Sarcocystis infection and identification of Sarcocystis species in pigs in Korea (국내(國內) 돼지의 Sarcocystis 감염과 Sarcocystis종의 동정에 관한 연구)

  • Five hundred and forty-eight samples of pig heart muscle were collected from the abattoirs of many regions in Korea to reveal the frequency of Sarcocystis infections and to identify the species from June 1988 to April 1989. Heart muscle of the pigs was inspected for sarcocysts by the direct detection technique and for bradyzoites by the trypsin digestion technique. For examination of development of the parasites in the final host, 5 cross bred mature dogs, 5 puppies and 5 kittens were fed 100g, 50g and 50g of the infected meat respectively, four times in 2 days. Of 402 fattened and 146 older culled breeding pigs, 3 fattened pigs and 39 culled pigs were positive for Sarcocystis. Sarcocystis cysts from heart muscle measured an average of $425{\times}169{\mu}m$ and bradyzoites an average of $15.6{\times}3.5{\mu}m$. Of 15 animals, only 2 puppies were infected with Sarcocystis. The prepatent period was 11 to 12 days and patent period was not examined since the puppies were infected with some another infections and one died on day 11 and another died on day 12 after ingestion of the meat. The sporulated oocysts were detected 11 days after ingestion of the meat and sporocysts 12 days from the puppy feces. The sporulated oocysts measured an average of $16.5{\times}11.5{\mu}m$ and sporocysts an average of $12.6{\times}7.9{\mu}m$. On scraping examination of the intestinal mucosa, fully sporulated oocysts were detected in the tip of the intestinal villi. Considering above all descriptions, Sarcocystis in pig heart muscle in Korea was identified with Sarcocystis suicanis.

Studies on the necrotic enteritis of chicken in Korea (국내(國內) 닭의 괴사성 장염 발생에 관한 연구)

  • This study was carried out to investigate the necrotic enteritis of chicken in Korea. Epidemiological, pathological and microbiological examination were done on 12 naturally occurred cases of necrotic enteritis of chicken. And the susceptibility of isolated bacteria to antimicrobial agents was also examined. The results obtained were as follows; 1. The Clostridium perfringens infection, necrotic enteritis of chicken, occurred in the chickens mainly raised in floor pens. Necrotic enteritis of chicken was occurred coincidently with coccidiosis and Gumboro disease frequently. And several cases were recurred at 2-3 weeks after recovery. 2. Clinical signs of the infected chickens were depression, decreased appetite, reluctance to move, diarrhea, ruffled feathers and acute death within several hours. 3. The characteristic biochemical properties of isolates were 2-band hemolysis, no motility, positive reaction of reverse CAMP test and the formation of LV precipitate in egg yolk medium. 4. Gross lesions of the infected chickens were distention of intestine with gas, thickened mucosa and formation of thick pseudomembrane in intestine. Livers were friable with yellowish brown color and, in some case, showed demarcated necrotic foci. 5. Histopathological findings of the infected chickens were severe necrosis of the intestinal mucosa and attachment of numerous large bacilli to the mucosal surface of necrotic villi. In liver, necrosis of liver tissue and numerous large bacilli in the necrotic foci were also observed. 6. In susceptibility test to antimicrobial agents, 12 isolates of Clostridium perfringens were highly sensitive to ampicillin, baytril, cephalothin and penicillin.

Dietary Supplementation of Citrus and Fermented Citrus By-product for Juvenile Red Seabream Pagrus major at Low Water Temperature (저수온기(13-15℃) 치어기 참돔(Pagrus major) 사료 내 비타민 C 대체제로써의 감귤착즙박 및 발효감귤착즙박의 이용 가능성)

  • This study compared the effects of dietary supplementation of citrus by-product (CBP) and CBP fermented with Bacillus subtilis (F-CBP) on growth performance, feed utilization, intestinal histology and innate immunity of red seabream Pagrus major with three commercial immune-boosting products. The six experimental diets were supplemented with L-ascorbyl-2-polyphosphate (LAPP; the control diet), CBP or F-CBP at a concentration of 100 mg vitamin C equivalent/kg diet or one of three commercial immune boosters. The Experimental diets were fed to triplicate groups of 17 fish (initial body weight, 116 g) for 8 weeks. The water temperature during the feeding trial was maintained at $13-15^{\circ}C$. Growth and feed utilization did not differ significantly among the six dietary treatments, nor did the phagocytic activity, superoxide dismutase or total immunoglobulin concentrations. However, myeloperoxidase activity was significantly higher in the CBP groups. For the intestinal histology, the intestine diameter, villi and enterocyte heights and number of goblet cells did not differ significantly among groups. Therefore, CBP or F-CBP can be used as a valuable eco-friendly byproduct in diets for fishes including red seabream to maintain their normal growth and health.

