Cowpea (Vigna unguiculata (L.) Walp.) is recognized as a potential source of protein and other nutrients. The genus Vigna includes 100 wild species of plants. Especially, Vigna unguiculata includes annual cowpeas (ssp. unguiculata) and ten wild perennial subspecies. DNA sequence of internal transcribed spacer (ITS) region was determined for Vigna sinensis, one of native plant, which was found in recent but thought to have gone extinct in Korea. The seeds of Vigna sinensis used in this study were donated from Dong-Young Jo. The DNA sequence of ITS-5.8S-ITS2 for Vigna sinensis obtained from this study was deposited as Vigna sinensis AY195581 on GenBank of NCBI (National Center for Biotechnology Information). We investigated the sequence-based phylogenetic relationships of plants related and clarified its taxonomical position. DNA similarities among subspecies including Vigna unguiculata showed the range 98 to 100% in sequence-based phylogenetic analysis using total 507 base pairs of ITS1, 5.8S and ITS2. Vigna unguiculata and subspecies were grouped independently as one cluster from other Vigna species used in the phylogenetic analysis. In this study, based on the phylogenetic analysis using the ITS1-5.8S-ITS2 sequence of Vigna sinensis, it may be concluded to be classified to one of Vigna unguiculata substrains.
Macronutrients ($Na^+$, $K^+$, $Ca^{2+}$, $Mg^{2+}$), yield and yield components, bioaccumulation and translocation of metal in plant parts of three Vigna species (V. cylindrica, V. mungo, V. radiata) were evaluated at 0, 50, 100 and $150mgkg^{-1}$ soil of Nickel (Ni). A marked inhibition (p < 0.001) in the distribution of various macronutrients was noticed in these Vigna species except for $Mg^{2+}$ content of the shoot and leaves. Similarly, all species retained more $Ca^{2+}$ in their roots (p < 0.05) as compared to the aerial tissues. Ni induced a drastic decline (p < 0.001) for various yield and yield attributes except for 100 seed weight. Toxicity and accumulation of Ni in plant tissues considerably increased in a concentration dependent manner. Vigna species signify an exclusion approach for Ni tolerance as both bioaccumulation factor (BF) and translocation factor (TF) were less than 1.0. The Ni content of plants being root > shoot > leaves > seeds. Scoring for percentage stimulation and inhibition (respective to control) at varying levels of Ni revealed tolerance of the species in an order of V. radiata > V. cylindrica > V. mungo. The acquisition of Ni tolerance in V. radiata seems to occur through an integrated mechanism of metal tolerance that includes sustainable macronutrients uptake, stronger roots due to greater deposition of $Ca^{2+}$in the roots, restricted transfer of Ni to above ground tissues and seeds as well as exclusion capacity of the roots to bind appreciable amount of metal to them. Thus, metal tolerant potential of V. radiata could be of great significance to remediate metal contaminated soil owing lesser impact of Ni on macro-nutrients, hence the yield.
Blackgram (Vigna mungo) is more salt tolerant than mungbean (Vigna radiata). This study was initiated to know whether the accumulation pattern of mineral ions in different plant parts plays a significant role in the differences in salt tolerance between the two Vigna species. Different mineral ions, viz. N, Cl, Na, K, Mg and Ca in different organs of two varieties of each of blackgram- Barimash-l (susceptible one) and Barimash-2 (tolerant one), and mungbean-Barimung-3 (tolerant one) and Barimung-4(susceptible one), were analyzed after growing with 0, 50, 75 and 100 mM NaCl solutions. The two crops showed a decreased but similar pattern of total N accumulation under saline conditions. The tolerant variety of both the crops showed a less reduction in total N than the susceptible one. Leaves showed the maximum while stem the minimum N, irrespective of levels of salinity. C $l^{[-10]}$ and N $a^{+}$ accumulation increased with the increasing salinity levels. Interestingly, similar to a halophyte, the salt tolerant blackgram exhibited conspicuously higher amount of N $a^{+}$ in the shoot than the salt-susceptible mungbean. However, the tolerant varieties showed less amount of N $a^{+}$ than the susceptible one, especially in blackgram. Seeds of both Vigna spp. accumulated the minimum amount of N $a^{+}$ than other plant parts. $K^{+}$ accumulation decreased by salinity in most of the plant parts, except seeds. Blackgram showed larger reduction in K than mungbean. The $Mg^{++}$ increased in leaves, petioles and stem by salinity while decreased in the roots, podshells and seeds in both the crops. Salinity increased $Ca^{++}$ accumulation in all plant-parts except roots of both Vigna spp. Apparently, the leaves of mungbean accumulated higher concentration of $Ca^{++}$ than blackgram. Varietal differences in the accumulation pattern of $K^{+}$, $Mg^{++}$ and $Ca^{++}$ were not clear. It was concluded that blackgram, presumably, possesses a similar salt tolerance mechanism to halophyte, and the pattern of accumulation of mineral ions in blackgram and mungbean was not fully ascribed to the differences in salinity tolerance between the two Vigna species.gna species.ies.s.ies.

