Evaluation of Soybean Oil as a Lipid Source for Pig Diets

  • An experiment was conducted to determine the effects of soybean oil supplementation replacing tallow in pig diets at different stages of growth. One hundred and twenty crossbred (Landrace${\times}$Yorkshire${\times}$Duroc) pigs weighing 18 kg on average were selected. Pigs were randomly allotted to 12 pens of 10 pigs (5 pigs of each sex) each. Three pens were assigned to each of the four treatments: TA; tallow diet, TA-SO-80; switched from tallow to soybean oil diet at 80 kg average body weight, TA-SO-45; switched from tallow to soybean oil diet at 45 kg average body weight, and SO; soybean oil diet. Treatment SO was significantly lower in ADG than tallow diets (TA, TA-SO-80 and TA-SO-45) during the grower period (18 to 45 kg). However, treatment SO showed greatest compensation in ADFI and ADG during the finisher-2 period (after 80 kg body weight). ADFI and ADG and Gain/Feed for the total period were not significantly different among treatments. Loin area, back fat thickness, firmness and melting point of back fat were not significantly different. The levels of total cholesterol and low density lipoprotein+very low density lipoprotein cholesterol in serum were significantly lower in treatment SO than in treatments TA-SO-45, TA-SO-80 and TA. The level of serum triglyceride linearly increased as the length of the tallow feeding period increased. Serum immunoglobulin-G (IgG) level was significantly higher in the soybean oiltreatment than in other treatments. Major fatty acid composition of short rib muscle and back fat were significantly influenced by treatments. Contents of ${\alpha}$-linolenic acid (C18:3) and docosahexaenoic acid (DHA, C22:6) linearly increased as the soybean oil feeding period increased. In conclusion, soybean oil can be supplemented to the diet of pigs without significant effects on growth performance and carcass characteristics. The level of polyunsaturated fatty acids (PUFA), especially $\omega-3$ fatty acids in the carcass was increased by soybean oil supplementation.

Protective effect of silymarin in streptozotocin-induced diabetic dyslipidaemia in rats

  • The present study investigated the effect of silymarin, a flavonoid, on streptozotocin (STZ) - induced diabetic dyslipidaemia in rats. Experimental diabetes was induced by a single intraperitoneal injection of STZ (60 mg/kg). Silymarin (25 mg/kg and 50 mg/kg) was orally administered to diabetic rats for a period of 15 days. Blood glucose levels, serum lipid profile and liver glycogen levels were estimated following the established procedures. Biochemical observations were supplemented with histological examination of liver sections. Oral administration of silymarin to diabetic rats significantly (P < 0.001) decreased the blood glucose levels ($259.99{\pm}23.64$ vs. $99.90{\pm}2.62$ [25 mg] & $89.17{\pm}3.32$ [50 mg]). The most interesting finding was the significant (p < 0.001) increase in HDL-cholesterol levels ($26.99{\pm}0.61$ vs. $40.55{\pm}0.52$ [25 mg] & $41.12{\pm}0.37$ [50 mg]) whereas, there was a significant decrease in serum total cholesterol (TCh), triglycerides (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) cholesterol levels observed in silymarin treated diabetic rats. STZ treatment caused significant degeneration of liver parenchyma, which was normalized to near normal morphology by administration of silymarin. The findings indicate that silymarin effectively improved the overall lipid profile and restored the glycogen stores in the liver of STZ-induced diabetic rats, in a dose dependent manner. The results indicate existence of abnormalities in lipid metabolism in STZ-induced diabetic rats and suggest a protective effect of silymarin in this animal model.

