Vibrio damsela의 분리연구 (Studies on the Isolation of Vibrio damsela)

  • Authors studied on the isolation of V. damsela from sea water, fish and shellfish at the Keoje Hae keumkang on the southern sea and at Hongdo island and Heucksan island on the western sea of Korea from May to September in 1986. Authors investigated for the isolated strains to bacteriological identification, hemolysis about various erythrocytes and antibiotic susceptibilities. The results obtained were as follows: 1. V. damsela was isolated 14 strains from total 383 specimens; 233 cases of sea water, 40 cases of fish and 110 cases of shellfish, respectively. Eight strains were isolated from sea water and 6 strains were isolated from shellfish. 2. The biochemical characteristics which differentiate it from other Vibrio species were indole negative, ornithine negative, Voges-Proskauer positive, arginine positive, galactose positive, glucose positive, maltose positive, mannose positive, trehalose positive, and growth in nutrient broth with 1% to 6% NaCl. 3. On hemolysis reaction on blood agar media using human, rabbit and guinea pig erythrocytes, human erythrocytes were 11 strain positive, rabbit erythrocytes were 12 strain positive and guinea pig erythrocytes were 13 strain positive. 4. Senistivity test using with chemotherapeutic agents of "BioLab" Microbial Sensitivity Test Discs were generally sensitived to amikacin, ampicillin, cephalothin, chloramphenicol, clindamycin, erythromycin, gentamycin, kanamycin, methicillin, penicillin, streptomycin, tetracycline and tobramycin, respectively, but were resistant to lincomycin.

비브리오 균속이 생산하는 식중독 유발 Toxin 유전자의 검출 (Detection of Food Poisoning Toxin Genes Produced by Vibrio sp.)

  • Recently Vibrio sp. are the most frequently isolated microorganism, which causing food poisoning. We investigated the detection of toxin genes and effect of chitosan to toxin genes with PCR. Thirty strains of Vibrio sp. were isolated from sea water and sea products through biological and biochemical tests. Out of 30 strains, 8 were identified as V. parahemolyticus, 7 as V. mimcus, 6 as V. damsela, 5 as V. vulnificus, 4 as V. alginoyticus. In detection of ctx, tdh, and t고 as food poisoning-causing toxin genes, ctx from 7 strains, trh from 4 strains and tdh from 6 strains were detected. Among toxin genes detected strains, we selected V. vulnificus-2 V. damsela-1 and V. parahemolyticus-7. As adding chitosan solution to PCR product of 3 strains, the amplified DNA bands were not detected over 450$\mu\textrm{g}$/$m\ell$ concentrantion of chitosan. Over the result, chitosan is thought to influence the detection of toxin gene.

한국 울릉도 근해의 비브리오속의 분리연구 (Studies on the Isolation of Vibrio Species from Sea Water and Marine Products at Ulleung Island of Korea in 1985)

  • Authors studies on the isolation of non-sucrose fermentation Vibrio species such as V. parahaemolyticus, V. vulnificus and V. damsela from sea water, shellfishes and various algae at Ulleung island in the east of Korea on summer in 1985. Authors carried out test for isolated strains to biochemical characteristics, halophilism, hemolysis and serological reaction. The results obtained were as follows: 1. Strains isolated from total specimens were 25 strains of V. parahaemolyticus, 2 strains of V. vulnificus and 1 strain of V. damsela, respectively. 2. Isolated frequency of Vibrio species from sea water and marine products was lower than on specimens in the southern sea of Korea from 1980 to 1985. 3. On hemolysis reaction on blood agar media using human and rabbit erythrocytes, 14 strains among 25 strains of V. parahaemolyticus isolated were positive, and V. vulnificus and V. damsela were positive, respectively. 4. The distributions of serotypes of V. parahaemolyticus isolated were from O1 group to O10 group, and from K 10 to K 69. The results of K serotypes were as follows ; serotype K 10 and K 33 were 2 strains, serotype K 59 and K 69 were 1 strain, respectively. And 2 strains of V. vulnificus isolated were not agglutinated by antiserum of V. vulnificus. Therefore, the probability is that 2 strains of V. vulnificus isolated were other serotypes distributed on sea weater and marine products of Ulleung island of Korea.