  • Intestinal lymphangiectasia, one of the protein-losing gastroenteropathies, is an uncommon disease characterized by dilated intestinal lymphatics, enteric protein loss, edema, hypoalbuminemia, and lympocytopenia. Small bowel biopsy and CT have been used to confirm the diagnosis of intestinal lymphangiectasia. Small bowel biopsy shows collections of abnormal dilated lacteals in submucosa with distortion of villi and CT findings have been described as diffuse nodular thickening of the small bowel and as linear hypodense streaking densities in the small bowel caused by dilated lymphatic channels. Demonstration of increased enteric protein loss using $^{51}Cr-$, $^{131}I-$ or $^{99m}Tc-labeled$ albumin, timed measurement of fecal excretion of radioactivity or by measuring fecal clearance of alpha 1-antitrypsin can also help the diagnosis. We experienced a rare case of intestinal lymphangiectasia in an eight year old boy who presented with facial edema, abdominal distension and intermittent diarrhea. We report a patient with intestinal lymphangiectasia, in whom abdominal CT, $^{99m}Tc-labeled$ albumin scintitigraphy, and stool alpha 1-antitrypsin measurement played key roles in determining the diagnosis. A brief review of literature was made.

  • The purpose of this study was to evaluate the effect of a subsequent infection of PCV2 on piglets with PEDV. In clinical signs, the signs observed in dual-infected with PEDV and PCV2 piglets and alone infected with PEDV piglets ranged from diarrhoea to vomiting and dehydration. Dual-infected piglets developed signs of anorexia, vomiting and watery diarrhoea within 12 hpi. Nevertheless alone -infected piglets caused pasty diarrhea at first. In mortality, dual infections showed 25%, but alone -infections showed 8.3%, respectively. In gross findings, piglets dual-infected with PEDV and PCV2 appeared the severe findings of congestion, distension of lumen, milder curdes of undigested milk in stomach than those of single-infected piglets. In histopathological findings, piglets of dual-infection group appeared the more severe findings of villous atrophy and fusion, congesion, exfoliation, vacuolation, squamation, loss of cilia and proliferation of crypt. Significant (P<0.05) decrease in VH:CD ratio in dually infected piglets compared to piglets from alone-PEDV infections. In immunohistochemical findings, strong hybridization signals in dual-infected piglets observed moderate to severe villous atrophy or vacuolation with positive cells arranged continuously over the villi. In the lumen, exfoliated enterocytes were strongly positive in dual-infected piglets. A number of PEDV-positive cells in dual-infected pigs were significantly higher than that in alone PEDV-infected piglets.

Effects of Backchulsan and Backpeundu on the mucosubstance of the goblet cells in the small and large intestines of the mouse (백출산(白朮散)과 백편두(白扁豆)가 백서(白鼠) 소장(小腸)과 대장(大腸) 배상세포내(杯狀細胞內) 점액질(粘液質)에 미치는 영향(影響))

  • This study was performed to identify the effect of Backchulsan and Backpeundu, on the muco-substances of the goblet cells in the small and large intestines of the mouse. After injected castor oil into oral cavity, the experimental animals were administrated with 5.952mg per 30g body weight of Backchulsan, with 0.519mg per 30g body weight of Backpeundu. The specimens used in this study were obtained from the small and large intestines of the mouse. All specimens were stained with periodic acid-Schiff reaction, alcian blue pH 2.5 and alcian blue pH 1.0 procedures. The results were as follows : 1. The most remarkable decrease of neutral and acid mucosubstance was observed in the control group 1 and 6 hours after castor oil administration. 2. Increase of neutral and mucosubstance was observed in Backchulsan administration group 3 hours in comparision with that of control group administrated of castor oil, especially of the upper and lower villi regions. 3. Increase of neutral and acid mucosubstance was observed in Backpeundu administration group 6 hours in comparision with that of control group administration of castor oil. 4. The most remarkable increase of neutral and acid mucosubstance was observed in Backchulsan administration group after 3 hours in comparision with that of Backpeundu administration group.

Isospora belli Infection with Chronic Diarrhea in an Alcoholic Patient

  • Chronic diarrhea with a 35 kg weight loss (75 kg to 40 kg) occurred during 2 years in an alcoholic patient was diagnosed with Isospora belli infection in the Republic of Korea. The patient, a 70-year old Korean male, had been a heavy drinker for more than 30 years. He was admitted to the Seoul National University Hospital because of long-standing diarrhea and severe weight loss. He had an increased white blood cell (WBC) count with high peripheral blood eosinophilia (36.8-39.9%) and lowered protein and albumin levels but without any evidence of immunosuppression. A parasitic infection was suspected and fecal examination was repeated 3 times with negative results. Peroral endoscopy with mural biopsy was performed in the upper jejunum. The biopsy specimens revealed villous atrophy with loss of villi together with various life cycle stages of I. belli, including trophozoites, schizonts, merozoites, macrogamonts, and microgamonts. The patient was treated successfully with oral doses of trimethoprim 160-320 mg and sulfamethoxazole 800-1,600 mg daily for 4 weeks. A follow-up evaluation at 2.5 years later revealed marked improvement of body weight (68 kg), increased protein and albumin levels, and normal WBC count with low eosinophils (3.1%). This is the first clinical case of isoporiasis with demonstration of various parasitic stages in the Republic of Korea.

