Apple Virus Diagnosis Using Simplified RNA Extraction Method (사과바이러스 간편 진단을 위한 RNA추출법 개선)

  • Kyungsan nursery complex which has a vast area for the production of various species of fruit tree stocks is in a high demand of virus-free saplings. Apple tree stocks, the most important products, urgently need more rapid and reliable viral diagnosis. In this study, a bead beater was tested because of convenience in dealing with large number of samples. Also, industrial glass bead abrasive (0.4 mm in diameter) at very low cost was used in a disposable way. For bead beater-aided RNA extraction from apple stem tissues, the guanidine thiocyanate method was confirmed to be very reliable. Silca membrane filter tube in connection to vacuum filtering device was strongly suggested for simplifying RNA capture and washing steps. Apple virus detection was confirmed by RT-PCR.

  • The mumps virus is a single-stranded, non-segmented, negative-sense RNA virus belonging to the $Paramyxoviridae$ family. Mumps is characterized by bilateral or unilateral swelling of the parotid gland. Aseptic meningitis is a common complication, and orchitis is also common in adolescents and adult men. Diagnosis is based on clinical findings, but because of high vaccination coverage, clinical findings alone are not sufficient for diagnosis, and laboratory confirmation is needed. Mumps is preventable by vaccination, but despite high vaccination coverage, epidemics occur in several countries, including Korea. Many hypotheses are suggested for these phenomena. In this review, we investigate the reason for the epidemics, optimal methods of diagnosis, and surveillance of immunization status for the prevention of future epidemics.

Analysis of Patent Trend on Dengue Virus Detection Technology (뎅기 바이러스 검출기술 관련 특허동향 분석)

  • Dengue virus is a typical mosquito-borne virus, and the half of the world's population is exposed to infection. Dengue virus causes relatively mild symptoms such as dengue fever. However, when not treated properly, it is known to cause severe symptoms such as dengue hemorrhagic fever and dengue shock syndrome with a mortality rate of over 20%. Development of dengue virus detection technology is very important because it is reported that early diagnosis of dengue fever can lower the mortality rate to less than 1%. In this study, patent search related to dengue virus detection technology was conducted in Korea, USA, Europe, Japan, and China. The quantitative analysis of 69 validated patents from the searched patents was conducted by country, year, and patent holder. In addition, in-depth analysis was carried out by classifying into three categories: molecular diagnostics, immuno-diagnostics, and cell culture-based diagnostics from all validated patents. From these results, we analyzed the patent trend related to dengue virus detection and dengue fever diagnosis technology and discussed the features and limitations of molecular diagnostics and immuno-diagnostics at present level. Furthermore, we discussed the direction of technology development and future prospects to overcome limitations.

Occurrence and Multiplex PCR Detection of Citrus Yellow Vein Clearing Virus in Korea

  • Citrus yellow vein clearing virus (CYVCV) is a member of the Alphaflexiviridae family that causes yellow vein clearing symptoms on citrus leaves. A total of 118 leaf samples from nine regions of six provinces in Korea were collected from various citrus species in 2020 and 2021. Viral diagnosis using next-generation sequencing and reverse transcription polymerase chain reaction (RT-PCR) identified four viruses: citrus tristeza virus, citrus leaf blotch virus, citrus vein enation virus, and CYVCV. A CYVCV incidence of 9.3% was observed in six host plants, including calamansi, kumquat, Persian lime, and Eureka lemon. Among the citrus infected by CYVCV, only three samples showed a single infection; the other showed a mixed infection with other viruses. Eureka lemon and Persian lime exhibited yellow vein clearing, leaf distortion, and water-soak symptom underside of the leaves, while the other hosts showed only yellowing symptoms on the leaves. The complete genome sequences were obtained from five CYVCV isolates. Comparison of the isolates reported from the different geographical regions and hosts revealed the high sequence identity (95.2% to 98.8%). Phylogenetic analysis indicated that all the five isolates from Korea were clustered into same clade but were not distinctly apart from isolates from China, Pakistan, India, and Türkiye. To develop an efficient diagnosis system for the four viruses, a simultaneous detection method was constructed using multiplex RT-PCR. Sensitivity evaluation, simplex RT-PCR, and stability testing were conducted to verify the multiplex RT-PCR system developed in this study. This information will be useful for developing effective disease management strategies for citrus growers in Korea.

  • An enzyme-linked immunosorbent assay (ELISA) was studied for the rapid diagnosis of the flacherie virus (FV) of the silkworm, Bombyx mori. The optimised concentration of rabbit anti-FV IgG and enzyme conjugate for the this technique were 15$\mu\textrm{g}$/$m\ell$ and 1:100 dilution, respectively. In ELISA, the detectable concentation of purified FV was 15ng/$m\ell$, and the flacherie viral antigens in the larval extracts were detected as early as 24 hours after the experimental infection. The results indicated that ELISA technique proved to be applicable for the rapid diagnosis of flacherie virus disease.

