넙치 (Paralichtys olivaceus) 유래 viral hemorrhagic septicemia virus (VHSV, genotype IVa)의 무지개송어 (Oncorhynchus mykiss) 감염 실험 (Experimental Infection of Rainbow Trout (Oncorhynchus mykiss) with Viral Hemorrhagic Septicemia Virus (VHSV, Genotype IVa) from Olive Flounder (Paralichtys olivaceus))

  • 본 연구에서는 넙치 유래 VHSV (genotype IVa)에 의한 무지개송어의 영향을 조사하기 위하여 다양한 조건에서 감염 실험을 실시하였다. 3개의 다른 로트의 담수 무지개송어 치어에 VHSV ($10^{6.3-7.3}TCID_{50}$/fish)를 복강 주사한 결과, 15% 이하의 누적 폐사율이 관찰되었다. 무지개송어 (in vivo)를 사용하여 VHSV를 계대 배양한 후 감염 실험을 실시한 결과, VHSV를 5회 계대 배양하여도 독력의 변화는 관찰되지 않았다. 해수에 사육 중인 송어 (성어)에 넙치 유래 VHSV ($10^{5.3}TCID_{50}$/fish와 $10^{6.3}TCID_{50}$/fish)를 복강 주사한 결과, 폐사어는 관찰되지 않았고, 어체 내에서도 VHSV는 검출되지 않았다. 이상의 결과, 해수 송어 (성어)에서는 넙치 유래 VHSV의 병원성이 확인되지 않았으나 담수 무지개송어 치어에서는 낮은 독력을 나타내었다.

  • Viral Hemorrhagic Septicemia Virus (VHSV)는 유럽과 북미지역의 담수 무지개송어에 심각한 바이러스성 질병을 야기하는 원인체였으나 담수어류 뿐만 아니라 해산어류에서도 심각한 질병을 유발하며 최근 일본과 우리나라 양식 넙치에서 VHSV에 의한 대량 폐사 발생이 보고되고 있다. 본 연구는 VHS에 대한 예방학적 접근의 일환으로서 자연산 어류로부터의 VHSV 검출 및 지리적 분포, 보균 어종에 대한 유전학적인 조사를 목적으로 2003년과, 2005년도에 동중국해역 3지점, 서해안 5지점 그리고 남해안 5지점에서 어류를 채집하였다. RT-PCR을 이용한 연구결과 자연산 해산어류 160마리 중 12종 17마리에서 VHSV가 검출되어 검출율 10.6%를 보였으며 조사된 12종의 어류 중 특히 상어목에서 가장 높은 검출율을 보였다. VHSV G gene을 이용한 계통분류학적 분석결과 염기서열 분석에서 양식산어류의 분리주 (KVHS01-1)와 98.4%~99%, 아미노산 분석에서 97~99% 유사성을 나타내어 본 연구에서 조사되어진 자연산 어류에서 분리된 VHSV는 GenogroupⅠ에 속하였다.

  • Viral haemorrhagic septicaemia virus (VHSV) is an epidemic virus in olive flounder Paralichthys olivaceus farms in Korea, since the virus have first isolated in 2001. In the present study, partial glycoprotein (G) gene nucleotide sequences of seven Korean VHSV from cultured olive flounder and wild marine fishes in coastal areas of Korea were analyzed to evaluate their genetic relatedness to worldwide isolates. Phylogenetically, all Korean VHSV formed only one minor cluster including Japanese isolates, in genotype IVa, while the North America isolates formed a different minor cluster in genotype IVa. These results suggest that Korean VHSV could be an indigenous virus in Korean and Japanese coastal areas, but have not been introduced from North America.

  • Viral hemorrhagic septicemia virus (VHSV) is a fish pathogen responsible for causing enormous economic loss to the aquaculture industry not only in Korea but worldwide. Thus, it is necessary to identify natural compounds that can be used to control the spread of VHSV. In this study, the anti-VHSV activities of five catechol derivatives, i.e., catechol, pyrogallol, 3-methyl catechol, veratrole, and 3-methyl veratrole-extracted from green tea-were assessed. The antiviral activities of these derivatives were found to be dependent on their structure, i.e., the hydroxyl or methoxyl group and their substituent groups-on the benzene ring. Catechol, pyrogallol, and 3-methyl catechol exhibited relatively high 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities than veratrole, and 3-methyl veratrole. Moreover, 3-methyl catechol harboring a methyl substituent group increased the viability of the virus-infected cells and resulted in a 2.86 log reduction in the gene copies of VHSV N (per mL) in real-time PCR analysis. In conclusion, the catechol derivatives harboring hydroxyl groups in their benzene ring exhibited higher antioxidant activities than those harboring the methoxyl groups. However, catechol derivatives with a methyl group at the 3'-position of the benzene ring exhibited higher antiviral activity than those harboring a hydroxyl group. To our knowledge, this is the first study to evaluate the relationship between the structure and the anti-VHSV activity of catechol derivatives.

