Induction of Cytotoxic T Lymphocyte Response against the Core and NS3 Genes of the Hepatitis C Virus in Balb/c Mice

  • Hepatitis C virus (HCV) is a positive strand RNA virus of the Flaviviridae family and the major cause of post-transfusion non-A, non-B hepatitis. Vaccine development for HCV is essential but has been slowed by poor understanding of the type of immunity that naturally terminates HCV infection. The DNA-based immunization technique offers the potential advantage of including cellular immune responses against conserved internal proteins of a virus, as well as the generation of antibodies to viral surface proteins. Here, we demonstrate that cell lines expressing the HCV core and/or NS3 proteins can induce a specific CTL response in mice, and these results suggest a possibility that the HCV core and NS3 DNA can be used to induce CTL activity against the antigen in mice and can be further developed as a therapeutic and preventive DNA vaccine.

  • Expression in yeast may prove more amenable to generating large amounts of viral antigens for a vaccine candidate. We, therefore, cloned the gene encoding the Hepatitis C virus (HCV) structural proteins (C-El-E2, c740) fused in-frame with, and immediately 3' to, the chicken-lysozyme signal peptide (C-SIG) gene and under the control of the yeast glyceraldehyde-3-phosphate dehydrogenase gene promoter. In yeast, the HCV structural proteins were expressed in two different forms: a processed and a nonprocessed aggregated form. Biophysical characterization by sucrose linear gradient centrifugation revealed that both forms were present in the same fractions with a buoyant density of 1.127-1.176 g/$cm^3$. These findings suggest that the efficient synthesis of HCV structural proteins in yeast may be an important tool to study virus assembly and may lead to the development of an HCV vaccine.

Synergistic effect of ribavirin and vaccine for protection during early infection stage of foot-and-mouth disease

  • In many countries, vaccines are used for the prevention of foot-and-mouth disease (FMD). However, because there is no protection against FMD immediately after vaccination, research and development on antiviral agents is being conducted to induce protection until immunological competence is produced. This study tested whether well-known chemicals used as RNA virus treatment agents had inhibitory effects on FMD viruses (FMDVs) and demonstrated that ribavirin showed antiviral effects against FMDV in vitro/in vivo. In addition, it was observed that combining the administration of the antiviral agents orally and complementary therapy with vaccines synergistically enhanced antiviral activity and preserved the survival rate and body weight in the experimental animals. Antiviral agents mixed with an adjuvant were inoculated intramuscularly along with the vaccines, thereby inhibiting virus replication after injection and verifying that it was possible to induce early protection against viral infection prior to immunity being achieved through the vaccine. Finally, pigs treated with antiviral agents and vaccines showed no clinical signs and had low virus excretion. Based on these results, it is expected that this combined approach could be a therapeutic and preventive treatment for early protection against FMD.

Ginseng, a promising choice for SARS-COV-2: A mini review

  • The current Covid-19 pandemic has changed the entire world and bought so many unprecedented challenges to the scientific community. More than 5 million people died due to the SARS-COV-2 outbreak. For many thousands of years, ginseng, the traditional herb has been used for various infectious diseases by traditional healers. Ginseng showed promising antiviral effects by modulating both natural and acquired immunity. Ginseng might be used as a potential therapeutic agent to prevent SARS-CoV-2 infection along with the vaccine. In this current review, we offer an alternative approach for SARS-COV-2 prevention during this unprecedented pandemic.

  • The ongoing global pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has not only influenced over 1.26 billion people but also caused 2.77 million deaths worldwide (as of March 28, 2021). The vaccination could be the most efficient strategy to prevent SARS-CoV-2 infection. However, the continuous emergence of novel variants such as VUI-202012/01 (United Kingdom) and 501.V2 (South Africa) raises huge concerns about the effectiveness of the vaccine designed to target the original virus strain. Since ancient times regardless of the East and West, the plants which refered in this presentation have been consumed not only as food but also as a natural medicine to treat diverse diseases including infectious diseases. Importantly, these plants contain secondary metabolites that display antiviral activity involved in the inhibition of viral adsorption, penetration, and replication. Also, plant-derived natural medicines are expected to have a wider range of efficacy and fewer side effects than synthetic medicine, discovering novel plant-based viral agents would be a promising strategy to fight against SARS-CoV-2.

