The purpose of this study was to evaluate the effect of a subsequent infection of PCV2 on piglets with PEDV. In clinical signs, the signs observed in dual-infected with PEDV and PCV2 piglets and alone infected with PEDV piglets ranged from diarrhoea to vomiting and dehydration. Dual-infected piglets developed signs of anorexia, vomiting and watery diarrhoea within 12 hpi. Nevertheless alone -infected piglets caused pasty diarrhea at first. In mortality, dual infections showed 25%, but alone -infections showed 8.3%, respectively. In gross findings, piglets dual-infected with PEDV and PCV2 appeared the severe findings of congestion, distension of lumen, milder curdes of undigested milk in stomach than those of single-infected piglets. In histopathological findings, piglets of dual-infection group appeared the more severe findings of villous atrophy and fusion, congesion, exfoliation, vacuolation, squamation, loss of cilia and proliferation of crypt. Significant (P<0.05) decrease in VH:CD ratio in dually infected piglets compared to piglets from alone-PEDV infections. In immunohistochemical findings, strong hybridization signals in dual-infected piglets observed moderate to severe villous atrophy or vacuolation with positive cells arranged continuously over the villi. In the lumen, exfoliated enterocytes were strongly positive in dual-infected piglets. A number of PEDV-positive cells in dual-infected pigs were significantly higher than that in alone PEDV-infected piglets.
The objective of this study was to evaluate Solpro500 (a wheat hydrolysate containing a high level of glutamine) as a replacement for spray dried porcine plasma (SDPP) in diets fed to nursery pigs. One hundred and eight pigs (Dalland, $5.39{\pm}0.80$ kg BW) weaned at 21 days were assigned to one of three treatment groups for a 28 day feeding trial. The experimental diets were based on corn and soybean meal and were supplemented with either 8% SDPP, 4% SDPP plus 4% Solpro500 or 8% Solpro500. Each treatment was fed to six pens with six pigs per pen (4 barrows and 2 gilts). The experimental results indicated no significant difference (p>0.05) in daily gain, feed intake or feed efficiency for pigs fed the three experimental diets. However, the diarrhea index for pigs fed either 4% SDPP and 4% Solpro500 or 8% Solpro500 was lower (p<0.05) than that for pigs fed 8% SDPP. No differences (p>0.05) were found in the apparent fecal digestibility of dry matter, organic matter or crude protein between pigs fed the three diets. The intestinal morphology (villous height, villous width and crypt depth) was not affected by diet treatments (p>0.05). In conclusion, Solpro500 SDPP can replace SDPP without any negative effects on nursery pig performance.

The morphologic changes of small intestinal epithelium in pigs diagnosed as porcine epidemic diarrhea(PED} by virus isolation and immunohistochemistry were studied through light microscope and transmissible electron microscope. On semi-thin section, the histologic findings showed severe villous atrophy and fusion with hyperplasia of cuboidal epithelium in the villi, inflammatory cell infiltration in lamina propria, and increased mitotic figures in the crypt. The structural changes were mostly restricted to the cytoplasm of affected absorptive epithelium of villi. 3 types of epithelial changes were found; degenerated virus-affected cells, undifferentiated cuboidal cells, and normal columnar cells. On electron microscopy, round to spherical viral particles of 50∼l00nm in diameter were found within the dilated vesicles and endoplasmic reticulums of degenerated cells, which had decreased their cytoplasmic electron density due to dilated and missing organelles(e.g. mitochondria, ERs, etc.). Microvilli were shortened and sparse, leaving denuded terminal web of the villous epithelial cells. Fat globules were often found within slightly degenerated enterocytes. On the tip of villi, severely damaged cells were exfoliated and replaced by undifferentiated cuboidal cells We found distinct ultrastructural changes in the jejunal epithelium confirming PED virus infection is involved in malabsorptive diarrhea.

Total parenteral nutritional effect was induced by surgical creation of Thiry-Vella fistula(TVFs) in rats. Glutamine, glycine or nucleosides/nucleotide mixture in solution was injected into the loops for 2, 4, 6, 8 days. Control animals received a 0.9% saline solution. Results include weight gain, total protein, DNA, [$^3$H] thymidine incorporation into DNA, morphometry of the intestine in both TVFs and intestine in continuity. Perfusion of nucleosides/nucleotide mixture into the bypassed loops caused an increase in total protein, DNA content, villous height, villous surface area in loops. The injection of glycine into loops caused an increase in [$^3$H] thymidine incorporation but the mean values of the protein and DNA contents were not significantly different from those in group Cont and group Nuc. Overall values for group Gln were slightly higher than those of the control but the differences were not statistically significant. This study suggests that this animal model may be useful for studying the effect of dietary factors on intestinal growth and maturation, separating the direct effect of diet from systemic effect on the intestine.