Efficacy of nobiletin in improving hypercholesterolemia and nonalcoholic fatty liver disease in high-cholesterol diet-fed mice

  • BACKGROUND/OBJECTIVES: Nobiletin (NOB), a citrus flavonoid, is reported to have beneficial effects on cardiovascular and metabolic health. However, there is limited research investigating the effect of long-term supplementation with low-dose NOB on high-cholesterol diet (HCD)-induced hypercholesterolemia and non-obese nonalcoholic fatty liver disease (NAFLD). Therefore, we investigated the influence of NOB on hypercholesterolemia and NAFLD in HCD-fed mice. SUBJECTS/METHODS: C57BL/6J mice were fed a normal diet (ND) or HCD (35 kcal% fat, 1.25% cholesterol, 0.5% cholic acid) with or without NOB (0.02%) for 20 weeks. RESULTS: HCD feeding markedly reduced the final body weight compared to ND feeding, with no apparent energy intake differences. NOB supplementation suppressed HCD-induced weight loss without altering energy intake. Moreover, NOB significantly decreased the total cholesterol (TC) levels and the low-density lipoprotein (LDL)/very-LDL-cholesterol to TC ratio, and increased the high-density lipoprotein-cholesterol/TC ratio in plasma, compared to those for HCD feeding alone. The plasma levels of inflammatory and atherosclerosis markers (C-reactive protein, oxidized LDL, interleukin [IL]-1β, IL-6, and plasminogen activator inhibitor-1) were significantly lower, whereas those of anti-atherogenic adiponectin and paraoxonase were higher in the NOB-supplemented group than in the HCD control group. Furthermore, NOB significantly decreased liver weight, hepatic cholesterol and triglyceride contents, and lipid droplet accumulation by inhibiting messenger RNA expression of hepatic genes and activity levels of cholesterol synthesis-, esterification-, and fatty acid synthesis-associated enzymes, concomitantly enhancing fatty acid oxidation-related gene expression and enzyme activities. Dietary NOB supplementation may protect against hypercholesterolemia and NAFLD via regulation of hepatic lipid metabolism in HCD-fed mice; these effects are associated with the amelioration of inflammation and reductions in the levels of atherosclerosis-associated cardiovascular markers. CONCLUSIONS: The present study suggests that NOB may serve as a potential therapeutic agent for the treatment of HCD-induced hypercholesterolemia and NAFLD.

Dietary Protein Restriction Alters Lipid Metabolism and Insulin Sensitivity in Rats

  • Dietary protein restriction affects lipid metabolism in rats. This study was performed to determine the effect of a low protein diet on hepatic lipid metabolism and insulin sensitivity in growing male rats. Growing rats were fed either a control 20% protein diet or an 8% low protein diet. Feeding a low protein diet for four weeks from 8 weeks of age induced a fatty liver. Expression of acetyl-CoA carboxylase, a key lipogenic enzyme, was increased in rats fed a low protein diet. Feeding a low protein diet decreased very low density lipoprotein (VLDL) secretion without statistical significance. Feeding a low protein diet down-regulated protein expression of microsomal triglyceride transfer protein, an important enzyme of VLDL secretion. Feeding a low protein diet increased serum adiponectin levels. We performed glucose tolerance test (GTT) and insulin tolerance test (ITT). Both GTT and ITT were increased in protein-restricted growing rats. Our results demonstrate that dietary protein restriction increases insulin sensitivity and that this could be due to low-protein diet-mediated metabolic adaptation. In addition, increased adiponectin levels may influences insulin sensitivity. In conclusion, dietary protein restriction induces a fatty liver. Both increased lipogenesis and decreased VLDL secretion has contributed to this metabolic changes. In addition, insulin resistance was not associated with fatty liver induced by protein restriction.

백서에서 신선초 8주 투여에 의한 quercetin, isoquercitrin 및 hyperoside의 체내농도 및 혈중 지질함량 변화 (Tissue Concentration of Quercetin, Isoquercitrin and Hyperoside, and Lipid Profile Changes Following 8-Week Feeding of Angelica keiskei Powder in Rats)

  • 신선초의 건조분말을 사료에 5%수준으로 혼합하여 백서에 8주간 투여하였을 때, 총 cholesterol, LDL-cholesterol, VLDL-cholesterol의 농도를 낮추며, HDL-cholesterol의 농도는 높이는 효과가 있음이 관찰되었다. 신선초에 존재한다고 알려진 항산화성 flavonoid들 중 quercetin과 그 배당체의 하나인 isoquercitrin이 백서의 체내에서 측정된 반면, 다른 quercetin 배당체인 hyperoside는 측정되지 않았다. 이 결과는 장기간 신선초투여에 의해 신선초에 함유된 quercetin 또는 그 배당체가 실제로 체내로 흡수되어 약리작용을 발휘할 수 있을 가능성을 제시해 준다.