Antiviral Efficacy of an Aquatic Disinfectant Tablet Composed of Calcium Hypochlorite Against Viral Hemorrhagic Septicemia Virus

  • Objectives: The objectives of this study were to identify the virucidal efficacy against the viral hemorrhagic septicemia virus (VHSV) of an aquatic disinfectant tablet composed of calcium hypochlorite. Methods: Virucidal efficacy was determined through the viability of VHSV contacted with the disinfectant in a viral stock cultured in a fathead minnow cell line. An aquatic disinfectant tablet and VHSV were reacted under distilled water (DW), hard water (HW) or organic matter suspension (OM) conditions. Results: Under DW and HW conditions, VHSV was inactivated with 24,000- and 2000-fold dilutions of the aquatic disinfectant tablet, respectively. With the investigation of the antiviral effect of the disinfectant under OM conditions, VHSV was inactivated with a 16,000-fold dilution of the aquatic disinfectant tablet. Conclusions: The results from this study showed that the aquatic disinfectant tablet was a highly effective disinfectant against VHSV. In the future, a controlled field trial is required to determine whether the use of an aquatic disinfectant tablet will be able to reduce VHSV in a cultured marine fish farm.

  • 우리나라 양식 넙치에서 분리한 VHSV (KR'01-1)의 glycoprotein 아미노산 배열을 기초로 단백질 전환 구조상 유연성, 친수성 및 항원성이 높고, 표면에 노출되어 있을 가능성(surface probability)이 높다고 분석되는 3개의 peptide (Gp1, Gp2, Gp3)를 선정하였다. 정제한 바이러스 입자 및 3개의 합성 peptide에 대한 polyclonal 항체를 제작하여 항원성 분석을 실시한 결과, 합성 peptide로 제작한 모든 항혈청은 Western blotting 결과 VHSV의 구조단백질과 특이적으로 결합하는 나타났으며, 이 가운데 Gp1 및 Gp2 합성 peptide에 대한 항혈청은 양식 넙치에서 분리한 VHSV를 중화시키는 것으로 나타나, peptide-based antigen의 이용 가능성을 제시하였다.

Evaluation of enzyme-linked immunosorbent assay (ELISA) for detection of olive flounder antibodies to viral hemorrhagic septicemia virus (VHSV, genotype IVa) using two Novirhabdovirus antigens

  • An enzyme-linked immunosorbent assay (ELISA) with two Novirhabdovirus antigens (viral hemorrhagic septicemia virus, VHSV and infectious hematopoietic necrosis virus, IHNV) was used to detect specific antibodies against VHSV from olive flounder (Paralichthys olivaceus) sera. In ELISA plates with VHSV culture supernatants (VHSV-Ag plate), optical density (OD) values for sera from olive flounder with VHS history (VHS sera) ranged from 0.64±0.36, and those of sera from fish without VHS history (non-VHS sera) ranged from 0.26±0.26. In IHNV-Ag plate, the OD values (0.43±0.28) for VHS sera were quite low compared to those in VHSV-Ag plates, while the OD values for non-VHS sera were almost similar. When the OD values for each serum were calculated by subtracting the OD values in the IHNV-Ag plate from those in the VHSV-Ag plate, the corrected OD values were significantly different between VHS sera and non-VHS sera. The results were completely in line with fish histories of VHS epizootics. It was considered that the corrected OD values may represent the true values recognized by VHSV-specific antibodies.

  • 국내외에서 80여 종이 넘는 담수 및 해산어류를 감염시켜 대량폐사를 발생시키는 바이러스성 출혈성 패혈증 바이러스(VHSV) 진단검사를 위해 넙치의 여러 조직의 바이러스 발현량에 대한 정량적 데이터를 시간순서에 따라 분석하였다. 무작위 선별된 넙치에 3.0E+07 TCID50 per 0.1mL per fish의 VHSV를 복강 주사하여 시간순서(0시간, 6시간, 12시간, 1일, 2일, 3일, 5일, 7일)에 따라 조직(아가미, 간, 신장, 비장, 근육)을 채취하였다. Real-time PCR 법을 통해 상대 정량한 결과 5일차 아가미, 간, 신장, 비장에서 바이러스의 발현량이 가장 높게 나타났다. 이번 연구를 통해 감염 초기단계에서 비장이 VHSV 확정진단을 위한 적정조직임을 입증하였으며, 국내 법정전염병 진단에 중요한 정보를 제공할 것이다.

Interleukin-8 (IL-8) Expression in the Olive Flounder (Paralichthys olivaceus) against Viral Hemorrhagic Septicemia Virus (VHSV) Challenge

  • Interleukin-8 (IL-8) is an inflammatory cytokine that plays an important role in the inflammatory response through the activation of neutrophil cells. The expression of IL-8 was investigated in early developmental stages of the olive flounder and in tissues of 8-month-old individuals. The expression of IL-8 increased after the initiation of the immune system rather than at the early stage of development, and high expression was observed in the gills and spleen, the organs associated with immunity and metabolism. In addition, IL-8 expression after infection by viral hemorrhagic septicemia virus significantly increased in the fin, gill, muscles, and spleen. These results suggest that IL-8 is closely related to inflammation and immune regulation in the immune response of the olive flounder and may be used as a basis for studies on the immune systems of other fish.

  • The caspase10 encodes an initiating caspase that plays an important role in the maintaining the cellular homeostasis by regulating the steps involved in the immune response and cell death. We investigated the expression of caspase10 during the different developmental stages and in olive flounder tissues. Caspase10 increased in the late stage of the formation of immune tissue, and high expression was observed in the gills, kidney, skin, and spleen. The current study analyzed the expressional changes of caspase10 in olive flounder infected with viral hemorrhagic septicemia virus (VHSV). One of the major causes of mass mortality, VHSV infection in olive flounder attributes to significant expression of caspase10 in the gills, spleen, skin, and kidneys. The results indicate a close association of caspase10 expression with the immune response to VHSV infection in olive flounder. The observations could form the basis data for exploration of other fish immune system.
