젤라틴입자응집반응법을 이용한 국내 시판 고추종자의 Tobamovirus 검출 및 오염률 조사 (Detection of Tobamoviruses and Survey on Contamination Rate in Commercial Pepper Seeds Using Gelatin Particle Agglutination Test)

  • 시판고추 종자로부터 Tobamovirus를 검출하고 Tobamovirus 오염률을 조사하고자 gelatin particle agglutination test (GPAT)의 최적조건을 조사하였다. Gelatin 입자에 감작할 담배 모자이크 바이러스 고추 계통에 대한 ${\gamma}$-globulin G 의 최적농도는 100$\mu\textrm{g}$/$m\ell$이었고, GPAT의 바이러스 검출 감도는 효소면역항체법 및 dot immuno-binding assay와 유사하였으며, Tobamovirus 검출을 위한 종자시료의 최적 희석배수는 5배에서 25배였다. 그리고 최적화된 GPAT에 의한 국내시판 고추종자의 Tobamovirus 오염률은 평균 79.1%인 것으로 판명되었다.

  • In the course of our search for anti-Herpes simplex virus type I (HSV 1) substances from natural sources, we screened crude drugs for their antiviral activity using SRB assay. T he methanol extract from herb of Brassica rapa (Cruciferae) was found to inhibit HSV-1. Though bioassay-directed fractionation of the extract, anti-HSV-1 agent was isolated by chromatographic separation using Amberlite XAD-4 and Sephadex LH-20. The structure of compound I was elucidated by spectral means including $^1H-^1H$ COSY, HMQC and HMBC to be isorhamnetin 3-O-${\beta}$-D-glucopyranoside (compound I). Compound I was active against HSV-1 with the 50% effective concentration of O.42mg/ml and the 50% cytotoxicity of 5.0mg/ml.

화학적 제어제에 의한 담배모자이크 바이러스의 불활성화 (Inactivation by Chemical Disinfectants in vitro against Tobacco Mosaic Virus)

  • 담배모자이크 바이러스에 대한 항바이러스 활성을 검사하기 위해 다양한 화학제를 처리한 결과, 1NHCl과 0.1-1N NaOH에 바이러스와 외피에 보호된 RNA가 완전하고 신속하게 분해되었다. TMV를 0.1N HCl에 처리하였을 때 바이러스의 외피단백질은 산의 가수분해에 의해 부분 분해되었으나, 0.01N HCl 혹은 0.01N NaOH에 처리했을 때는 분해되지 않았다. 위의 결과에서 적절한 산이나 알칼리에 처리하는 것은 바이러스를 제거하는데 효과적인 가치가 있음이 판명되었다. 50% isopropanol과 UV처리는 바이러스와 외피화된 RNA에 어떤 효과도 미치지 않았다. 농장의 농기구와 실험실 기구를 바이러스의 오염으로부터 방지하기 위해서는 적절한 화학제의 적용이 필요하다.

  • 고양이 백혈병 바이러스에서 reverse transcriptase를 분리하여 생화학적 및 면역학적 연구를 하였다. 분자량은 72,000이고, DNA polymerase와 RNase H의 활성은 0.05-1 mM $M_n^2+$와 50-80 mM KCl에서 가장 좋았다. DNA polymerase와 RNase H는 같은 단백질 분자에 있으며, chymotrypsin 처리로서 RNase H를 쪼개낼 수 있으며, 이 RNase H도 reverse transcriptase의 항체에 의해서 활성이 거의 억제 된다. Reverse transcriptase의 항체 결합위치와 활성을 내는 위치는 다른 것 같다.