오메가-3 지방산 함유 복분자종자유에 의한 고지방식이 유도 고지혈증 마우스의 혈중 중성지방 및 콜레스테롤 감소 효과 (Reduction of Plasma Triglycerides and Cholesterol in High Fat Diet-Induced Hyper-Lipidemic Mice by n-3 Fatty Acid from Bokbunja (Rubus coreanus Miquel) Seed Oil)

  • 본 실험의 복분자종자유는 리놀렌산 238.3 mg/g, 리놀레산과 감마리놀렌산 427.1 mg/g을 포함하며 이는 고시된 범위내의 섭취량이므로 독성시험 없이 4주 동안 매일 경구 투여로 암컷과 수컷 쥐 모두에게 섭취하도록 하였다. 또한 성인 정상체중을 약 60 kg으로 설정하고 60 kg의 성인이 하루 1 g, 2 g을 각각 섭취할 때의 혈중 지방의 감소 효과를 알아보기 위해 급이군의 설정을 1 g/60 kg BW/d(BSO 1 g), 2 g/60 kg BW/d(BSO 2 g)로 정하였다. 양성대조군의 경우에 판매되는 연어유 1,000 mg 캡슐을 2정 섭취하는 것을 권장하였고, 이에 맞추어 2 g/60 kg BW/d로 설정하게 되었다. 실험기간 종료 후 마우스를 희생시켜 혈액을 얻었으며, 복분자종자유(BSO 2 g)를 섭취한 마우스에서 총콜레스테롤 및 HDL, LDL/VLDL-콜레스테롤과 혈중 중성지방이 유의적으로 감소하는 것을 확인할 수 있었다(P<0.05). 이를 종합해 보았을 때 복분자종자유는 고지혈증 상태를 개선하고 미약하지만 항응고 활성을 통해 혈액의 항상성을 유지하도록 돕는 작용을 한다고 할 수 있다. 따라서 앞으로 동물 유래가 아닌 식물유래 복분자종자유를 이용한다면 고지혈증 개선 효과와 혈액 항응고 활성을 조절할 수 있을 것이며, 더욱 다양한 표적 인자 분석을 통하여 복분자종자유의 고지혈, 항응고 및 혈행개선 기전연구가 가능할 것이라 판단된다.

태음조위탕 엑스과립이 비만을 포함한 대사증후군 위험인자에 미치는 영향에 대한 연구: 단일군, 전향적, 다기관 임상연구 (The Effects of Taeeumjowi-tang Extract Granule on Metabolic Syndrome Risk Factors with Obesity: A Single Group, Prospective, Multi-Center Trial)

  • Objective The purpose of this study is to study the effects of Taeeumjowi-tang extract granule on metabolic syndrome with obesity in Taeeumin. Methods We studied 206 subjects who diagnosed metabolic syndrome with obesity and classified Taeeumin by a Sasang constitution analysis tool. The subjects were administered by Taeeumjowi-tang for 12 weeks. We investigated the changes of body mass index, abdominal circumference, the number of risk factors for metabolic syndrome, lipid, fasting blood sugar, Hemoglobin A1c, free fatty acid, insulin, and blood pressure. Results Body mass index and abdominal circumference, systolic and diastolic blood pressure were significantly reduced after 4 weeks, 8 weeks, and 12 weeks of administration compared to before administration. Triglyceride significantly decreased after 8 weeks and 12 weeks of dosing compared to before taking. Low density lipoprotein cholesterol, fasting blood sugar were significantly increased after 8 weeks of administration compared to before administration. High density lipoprotein cholessterol was decreased significantly after 4 weeks of administration compared to before administration. The number of risk factors for metabolic syndrome, Hemoglobin A1c increased significantly after 12 weeks of administration compared to screening. Conclusions In this study we knew that Taeeumjowi-tang extract granule is very efficient in metabolic syndrome, especially in obesity and hypertension, triglyceride, insulin resistance.

