Evaluation of Pollution Level for Organic Matter and Trace Metals in Sediments around Taehwa River Estuary, Ulsan (울산 태화강 하구역 퇴적물의 유기물 및 미량금속 오염도 평가)

  • Grain size, the content of ignition loss (IL), and the concentrations of chemical oxygen demand (COD), acid volatile sulfide (AVS), and trace metals (Fe, Mn, Cu, Pb, Zn, Cd, Cr, As, and Hg) in surface sediments from the Taehwa River estuary, Ulsan, were measured to evaluate pollution levels and potential ecological risks of organic matter and trace metals in estuarine sediment. The mean grain size (Mz) of sediments in the study region ranged from $-0.8-7.7{\varphi}$ (mean $2.8{\pm}2.4{\varphi}$). Surface sediments in the upstream region of the Taehwa River were mainly composed of coarse sediments compared to the downstream region. The concentrations of IL, COD, AVS and trace metals in the sediment were much higher at downstream sites of Myeongchon Bridge in the vicinity of industrial complexes than at upstream sites of those in the vicinity of the residential areas due to the anthropogenic input of organic matter and trace metals by industrial activities. On the basis of several geochemical assessment techniques [sediment quality guidelines (SQGs), enrichment factor (EF), geoaccumulation index ($I_{geo}$), pollution load index (PLI) and ecological risk index (ERI)], the surfaces sediments in the study region are not highly polluted for trace metals, except for As. However, the higher concentrations in downstream study regions of the Taehwa River could impact benthic organisms including shellfish (i.e. Manila clam) in sediments.

Distributions of Organic Matter and Trace Metals in Intertidal Surface Sediment from the Mokpo-Haenam Coast (목포-해남 연안 조간대 퇴적물중 유기물 및 미량금속 분포 특성)

  • To evaluate the organic matter and trace metal pollution in intertidal sediment of the coastal zone, various geochemical parameters (grain size, ignition loss [IL], chemical oxygen demand [COD], acid volatile sulfide [AVS], and metals [Al, Fe, Cu, Pb, Zn, Cd, Cr, Mn, Hg, and As]) were measured for the intertidal surface sediment of the mainland and islands between Mokpo and Haenam in the southwestern coast of Korea. The surface sediments consist mainly of finer sediments, such as mud and silt. The concentrations of IL, COD, and trace metals in intertidal sediment were relatively high in the shoreline of the mainland than in that of islands and those in some stations exceeded the sediment quality guidelines (SQGs). Moreover, the concentrations of IL, COD, and trace metals (except As) in sediment showed relatively good positive correlations with mean grain size, indicating that the concentrations of organic matter and trace metals in intertidal sediment of the study region are dependent on grain size of sediment. Pollution evaluation for trace metals using geochemical assessment techniques, such as enrichment factor, geoaccumulation index, and SQGs, suggested that the intertidal sediments in the study region show light pollution with Cr and moderate pollution with As. More extensive interdisciplinary studies are required to determine the potential causes of As pollution in intertidal sediment.

Synthesis of AlPO4-type Mesoporous Materials Using Alum Sludge (Alum 슬러지를 이용한 AlPO4-계 다공성 물질의 합성)

  • In this study, the formation of $AlPO_4$-type porous materials from alum sludge was investigated. The materials were synthesized by the reaction of aluminum hydroxide and phosphoric acid with an organic template. Cationic surfactant, natural humic acid, and amino acids were used for the organic template. The residual organic templates were removed by calcination at $600^{\circ}C$ in the air. Powder X-ray diffraction patterns showed the charicteristic patterns of the $AlPO_4$-type porous materials. The morphology of the material was examined using a scanning electron microscopy. The coordination environment of $Al^{3+}$ ion was investigated by $^{27}Al$ MAS NMR technique. Both tetrahedrally and octahedrally coordinated$Al^{3+}$ ions were found in the as-synthesized samples while all $Al^{3+}$ ions were tetrahedrally coordinated in the calcined products. The development of mesopore in the solid material was confirmed by the measurement of BET specific surface area. Finally, they were used for removal of toxic formaldehyde from the air and the formaldehyde molecules were adsorbed on the surface of pores. In conclusion, $AlPO_4$-type porous materials from alum sludge might be applicable in the removal of toxic volatile organic compounds from the air.

Effects of Pre-heating on the Flavor of Garlic (예열처리가 마늘의 향미성분에 미치는 영향)

  • Garlic pieces without peeling were subjected to hot air heating at $60{\sim}90^{\circ}C$ for 30 min. and microwave oven heating for 10 sec to reduce the strength of garlic flavor. Volatile compounds of the treated garlic were decreased, as the heating temperature was raised. Garlic heated at 70, 80 and $90^{\circ}C$ had 20%, 31% and 49% less volatile compounds, respectively, compared to those of the control. Microwave oven heating revealed a similar trend to those heated at $60{\sim}80^{\circ}C$. As the heating temperature was raised, glucose, fructose and fructooligosaccharide contents in the heated garlic were decreased. Pyruvic acid contents in garlic, a degradation product of alliin, was decreased as the heating temperature was raised, but contents of the other organic acids were not affected. Arginine, the main amino acid in garlic was decreased by the hot air heating, but was not affected by the microwave heating. Glutamic acid and tyrosine contents were decreased in all the treated garlic samples.

