬러리의 온도가 높을수록 슬러리로 부터 발생하는 암모니아 농도는 급격히 증가하는 것으로 나타났고, 슬러리의 pH를 5, 6, 7, 8의 4수준으로 조절하여 각각의 온도에 따라 발생하는 암모니아 농도를 측정한 결과 pH 5, 6에서는 암모니아 가스가 검출되지 않았으나 pH 7, 8에서는 많은 양의 암모니아 가스가 검출되었고 양돈 슬러리의 pH가 높을수록 발생하는 암모니아 가스가 많이 발생하는 것으로 분석되었다. 본 연구결과 슬러리의 온도를 낮추어 주거나 슬러리의 pH를 낮춤으로써 슬러리로부터 발생하는 악취가스를 크게 줄일 수 있을 것으로 판단되었다.

약물 독성 평가용 생체외 각막 모델 제작 연구 (Fabrication of Ex vivo Cornea Model for a Drug Toxicity Evaluation)

  • To evaluate the toxicity of ophthalmic drug, the Draize test and Bovine Corneal Opacity and Permeability (BCOP) test commonly used. In Draize test, experimental animals were under stress and pain due to long-term exposure of drug. In addition, regarding physiological functions, animal model is not perfectly reflected a human eye condition. Although some models such as $EpiOcular^{TM}$, HCE model, LabCyte Cornea-Model, and MCTT $HCE^{TM}$ were already presented advanced cornea ex-vivo model to replace animal test. In this sense, cornea tissue structure mimicked ex-vivo toxicity model was fabricated in this study. The corneal epithelial cells (CECs) and keratocytes (CKs) isolated from rabbit eyeball were seeded on non-patterned silk film (n-pSF) and patterned silk film (pSF) at $32,500cells/cm^2$ and $6,500cells/cm^2$. Sequentially, n-pSF and pSF were stacked to mimic a multi-layered stroma structure. The thickness of films was about $15.63{\mu}m$ and the distance of patterns was about $3{\mu}m$. H&E stain was performed to confirm the cell proliferation on silk film. F-actin of CKs was also stained with Phalloidin to observe the cytoskeletal alignment along with patterns of the pSF. In the results, CECs and CKs were shown the good cell attachment on the n-pSF and pSFs. Proliferated cells expressed the specific phenotype of cornea epithelium and stroma. In conclusion, we successfully established the ex-vivo cornea toxicity model to replace the eye irritation tests. In further study, we will set up the human ex-vivo cornea toxicity model and then will evaluate the drug screening efficacy.

Localization of Sop Proteins and Interaction of Plasmid DNA with the Cell Membrane of Host Bacteria in Partitioning

  • A sopA protein (41K) encoded by plasmid pXX288 was observed in the cytoplasm, whereas a sopB protein (37K) encoded by plasmid pXX157 was observed in the membrane fraction. Most of the sopB protein was solubilized from the crude membrane by treatment with Sarkosyl, which suggested that the protein may be located in the inner membrane. The sopA protein was precipitated at the concentration of 30 to 60% ammonium sulfate. The sedimentation profile of the crude membrane fraction showed a little difference according to culture media used, and the sopB protein existed in all fractions of inner membrane. The DNA of plasmids, pXX157, pXX300, and pXX167 co-sedimented with inner membrane fraction.

  • The swelling behavior of hyddrogels has been interested in many applications of drug carriers. These gels show reversible swelling changes in response to pH, electric currcnt, and temperature. Among others, the temperature-responsive behavior of poly(N-isopropylacrylanxide) (p(NIPAAm)) was studied, because a lower critical solution temperature(LCST) is in the vicinity of 32$\circ$C, and remarkable temperature-response can be obtained. We propose a novel composite membrane, which is appropriate for transporting drug ingredients above the transition temperature. Our object was to design a high permeation system above the shrinking temperature of p(NIPAAm). The membrane was composed of a matrix polymer and thermosensitive p(NIPAAm) hydrogel. The flux pattern of 4-acctamidophen through membrane in response of temperature was opposite to that of p(NIPAAm) membrane.

자외선분광기를 이용한 미생물 세포 생촉매의 에폭사이드 가수분해효소 활성평가 (UV Spectrometric Assay of Epoxide Hydrolase Activity of Microbial Cell Biocatalysts)

  • 미생물 세포 유래의 에폭사이드 가수분해 효소 활성을 효율적으로 분석하기 위하여 UV 분광기 측정법을 최적화하였다. 세포 생촉매의 에폭사이드 가수분해 활성에 의해 p-nitrostyrene oxide (pNSO) 기질이 p-nitrostyrene diol로 전환된 양을 측정함으로써 에폭사이드 가수분해 활성을 평가하였다. Rhodosporidium toruloides를 생촉매로 사용하고 pNSO에 대한 입체선택적 가수분해 동력학을 UV 분광기 측정법으로 분석한 결과, $K_m$$V_m$ 값을 $2.457nmol/min{\cdot}mg$ 및 1.078 mM로 각각 결정할 수 있었다.