In vivo에서 Photobacterium damselae subsp. damselae와 Vibrio 속 세균의 병원성 비교 (Comparative study of Photobacterium damselae subsp. damselae and Vibrios on pathogenicity in vivo)

  • 넙치 병어에서 Photobacterium damselae subsp. damselae와 Vibrio spp.(V. anguillarum, V. splendidus, V. harveyi and V. ordalii)를 분리한 후 in vivo에서 병원성을 비교하였다. 분리균을 인위감염 한 결과, 폐사율, 혈청의 ALT, AST, cortisol 농도가 vibrio 감염구에 비하여 P. damselae 감염구에서 높게 나타났다. 또한, P. damselae 감염구에서 비특이적 면역반응인 식세포의 nitroblue tetrazolium(NBT) 반응과 혈청의 라이소자임 활성이 유의적으로 낮게 나타났다. 따라서, P. damselae는 특이적 효소인 damselysin의 분비로 높은 용혈능과 phospholipase 활성, 감염 시 내부 장기의 손상에 의한 ALT와 AST 상승, cortisol 농도의 증가로 인한 면역기능 저하가 높은 병원성을 나타낸 것으로 사료된다.

동절기 한국 남해안의 궤양증 및 복수증 양식 넙치로부터 vibrio종의 분리 (Isolation of vibrio species from cultured flounders (paralichthys olivaceus) with uicers and ascites in the southern coast of Korea during the winter season)

  • this study was performed for the purpose of surveying the distribution and characteristics of the Vibrio species in the diseased farm flounders (Paralichthys olivaceus) during the winter season in Korea. Samples were collected along the southern coast in the Sinam, Keoje Island, and Teosu areas from November '90 to Feburary '91. Nine species of Vibrio were identified as V. tubiashii (85 strains), V. damsela (78 strains), V. anguillarum (21 strains), V. compbelli (9 strains), V. fluvialis (9 strain), V. costicola (1 strain), V. alginolyticus (1 strain), V. gazogenes (1 strain), V. marinus (1 strain), and unidentified 3 groups of bacteria (154 strains) which may have related with V. tubiashii were isolated. Eleven kinds of discs (BBL Co.) were used for antibiotic susceptibility test and 116 strains were selected for the experiment. Most bacteria were powerfully inhibited their growth by 4 antibiotics such as cephalothin, chloramphenicol, colistin, and tetracycline, but they were poorly inhibited by 4 antibiotics such as carbenicillin, kanamycin, penicillin G, and streptomycin.

2002-2003년도 복수증 양식산 넙치로부터 동정된 미생물상 (Microbial Flora in Ascitic Cultivated Olive Flounder (Paralichthys olivaceus) in Koe-je Island in Korea During 2002-2003.)

  • 본 연구는 2002년 6월부터 2003년 11월까지 복수증을 나타내는 양식넙치의 세균상과 virus를 동정한 것이다. 생화학적 성상을 기초로 한 표현형으로, 병어체의 복수, 간, 신장, 내장으로부터 Vibrio, Edwardsiella, 및 stonella 등 3개 속이 동정되었다. Vibrio속은 5종으로 우점이었고 V. harveyi (32균주), V. parahaemolyticus (23균주), V. alginolyticus (10균주), V. carcariae (2균주), V. metschnikovii (5균주)였다. 그밖에 Photobacterium damselae (6균주)와 장내세균인 Edwardsiella tarda (15균주)가 동정되었다. 감염어의 내부 장기로부터 해산어 병원성 바이러스로 알려진 marine birnavirus (MABV)가 분리되었다.

경남 남부지역 양식어류 질병에서 항생제 감수성에 관한 연구 (The Study on Fish Diseases with Reference to Bacterial Susceptibility to Antibiotics in the Southern Area of Kyeognam)

  • The examination with 96 bacteria isolates for antibiotics susceptibility was resulted in exploring many antibiotics resistance isolates from the diseased fishes. Vibrio sp. isolates were demonstrated over 90% resistances for Penicillin (P), Amoxicillin (Amc), Erythromycin (I), Colistin (CL). Streptomycin (S). Novobiocin (NV) and Neomycin(N), these isolates were, however, appeared over 80% susceptibilities for Norfloxacin (Nor), CE and UB. In Edwardsiella tarda case, 90 to 100% high resistance was observed for P, Doxycycline (Do), Tetracyclin (Te), Oxytetracycline (T), E, CL, Sulfamethoxasoletrimetoprim (SXT), S, Oxolinic acid (OA), W, Neomycin(N), on the contrary, 90 to 100% high susceptibilities was found for Amc, Nor, Ciprofloxacin (Cip), Orbifloxacin (ORB), Enrofloxacin (ENR), Flumeguine (UB) and NA. CL, Pefloxacin (PEF),S, Flumeguine (UB), OA, NA, NV N was exhibited 90 to 100% resistances for Streptococcus sp., on the other hand, 100% susceptible to AMC and 80% susceptible to Do, Te, ENR and UB was recognised. Lastly, Photobacterium damsela subsp. piscicida was showed 100fe susceptible to Amc and 86% susceptible to NOR, CIP ENR and UB. As a consequence, fish bacterial pathogens isolated from Kyeongnam area, especially Tongyeong-si, Geoje-si, and Goseong-gun, were showed highly resistant to a variety of antibiotics available in the field.