성인의 심혈관계 위험인자를 적용한 고저밀도지단백-콜레스테롤혈증과 건강행태의 관련성 연구 : 국민건강영양조사 제6기 (2013 ~ 2015) 자료 이용 (Relationship between health behaviors and high level of low density lipoprotein-cholesterol applying cardiovascular risk factors among Korean adults: based on the sixth Korea National Health and Nutrition Examination Survey (KNHANES VI), 2013 ~ 2015)

  • 본 연구는 국민건강영양조사 제6기 (2013 ~ 2015) 원시자료를 이용하여 심혈관계 위험인자를 적용한 고LDL-콜레스테롤혈증과 건강행태의 관련성을 조사하기 위해 수행되었으며, 19세 이상 남자 5,939명, 여자 7,902명의 총 13,841명을 대상으로 하였다. 심혈관계 위험인자는 뇌졸중, 심근경색증 또는 협심증, 당뇨병, 고혈압, 흡연, 저HDL-콜레스테롤 (40 mg/dL 미만), 연령 (남자 45세 이상, 여자 55세 이상), 고HDL-콜레스테롤 (60 mg/dL 이상)을 포함하였다. 뇌졸중이나 심근경색증 또는 협심증은 초고위험군, 당뇨병은 고위험군, 이외 위험인자가 2개 이상은 중등도위험군, 1개 이하는 저위험군으로 구분하였다. 각 위험군 별 목표 수치를 기준으로 한 고LDL-콜레스테롤혈증 유병자는 남자 25.5%, 여자 21.7%이었다. 건강행태와 고LDL-콜레스테롤혈증 유무의 연관성을 복합표본 교차분석으로 분석한 결과, 남녀 모두 연령, 교육수준, 결혼상태, 체질량지수, 탄수화물 에너지 섭취 비율, 지방의 에너지 섭취비율, 포화지방 에너지 섭취비율, n-6 지방산의 에너지 섭취 비율에 따라 고LDL-콜레스테롤혈증 유무는 유의한 연관성이 있었다. 그리고 흡연은 남자에서, n-3계 지방산의 에너지 섭취 비율은 여자에서 고LDL-콜레스테롤혈증 유무와 각각 유의한 연관성이 있었다. 건강행태와 고LDL-콜레스테롤혈증 발생 위험과의 관련성을 확인하기 위해 복합표본 다중 로지스틱 회귀분석을 실시한 결과, 교란인자 보정 후 남자에서는 현재 흡연 (OR: 1.66, 95% CI: 1.40-1.99), 비만 (OR: 1.95, 95% CI: 1.64-2.31) 에서 고LDL-콜레스테롤혈증 유병 오즈비가 유의하게 높았다. 여자의 경우 현재 흡연 (OR: 1.73, 95% CI: 1.19-2.52), 비만 (OR: 1.63, 95% CI: 1.39-1.90)에서 고LDL-콜레스테롤혈증 유병 오즈비가 유의하게 높았고, n-3 지방산 에너지 섭취 비율이 '하'(Q1)에 비해 '중 하' (Q2) (OR: 0.77, 95% CI: 0.62-0.96), '중 상' (Q3) (OR: 0.73, 95% CI: 0.56-0.94), '상' (Q4) (OR: 0.67, 95% CI: 0.51-0.87) 에서 고LDL-콜레스테롤혈증 유병 오즈비가 유의하게 낮았다. 따라서 고LDL-콜레스테롤혈증 개선을 위한 전략 수립에 금연, 정상 체중 유지가 필요하며, 영양소별 적정한 에너지 섭취 비율 유지를 고려해야 한다.