The esterase isozyme in tissue of wild legume plants were separated by horizontal starch gel electrophoresis. Extracts used in this study were prepared from fully expanded young leaf, cotyledon and radicle of seedling and root-nodule of Glycine sola, Vigna vexillata var. tsuscmensis and Trifoliwn repens. The results are as follows; 1. Each tissue examined had a characteristic banding pattcrn. Number of bands in each species, G. soja, V. vexillata, and T . repens, were 14, 8 and 1 1 bands, respectively. And difference in esterase isozyme bands were greater from tissue to tissue than difference between habitat. 2. Est-I, Est-2. Est-3 and Est-4 in G. soja, Est-I in V. vexillata and Est-l and Est-2 in T. repens showed strong cnzyme activity than other enzyme. 3. Esterase isozyme variation in G. soja and T . repens showed more variety than V. vexillata. This is resulted from many genotypic differences within species. 4. The main enLyme among thc esterase isozyme were Est-I. Est-2, Est-3 and Est-4.
Samples showing mosaic symptom of cowpea (Vigna sesquipedalis) with vein banding, chlorotic spot, vein yellow were collected from Chinju areas in Korea, Two viruses were distinguishable by stability in sap, host range, and relations with cells and tissues were examined under an electron microscope, Blackeye cowpea mosaic(BICMV) was sap-transmissible to 7 plant species in 2 families, Of the plants, only leguminous species were systemically infected. This virus was inactivated by heating at $50-65^{\circ}C$ for 10 min, by diluting at $10^{-4}-10^{-5}$, and aging at room temperature for 1-6 days. Preparations examined under the electron microscope by direct negative staining method(DN -method) always showed particles of flexuous filament bout 750nm in length and cytopasmic inclusions. Cytoplasmic inclusions and virus particles were also confirmed to present in the cytoplasm of a mesophyll cell by ultrathin sections of BICMV infected cowpea leaves. Cucumber mosaic virus (CMV) was transmitted by sap- inoculation on inoculated leaves of Chenopodium amaranticolor, C. quinoa producing local lesions, but non-inoculated upper leaves of Nicotiana glutinosa, Cucurbita pepo and Vigna sesquipedalis producting systemic mosaic symptoms. Electron microscopic examination of virus preparation by direct negative staining showed spherical particles of about 30nm in diameter. In ultrathin sections of CMV infected tissues, virus particles of crystalline array were found in the vacuole and a large number of virus particles were found in the cytoplasm and the plasmodesmata of mesophyll cells.
To investigate the growth response of various crop species to mycorrhizal inoculation, arbuscular mycorrhizal fungi were applied to Glycine max, Vigna angularis, Senna tora, Hordeum vulgare var. hexastichon. Zea mays, Sorghum bicolor, Allium tuberosum, Solanum melongena, and Capsicum annuum. The biomass of the inoculated crops was measured every two weeks for the 12-week growth period. By measuring biomass, we calculated the mycorrhizal responsiveness of the nine crop species. Among the nine crop species, four species showed a significant response to mycorrhizal inoculation. The shoot biomasses of V. angularis, C. annuum, A. tuberosum, and S. tora significantly increased with mycorrhizal inoculation.