The effect of two doses of different sources of zinc, inorganic (zinc oxide) or chelated (zinc glutamate chelate), on morphology and turn-over of the small intestine was assessed in early-weaned pigs orally challenged with enterotoxigenic E. coli K88 (ETEC). Sixty pigs weaned at 21 days were assigned to one of the following 5 diets: control (C); C+Zinc oxide (ZnO), either a 200 or a 2,500 mg Zn/kg dose; or C+zinc chelate with glutamic acid (Glu-Zn), either a 200 or a 2,500 mg Zn/kg dose. On d 2, the pigs were orally inoculated with 1.5 ml of a $10^{10}$ CFU/ml E. coli K88ac O148 suspension. Zinc supplements did not improve the performance of the pigs, but on d 5 faecal excretion of ETEC was reduced, and this was mainly due to high zinc doses (p<0.05). The villous height in the duodenum was improved by the zinc supplements (p<0.01) whatever the source and the level, whereas no effect was seen in the other two tracts of small intestine. The diet did not affect apoptosis and mitosis counts, while ETEC-susceptible pigs had more mitotic cells in the villi than non-susceptible pigs, particularly in the jejunum (p<0.01). The duodenum had fewer mitotic cells in the villi (p<0.05) and in the crypts (p<0.01) and more apoptotic cells in the villi. High dietary doses of ZnO or Zn-Glutamate improve villous height of the duodenum, but not of the jejunum and the ileum, and do not affect the epithelial proliferative activity and apoptotic index of intestinal mucosa of early-weaned pigs orally challenged with ETEC.
Objective: Gut health improvements were monitored with respect to growth performance, diarrhea incidence, fecal bacterial population and intestinal morphology of suckling pigs orally supplemented with live Lactobacillus salivarius (L. salivarius) oral suspensions and challenged with $F4^+$ enterotoxigenic Escherichia coli (ETEC). Methods: Two groups of newborn pigs from 18 multiparous sows were randomly designated as non-supplemented (control: n = 114 piglets) and L. salivarius supplemented groups (treatment: n = 87 piglets). Treatment pigs were orally administered with 2 mL of $10^9$ colony-forming unit (CFU)/mL L. salivarius on days 1 to 3, then they were orally administered with 5 mL of $10^9CFU/mL$ L. salivarius on days 4 to 10, while those in control group received an equal amount of phosphate buffered saline solution. On day 24 (2 weeks post supplementation), one pig per replicate of both groups was orally administered with $10^8CFU/mL$$F4^+$ ETEC, then they were euthanized on day 29 of experiment. Results: Results revealed that pigs in treatment group had a statistically significant increase in average daily gain, body weight and weight gain, and tended to lower diarrhea throughout the study. Numbers of Lactobacillus population in feces of treatment pigs were higher than control pigs, especially on day 10 of study. Numbers of total bacteria in intestinal contents of control pigs were also increased, but not Coliform and Lactobacillus populations. Histological examination revealed statistically significant improvements of villous height and villous/crypt ratio of duodenum, proximal jejunum and distal jejunum parts of treatment pigs compared with controls. Duodenal pH of treatment group was significantly decreased. Conclusion: Oral supplementation of live L. salivarius during the first 10 days of suckling pig promoted growth performance and gut health, reduced diarrhea incidence, increased fecal Lactobacillus populations and improved intestinal morphology.

Background/Aims: There has been no evidence for the necessity of endoscopy in asymptomatic young men with iron deficiency anemia (IDA). To determine whether endoscopy should be recommended in asymptomatic young men with IDA, we compared the prevalence of gastrointestinal (GI) lesions between young men (< 50 years) with IDA and those without IDA. Methods: We conducted a case-control study on asymptomatic young men aged < 50 years who underwent both esophagogastroduodenoscopy (EGD) and colonoscopy as part of a health checkup between 2010 and 2014. Results: Of 77,864 participants, 128 (0.16%) had IDA and 512 subjects without IDA were matched for several variables including age. Young men with IDA had a significantly higher proportion of colorectal cancer (CRC) (0.8% vs. 0.0%, p = 0.045), villous adenoma (0.8% vs. 0.0%, p = 0.045), and inflammatory bowel disease (IBD; 2.3% vs. 0.4%, p = 0.025) than those without IDA. Additionally, the prevalence of advanced colorectal neoplasia (ACRN) tended to be higher in subjects with IDA than in those without IDA (3.1% vs. 1.0%, p = 0.084). The prevalence of significant lower GI lesions including ACRN and IBD was higher in subjects with IDA than in those without IDA (5.5% vs. 1.4%, p = 0.011). Regarding upper GI lesions, a positive association with IDA was observed only for gastric ulcer (4.7% vs. 1.0%, p = 0.011). Conclusions: GI lesions including CRC, villous adenoma, IBD, and gastric ulcer were more common in asymptomatic young men with IDA. Our results suggest that EGD and particularly colonoscopy should be recommended even in asymptomatic young men with IDA.