일개 농촌지역 저소득층 당뇨병환자의 관리 상태 (Management of Diabetic Mellitus in Low-income Rural Patients)

  • Objectives : Knowledge about the management status of diabetic melitus (DM) is essential to improve diabetic management. Moreover, low income is associated with poor adherence to treatment and increased mortality. This study was performed to evaluate the management status of DM in low-income patients in a rural area. Methods : We enrolled 370 patients with type 2 DM living in Gokseong county, JeollaNamdo. A well-trained examiner measured the height, weight, waist circumference, blood pressure, total cholesterol, triglyceride, high density lipoprotein cholesterol, fasting blood sugar and glycosylated hemoglobin (HbA1c) levels. Carotid ultrasonography was used to measure carotid artery carotid artery intima media thickness (IMT) and plaque. anklebrachial index (ABI) was used to evaluate peripheral artery disease. A fundoscopic examination was performed to evaluate diabetic retinopathy. A history of diabetes complications and health-related questionnaires were also completed. Results : The age of diabetic subjects was 68.7$\pm$8.7 years and the duration of diabetes was 8.9$\pm$8.2 years. Most (63.5%) had hypertension, and 45.7% had triglycerides below 150 mg/dl, 38.1% had low density lipoprotein cholesterol (LDL) cholesterol below 100 mg/dl, 48.7% had urine albumin to creatinine ratio (UACR) below 30 mg/g. Less than half (45.9%) achieved the goal of HbA1c less than 7% suggested by the American Diabetes Association (ADA). 10.6% had peripheral vascular disease, 11.9% had retinopathy, and 60.8% had chronic kidney disease. Conclusions : DM management in low income patients is very poor and requires further work to improve.

Purple perilla frutescens extracts containing α-asarone inhibit inflammatory atheroma formation and promote hepatic HDL cholesterol uptake in dyslipidemic apoE-deficient mice

  • BACKGROUND/OBJECTIVES: Dyslipidemia causes metabolic disorders such as atherosclerosis and fatty liver syndrome due to abnormally high blood lipids. Purple perilla frutescens extract (PPE) possesses various bioactive compounds such as α-asarone, chlorogenic acid and rosmarinic acid. This study examined whether PPE and α-asarone improved dyslipidemia-associated inflammation and inhibited atheroma formation in apolipoprotein E (apoE)-deficient mice, an experimental animal model of atherosclerosis. MATERIALS/METHODS: ApoE-deficient mice were fed on high cholesterol-diet (Paigen's diet) and orally administrated with 10-20 mg/kg PPE and α-asarone for 10 wk. RESULTS: The Paigen's diet reduced body weight gain in apoE-deficient mice, which was not restored by PPE or α-asarone. PPE or α-asarone improved the plasma lipid profiles in Paigen's diet-fed apoE-deficient mice, and despite a small increase in high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein (LDL)-cholesterol, and very LDL were significantly reduced. Paigen's diet-induced systemic inflammation was reduced in PPE or α-asarone-treated apoE-deficient mice. Supplying PPE or α-asarone to mice lacking apoE suppressed aorta atherogenesis induced by atherogenic diet. PPE or α-asarone diminished aorta accumulation of CD68- and/or F4/80-positive macrophages induced by atherogenic diet in apoE-deficient mice. Treatment of apoE-deficient mice with PPE and α-asarone resulted in a significant decrease in plasma cholesteryl ester transfer protein level and an increase in lecithin:cholesterol acyltransferase reduced by supply of Paigen's diet. Supplementation of PPE and α-asarone enhanced the transcription of hepatic apoA1 and SR-B1 reduced by Paigen's diet in apoE-deficient mice. CONCLUSIONS: α-Asarone in PPE inhibited inflammation-associated atheroma formation and promoted hepatic HDL-C trafficking in dyslipidemic mice.
