Levels of Viral Glycoprotein Provide a Measure of Modulated Chemotherapeutic Effect

  • A chemosensitivity assay with small replicate Mm5mt/cl C3H mammary tumor cell cultures was developed to determine whether changes in viral antigen expression and release into culture fluids could be utilized as an in vitro measure of modulating drug effect. The 52,000 MW viral envelope glycoprotein (gp52) of the mouse mammary tumor virus (MMTV) was measured in culture fluids of control and drug-treated cultures while cell density was simultaneously determined by cell staining and OD 664 nm determination. While extra-cellular gp52 levels and cell density progressively increased over 72 hours for control cultures, declines in both parameters provided dual measures of effect for combination [N(phophonacetyl-L-aspartic acid)+5-fluorouracil], combination 〔N(phophonacetyl-L-aspartic acid )+5-fluoro-5'-deoxyuridine〕and single component treatment of this combination. At each treated time point, thesecombinations begin to produce a greater decline in both cell density and gp52 levels as compared to single drug treatments. These results indicate that N(phopho-nacetyl-L-aspartic acid) in combination can enhance the effectiveness of single drug.

Analysis of Polymeric Immunoglobulin Receptor Expression in Olive Flounder (Paralichthys olivaceus) against Viral Hemorrhagic Septicemia Virus

  • Polymeric immunoglobulin receptor (pIgR) mediates the transfer of polymeric immunoglobulin to protect organisms and is one of the most important mucosal effectors. In this study, the developmental stage- and tissue-specific expression of pIgR were observed before virus inoculation in olive flounder. pIgR was gradually expressed until the formation of immune tissue, exhibiting high expression in the late juvenile period; thereafter, pIgR expression gradually decreased and exhibited high expression in the spleen and skin. Moreover, pIgR expression after viral hemorrhagic septicemia virus infection was high in the kidney and spleen tissues at high density and low at low density. The results of this study can provide a basis for future studies on breeding density, virus expression, and immune system studies in fish.

대용량 입자 발생 장치 개발 및 이를 이용한 항바이러스 공조용 공기필터 제조 (Development of mass aerosol particle generator and fabrication of commercial anti-viral air filter)

  • Since airborne viruses have been known to aggravate indoor air quality, studies on development of anti-viral air filter increase recently. In this study, a mass aerosol particle generator for coating a commercial air filter (over $300{\times}300mm^2$) was built, and evaluated by comparing a commercial particle generator. Then, via this device, a commercial air filter was coated with anti-viral material ($SiO_2-Ag$ nanoparticles in this study), so fabrication of commercial anti-viral air filter was performed and the pressure drop, filtration efficiency, and anti-viral ability of the filter were evaluated against aerosolized bacteriophage MS2 in a continuous air flow condition. The result showed that the particle generation of the new generator was more than about 8.5 times over which of the commercial one. Consequently, $SiO_2-Ag$ particle coating on a filter does not have significant effects on the filtration efficiency and pressure drop with different areas, and the average anti-viral efficiency of the $SiO_2-Ag$ filter was about 92% when the coating areal density was $1.0{\times}10^{12}particles/m^2$.

Infectious RNA Viruses in the Edible Mushroom Pleurotus spp.

  • Double-stranded RNA (dsRNA) viruses and single-stranded RNA(ssRNA) viruses were detected in a strain of Pleurotus mushroom cultivated in a farm. Those fungal virsus were purified in the pH 6.0 or pH 7.2 using CsCI or Cs$_{2}$SO$_{4}$ buoyant density centrifugation. Each viral particles were not completely separated at any trials. However, mushroom bacili-form virus contains a single major nucleic acid with 0.7 Kb ssRNA, which might code for 20 Kd viral capsid protein. The dsRNAs are encapsidatred into spherical-form viruses, whereas ssRNA viral genomes are encapsidated into two different sizes of bacili-form particles. A healthy-looking mushroom also contained some spherical-form viruses with dsRNAs. Laboratory strains of Pleurotus ostreatus and a cultivated strain of P. sajor-caju did not show any viral particles. Mushrooms with specific disease symptoms. however, contained at least four different sizes of spherical-form viruses. Thus, we concluded that a bacilli-form virus case a severe disease symptoms of adnormal on mushroom development.

세포 비병원성 소 설사병 바이러스의 이화학적 성상 조사 (Biophysical characteristics of a noncytopathic bovine viral diarrhea virus)

  • 세포 비병원성 소 설사병 바이러스가 감염된 우태아 폐세포를 이용 바이러스를 순수분리하여 이화학적 성상을 검사하였던 바 다음의 결과를 얻었다. 바이러스의 비중은 $1.090{\sim}1,114gm/cm^3$로 검출되었으며 $1,098gm/cm^3$에서 최대 감염가를 나타내었다. 면역 전자현미경을 이용한 형태학적 분석결과 30~80nm의 바이러스성 입자를 관찰할 수 있었으며 유전인자의 추출 분석결과 병원성 바이러스와의 전기영동상 유전자 규모에 있어서 차이는 인정되지 않았다. 그러나 비세포 병원성 소 설사병 바이러스는 병원성 바이러스에 비하여 세포외 배출이 적었으며 세포내에서 보다 많이 존재하는 것으로 관찰되었다.