Serological characterization of bovine viral diarrhea virus isolates

  • Bovine viral diarrhea viruses (BVDVs) were isolated from cattle with respiratory and diarrhea signs as well as persistently infected cattle. These isolates were analysed serologically to characterize serogroups and to compare serological relationship with reference viruses of type I and II. Most isolates from calf diarrheal cases and persistently infected individuals showed a significant difference in cross-neutralization test with the viruses isolated from nasal discharges showing severe respiratory signs. Serologically most of the commercial vaccine strains could be classified into classical BVDV (type I) such as NADL strain. This serological difference among BVDV isolates suggested the need for new vaccines to protect cattle from both respiratory and enteric BVDV infections in field. The immunogenicity of BVDVs which showed a good propagation capability in MDBK cells and high rates of neutralizing activity (isolate : KD26-1, PHG, B5 and 95002) against all viruses used in this study, was confirmed in guinea pig when treated as single or combined groups.

DISTRIBUTION OF VIRUSES IN WATER ENVIRONMENT

  • We investigated the viral contamination of water environment including tap water in Korea. River water used for source water was analyzed about monthly between 1997 and 1999 over a period 26 months. A total of 22 tap water samples were collected in 10 sites in 2 urban areas between 1997 and 1998 over a 11 months. All samples were examined for infectious enteroviruses and adenoviruses by a cell culture technique followed by PCR amplification. To identify the recovered viruses from tap water, sequence analysis of PCR products was performed. Infectious viral particles were detected in river water all year round, ranging from 0.93 to 17.3 Most Probable Number of Infectious Unit (MPNIU) /100L. Tap water samples also contained infectious viral particles. The frequency of enteroviruses and adenoviruses in tap water were $50.0\%$ (11/22) and $36.7\%$ (8/22), respectively. Both enteroviruses and adenoviruses were detected in five tap water samples $(22.7\%)$. The level of viral contamination in tap water was quite high, ranging from 0.2 to 2.9 MPNIU/100L, far above the recommended virus level in drinking water set by the U.S. EPA. Poliovirus type 1 derived from vaccine was frequently detected and the remainder comprised coxsackievirus B type or echovirus type 6, which were causative agents of aseptic meningitis in Korea in 1997 and 1998, respectively. Several types of adenovirus were detected in tap water samples and some water samples were found to contain adenoviruses which were closely related to enteric adenovirus type 40 and 41. This stusy shows that surface water and tap water in Korea may be exposed to the risk of viral contamination, especially from recently recognized viruses and this constitutes a potential public health hazard.

2000년 영덕군 홍역 유행시 유행 차이에 따른 2개 초등학교 일부 학생에 대한 역학 조사 (Comparative Epidemiologic Survey of Measles in Two Primary Schools)

  • Objectives : During March-May, 2000, a measles outbreak occurred at Youngduk, Korea. This county is divided into two areas with different historical and socioeconomic background. The outbreak occurred in one of these areas. We conducted a comparative epidemiologic study on the two areas in order to evaluate the factors related to the epidemic. Materials and Methods : We selected two groups, grades 3 and 5 in a primary schools in each area. We investigated outbreak-related factors using parent-questionnaires, the vaccination history from the student's health record and the records concerning the recent measles-outbreak from the local health center. Serologic test on measles-IgG and -IgM antibody was done. Results : The infection rate was 31.5% for the epidemic area and 3.7% for non-the epidemic area according to clinical or serological criteria (p<0.001). No difference was seen in the measles vaccination rate, residence at the time of vaccination or past measles infection history between the two areas. In the epidemic area, the attack rate for the 4-6 year-old MMR booster group(20.5%) was higher than the non-booster group(32.4%), but was not found significantly. Vaccine efficacy was 29.6% in the epidemic area and 87.0% in the non-epidemic area (p<0.001). The IgG level and positive rate were significantly different between the two areas (median 10727 IU/ml, 98.9% in epidemic area; median 346 IU/ml, 85.9% in the non-epidemic area, p<0.001). However, the IgG level and positive rate between the measles-cases and non-cases were not significantly different. Conclusions : This outbreak took place in mostly vaccinated children. These results suggest that a reduction of herd immunity for immunity failure after vaccination may be one of the feasible factors related to the outbreak pattern in the two areas. The results of the IgG level and positive rate suggest that re-establishment of a normal value for IgG level and of a qualitative method for IgG are needed.