Synthesis and pH-Dependent Micellization of a Novel Block Copolymer Containing s- Triazine Linkage

  • Novel pH-sensitive moieties containing an s-triazine ring were synthesized with sulfonamide and secondary amino groups. The synthesized pH-sensitive moieties were used for the synthesis of a pH-sensitive amphiphilic ABA triblock copolymer. The pH-sensitive triblock copolymer was composed of diblock copolymers, methoxy poly(ethylene glycol)-poly ($\varepsilon$-caprolactone-co-D,L-lactide) (MPEG-PCLA), and pH-sensitive moiety. These copolymers could be dissolved molecularly in both acidic and basic aqueous media at room temperature due to secondary amino and sulfonamide groups. The synthesized s-triazine rings containing pH-sensitive compounds were characterized by ${^1}H-NMR,\;{^13}C-NMR$, and LC/MSD spectral data. The synthesized diblock and triblock copolymers were also characterized by ${^1}H-NMR$ and GPC analyses. The critical micelle concentrations at various pH conditions were determined by fluorescence technique using pyrene as a probe. Furthermore, the micellization and demicellization study of the triblock copolymer was done with pH-sensitive groups. The sensitivity towards pH change was further established by acid-base titration.

Alloy 600에 전기 도금한 Ni-P-Fe 및 Ni-P-B 층의 열적 안정성 연구 (Study on Thermal Stability of Ni-P-Fe and Ni-P-B Layers Electroplated on Alloy 600)

  • In this study, thermal stability of the mechanical properties of Ni-P-B and Ni-P-Fe layers electroplated on Alloy 600 material was evaluated by measuring their microhardness, tensile strength, and elongation after heat treatment at $325^{\circ}C$ and $400^{\circ}C$. According to the results, there was no noticeable change in microhardness of the two electrodeposits before and after heat treatment at the temperatures for 30 days. In the case of a Ni-P-B electrodeposit, ultimate tensile strength (UTS) slightly increases with heat treatment time, while its elongation decreases, showing good thermal stability in the mechanical properties at high temperature. On the other hand, UTS and elongation of Ni-P-Fe decrease with heat treatment time, which is very unusual observation. This result was attributed to the bad microstructure of Ni-P-Fe having many defects in the deposit formed early stage of an electroplating process and their redistribution to link to become large ones during heat treatment.

Purification and Characterization of an Alkaliphilic Alginate Lyase AlgMytC from Saccharophagus sp. Myt-1

  • In a previous study, we isolated and reported a second species of the Saccharophagus genus, Saccharophagus sp. strain Myt-1. In the present study, an alginate lyase gene (algMytC) from the genomic DNA of Myt-1 was cloned and characterized. The DNA sequence fragment obtained contained an open reading frame of 1,032 bp that encoded a protein of 343 amino acids with an estimated molecular mass of 37.6 kDa and a pI of 6.60. The deduced protein, AlgMytC, had the conserved amino acid sequences (RTELREM, QIH, YFKAGVYNQ) of the polysaccharide lyase family 7. A BLAST homology search indicated that AlgMytC shared an amino acid sequence identity of 95.9% with alg7A of S. degradans 2-40. The cloned and purified AlgMytC protein showed optimal activity at $40^{\circ}C$, and retained more than 90% of its total activity even after treatment at $25^{\circ}C$ for 24 h. AlgMytC was very alkaliphilic with an optimal pH of 9.0, and more than 90% of its activity was retained in the pH range 8.5-10.0. Moreover, AlgMytC was stable over a wide pH range. The activity of AlgMytC was also stable in the presence of various detergents.

Extracellular Overproduction of $\beta$-Cyclodextrin Glucanotransferase in a Recombinant E. coli Using Secretive Expression System

  • $\beta$-Cyclodextrin glucanotransferase ($\beta$-CGTase) was overproduced extracellularly using recombinant E. coli by transforming the plasmid pECGT harboring a secretive signal peptide. The $\beta$-CGTase gene of alkalophilic Bacillus firmus var alkalophilus was inserted into the high expression vector pET20b(+) containing a secretive pelB signal peptide, and then transformed into E. coli BL2l(DE3)pLysS. The optimum culture conditions fer the overproduction of $\beta$-CGTase were determined to be TB medium containing 0.5% (w/v) soluble starch at post-induction temperature of $25^{\circ}C$. A significant amount of $\beta$-CGTase, up to 5.83 U/ml, which was nine times higher than that in the parent strain B. firmus var. alkalophilus, was overproduced in the extracellular compartment. A pH-stat fed-batch cultivation of the recombinant E. coli was also performed to achieve the secretive overproduction of $\beta$-CGTase at a high cell density, resulting in production of up to 21.6 U/ml of $\beta$-CGTase.

DEPHANOX 공정 내 알루미늄 첨가에 따른 질소 및 인 제거 효율 평가 (Nitrogen and Phosphorus Removal Efficiency of Aluminum Usage in DEPHANOX Process)

  • 본 연구는 DEPHANOX 공정내 분리조에 Al(III) ($Al_2(SO_4)_3{\cdot}17H_2O$)를 주입하여 유출수의 T-P 농도를 0.2 mg/L 이하로 유지하기 위한 주입량을 도출하고 Al(III) 첨가에 따른 질소 및 인 제거 효율을 평가하였다. 반응조 내에 Al(III)을 5, 10, 15 mg/L주입하여도 pH와 알칼리도 저감에 대한 영향은 나타나지 않았으며 반응조 내의 pH는 7~8 사이로 유지하였다. 응집제의 주입량이 증가할수록 유기물과 T-N 제거효율은 감소하였으나, T-P 제거효율은 주입량이 5, 10, 15 mg/L로 증가할수록 76.28, 84.02, 94.66% 증가하였다. Al(III)주입량 15 mg/L에서 유출수의 T-P 농도는 0.17 mg/L로 관찰되어 유출수의 T-P 농도를 0.2 mg/L 이하로 유지하기 위한 Al(III)의 주입량은 최소 15 mg/L 이상임을 확인하였다.