Analysis of Tissue-Specific Interferon Regulatory Factor 3 (IRF3) Gene Expression against Viral Infection in Paralichthys olivaceus

  • Interferon Regulatory Factor 3 (IRF3) is a member of interferon-regulated transcription factor family and is known to play an important role in the innate immune response against viral infections. In this study, the expression of IRF3 in different tissues, developmental stages, and stocking densities of olive flounder was investigated. The expression of IRF3 was observed to gradually increase in early-stage juvenile fish. The highest expression was observed in later-stage juvenile fish when immune tissues were formed. High IRF3 expression was observed in the muscles and the brain tissues. The expression of IRF3 was observed in fish at different stocking densities after viral hemorrhagic septicemia virus (VHSV) infection. It yielded an interesting expression pattern in the muscles and the brain tissues of fish stocked at low density. These observations can be used as basic data for the study of the expression of immune response-related genes against viruses based on stocking density and immune systems in other fish species.

  • The viral hemorrhagic septicemia virus (VHSV) has an extensive host range, and infects farmed and wild fish inhabiting both freshwater and marine ecosystems. Enzyme-linked immunosorbent assay (ELISA) is highly useful in diagnosing viral hemorrhagic septicemia. However, ELISA shows high, non-specific background reaction with fish antibodies. In this study, we optimized the antigen and antibody concentrations used for detecting specific antibodies in VHSV-infected olive flounder to reduce non-specific binding, and improve the sensitivity of ELISA. The results suggested that OD (optical Density) values were valid when ELISA was performed with $0.1{\mu}g/well$ of virus, involving blocking with blocking buffer (Roth, Roti-Block), 1:300-1:600 dilution with flounder antisera, and 1:1000 dilution with anti-flounder IgM and HRP-conjugated goat anti-mouse IgG for detecting the VHSV antibody in flounder sera. Furthermore, 11 different VHSV strains isolated in Korea from 2012 to 2016 were used to infect the fish. The results showed no correlation between viral pathogenicity and antibody production. This research is a basic study on the application of antibody detection in the diagnosis of viral hemorrhagic septicemia in the olive flounder.

개 REM134 유선종양세포의 sphere 형성을 통한 암 줄기세포 특성 분석 (Tumorsphere formation and cancer stem cell characterization of REM134 canine mammary carcinoma cells)

  • Canine mammary tumors are among the most frequently observed cutaneous tumors in female dogs. Cancer stem cells (CSCs), referred to as tumor-initiating cells, are thought to have properties similar to normal stem cells such as the ability to self-renewal and to differentiate into various cell types. Biological understanding of CSCs and the critical pathways involved in their maintenance are important in research and therapy for mammary tumors. We conducted the present study on sphere formation from REM134 cells by using methylcellulose to produce tumorspheres on a large scale and compared the specific markers of the spheres-formed and plating-cultured REM134 cells. The results revealed that the tumorspheres cultured in methylcellulose had higher seeding density and improved morphology compared to those produced in normal sphere formation medium. Expression levels of stemness markers and CSC-related markers were higher in tumorsphere-forming cells than in plating-cultured cells. Subsequently, we transplanted the tumorsphere-forming and plating-cultured cells into female nude mice to examine their tumorigenic potential. Tumor volume increased rapidly in mice transplanted with tumorsphere-derived cells compared to plating-cultured cells. We observed a novel sphere-forming condition for REM134 cells and showed that REM134 cell tumorspheres can exhibit improved CSC properties.

광양만 해양바이러스 개체수와 물리화학적 요인의 계절적 변동 (Seasonal Fluctuations of Marine Viral Abundances and Physicochemical Parameters in Gwangyang Bay)

  • 광양만의 3개의 다른 지역에서 바이러스 개체수와 물리화학적 요인의 계절적 분포도를 분석하였다. 또한 바이러스의 형태적 구조를 투과전자현미경을 이용하여 관찰하였다. 대부분의 해양바이러스는 박테리오파지와 유사한 구조를 갖고 있었으나, 일부는 진핵 미세조류 바이러스인 Ectocarpus siliculosus virus (EsV)와 유사하였다. 그룹 1 (폐쇄된 부영양 지역)에서의 바이러스 개체수는 그룹 2 (폐쇄된 빈영양 지역) 및 그룹 3 (개방된 빈영양 지역)에서의 바이러스 개체수보다 높았다. 그룹 1 바이러스의 계절적 개체수는 크게 변화하였다. 바이러스 개체수는 8월에 가장 높은 $7.33{\times}10^8ml^{-1}$에서 12월에 가장 낮은 $0.3{\times}10^8ml^{-1}$로 변화하였고, 봄과 가을에는 중간값을 유지하였다. 질소원 및 인산원의 농도는 바이러스 개체수에 상관관계를 보였다. 특히 용존산소의 계절적 변화는 바이러스 개체수와 반대 경향을 보였다. 이러한 결과는 광양만에서의 해양바이러스 개체수가 물리화학적 환경과 밀접하게 연관되어 있음을 보여준다.

경북지방에서 사육되고 있는 한우에서 소 바이러스성 설사 바이러스의 유전적 특징 (Genetic Characterization of Bovine Viral Diarrhea Virus from Korean Indigenous Calves in Gyeongbuk Province)

  • 소 바이러스성 설사 바이러스는 전세계 축산업에서 중요한 질병이다. 대한민국의 최대 소 사육지역인 경북지방에서 유행하고 있는 소 바이러스 설사 바이러스를 조사하기 위해, 소 바이러스성 설사 바이러스의 임상증상을 보이는 한우 송아지로부터 365개의 분변을 채취하였다. 가검물을 채취한 365 마리 송아지 가운데 50 마리가 소 바이러스성 설사 바이러스에 양성반응을 보였다. 5'-UTR을 이용한 계통발생 분석에서 48두 송아지가 BDVD-2a에 속했고, 2두에서만 각각 BVDV-1a와 BVDV-1b로 분류되었다. 이들 결과는 BVDV-2a가 경북지역의 한우 송아지에서 유행하고 있음을 제시하고 있다.