Effects of vitamin A supplementation in the diet of breeding geese on offspring intestinal tissue morphology and immune performance

  • Objective: The effects of maternal and offspring dietary vitamin A (VA) supplementation on early body weight, digestive tract function and immune function in goslings were studied. Methods: Yangzhou geese (180 d old) were randomly divided into 5 experimental groups of 15 females and 3 males (the males were kept until slaughter). Eggs were collected for hatching during the peak laying period. A total of 96 goslings were selected from each treatment group (each fed a basic diet supplemented with 0, 4,000, 8,000, 12,000 or 16,000 IU/kg VA) and randomly divided into 2 groups, with 6 replicates in each group and 8 goslings in each replicate. The gosling diet was supplemented with 0 or 9,000 IU/kg VA. Results: i) Villus length, villus width and the muscle thickness of the duodenum, jejunum and ileum were increased and the crypt depth was reduced after adding 12,000 IU/kg VA to the goslings' diet (p<0.05). Adding 9,000 IU/kg VA to the offspring diet increased the length of the duodenal villi and width of the ileum and decreased the crypt depth of the ileum (p<0.05). ii) Supplementing the maternal diet with 12,000 IU/kg VA increased immune organ weight, the immune organ index and immunoglobulin content in goslings (p<0.05). The bursa weight and immunoglobulin G content of offspring were higher in the 9,000 IU/kg VA supplementation group than in the group with no supplementation (p<0.05). Conclusion: Offspring growth and development were affected by the amount of VA added into maternal diet. The negative effect of maternal VA deficiency on offspring can be compensated by adding VA to the offspring diet. Continued VA supplementation in the offspring diet after excessive VA supplementation in the maternal diet is unfavorable for gosling growth and development.

Role of P57KIP2 Immunohistochemical Expression in Histological Diagnosis of Hydatidiform Moles

  • Purpose: To determine the significance of P57KIP2 immunohistochemistry expression in the histopathological diagnosis of hydatidiform mole. Materials and Methods: Hydatidiform mole patients at King Chulalongkorn Memorial Hospital between January 1999 and December 2011 were recruited. Two gynecologic pathologists reviewed histopathologic slides to confirm diagnosis. Formalin-fixed, paraffin-embedded tissue sections were stained using a bstandard immunostaining system with monoclonal antibodies against P57KIP2 protein. Correlations among pathological features, immunohistochemical expression and clinical data were analyzed. Results: One hundred and twenty-seven hydatidiform mole patients were enrolled. After consensus review, 97 cases were diagnosed as complet (CHM) and 30 cases as partial (PHM). Discordance between the first and final H&E diagnoses was found in 19 cases (14.9%, k= 0.578). Significant pathological features to classify the type of hydatidiform mole are central cisterns, trophoblastic proliferation, trophoblastic atypia, two populations of villi, fetal vessels and scalloped borders. After performing immunohistochemistry for P57KIP2, 107 cases were P57KIP2 negative and 20 cases positive. Discordant diagnoses between final H&E diagnosis and P57KIP2 immunohistochemistry was identified in 12 cases (9.4%). Sensitivity of final H&E diagnosis for CHM was 89.7%; specificity was 95.0%. PHM sensitivity and specificity of final H&E diagnosis was 95.0% and 89.7%, respectively. Conclusions: Histopathological diagnosis alone has certain limitations in accurately defining types of hydatidiform mole; P57KIP2 immunohistochemistry is practical and can be a useful adjunct to histopathology to distinguish CHM from non-CHM.

On-farm evaluation of dietary animal and plant proteins to replace fishmeal in sub-adult olive flounder Paralichthys olivaceus

  • Background: High demand and low supply of fishmeal due to overexploitation of fisheries resources have resulted in a dramatic increase in the price of this ingredient. Olive flounder (Paralichthys olivaceus) commercial feed contains approximately 60% fishmeal and limited success has been achieved in identifying sustainable alternative protein sources for this species. Methods: An on-farm feeding trial was conducted to compare a basal diet containing 65% as the control (CONT) with two experimental diets replacing 10% of fishmeal by animal protein (AP10) or 20% of fishmeal by animal and plant protein (APP20). Sub-adult olive flounder averaging 327 ± 9.3 g (mean±SD) were fed one of the three diets in triplicate groups for 16 weeks. Results: Weight gain, specific growth rate, feed efficiency, protein efficiency ratio, and survival were not significantly different among fish fed all the experimental diets (P > 0.05). Also, non-specific immune responses (superoxide dismutase and lysozyme activity), serum biochemical parameters, and intestinal villi length were not significantly different among fish fed all the experimental diets (P > 0.05). Conclusions: Therefore, based on growth performance, non-specific immune responses, serum biochemical parameters, and intestinal histology, dietary animal and plant protein mixtures could replace up to 20% of fishmeal in the diet of sub-adult olive flounder.