Disinfectant effect of monopersulfate (MPS) compound to white spot syndrome virus (WSSV) of shrimp

  • This study investigated the disinfection effect of monopersulfate (MPS) compound against white spot syndrome virus (WSSV) by bioassay using kuruma shrimp (Marsupenaeus japonicas). A WSSV stock was prepared with muscle homogenate from WSSV-infected whiteleg shrimp (Penaeus vannamei) and its lethal dose 50% endpoint (LD50) and infectious dose 50% endpoint (SID50) were respectively determined as 10-5.63 and 10-6.79 in bioassay using kuruma shrimp, followed by PCR assays. The disinfective effect of MPS compound (1.2 ppm, 2.4 ppm, 4.8 ppm) was performed by bioassay using about 10-fold higher dilution (10-4) of WSSV homogenate. The compound resulted in WSSV inactivation by a concentration-dependent manner. In addition, 4.8 ppm of MPS completely prohibited WSSV infection. To our knowledge, this study is the first report about the usefulness of MPS as a disinfectant to WSSV.

  • Biologically active porcine Interleukin-2(poIL-2) was produced from in vitro and in viva baculovirus expression system, namely the Autographa californica nuclear polyhedrosis virus (ACNPV)-cell culture system and the Hybrid nucler polthedrosis virus (HyNPV)-sillkworm larva system. The concentration of the recombinant poIL-2(rpoIL-2) in the larvae hemolymph was 1 to 3 mg/mL, which was about 7 to 20 times those of the cell culture systems. The level of this expression efficiency is equal to that with transgenic livestock, secretion products in milk.

  • HRccine was administered subcutaneously to rats for 4 weeks at dose levels of 300, 60 and 12 times the expected clinical dose to evaluate the subacute toxicity. There were no treatment-related effects in clinical signs, body weight changes, food consumption, water consumption, urinalysis and blood biochemistry in any dose groups. In hematological examinations, increase of leucocyte counts and decrease of hemoglobin concentration were observed in the high dose-treated group. However, no treatment-attributable pathological changes were observed in microscopic examinations. The no-effect dose in subacute toxicity study of rats was considered to be 300 times the expected clinical dose.

오이 모자이크 바이러스 위성RNA의 cDNA가 도입된 형질전환 담배의 육성 (Transgenic Tobacco Plants Introduced with cDNA of Cucumber Mosaic Virus Satellite RNA)

  • The cDNA of CMV-As satellite RNA was introduced into tobacco plants (Nicotiana tabacum cv. Samsun NN) using a binary Ti plasmid vector system of Agrobacterium tumefaciens. The cDNA of satellite RNA introduced into tobacco plants was detected by polymerase chain reaction (PCR) and molecular hybridization analyses. Symptom development was distinctly suppressed in the transgenic tobacco plants when inoculated with CMV-Co. CMV concentration in the transgenic tobacco plants was decreased to 1/40 of non-transgenic tobacco plants. The kanamycin resistance gene of the transgenic tobacco plants was also detected in the progeny.

  • A DNA chip that rapidly and accurately detects the viral genes in rhabdovirus-infected fish was developed. The N, Ml, and G proteins of three rhabdovirus strains, infectious hematopoietic necrosis virus (IHNV), viral hemorrhagic septicemia virus (VHSV), and flounder rhabdovirus (HIRRV), were selected for use as probes. The sequences of the corresponding genes were obtained, and probes were prepared by PCR using specific primer sets. The specificity of the probes was confirmed by cross PCR. The prepared probes were spotted on poly-L-lysine- or aminosilane-coated glass slides and hybridized with target DNA under several different conditions in order to determine the optimal hybridization temperature, glass-slide coating, and target cDNA concentration.

  • For the development of anti-AIDS agents, thirty-seven methanol extracts of Korean plant materials were tested for their inhibitory effects on human immunodeficiency virus type-1 (HIV-1) protease. Extracts of seven plants showed more than 30% inhibitory activities on HIV-1 protease at a concentration of $100\;{\mu}g/ml$. The bark of Berchemia berchemiaefolia, the leaf of Lindera erythrocarpa and the whole plant of Siegesbeckia pubescens exhibited significant inhibititory activities on HIV-1 protease with 56.2, 50.8, and 46.6%, respectively.