Application of lactic acid bacteria producing antifungal substance and carboxylesterase on whole crop rice silage with different dry matter

  • Objective: This study was conducted to investigate effects of antifungal substance and carboxylesterase-producing inoculant on fermentation indices and rumen degradation kinetics of whole crop rice (WCR) silage ensiled at different dry matter (DM) contents. Methods: Dual-purpose inoculants, Lactobacillus brevis 5M2 and Lactobacillus buchneri 6M1, confirmed both activities of antifungal and carboxylesterase in the previous study. The WCR at mature stage was chopped, and then wilted to obtain three different DM contents consisting of 35.4%, 43.6%, and 51.5%. All WCR forages were applied distilled water (CON) or mixed inoculants with 1:1 ratio at 1×105 colony forming unit/g (INO), and ensiled into 20 L mini silo (5 kg) in quadruplicates for 108 d. Results: The INO silages had lower lactate (p<0.001) and butyrate (p = 0.022) with higher acetate (p<0.001) and propionate (p<0.001) than those of CON silages. Ammonia-N (p<0.001), lactate (tendency; p = 0.068), acetate (p = 0.030), and butyrate (p<0.001) concentrations of INO silages decreased linearly with increasing DM content of WCR forage. The INO silages presented higher lactic acid bacteria (p<0.001) with lower molds (p<0.001) than those of CON silages. Yeasts (p = 0.042) and molds (p = 0.046) of WCR silages decreased linearly with increasing DM content of WCR forage. In the rumen, INO silages had higher the total degradable fraction (p<0.001), total volatile fatty acid (tendency; p = 0.097), and acetate (p = 0.007), but lower the fractional degradation rate (p = 0.011) and propionate (p<0.001) than those of CON silage. The total degradable fraction (p<0.001), total volatile fatty acid (p = 0.001), iso-butyrate (p = 0.036), and valerate (p = 0.008) decreased linearly with increasing DM content of WCR forage, while the lag phase (p<0.001) was increased linearly. Conclusion: This study concluded that application of dual-purpose inoculants on WCR silage confirmed antifungal and carboxylesterase activities by inhibiting mold and improving rumen digestibility, while increase of wilting times decreased organic acids production and rumen digestibility.

The Effects of Additives in Napier Grass Silages on Chemical Composition, Feed Intake, Nutrient Digestibility and Rumen Fermentation

  • The effect of silage additives on ensiling characteristics and nutritive value of Napier grass (Pennisetum purpureum) silages was studied. Napier grass silages were made with no additive, fermented juice of epiphytic lactic acid bacteria (FJLB), molasses or cassava meal. The ensiling characteristics were determined by ensiling Napier grass silages in airtight plastic pouches for 2, 4, 7, 14, 21 and 45 d. The effect of Napier grass silages treated with these additives on voluntary feed intake, digestibility, rumen fermentation and microbial rumen fermentation was determined in 4 fistulated cows using $4{\times}4$ Latin square design. The pH value of the treated silages rapidly decreased, and reached to the lowest value within 7 d of the start of fermentation, as compared to the control. Lactic acid content of silages treated with FJLB was stable at 14 d of fermentation and constant until 45 d of ensiling. At 45 d of ensiling, neutral detergent fiber (NDF) and acid detergent fiber (ADF) of silage treated with cassava meal were significantly lower (p<0.05) than the others. In the feeding trial, the intake of silage increased (p<0.05) in the cow fed with the treated silage. Among the treatments, dry matter intake was the lowest in the silage treated with cassava meal. The organic matter, crude protein and NDF digestibility of the silage treated with molasses was higher than the silage without additive and the silage treated with FJLB. The rumen parameters: ruminal pH, ammonia-nitrogen ($NH_3$-N), volatile fatty acid (VFA), blood urea nitrogen (BUN) and bacterial populations were not significantly different among the treatments. In conclusion, these studies confirmed that the applying of molasses improved fermentative quality, feed intake and digestibility of Napier grass.

Changes in the Components of Persimmon Vinegars by Two Stages Fermentation (II) (2단계 발효에 의한 감식초의 성분 변화 (II))

  • This study was determined changes of components of sweet and astringent persimmon vinegars by two stages fermentation. Free sugars of persimmon juices before alcohol fermentation were mainly composed of glucose, fructose and sucrose. The content of glucose, fructose and sucrose of sweet persimmon juice was 6.60, 6.12 and 1.74%, respectively, and those of astringent persimmon was 5.63, 5.21, 0.62%, respectively. The contents of free sugar decreased continuously during fermentation. Major organic acids of persimmon juices were acetic, galacturonic, malic, citric and ascorbic acid. Alcohols of persimmon juices was detected methanol, ethanol, iso-propylalcohol, n-propylalcohol and iso-butylalcohol at the initial fermentation. The contents of alcohols increased continuously up to 4days of fermentation but their contents except ethanol decreased slightly at 5th day of fermentation. Contents of free amino acid were higher in sweet persimmon than those in astringent persimmon. Volatile components increased during acetic acid fermentation.

