The restriction clevage map of multi-copy recombinant plasmid, pJY712(8.1kb), carrying the thiostrepton resistance gene(tsr) was determined. pJY712 had a broad host range in Streptomyces and contained single BglII site for cloning purpose. The plasmid showed the phenomenon of lethal zygosis ($Ltz^{+}$). Transformation frequency of pJY712 was $5.0\times 10^{4}$ transformants per ug plasmid DNA (TFU) in S. lividans. Plasmid pJY713 was constructed by inserting the tyrosinase gene(mel) into the BclI site of pJY712. Recombinant plasmid pJY714 carrying the mel gene was constructed by in vitro deletion of a segment (1.9kb BglII-BclI fragment) from pJY713.
The purpose of this study was to determine whether vitamin B6(B6) deficiency affects fuel utilization and blood cholesterol profile with exercise-training. Twenty-four rats were fed a B6 deficient(-B6) diet or a control (+B6) diet for 5 weeks and either exercised(EX) or nonexercised (NE). EX rats were exercised on treadmill(10$^{\circ}$, 0.5-0.8km/h) for 20 minutes everyday. Glucose(GLU), glycogen (GLY), protein(PRO), trglyceride(TG), free fatty acid(FFA), total cholesterl(TC), HDL-cholesterol(HDL-C) and LDL-choleterol(LDL-C) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. There was a vitamin effect on the level of P-GLU, P-TG, M-TG, L-GLY, L-PRO and an exercise effect on the level of P-PRO, P-FFA, M-PRO, L-GLY, L-TG, P-TC, P-HDL-C, P-LDL-C. Compared to +B6 rats were lower and there were no differences in P-GLU, P-FFA, P-TG. M-GLY, L-TG, P-TC and P-HDL-C. In EX group, the level of P-TG was higher and M-PRO was lower in -B6 rats. There were no differences in M-GLY, L-TG, P-TC and P-HDL-C. These results suggest that a lowered intake of vitamin B6 may impair the adaptation of animals to fuel metabolism related to a decrease of fatty acid oxidation and attenuates the exercise-traning effect on blood lipid profile.
Partial nucleotide sequence (nt 1-1842) of chloramphenicol resistance plasmid pKH7 has been reported previously and residual nucleotide sequence (nt 1843-4118) of pKH7 was determined and then the complete nucleotide sequence of pKH7 was obtained. pKH7 consists of 4118 bp and has three ORFs. Besides Rep and CAT proteins described in previous paper, Pre protein which mediates site-specific recombination in Staphylococcus aureus was found to be on pKH7. R $S_{A}$, a site-specific recombination site of Pre protein, and palA, a specific lagging-strand conversion signal, was also found in pKH7. Amino acid sequence of Pre protein of pKH7 was compared with those of other antibiotic resistant Staphylococcus aureus plasmids.s.
The experiment was conducted to investigate accumulation o( phosphate in tobacco production area. The results are as follows: 1) The content of available P in burley production area was about 200ppm higher than that in flue-cured tobacco production area, the available P content of 20% tobacco fields surveyed was above 1000ppm P. 2) Tobacco fields having 200ppm of available P which were grouped as medium category for phosphate recommendation were more than 90% out of tobacco fields surveyed. 3) Total phosphate content was above 4000ppm in 42.5% burley tobacco fields before transplanting, and 2000~3000ppm in about 40% flue-cured tobacco fields. 4) Phosphate fractions in soil increased in order of Al-P > Fe-P > Ca-p, and their content was about 50% of total phosphate in tobacco fields surveyed. 5) pH value in most of tobacco fields surveyed ranged from 4.5 to 5.0 and, field soils with pH value below 5.5 were 64 %.
This study was attempted to investigate the effects of enzymatic modification of milk protein with Meju protease on its acid clotting and digestibility. The proteases used in this study were isolated from Meju(fermented soybeans) and had specific acticity of 250 units/mg protein at pH 7.0. These proteases were found to be at least 3 different isoenzymes of different pH optima(pH 4.0, 6.0, 10.0). The optimum temperature was 5$0^{\circ}C$. Hydrolyzed skim milk showed 30.5% degree of hydrolysis for 1 hr. and 36.4% degree of hydrolysis for 3.5 hrs. of protease treatment at pH 7.0. Upon acidification to pH 4.0, skim milk produced large and dense coagulum, but the coagulum was getting smaller by protease treatment. Generally, digestability of skim milk at pH 4.0 was lower than pH 2.0. At pH 4.0, native skim milk and control group had problem with hydrolysis of skim milk protein. Among protease treated groups, 1 hour treated skim milk was most effectively hyrolyzed at pH 4.0.