질병의 증상을 보이는 해수 양식 어류에서 분리한 비브리오속 세균 (Vibrios Isolated from Diseased Marine Culturing Fishes in Korea)

  • 본 연구에서는 2002년에서 2004년간 우리 나라 해수 어류 양식장에서 분리되는 세균 중 비브리오 속에 속하는 세균의 종 조성을 조사하였다. 질병의 증상을 보이는 어류로부터 166개의 비브리오속 세균 균주를 수집하였으며, 이들 균주는 넙치 (133 균주), 조피볼락 (8 균주), 참돔 (6 균주), 대하 (5 균주), 감성돔 (4 균주), 전복 (3 균주) 및 기타 해수 어류 (7 균주) 등에서 분리한 균주를 대상으로 하였다. 모든 분리 균주에 대하여 각종 생화학적 성상을 조사하고 그 특성에 따라 분리 균주를 구분하였다. 각 생화화적 성상 group에서 대표 균주를 선정하여 16S rRNA 및 16S-23S rRNA 유전자 서열을 분석하고 이들 결과를 종합하여 분리 균주을 동정하였다. 그 결과 14종 이상의 비브리오 종으로 동정할 수 있었으며, 그 종 조성은 V. ichthyoenteri (45 strains), V. alginolyticus (34 strains), V. harveyi (32 strains), Ph. damselae subsp. damselae (Formerly V. damsela, 10 strains), V. campbellii (6 strains), V. costicola-like (6 strains), V. fisheri (5 stains), V. fluvialis (4 strains) 및 Vibrio spp. (24 strains) 등이었다.

새로운 conjugation 방법을 응용한 R plasmid 함유 어병세균의 분리와 양식장 내성균의 현황 분석 (Application of a New Conjugation Method to Fish Pathogenic Bacteria Containing R Plasmid for the Analysis of Drug-Resistant Status in Aquaculture)

  • R plasmid의 분포와 새로운 conjugation 방법 개발을 위하여, 한국의 남해안과 동해안의 양식현장의 어류질병관련 세균으로부터 항균제 다제 내성균을 분리하였다. 분리균 134종에서 10균주가 chlorarnphenicol, tetracycline, streptomycin, ampicillin, colistin, nalidixic acid, oxolinic acid, kanamycin 등에 대한 다제내성의 특성을 가지고 있었으며 이중 V. damsela JE1 한 균주는 chloram-phenicol과 tetracycline에 대한 내성전달 특성을 갖는 R plasmid 와 ampicillin과 kanamycin에 대한 유전자를 chromosome 등에 함유하고 있었다. 분리된 어병세균의 다제내성 특성이 R plasmid에 기인하는 것인지의 확인은 다양한 장내세균의 선택배지로 최근 개발된 CC 배지에서 각기 다른 균주들이 나타내는 증식여부 또는 집락 색깔의 차이를 이용하여 donor, recipient 그리고 transconjugant를 구별할 수 있는 방법을 적용하였다. Conjugation의 최적화를 위해 여러 가지 조건을 비교한 결과, V. damsela JE1의 R plasmid의 전달은 filter mating법을 사용하는 것이 broth mating법보다 약 100배 이상의 높은 성공률을 보여 주었으며 이때 사용온도는 30$^{\circ}C$ 이상, mating 시간은 24시간에서 가장 높은 전달 빈도를 나타내었다. R plasmid를 함유한 V. damsela JE1이 분리된 4개월 후, 동일 양식장의 동일 그룹의 넙치 장내세균에서 분리한 다제내성균 Sphingomonas sp. 3균주는 V. damsela JE1에서 분리된 것과 같은 크기의 R plasmid를 함유하고 있었으며, 그 특성 또한 V. damsela JE1의 R plasmid와 마찬가지로 다제 내성 중 Cm과 Tc에 대한 유전자만이 recipient로 전달되어 장내 세균총이 R plasmid의 reservoir로서의 역할을 하고 있음을 확인하였다.