The toxic effects of aluminium (Al) on growth, chlorophyll content, $\delta-aminolevulinic$ acid dehydratase (ALAD) activity and anatomy of root and shoot were investigated in 7-day-old azuki bean (Vigna angularis) seedlings. Significant depressions in root elongation was observed in the low concentrations of Al (50, 100 $\muM)$ and increasing Al concentrations caused a sharp decline of root and shoot growth. The degree of inhibition was dependent upon Al supply. Exposure to 50 $\muM$ Al or more inhibited root elongation within 1 day. In the 50 $\muM$ Al treatments, a recovery of root growth was seen after 7 days exposure. In contrast, lateral root initials was little affected by Al exposure. Al toxicity symptoms and growth responses were more well developed in the roots than in the shoots. Analysis of Al localization in root cells by hematoxylin stAlning showed that Al entered root apices and accumulated in the epidermal and cortical cells immeadiately below the epidermis. There was a good positive correlation between the level of chlorophyll and ALAD activity. Increasing Al concentrations caused a decrease in total chlorophyll contents, accompanied by proportional changes in ALAD activity, suggesting a cootr-dinated reduction of a photosynthetic machinery. Al exerted specific influence on the morphology of root ann shoot. At higher concentrations of Al the roots induced drastic anatomical changes. The epidermal cells were disorganized or destructed while the cortical cells exhibited distortion of cell shape and/or disintegration. The diameter of root and transectional area of cortical cells decreased considerably with Al treatment. In the shoot Al also enhanced reduction of diameter of shoot and cell size. Gross anatomy of leaves treated with Al did not differ significantly from the controls, except for fewer and smaller chloroplast. Our results indicate that toxic effect of Al appear to be manifested primarily in roots and secondarily on shoots, and changes in root morphology are related to changes in the root growth patterns. Results are further discussed in re181ion to the findings in other plant species, and it is concluded that Al causes morphological, structural and, presumably, functional damage to the roots of the species investigated.
Some indices of species of the Arundinella hirta communities in Cheolwon area were studied and the following results were obtained. The vegetational date was analyzed from six grass stands selected with the distance away from the DMZ in Cheolwon area. Species diversity was higher in the stand away from the DMZ than in that neat to it. All six stands are not considered to be free of disturbance. Species composition of the stands might be related to the degree of disturbance. It can be said that the destruction of the vegetation by periodical fire or cutting simplifies the species composition. In the rear district the human inter ference is supposed to be more frequent. But it is supposed to be slight in near the DMZ and that the degree of disturbance is not so destructive. This could be the explanation of high diversity in these stands.
Anthracnose fungi associated with soybean (Glycine max), small red bean (Vigna angularis), and green bean (Phaseolus radiatus var. aurea) were compared with respect to cultural characteristics, morphological aspects, and pathogenicity. From the legumes, four different species of Colletotrichum were identified based on their morphological characters : C. truncatum, C. destructivum, C. trifolii and C. gloeosporioides. Colonies of Colletotrichum spp.on culture media varied in colour and growth habits depending upon the media and isolates. In general, formation of conidia, setae, and perithecia on water agar leaf medium was better than that on potato dextrose agar medium under near ultraviolet light. In cross inoculation tests, isolates of C. truncatum, c. destructivum, C. trifolii and C. gloeosporioides could infect soybean, small red bean, and green bean at different growth stages. There were some differences in pathogenicity or susceptibility depending on crops tested. C. truncatum was the most virulent and major pathogenic species among the four species.
In order to investigate the host range of the mosaic disease of black locust in the Chunchon area, the sap of the mosaic-diseased leaves of black locust itself and the cowpea leaves infected with the above mentioned sap, were inoculated to 53 species of plants belong to 12 families. As to the result, no difference in infection was found as related to the virus sources, and the infection was recognized in 4 species of the family Chenopodiaceae and 8 species of the family Leguminosae. The plants recognized as hosts are as follows: the plants which showed local infection are Chenopodium album, Ch. ambrosioides, Ch. quinoa; the plants which showed systemic infection are Chenopodium amaranticolor, Phaseolus vulgaris, Robinia pseudo-acacia, Vigna sinensis; and Astragalus sinicus, Melilotus indicus, Phaseolus angularis, Pisum sativum and Vicia faba were recognized as carriers. Through investigating its host ranges and symptoms, this mosaic virus of black locust seems not to be regarded as the group of the black locust mosaic virus in southeastern Europe reported by Milinko et al (1961). And, too, it is thought hardly to exist in combination with the cowpea mosaic virus. It appears, therefore, that this mosaic virus was confined to that of black locust.