Simple and Rapid Detection for Rice stripe virus Using RT-PCR and Porous Ceramic Cubes (RT-PCR과 다공성 세라믹 큐브를 이용한 벼줄무늬잎마름바이러스 간편 진단)

  • A rapid and simple RT-PCR diagnosis method for detection of Rice stripe virus (RSV), one of major virus infecting rice, was developed using porous ceramic cubes in this study. The porous ceramic cube can rapidly absorb biological molecules such as small-sized proteins and nucleic acid fragments into its pores. We examined whether this ability of porous ceramic cubes could be applied for isolating viral nucleic acids or particles from the RSV- infected plant tissues. In this study, we found that the porous ceramic cube was capable of absorbing a detection level of viruses from the rice tissues infected with RSV and established RT-PCR-based RNA diagnosis method using porous ceramic cubes.

  • In these days, malicious codes have become reality and evolved significantly to become one of the greatest threats to the modern society where important information is stored, processed, and accessed through the internet and the computers. Computer virus is a common type of malicious codes. The standard techniques in anti-virus industry is still based on signatures matching. The detection mechanism searches for a signature pattern that identifies a particular virus or stain of viruses. Though more accurate in detecting known viruses, the technique falls short for detecting new or unknown viruses for which no identifying patterns present. To cope with this problem, anti-virus software has to incorporate the learning mechanism and heuristic. In this paper, we propose a fuzzy diagnosis system(FDS) using fuzzy c-means algorithm(FCM) for the cluster analysis and a decision status measure for giving a diagnosis. We compare proposed system FDS to three well known classifiers-KNN, RF, SVM. Experimental results show that the proposed approach can detect unknown viruses effectively.

  • Ten pigs infected with Aujeszky's disease virus (ADY) or hog cholera virus(HCV) were tested for the detection of virus antigens in frozens or paraffin-embedded sections by avidin-biotin-peroxidase complex(ABC) method. Tonsils, spleens, cerebra and buffy coats were examined for the immunohistochemical test. Where ADV antigen was detected by ABC, a dark brown deposit occurred in both the nucleus and the cytoplasm of lymphocytes and macrophages, however, HCV antigen was demonstrated in the cytoplasm of the infected cells. ADV-positive cells were most frequently detected in tonsils and cerebra, whereas, HCV -positive cells were frequently observed in spleens. And buffy coat were also good for both virus detection. The results suggested that ABC method is considered as an excellent and reliable tool for confirmative diagnosis of these viral diseases.

Acute Epstein-Barr Virus Hepatitis in a 32 Month Old Female Manifesting as Henoch-Sch$\ddot{o}$nlein Purpura

  • Henoch-Sch$\ddot{o}$nlein purpura can result from exposure to an antigen after infection with several types of organisms. However, Henoch-Sch$\ddot{o}$nlein purpura caused by a primary Epstein-Barr virus infection has been rarely reported. Here, we report the case of a 32-month-old female patient who presented with Henoch-Sch$\ddot{o}$nlein purpura. Based on abnormal liver function test results and positive results for Epstein-Barr virus infection markers, a diagnosis of Epstein-Barr virus hepatitis manifesting as Henoch-Sch$\ddot{o}$nlein purpura was made. Treatment with methyl-prednisolone and hydration improved the symptoms, and a switch to oral steroids was effective in completely alleviating the purpura. No recurrence was noted and no liver function abnormalities were detected during the follow up period.

Production of monoclonal antibodies against viral hemorrhagic septicemia virus (VHSV, genotype IVa) from olive flounder (넙치 유래 viral hemorrhagic septicemia virus (VHSV, genotype IVa)에 대한 단클론 항체 생산)

  • Mouse monoclonal antibodies (MAbs) were produced by using viral hemorrhagic septicemia virus (VHSV, genotype IVa) as an immunogen, isolated from diseased olive flounder (Paralichthys olivaceus). Four hybridoma clones secreting MAbs against VHSV were established. The MAbs were recognized the nucleoprotein (MAb 4), phosphoprotein (MAb 1) and matrix protein (MAbs 2 and 3) of VHSV by western blot analysis. Among them, the MAbs 1 and 4 strongly reacted with the VHSV-infected FHM cells, but not normal FHM cells. In enzyme linked immunosorbent assay, the four MAbs reacted with the VHSV, but not different six fish viruses (infectious hematopoietic necrosis virus, hirame rhabdovirus, spring viraemia of carp virus, infectious pancreatic necrosis virus, marine birnavirus and nervous necrosis virus). These results indicate that the MAbs are useful for diagnosis of VHSV infection.
