Optimization of Beverage Preparation from Schizandra chinensis Baillon by Response Surface Methodlogy (반응표면 방법에 의한 오미자 음료 제조의 최적화)

  • To develop the beverage from Schizandra chinensis Baillon, extraction condition was optimized, using the fractional factorial design with 3 variables and 3 levels, by a RSM computer program. The optimum extraction time, extraction temperature and alcohol concentration levels were 3 hrs, $80{\sim}85^{\circ}C$ and 0%(water) for a desired yield, color and non-volatile organic acid. And to obtain the optimum recipe, amounts of extract, sugar and citric acid were optimized, using the fractional factorial design with 3 variables and 3 levels, by a RSM program. The optimum extract, sugar and citric acid levels were 4%, 9.6%n and 0.06%, respectively, for a disired flavor and overall acceptance.

  • This study explored the analytical conditions for 21 veterinary antibiotics which have been popularly sold in South Korea in 2014 but have not yet been targeted in EPA method 1694. Most of the selected antibiotics were separated by a reverse-phase C18 column with a combination of (buffered) water and organic polar solvent, which was commonly methanol and acetonitrile in the gradient elution mode. Volatile additives such as formic acid, ammonium acetate and ammonium formate were usually added to the mobile phases to minimize asymmetrical and tailing of antibiotics' peaks and to increase their ionization in mass spectrometry. The analytical methods of aminoglycoside antibiotics were distinct from those of the other antibiotics in terms of adoption of ion-pair chromatography (IPC) and hydrophilic interaction liquid chromatography (HILIC) capable of retaining and separating extremely polar compounds due to their hydrophilicity. Trifluoroacetic acid or heptafluorobutyric acid was frequently added to the mobile phase as an ion-pair reagent for the IPC. Tandem mass spectrometry was numerously applied to the detection of antibiotics using positive electrospray ionization (ESI) and the selected reaction monitoring (SRM) mode. All reviewed analytical methods had been/were validated by evaluating recovery, limits of detection and quantification, decision limit or detection capability of the methods.

Production Behavior of Amino Acid from High Temperature and High Pressure Water Reaction of Fish Entrails (고온고압수 반응을 이용한 생선내장의 아미노산 생성거동)

  • The effect of operating parameters (reaction temperature and time) and reaction modes (batch and semi-batch) on the behavior of amino acid production from hydrothermal decomposition of fish-derived wastes was investigated. The amino acids obtained in batch experiments at temperature of 250$^{\circ}C$ were mainly alanine (Ala) and glycine (Gly) at maximum yield of 65 and 28mg/g-dry fish, respectively. At relatively lower temperature of 200$^{\circ}C$, the yield of high-molecular-weight amino acids such as aspartic acid (Asp) and serine (Ser) is high, but decreases as temperature increases. It is likely that high-molecular-weight amino acids decompose faster than low-molecular ones. Semi-batch mode of reaction suppressed decomposition of amino acids into organic acids (or volatile materials) by continuously removing the products from the reaction zone as soon as they are formed. Thus, large amount of high-molecular-weight amino acids such as Asp and Ser at this reaction mode was observed.

Ecotoxicological Effects of NaDCC injection method in Ballast Water Management system on Marine Environments (NaDCC 주입 선박평형수 처리기술의 해양생태위해성에 대한 연구)

  • Effluent treated by an NaDCC injection method in Ballast water management system (BWMS) contains reactive chlorine species and disinfection by-products (DBPs). In this study, we conducted whole effluent toxicity (WET) testing and ecological risk assessment (ERA) to investigate its ecotoxicological effects on marine environment. WET testing was carried out for four marine pelagic and freshwater organisms, i.e., diatom Skeletonema costatum, Navicula pellicuosa, chlorophyta Dunaliella tertiolecta, Pseudokirchneriella subcapitata, rotifer Brachionus plicatilis, Brachionus calyciflorus and fish Cyprinodon variegatus, Pimephales promelas. The biological toxicity test revealed that algae was the only biota that showed apparent toxicity to the effluent; it showed no observed effect concentration (NOEC), lowest observable effect concentration (LOEC) and effect concentration of 50% (EC50) values of 25-50%, 50-100% and >100%, respectively, at three water condition, but did not show any significant toxicities on other biota. Meanwhile, chemical analysis revealed that the BWMS effluent contained total residual oxidants (TROs) below $0.03{\mu}g/L$ and a total of 25 DBPs such as bromate, volatile halogenated organic compounds (VOCs), halogenated acetonitriles (HANs), halogenated acetic acids (HAAs), chloropicrin and Isocyanuric acid. Based on ERA, the 25 DBPs were not considered to have persistency, bioaccumulation and toxicity (PBT) properties. The ratio of predicted environmental concentration (PEC) to predicted no effect concentration (PNEC) of the other DBPs did not exceed 1 for General harbor environment. However, four substances (Isocyanuric acid, Tribromomethane, Chloropicrin and Monochloroacetic acid) were exceed 1 for Nearship environment. But observed toxicity in the test water on algal growth inhibition would be mitigated by normal dilution factor of 5 applied for nearship exposure. Thus, our results of WET testing and ERA showed that the BWMS effluent treated by NaDCC injection method would have no adverse impacts on marine environment.

