Knowledge, attitudes and practices toward antimicrobial usage: a cross-sectional study of layer and pig farm owners/managers in Chiang Mai, Lamphun, and Chonburi provinces, Thailand, May 2014 to February 2016

  • This study aimed at determining the current knowledge, attitude, and practices (KAP) of layer and pig farmers to antimicrobial usage and its consequences. Description of such KAP could provide insights useful for promoting the rational use of antimicrobials in livestock. From May 2014 to February 2016, a survey involving 251 respondents in Chiang Mai, Lamphun, and Chonburi provinces, Thailand, was conducted by using a validated questionnaire. More than half (51.0%) of the respondents incorrectly believed that antimicrobial drug efficacy could not be reduced by using sub-recommended dosages, 61.2% had misconceptions about non-therapeutic antimicrobial use, and 66.9% inaccurately felt that antimicrobials were also effective for virus and fungi. Over half (50.6-55.2%) did not see the need to follow instructions or advice of veterinarians. Moreover, only 10.4% regularly relied on responses to drugs sensitivity tests when evaluating the effectiveness of antimicrobials. Overall, assessment of KAP regarding antimicrobial usage indicated that the majority of respondents had low levels of knowledge of antimicrobials, neutral rather than positive attitudes, and employed poor practices in the use of antimicrobials. The results indicate improvements in KAP could be helpful in developing more effective interventions by farmers, reduce antibiotic usage, and slow the growth of antimicrobial resistance.

Long-Term Management of a Refractory Corticosteroid Responsive Tremor Syndrome

  • A 2-year-old intact female Maltese dog was presented with generalized involuntary tremors and nystagmus without regular direction. The dog was conscious the whole time while it was trembling. Its involuntary tremors were alleviated at rest or during sleep. Magnetic resonance imaging (MRI) revealed asymmetric hydrocephalus and caudal occipital malformation. In cerebrospinal fluid (CSF) analysis, a trace of protein was found and total nucleated cell count (TNCC) was slightly increased. However, infectious pathogens were not found. In complete blood count, there was a mild leukocytosis. After the patient received anticonvulsants (midazolam, phenobarbital, KBr), diuretics (furosemide) with an anti-inflammatory drug (prednisolone, 0.5 mg/kg PO bid), and a proton-pump inhibitor (omeprazole), it showed no improvement. The patient was tentatively diagnosed with corticosteroid responsive tremor syndrome. So the anticonvulsants and diuretics were discontinued and the dose of prednisolone was increased to an immunosuppressive dose (1 mg/kg PO bid). After administering the immunosuppressive dose of prednisolone, the patient did not show nystagmus. Its tremors were much alleviated. However, they did not disappear. Five weeks later, the patient showed gradual improvement but still was trembling when moving around. Nine weeks later, its tremors were similar to before. So diazepam (0.3 mg/kg PO sid) was added to the treatment. After that, its tremors were alleviated more. Prednisolone and diazepam were maintained for about five months, with tapering of the dose of prednisolone (until 0.5 mg/kg PO sid). About 7 months later after the treatment was started, the dog was trembling rarely except when it was excited. Therefore, diazepam was discontinued. This case describes a refractory white dog shaker syndrome successfully managed with long-term administration of a steroid and diazepam.

Antibacterial and Pharmacological Evaluation of Fluoroquinolones: A Chemoinformatics Approach

  • Fluoroquinolone (FQ) antibiotics are an important class of synthetic antibacterial agents. These are the most extensively used drugs for treating bacterial infections in the field of both human and veterinary medicine. Herein, the antibacterial and pharmacological properties of four fluoroquinolones: lomefloxacin, norfloxacin, ciprofloxacin, and ofloxacin have been studied. The objective of this study was to analyze the antibacterial characteristics of the different fluoroquinolones. Also, the pharmacological properties of the compounds including the Lipinski rule of five, absorption, distribution, metabolism, and excretion, LD50, drug likeliness, and toxicity were evaluated. We found that among all four FQ molecules, ofloxacin showed the highest antibacterial activity through in silico assays with a strong interaction (-38.52 kJ/mol) with the antibacterial target protein (topoisomerase-II DNA gyrase enzyme). The pharmacological and pharmacokinetic analysis also showed that the compounds ciprofloxacin, ofloxacin, lomefloxacin and norfloxacin have good pharmacological properties. Notably, ofloxacin was found to possess an IGC50 (concentration needed to inhibit 50% growth) value of $0.286{\mu}g/L$ against the Tetrahymena pyriformis protozoa. It also tested negative for the Ames toxicity test, showing its non-carcinogenic character.

치와와에서 발생한 외상적 뇌손상 증례; 연속적인 임상적 및 전산화 단층영상 소견 (A Case of Traumatic Brain Injury in a Chihuahua Dog: Serial Clinical and Computed Tomographic Findings)

  • A dog (Chihuahua, 2-year-old, intact female) was referred to us because of cluster seizure. She had history of falling from height few days before presentation. Brain computed tomography (CT) results demonstrated fracture line on right temporal bone and hypodense, edematous changes of the adjacent brain parenchyma on right cerebral parenchyma. Based on history, clinical signs, and diagnostic imaging findings, this patient was diagnosed to traumatic brain injury. After diagnosis, the patient was well controlled with anti-inflammatory drug and anti-epileptic drugs. When 30, 480, and 1260 days after initial brain CT examination, we performed serial brain CT rechecks. This case report describes serial clinical and brain CT findings after traumatic brain injury.