동물용 생 바이러스 백신에서 Mycoplasma 검출을 위한 PCR 기법 적용 (Application of a PCR Method for the Detection of Mycoplasma in Veterinary Live Viral Vaccines)

  • 동물용 생 바이러스 백신 내에 mycoplasma를 검출하기 위해 polymerase chain reaction (PCR)기법과 2가지의 상품화된 PCR 검출킷트를 평가하였다. PCR기법은 시험에 사용된 모든 mycoplasma를 특이적으로 검출할 수 있었으나, 2가지의 상품화된 PCR 검출킷트는 일부의 mycoplasma를 검출하지 못하였다. 또한, PCR기법의 검출 특이도는 조류 유래 mycoplasma에 속한 4주의 표준주 및 7주의 야외분리주를 모두 검출할 수 있었다. PCR기법의 민감도는 9 CFR Mycoplasma액체배지에서 배양한 Mycoplasma 속균 및 Acholeplasma속균에 대해 $1\~100$ colony forming units/ml까지 검출할 수 있었다. 동물용생 바이러스 백신에 대해 PCR기법의 적용가능성을 평가하기 위해, 돼지 전염성위장염 및 로타바이러스 흔합백신과 개 파보바이러스 백신내에 A. laidlawii를 인공적으로 접종한 후, PCR기법의 민감도를 조사하였을 때 배양액을 이용한 검출한계와 유사하였다. 본 연구에서 사용된 PCR 기법은 동물용 생 바이러스 백신내의 mycoplasma를 신속하고 민감하게 검출할 수 있을 것으로 판단되었다.

바이러스성 크루프로 입원하는 소아 환자의 역학적 특성과 임상적 중증도 평가 (Epidemiology and Clinical Severity of the Hospitalized Children with Viral Croup)

  • 목적: 본 연구는 바이러스성 크루프로 입원하는 환아의 임상적 및 역학적 특성을 분석하여 원인 바이러스 감염에 따른 중증도를 평가하고자 하였다. 방법: 2013년 5월부터 2016년 12월까지 원주세브란스기독병원 소아청소년과에 바이러스성 크루프로 입원한 10세 이하 환아 중 비강인두도말 검체 채취 및 다중 역전사중합효소연쇄반응 검사를 하여 호흡기 바이러스가 검출된 302명을 대상으로 의무기록을 후향적으로 검토하였다. 바이러스성 크루프의 중 증도를 평가하기 위하여 Westley의 점수제를 사용하였다. 결과: 전체 302명 중 중증 바이러스성 크루프로 입원한 환아는 149명(49.3%)이었으며, 이 중 남아가 88명, 여아가 61명으로 남녀 비는 1.44:1이었다. Parainfluenza virus가 110예(48.7%)로 거의 절반에 가까운 빈도를 보였으며, 이후로 influenza virus (15.5%), human rhinovirus (11.9%), respiratory syncytial virus (10.2%) 순이었다. 중증 바이러스성 크루프와 원인 바이러스와의 연관성에 대한 분석에서는 parainfluenza virus 2형에서만 위험도가 의미 있게 높은 것으로 나타났다. Parainfluenza virus 2형은 연령에 따라서는 발병 빈도에 차이가 없었으나 여름, 가을에 상대적으로 더 높은 감염 빈도를 보였다. 결론: 본 연구에서 중증 바이러스성 크루프와 연관이 있었던 바이러스는 parainfluenza virus 2형이 유일하였다. 추후 전향적, 다기관 연구 및 추가적인 변수들을 복합적으로 고려하여 원인 바이러스 감염에 따른 중증도를 재확인하고, 원인 바이러스에 대한 지역별, 시기별, 연령별 분석이 필요하다.