Naturally Occurring of Sulfur Dioxide in Medicinal Herbs(Crude Drug Materials) and Its Origin (생약재중 천연유래 이산화황 함유량 및 그 출처)

  • Naturally occurring sulfur dioxide in medicinal herbs(crude drug materials) and its origin was investigated. Sulfur dioxide content in 28 kinds (101 samples) of medicinal herbs was studied by two different methods, Monnier-Williams and acid-distillation ion chromatography. Generally, less than 5 ppm of sulfur dioxide was found from the tested herbs. In some herbs, Monnier-Williams method showed higher level of sulfur dioxide than acid-distillation ion chromatography probably due to the presence of volatile organic acid presence in herbs. Naturally occurring sulfur dioxide measured in medicinal herbs may be derived from sulfur compounds of medicinal herbs.

Nutrient composition and in vitro fermentability of corn grain and stover harvested at different periods in Goesan, a mountainous area

  • With South Korea's limited capability of feed production because of its relatively small cultivable area, the country is pushed to depend on foreign feed imports despite the immensely fluctuating price of corn. Hence, intensive efforts to increase the total cultivable area in Korea like extending of farming to mountainous area is being practiced. Corn was planted in Goesan County, a mountainous area in the country. Grain and stover were harvested separately in three harvest periods: early-harvest (Aug 8), mid-harvest (Aug 18), and late-harvest (Aug 28). The nutrient composition such as dry matter (DM), crude protein (CP), crude fat (EE), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), and non-fibrous carbohydrates (NFC) was determined after harvest. Effective degradability (ED) of the major nutrients (DM, NDF, ADF, and CP) were measured through in vitro fermentation of rumen fluid from Hanwoo (Korean cattle). pH, ammonia-N concentration, volatile fatty acid (VFA) concentration, and gas production were periodically measured at 0, 3, 6, 12, 24, 48, and 72 h. Corn grain showed higher nutrient content and ED than stover. It also had higher gas production but its pH, ammonia-N, and total VFA concentration were lower than corn stover. The best nutrient composition of corn grain was observed in early-harvest (high CP, EE, NDF, OM, NFC, and low ADF). Early-harvest of corn grain also had high effective degradability of dry matter (EDDM), effective degradability of neutral detergent fiber (EDNDF), effective degradability of acid detergent fiber (EDADF), and total VFA concentration. On the other hand, the best nutrient composition of stover was observed in mid-harvest (high DM, CP, NDF, and low ADF). EDDM, EDNDF, and EDADF were pronounced in early-harvest and mid-harvest of stover but the latter showed high total VFA concentration. Hence, early and mid-harvested corn stover and grain in a mountainous area preserved their nutrients, which led to the effective degradation of major nutrients and high VFA production.

Effects of Rice Koji Treatment and Salt Content on the Fermentation Characteristics of the Fish Sauce Made from the Pacific Sand Lance Ammodytes personatus (쌀코지 처리 및 식염함량이 동해안 까나리(Ammodytes personatus) 어간장의 발효특성에 미치는 영향)

  • This study investigated changes in the physicochemical and microbiological properties of fermented fish sauces based on the pacific sand lance Ammodytes personatus during fermentation according to rice koji treatments and salt contents. In total, six fish sauces were prepared, with rice koji treatment for 24 or 72 h before salt addition, and salt contents of 10 or 20%, including two controls to compare the response to different salt contents without koji treatment. The initial pH values of the sauces with koji treatments were notably lower than those of the controls. Over the 10 months of fermentation, the amino nitrogen contents of the sauces with 10% salt were higher than those with 20% salt, while the volatile base nitrogen value was also increased. In the microbial community, the initial numbers of lactic acid bacteria and mold were higher in the sauces with koji treatment than in the controls. After 8 months of fermentation, the free amino acid and organic acid contents were higher in the sauces with koji treatment than in the controls. Although the fermentation rate was affected to a greater extent by salt contents, the rice koji treatment also exerted a positive effect on the acceleration of fermentation.