Application of a solid-phase fluorescence immunoassay to determine ampicillin residues in muscle tissue of olive flounder (Paralichthys olivaceus)

  • $Parallux^{TM}$, a solid-phase fluorescence immunoassay(SPFIA) developed for antibiotics residue detection in milk, was applied for analysis of fish muscle. The recommended therapeutic dose of ampicillin(100 mg/kg body weight, withdrawal period 7 days) was orally administered to a group of 25 olive flounders(Paralichthys olivaceus) for consecutive five days. Muscle was sampled after drug treatment 1st, 2nd, 3rd, 4th and 5th day. The concentration of ampicillin in muscle, determined by SPFIA, was compared with that of internal standard(10 ppb as ampicillin). The absorbance ratio of sample to internal standard(Bs/Bo) was employed as an index to determine the muscle residue in olive flounder. To investigate the recovery rate, the standard solutions were added to muscle samples to give final concentrations in muscle of 10 and 50 ng/ml. The recovery rates of all spiked samples were > 89% of the spiked value. Ampicillin was detected in muscle of fishes treated until the 3rd day of withdrawal period. The present study showed that the SPFIA can be easily adopted in predicting tissue residues for ampicillin in farmed fishes.

  • This study was carried out the antimicrobial resistance of staphylococcus aureus isolated from bovine mastitic milk, chickens, korean native cattle, korean native goats, pigs, dog and mice in northern area of kyongbuk. The result were summarised as follows ; A total of 149 S aureus were isolated from bovine mastitic milk, chickens, korean native cattle, korean native goats, pigs, dog and mice. In 80 isolates of S aureus from bovine mastitic milk, 60% of isolates revealed resistance to penicillin and ampicillin, 19% to gentamicin, 6% to tetracycline. In 36 isolates of S aureus from chickens, 72% of isolates revealed resistance to tetracycline, 58% to penicillin and ampicillin, 42% to streptomycin, 31% to lincomycin, 25% to norfloxacin, 22% to gentamicin. In 17 isolates of S aureus from korean native cattle, 100% of isolates revealed resistance to penicillin and ampicillin, 88% to lincomycin, 76% to tetracycline. 2 MRSA were isolated from the isolates of S aureus from bovine mastitic milk and revealed multi-drug resistance.

Neonatal isoerythrolysis in Thoroughbred foals

  • Two 4-day-old Thoroughbred foals with acute clinical signs of depression, elevated pulse and respiratory rates, anemia, and jaundice were referred to the equine hospital of Korea Racing Authority. The foals were seemingly normal at birth, but showed clinical signs such as abnormal behavior, jaundice and anemia suddenly after ingestion of the mare's colostrum, followed by death at 4 days after birth. According to the results of the hematology, serum chemistry analysis, jaundice foal agglutination test, and blood groups test, the foals were diagnosed as neonatal isoerythrolysis (NI) caused by the mare's antibodies. These results suggested that the pregnant mares have a potential for NI.

랫트 뇌에서의 galectin-3의 검출 (Expression of galectin-3 in rat brain)

  • Galectin family, endogenous ${\beta}$-galactoside-binding animal lectins, is known for the role in cell differentiation, morphogenesis, apoptosis and tumorigenesis. Galectin-3, one of family member, has been studied for its role in cell differentiation and tumor metastasis, and for its expression on epithelial cells of colon and mast cells but not in brain. Several reports, however, suggest its expression in brain including as a prion binding protein. In this report we explored possibility of galectin-3 expression in brain tissue. With Western blot and RT-PCR with rat brain tissues, we could detect galectin-3 that was not shown by conventional immunohistochemistry. Our results indicated galectin-3 was expressed in brain, and substantiate the previous report on galecin-3 as a prion-related protein in brain.

Rhodococcus equi pneumonia in foals in Gyeonggi-do and characterization of the isolates from lesions and environment

  • We report here two cases of Rhodococcus (R.) equi-causing pneumonia of Throughbred foals in Gyeonggi-do in 2006. R. equi was isolated from the lung lesions of the dead foals, and from the feces and soils on the farms where the clinical cases of R. equi infection occurred. The isolates were characterized by biochemical properties, polymerase chain reaction for vapA gene and antimicrobial susceptibility. In drug susceptibility test, erythromycin, gentamycin, vancomycin, and rifampin were found to be the most susceptible for all isolates. These results suggest that R. equi pneumonia may be endemic in the horse-breeding farms in inland Korea and the farm environment may be widely contaminated with virulent R. equi.

Toxicity of disulfiram, a therapeutic agent for chronic alcoholism, to mouse spleen cells

  • Disulfiram is a drug used to treat alcohol dependence. Recent studies have shown that disulfiram also has anti-cancer effects. Considering that many anti-cancer agents have side effects, including immunosuppression, it is important to check if disulfiram has some cytotoxicity to immune cells. In this study, mouse spleen cells were treated with disulfiram and the metabolic activity was measured. Disulfiram increased the cell death of spleen cells according to annexin V-FITC/PI staining analysis. In addition, disulfiram decreased the mitochondrial membrane potential of spleen cells. The toxicity of disulfiram was concentration dependent. Interestingly, disulfiram affected the population of lymphocytes and the subset of spleen cells was altered. This study provides clinicians and researchers with valuable information regarding the toxicity of disulfiram to mouse spleen cells, particularly lymphocytes.
