The ecotoxicity detection device for preliminary test (Test jig) was manufactured to develop the biological early warning system using Vibrio fischeri. In this study, the ecotoxicity detection charateristics of the Test jig was investigated for 6 heavy metals (Cr, Zn, Pb, Cd, Cu and Hg). It was observed that relative luminescence unit (RLU) of Vibrio fischeri constantly decreased by the concentrations of the tested heavy metals. In contrast with other heavy metals, RLUs of Pb and Hg constantly decreased even at low concentrations. RLU of Hg drastically decreased when its concentration increased from 0.13 mg/L to 0.25 mg/L. $EC_{50}$ values of Cr, Zn, Pb and Cd gradually decreased with exposure time, whereas there was no significant change in $EC_{50}$ values of Cu and Hg with time. On the other hand, $EC_{50}$ values between the Test jig and Reference device were compared to evaluate the ecotoxicity detection performance of the Test jig. No big difference was found in $EC_{50}$ vlaues between the two devices, indicating that the Test jig could be applied as the ecotoxicity detection device for heavy metals.
This paper aims to evaluate the results of toxicity testing with Daphnia magna and Vibrio fischeri on wastewater samples which might be influenced by ion imbalance. The effluents from factories were found to be more toxic with high salinity levels than those from public wastewater treatment plant (WTP) and sewage treatment plant (SWP). Clion composition was highest in the effluent, in terms of percentage, which was followed by $Na^+$, $SO_4^{2-}$ and $Ca^{2+}$. $K^+$ and $Mg^{2+}$ ion was relatively low. The sensitivity of D. magna test results was higher than V. fischeri. Among samples which were proved by V. fischeri testing to be nontoxic, the composition ratio of each ion whether toxic samples or nontoxic samples which were decided by D. magna toxicity testing, were compared. $Na^+$, $K^+$, $Ca^{2+}$, $Mg^{2+}$ ion composition ratio showed high level in nontoxic samples whereas $SO_4^{2-}$ and $Cl^-$ ion composition ratio was high in toxic samples. Accordingly, $SO_4^{2-}$ and $Cl^-$ ion seemed to be considered the ions causing toxicity in effluent. Toxicity from some categories of industries (Mining of non-metallic minerals, Manufacture of basic organic petrochemicals, Manufacture of other basic organic chemicals, Manufacture of other chemical products etc.) seemed to be influenced by salinity. The Ion concentration in influent and effluent were similar. Concentration of $Na^+$, $Cl^-$, $K^+$, $Ca^{2+}$ ions were high in influent, however $Mg^{2+}$ and $SO_4^{2-}$ ions were high in effluent.

Four clays (both natural and commercial types) mainly used in Korea were tested for removal efficiency of Microcystis spp. and ecotoxicity on Daphnia magna and Vibrio fischeri. Four clays (clay A~D) were composed of 91.9~100% of sand (0.02~0.2 mm in particle size). Clay D consisted of lager particles than other clays. Major elements of the four clays were $SiO_2$ (45.3~62.8%), $Al_2O_3$ (18.5~29.7%) and $Fe_2O_3$ (5.4~7.9%). They contained kaolinite (clay mineral), quartz, muscovite, and so on. Clay C and D contained montmorillonite, one of the clay minerals improving clay-cell aggregation. For clay A, B and C, removal efficiency of Microcystis spp. was over 60% at 2 g/L. It reached about 100% at over 5 g/L. For clay D, it was over 60% and 95~100% at 5 g/L and 20 g/L respectively. After adding clays, pH decreased. The greatest drop of pH appeared at clay C. Except for addition of 100 g/L clay C, ecotoxicity on D. magna and V. fischeri didn't appeared at all dose of clays.

Toxicity evaluations of 3-tert-butyl-, 2-isopropyl-, 3-isopropyl- and 4-propyl-phenol and their binary mixtures were performed with the Microtox$\^$(R) / assay and compared to invertebrates and fish. The single chemical, 4-isopropylphenol, exhibited the greatest relative toxicity to the Microtox organism (Vibrio fischeri). The relative electrophilicity (LUMO) of the phenols, in contrast to the lipophilicity (Log P), was strongly correlated with toxicity to V fischeri (r$^2$=0.96, p<0.01). In contrast, relative electrophilicity alone could not explain variances in toxicity of the phenols to Ceriodaphnia dubia. Results suggest that electrophilicity in conjunction with lipophilicity provide better correlation with toxicity to C. dubia and Pimephales promelas. Microtox results from the binary mixture toxicity tests of selected phenolics indicate a mechanism of interaction governed by suppression/antagonism.

In addition to North America and Europe, Korea is also responding to the toxic damage caused by the production and distribution of chemicals. Methods for assessing bio-toxicity of harmful substances have been widely introduced, but it is required of quantitative and speedy information for modeling. For 6 heavy metals, as zinc, copper, chrome, cadmium, mercury and lead, bio-toxicity assessment and kinetics model were constructed using Vibrio fischeri which is widely used luminous bacteria. The degree of luminescence activity and the toxicity of heavy metals were relative limunescence unit, RLU measured as by using a photomultiplier embedded device. The toxicity was assessed by the concentration levels giving under 20% lethality and lethal concentration, $EC_{50}$. In the results, the toxicity order were followed from mercury, lead, copper, chrome, zinc and cadmium. $EC_{{50},{\infty}}$ obtained by trends of $EC_{50}$ by time follows had highly linear agreement with main parameters of bio-toxicity modelling. The average error rates of the reproduced lethality obtained from DAM and TDM model on the basis of body residue, were 10.2% for mercury, lead, copper, chrome and 20.0 for the all 6 methals.