Background: Hystricomorpha rodents display a similar placentation model to humans. The present study was carried out considering the scarcity of information concerning the placental development in agouti. Objective: Describe the microscopy of the placenta, subplacenta and yolk sac of agoutis in early pregnancy and report on the inversion of the yolk sac. Methods: Fifteen females between the 14th-32nd day of gestation were used following euthanasia. Gestational buttons were collected, fixed, processed, stained to optical microscopy or immunohistochemistry. Results: Chorioallantoic placenta (CP) ranged from conical to a half-sphere, as follows: from the 14th to 17th day, the CP displays an inverted "V" shape, predominantly formed by cytotrophoblasts; from 20 to 22 days, formed almost entirely by cytotrophoblasts; at 28 days, a half sphere, with distinct lobes and interlobular area, numerous maternal gaps delimited by syncytiotrophoblasts and trophoblast giant cells; at 32 days, globose and undergoing the maturation process. Subplacenta, located between decidua and CP, initially presents septa consisting of simple columnar epithelium and after 17 days, comprising stratified epithelium. Visceral yolk sac (VYS) is attached to two CP projections between 14 and 17 days, formed by a simple cubic epithelium and inverted. Between 20 and 22 days, the epithelium displays apical villous projections with cytoplasmic vacuoles and a vascularized mesoderm. After the 24th day, the VYS near the placenta is pleated, very vascularized and villous, with decreased villi sizes further away from the placenta. Conclusion: The agouti CP displays similar characteristics to other hystricomorpha, including placenta lobulation, a subplacenta and an inverted vitelline placenta.
Importance: Porcine epidemic diarrhea virus (PEDV) binds to particular cell surface receptors to penetrate cells. The virus specifically identifies certain carbohydrate structures present on the surface of the cell to facilitate the binding process. Nevertheless, the influence of viral infections on specific alterations of glycoconjugates in the small intestines remains unexplored. Objective: This work aimed to examine the alterations in glycoconjugates in the small intestines of piglets naturally infected with PEDV using lectin histochemistry. Methods: Six piglets including three PEDV-infected and three non-infected piglets were evaluated. Small intestinal samples were histopathologically examined, and lectin histochemistry was performed. Results: Piglets infected with PEDV had significant histological abnormalities in their small intestines, such as pronounced villous atrophy, varying degrees of villous fusion, and diverse mucosal alterations. Specific regions of the duodenum, jejunum, and ileum showed discernible variations in glycoconjugate distribution, as determined by lectin histochemistry. Compared with the controls, the PEDV-infected piglets showed significant changes in N-acetylglucosamine- and galactose-binding lectins (particularly wheat germ agglutinin and Arachis hypogaea (peanut) agglutinin) in multiple intestinal regions. Conclusions and Relevance: These findings can enhance understanding of how viruses such as PEDV impact the glycoconjugate composition of the small intestines and emphasize the potential connection between the pathogenesis of PEDV and glycoconjugate.
Background: Diagnosis and treatment are often successful in the setting of cardiac myxomas. However, cardiac myxomas can lead to catastrophic complications, due to intracardiac obstruction and embolism preoperatively, and can recur postoperatively. Material and Method: We retrospectively reviewed the clinical characteristics, surgical treatment, and recurrence data of 85 patients who underwent cardiac myxoma surgery at Asan Medical Center between November 1994 and June 2007. We analyzed the morphologic characteristics of 58 patients with left atrial myxomas and determined the development of functional mitral valve stenosis and systemic embolism through reviewing the results of preoperative echo-cardiograms to find potential preoperative risk factors. Result: Twenty-seven (31.8%) patients were men, and 58 (68.2%) were women. The mean patient age was $54.5{\pm}14.3$ years. Preoperative symptoms included obstructive symptoms in 41 (48.2%) patients, signs of embolism in 19 (22.4%), constitutional symptoms in 8 (9.4%), and no symptoms in 19 (20.0%). Among the 58 patients with left atrial myxomas, the mean maximal tumor diameter was $4.3{\pm}1.8$ (range $1.1{\sim}8\;cm$)cm. Twenty-six (44.8%) patients had a prolapsing type, defined as a tumor mobile enough to move down. to the mitral. annular plane during diastole, and 32 (55.2%) had villous type, defined as a tumor consisting of multiple fine villous extensions on the surface. Twelve (20.7%) patients had severe functional mitral valve stenosis, and 15 (25.9%) had systemic embolism preoperatively. The incidence of severe functional mitral valve stenosis was significantly higher in patients with the prolapsing type than in those with the non-prolapsing type (p=0.001). The mean maximal tumor diameter in patients with severe functional mitral valve stenosis was $5.1{\pm}1.0\;cm$, significantly larger than that seen in patients without severe functional mitral valve stenosis (p=0.041). The incidence of systemic embolism was significantly higher in patients with the villous type than in those with the smooth type (p=0.006). Postoperative complications were noted in 6 (7.1%) patients, and early mortality was noted in 1 (1.2%). The mean postoperative follow-up duration was $36.2{\pm}37.5$ months, with recurrence reported in 2(2.4%) patients during the follow-up period. The disease free interval were 48, 12 months, respectively. Conclusion: Surgical treatment for cardiac myxomas was performed safely, and long-term prognosis was good. In patients with left atrial myxoma, close attention should be maintained and surgery should be performed promptly in those of prolapsing type, those with large maximal diameter in order to prevent severe functional mitral valve stenosis, and those of villous type in order to prevent systemic embolism. Echocardiography should be followed serially in order to detect recurrence.