Purpose: To investigate the clinical value of serum miR-221-3p and miR-122-5p expression levels in the diagnosis and prognosis of gastric cancer. Materials and Methods: Serum samples from 141 gastric cancer cases (gastric cancer group), 110 gastric polyps (gastric polyp group), and 75 healthy people (healthy control) were used to detect miR-221-3p and miR-122-5p expression using real-time reverse transcription polymerase chain reaction. Results: Serum miR-221-3p expression was significantly higher in the gastric cancer group than in the gastric polyp group, and it was significantly lower than that before operation. The miR-221-3p expression was significantly higher in the death group than in the survival group. The proliferation and migration ability significantly increased and the apoptosis rate significantly decreased by miR-221-3p transfection in gastric cancer cells. In contrast, the function of miR-122-5p in gastric cancer cells was opposite of miR-221-3p. Serum miR-221-3p expression was negatively correlated with that of miR-122-5p in gastric cancer. Serum miR-221-3p and miR-122-5p expressions were significantly correlated with the degree of differentiation, tumor, node, metastasis stage, lymph node metastasis, and invasion depth. miR-221-3p and miR-122-5p expression levels were independent prognostic factors for postoperative gastric cancer. In the diagnosis and predicting prognosis of gastric cancer, receiver operating characteristic analysis revealed that the area under curve of combined detection of serum miR-221-3p and miR-122-5p expression had a greater diagnostic effect than either single maker. Conclusions: The miR-221-3p and miR-122-5p are involved in the development of gastric cancer, and they have important clinical values in gastric cancer diagnosis and prognosis.
The purpose of this study was to investigate the difference of body composition of the elderly according to the age. This study consisted of elder male(n=48) and elder female(n=58). The average age of male elder and female elder was 73.81, 73.05 years. The data were analyzed with t-test, using SPSSWIN 10.1 program. Body composition was measured using Inbody 3.0(seoul, korea) of segmental multifrequency impedance analyzer technique. The result of this study were the following : 1. The difference of body composition according to the age 1) The MV(Muscle Volume)of male elder was significant difference among group(p<.05). The MV of female elder was very significant difference among group(p<.01). 2) The %FAT of male elder and female elder was no significant difference among group. 3) The FFM(Free Fat Mass)of male elder was significant difference among group(p<.05). The FFM of female elder was very significant difference among group(p<.01). 4) The TBW(Total Body Water)of male elder was significant difference among group(p<.05) The TBW of female elder was very significant difference among group(p<.01). 5) The WHR(Waist to Hip Ratio)of male elder was significant difference among group(p<.05). The WHR of female elder was very significant difference among group(p<.01).
Antagonistic mode of action of fenoxaprop-P-ethyl [ethyl(R)2-4-{(6-chloro-2-benzoxazolyloxy) phenoxy}propionate] with bentazon was investigated with respect to absorption, translocation, metabolism, and change in target site response of fenoxaprop-P-ethyl using four-leaf stage of rice(Oryza sativa L.) and barnyardgrass [Echinochloa eras-galli (L.) P. Beauv.]. Shoots of rice and barnyardgrass was more sensitive to fenoxaprop-P-ethyl than the roots. More than 90% of fenoxaprop-P-ethyl was absorbed within 6 hours after treatment and 30% of the absorbed was acropetally and basipetally translocated at 24 hours after treatment. Fenoxaprop-P-ethyl was rapidly transformed to its acid form, fenoxaprop(2-[4-(6-chloro-2-benzoxazolyloxy)phenoxy]propionic acid), which was subsequently metabolized to polar conjugates. However, changes in absorption, translocation, and metabolism of fenoxaprop-P-ethyl by bentazon treatment were not found in both species. Background activity of acetyl-CoA carboxylase(ACCase) in rice and barnyardgrass was 26.5 and 23.2nmol/min/mg, respectively. Concentration required to inhibit fifty percent enzyme activity$(I_{50})$ in vitro was 6.5~7.4${\mu}M$ of fenoxaprop-P-ethyl and more than 500${\mu}M$ of bentazon. There were no significant differences in $I_{50}$ value between two treatments of fenoxaprop-P-ethyl alone and its bentazon mixture. However, bentazon reduced ACCase activity in vivo and inhibited electron transport in chloroplast thylakoid. Based on the results obtained, it is concluded that the antagonistic effect of bentazon occurs due not to direct effect on target site of fenoxaprop-P-ethyl, but to indirect involvement in reducing herbicidal activity of fenoxaprop-P-ethyl through physiological disturbances caused by bentazone at whole chloroplast level.

The purpose of this study was to systematically evaluate and compare the effects of 640 nm LED irradiation and aromatherapy on the skin condition of the neck. Twenty female participants were divided into Group A (640 nm LED irradiation + aromatherapy) and Group B (640 nm LED irradiation only). Total of 8 experiments were conducted on the 2 groups twice a week. As a result of the experiment, moisture content was significantly improved in both group A (p<0.01) and group B (p<0.001), and wrinkle index was significantly improved in both group A (p<0.01) and group B (p<0.05). Additionally, the change in skin density was significantly improved in both group A (p<0.001) and group B (p<0.05), and the change in elasticity was significantly improved in both group A (p<0.001) and group B (p<0.001) as well. However, the change in skin tone was not significant in group A (p>0.05), but the change was drastically improved in group B (p<0.05). Blemishes and pigmentation changes were significantly improved in group A (p<0.05), but was not significant in group B (p>0.05). These results conclude that combining aromatherapy with 640 nm LED irradiation can be highly effective in improving skin condition of the neck.
Simulations using two well-known commercial codes, WAsP and WindSIM, were performed to predict the wind resources in complex terrains of Korea. The predictions from the codes were compared with the measured data. Cross predictions were performed for two closely located measurement sites. The results from WindSIM were found to be more accurate than those from WAsP. The predictions for wind velocity and direction in five different sites of complex terrain from WAsP and WindSIM were also compared. It was found that if the self prediction of the wind velocity and direction from WAsP is close to the measured wind data, the discrepancies between WAsP results and WindSIM results are also close.