Six standard methods for marine ecotoxicological tests were established(or applicated) using marine decomposer, primary producers and consumers. Development processes referred to the standard methods established by USEPA(United States Environmental Protection Agency), international organizations and European methods. However, the standard test species were selected among the domestic species generally found in the Korean waters and sediments. The test methods provide the culture/maintenance of test species, test methods, reproducibility and quality acceptance criteria etc. A total of nine test species were designated including bioluminescent bacteria(Vibrio fischeri), diatom(Skeletonema costatum), seaweed(Ulva pertusa), rotifer(Brachionus plicatilis), benthic copepod(Tigriopus japonicus), benthic amphipods(Mandibulophoxus mai, Monocorophium acherusicum), and fishes(Oryzias latipes, Paralichthys olivaceus). These test species represent the decomposer, primary producer and consumers in marine trophic system in Korean coastal ecosystems, and we recommend the "battery test" including at least one species from the each trophic level for marine ecotoxicological test.

Luminescent bacterial toxicity test was first introduced in the early 1980s, registered as international standard method in 1998 and now widely used as a common toxicity test method. This toxicity test uses luminescent bacterium, Vibrio fischeri, originated from marine environment as a test organism. The degree of toxicity can be evaluated from the comparison of luminescent emission intensity between control and treatment groups to toxicants and materials from various environmental matrix for 30 min. This test can be carried out by using commercial products and its results are sensitive and precise. This research is on the feasibility of adopting luminescent bacterial test as a domestic standard test protocol. Using commercial products, a series of experiments were conducted to identify the precision and accuracy of injection volume and light emission, and to evaluate concentration-response relationship between chemical concentrations and light emissions. Also, the feasibility of the application to environmental media and quality assurance/quality control were checked. The results of serial toxicity tests revealed that the preliminary luminescent bacterial toxicity test was robust and suitable as a standard method.
In this study, we cloned and sequenced the 15,204-bp DNA region containing the gene cluster for urease production from the chromosome of the environmental Photobacterium sp. strain HA-2. We identified 15 open reading frames (ORFs) and the G+C content was 40.3%. The urease gene cluster of Photobacterium sp. strain HA-2 consisted of seven genes, namely, ureDABCEF and ureG. There were five ORFs of urease genes in the opposite direction, which were homologous to the nickel transport operons (nik) of Vibrio parahaemolyticus and Escherichia coli. The genetic organization and sequences of the urease genes of Photobacterium sp. strain HA-2 resembled those found in Vibrio fischeri and V. parahaemolyticus.

Bioassay using the marine bacteria, Vibrio fischeri and rotifer, Brachionus plicatilis, and chemical analyses were conducted to assess the toxicity of the various sewage sludges, one of the major ocean dumped materials in the Yellow Sea of Korea. Sludge elutriates extracted by filtered seawater were used to estimate the ecotoxicity of the sludge. Chemical characterization included the analyses of organic contents, heavy metals, and persistent organic pollutants in sludge. Bacterial bioluminescent inhibition (15 min), rotifer mortality (24 hr) and rotifer population growth inhibition (48 hr) assay were conducted to estimate the sludge toxicity. EC50 15 min (inhibition concentration of bioluminescence after 15 minutes exposed) values by Microtox(R) bioassay clearly revealed different toxicity levels depending on the sludge sources. Highest toxicity for the bacteria was found with the sludge extract from dyeing waste and followed by industrial waste, livestock waste, and leather processing waste. Clear toxic effects on the bacteria were not found in the sludge extract from filtration bed sludge and rural sewage sludge. Consistent with Microtox(R) results, rotifer neonate mortality and population growth inhibition test also showed highest toxicity in dyeing waste and low in filtration bed and rural sewage sludge. High concentrations of persistent organic pollutants (POPs) and heavy metals were measured in the samples from the industrial wastes, leather processing plant waste sludge, and urban sewage sludge. However, there was no significant correlation between pollutant concentration levels and the toxicity values of the sludge. This suggests that the ecotoxicity in addition to the chemical analyses of various sludge samples must be estimated before release of potential harmful waste in the natural environment as part of an ecological risk assessment.

The acute and chronic toxicity of ambient and effluent water discharged to Lake Shihwa were investigated by using Vibrio fischeri, Daphnia magna and Oryzias latipes. Physicochemical characteristics including biochemical oxygen demand (BOD) and nine heavy metals in a total of 15 water samples were evaluated and were satisfied with relevant Korean Water Quality Standards (KWQS) except for Hg in one sample. Acute toxicity was observed in five samples collected from three sampling locations. When impacts on reproduction and growth after chronic exposure were evaluated with D. magna, all the samples showed significant chronic effects. Reproduction appeared relatively more sensitive endpoint. In 21 days chronic tests on O. latipes, survival, mean egg number per female per day, hatching success rate and time to hatch were affected by increasing sample concentration. The organ-level changes such as gonadosomatic index (GSI), and hepatosomatic index (HSI), and molecular biomarker of vitellogenin (Vtg) induction that evaluated with O. latipes increased as exposure concentrations increased. It is noteworthy that the samples that did not exceed the KWQS resulted in acute and chronic toxicities. The results suggested that numeric criteria based on physicochemical parameters may not be protective of aquatic ecosystem. Acute and chronic toxicity tests with organisms representing different trophic groups should be supplemented in order to provide adequate level of environmental protection.