Improvement of Nutritive Value and In vitro Ruminal Fermentation of Leucaena Silage by Molasses and Urea Supplementation

  • Leucaena silage was supplemented with different levels of molasses and urea to study its nutritive value and in vitro rumen fermentation efficiency. The ensiling study was randomly assigned according to a $3{\times}3$ factorial arrangement in which the first factor was molasses (M) supplement at 0%, 1%, and 2% of crop dry matter (DM) and the second was urea (U) supplement as 0%, 0.5%, and 1% of the crop DM, respectively. After 28 days of ensiling, the silage samples were collected and analyzed for chemical composition. All the nine Leucaena silages were kept for study of rumen fermentation efficiency using in vitro gas production techniques. The present result shows that supplementation of U or M did not affect DM, organic matter, neutral detergent fiber, and acid detergent fiber content in the silage. However, increasing level of U supplementation increased crude protein content while M level did not show any effect. Moreover, the combination of U and M supplement decreased the content of mimosine concentration especially with M2U1 (molasses 2% and urea 1%) silage. The result of the in vitro study shows that gas production kinetics, cumulation gas at 96 h and in vitro true digestibility increased with the increasing level of U and M supplementation especially in the combination treatments. Supplementation of M and U resulted in increasing propionic acid and total volatile fatty acid whereas, acetic acid, butyric acid concentrations and methane production were not changed. In addition, increasing U level supplementation increased $NH_3$-N concentration. Result from real-time polymerase chain reaction revealed a significant effect on total bacteria, whereas F. succinogenes and R. flavefaciens population while R. albus was not affected by the M and U supplementation. Based on this study, it could be concluded that M and urea U supplementation could improve the nutritive value of Leucaena silage and enhance in vitro rumen fermentation efficiency. This study also suggested that the combination use of M and U supplementation level was at 2% and 1%, respectively.

Quality Characteristics of Korean Domestic Commercial Meoru Wines (국산 시판 머루 와인의 품질 특성)

  • This study was carried out to investigate quality characteristics of Korean domestic commercial meoru wines, 8 kinds of wines were purchased from the Korea Wine Festival in 2016 and we compared the characteristics and physiological activity of 8 domestic wine. The results of this study have shown that the alcohol contents of wines ranged from 9.8 to 14.3%, pH of wines ranged from 3.86 to 4.22 and the total acidity of wines ranged from 0.56~0.75%. The hue value of meoru wines ranged from 0.81 to 1.02, The brightness of meoru wines ranged from 0.77 to 5.55, the redness from 3.97 to 31.16, and the yellowness from 0.99 to 5.63. The organic acid analysis of wine revealed lactic acid content at 4.281~9.606 mg/mL, followed by malic acid, tartaric acid, and acetic acid. The concentrations of total polyphenol and anthocyanin contents in the samples were investigated by spectrophotometric methods. Total polyphenol contents of the M8 (172.24 mg%) wine was higher than those of the other wines and total anthocyanin contents represented from 356.69 to 601.33 mg/mL. The DPPH radical scavenging activity of meoru wines was between 57.23 to 73.98%. Volatile flavor component analysis of meoru wines identified 7 alcohols, 16 esters, 5 acids, 3 terpenes and 4 other compounds.

Effect of Lactic Acid Producing Bacteria on the Performance of Male Crossbred Calves Fed Roughage Based Diet

  • To study the effect of feeding lactic acid producing bacteria on the performance of cattle calves, twenty four, day old male crossbred cattle calves (Bos indicus${\times}$Bos taurus), were distributed into two groups of 12 animals each. The animals were fed on calf starter containing wheat bran and green berseem ad libitum and milk as per requirement upto 8 weeks of age. The diet of calves of Group 2 was supplemented with 500 ml culture of Lactobacillus acidophilus-15. Total duration of the experiment was 31 weeks. There was no significant difference in intake and digestibility of dry matter (DM), organic matter (OM), neutral detergent fibre (NDF), acid detergent fibre (ADF) and crude protein (CP) between the groups. The rumen pH, protozoa numbers, concentration of volatile fatty acids (VFA), ammonia nitrogen ($NH_3-N$), trichloroacetic acid precipitable nitrogen (TCA-ppt N) and activity of microbial enzymes (carboxymethylcellulase, xylanase, amylase and protease) were not affected due to probiotic supplementation. Average live weight gain of the calves was improved (about 10%) and feed:gain ratio was reduced (about 5%) in the animals given Lactobacillus culture. The data indicated that crossbred calves could be reared on a diet devoid of cereal grain and addition of Lactobacillus culture in the diet resulted in an added advantage in growth performance of the animals.