Effect of Transplanting Times on the Content of Aromatic Substances in Tobacco Cultivar, Hyangcho (이식시기(移植時期)가 향초(香草)(Nicotiana tabacum L.)의 향끽미(香喫味) 물질(物質) 발현(發現)에 미치는 영향(影響))

  • This study was conducted to establish the most appropriate transplanting time which can attribute to increase of aromatic substances in tobacco variety, Hyangcho, maintaining its original aroma. The highest content of total alkaloid, total nicotine and total nitrogenous compounds were observed in the late transplanting time such as July 5, showing 4.16%, 3.83% and 3.68%, respectively. However, the petroleum ether extract was highest in the early transplanted one such as April 5, showing 5.77% and total sugar content, 12% in April 20. The content of petroleum ether extract decreased as the transplanting time delayed. The early transplanting of Hyangcho on April 20 under vinyl mulching showed the increase of fatty acid content, but the decrease of non-volatile organic acids such as malic and citric acids than those of the conventional transplanting time on March 5. The earlier transplanting time also increased the content of volatile acids such as 2,3-methylbutanoic acid and 3-methyl-pentanoic acid, which seem to be related to tobacco aroma.

Effect of Season on Volatile Organic Compounds and Volatile Fatty Acids Concentration in finishing Pig Slurry to Grassland (초지환원용 비육돈 슬러리의 계절에 따른 휘발성유기물과 휘발성지방산 농도 분석연구)

  • The objective of this study is to investigate the changes in levels of odorous compounds in pig slurry during different seasons. Slurry from pens of finishing pigs was sampled every 4-wk and concentration of odorous compounds was analyzed. There was no difference in the range of phenols level (123 to 156 ppm) during spring (April to May), summer (July to August) and fall (October to November). The concentration of indoles was higher (P<0.05) during spring (14.3 ppm) than summer and fall (5.4~7.6 ppm). Level of BCFA ranging from 727 to 1,194 ppm was not different at any season. Among SCFA, there was no difference in propionic acid during any season but levels of acetic acid and butyric acid were highest (P<0.05) during spring season. Concentration of odorous compounds in pig slurry was highest during spring season. Interestingly, it tended to be lower during summer season compare to fall. This result might be due to relatively lower ventilation rate in order to maintain a constant temperature during spring season. Further study will be necessary to determine the relationship between the concentration of odorous compounds and ventilation system.

  • This study was carried out to develop the powdered Katsuobushi (a kind of boiled, smoked, and dried fish product which is used for seasoning soup as it is.) using skipjack as a natural flavoring substance. The processing conditions of the powdered Katsuobushi and the changes of taste compounds during processing of the products were examined. In preparation of the powdered Katsuobushl, frozen skipjack was thawed, beheaded, gutted, filleted and then sliced to 1cm of thickness. The silted meats were boiled in skipjack extract for 20 minutes, and then it was smoked for, 3 times to $10\~12\%$ moisture content at $80^{\circ}C$ for 8 hours. The smoked - dried meats were followed to be 50 mesh of particle size. The effect of slicing and boiling in skipjack meat extract on enhancing flavor and on preventing lipid oxidation of product during processing were observed. The moisture content and crude lipid content of the powdered Katsuobushi was in the range of 11 to $12\%$ and 4.3 to $4.8\%$, respectively. The taste compounds of the product were nucleotides and their related compounds, 1135.8mg/100g ; free amino acid and related compounds, 2210.2mg/100g ; non-volatile organic acids, 1148.0mg/100g ; and total creatinine. 592.1mg/100g on dry basis, and small amount of betaine and TMAO. The major elements of mineral in the product were found to be K, Mg, Na, and Ca. The content of IMP was 542.0mg/100g, and major free amino acids were found to be histidine, anserine, taurine, carnosine and alanine of which occupied to $83.6\%$ of total free amino acids. In non -volatile organic acids, major ones were lactic acid, succinic acid, pyroglutamic acid and $\alpha-ketoglutaric$ acid. From the results of the chemical experiments and sensory evaluation, we may conclude that the flavor of the product from present experiment is more desirable than that of conventional products although the processing time used were much shortened than that of conventional method, and it can be commercialized as a seasoning powder.
