Distribution of actin and tropomyosin in Cryptosporidium muris (쥐와포자충에서 acin과 tropomyosin의 분포)

  • Actin and tropomyosin of Cryptosporidium muris were localized by immunogold labeling. Two kinds of antibodies for actin labeling were used. The polyclonal antibody to skeletal muscle (chicken back muscle) actin was labeled on the pellicle and cytoplasmic vacuoles of parasites. The feeder organelle has showed a small amount of polyclonal actin antibody labeling as well. Whereas the monoclonal antibody to smooth muscle (chicken gizzard muscle) actin was chiefly labeled on the filamentous cytoplasm of parasites. The apical portion of host gastric epithelial cell cytoplasm was also labeled by smooth muscle actin together. The polyclonal antibody to tropomyosin was much more labeled at C. muris than host cells, so it could be easily identified even with low magnification (${\times}2,000$). The tropomyosin was observed along the pellicle, cytoplasmic vacuoles, and around the nucleus also. The skeletal muscle type actin seems to play a role in various celluar functions with tropomyosin in C. muris; on the other hand, the smooth muscle type actin was located mainly on the filamentous cytoplasm and supported the parasites firm attachment to host cells. Tropomyosin on the pellicle was thought to be able to stimulate the host as a major antigen through continuous shedding out by the escape of sporozoites or merozoites from their mother cells.

Studies on the Light Organ of the Firefly, Luciola lateralis Motschulsky (애반딧불이의 발광기관 구조)

  • Studies were carried out to investigate structural characteristics of the larval and adult light organs of Luciola lateralis Mot. and to observe the relation between the light organ and nerve. The larval light organs, (LLO)existed at a paired dorsal lateral positions of the 8th abdominal segment. The organ was spherical or sub-spherical in shape. There were many vacuoles around the LLO. As larva grew, the number of vacuoles increased. LLO had the muscles in its interior part and their role seemed to fix the LLO position in space. Also, there were the tracheae and tracheoles in LLO. The Adult light organs (ALO) were at the ventral portions of the 5th and 6th abdominal segments in the male, but only on the 5th abdominal segment in the female. ALO had two functional layers, i.e., photocyte and dorsal layer. Tracheal end organs existed in both layers but their arrangements were irregular. Rod-shaped photocytes and spherical photocytes were observed in the ALO of male and female, but the rod-shaped types were rarely found in the female. In the ALO of the 5th abdominal segment, two paired peripheral nerves were originated from the anterior part of the last abdominal compound ganglion. A pair of peripheral nerves were originated from the posterior part of the compound ganglion to innervate the 6th abdominal segment ALO. And LLO was innervated by a paired peripheral nerves from the last abdominal ganglion.

Ultrastructural Study of the Effect of Activated Carbon Treatment on the Mouse Kidney Treated with Lead (납 투여된 마우스의 신장에서 활성탄 처리 효과에 대한 전자현미경적 연구)

  • For investigation of the activated carbon on the mouse kidney treated with Pb, the activated carbon (40 mg/kg) and Pb (30 mg/kg) were treated orally for three and six weeks, respectivelly, and observed by the electron microscope. On the glomerulus of the group with only Pb, the basal membrane thicked, projected, and the foot processes fused. On the proximal convoluted tubules, the number of microvilli were decreased and the number of vacuoles and lysosome increased on the cytoplasm. The mitochondria and endoplasmic reticulum were extended and ribosomes dropped from the ER. On the giomerulus of the group with Pb-activated carbon, the basal membrane and the foot processes were merely changed. On the proximal convoluted tubules, the shapes and number of microvilli were not changed and the number of vacuoles, microbodies, and lysosomes decreased. The shapes of mitochondria and endoplasmic reticulum observed almost similar with control group. Th at is, elongated mitochondria and attached ribosomes to endoplasmic reticulum. As result, the activated carbon has positive effect on reducing toxicity of lead in the mouse kidney in the view of electron microscope.

  • Fine structures of retina of an ommatidium in dragonfly, having eyes of closed rhabdom type, were studied under light and electron microscopes. The ommatidia consisted of eight retinular cells distributed in a circular pattern and the retinular layer in turn can be divided into three sublayers according to the number of cells in the retina. Each retinular cell has different starting points in the retina and the length of retinular cells is varied greatly; the length of one distal retinular cell shows one half of that of others. In the middle layer, three proximal retinular cells interconnect the adjacent two rhabdoms which are triangular in the appearence of the cross section which in turn consisted of tubular, parallel and lamellated microvilli. The rhabdom is formed by three rhabdomeres, each of which is separated by $120^{\circ}$ between them, but they can be distinguished into two parts according to electron density. Around the outer part of microvilli composing rhabdom, electron density was much less than the inner part of the structure. The microvilli of the inner part appear to be connected to the cytoplasm of retinular cells. Rough endoplsmic reticulum with enlarged cisternae runs through the vacuoles in the outer part of distal retinular cells. Abundant mitochondria concentrated in the vicinity of rhabdom are found at the central part of the retinular cells, while in the area of immediate vicinity of the rhabdom, prominent vacuoles are observed. Above the rhabdom of an ommtidium stands a crystalline cone which is consisted of four cone cells arranged radially along the axis. The crystalline cone is surrounded by cells containing pigment granules. The outermost photoreceptor element of an ommatidium is corneal lens.

Factors Indicating Culture Status During Cultivation of Spirulina (Arthrospira) platensis

  • Factors indicating culture status of two Spirulina platensis strains were monitored in a batch mode cultivation for 36 days. Changing mode in all factors showed a common turning point, indicating shift of cell or culture status. Mean biomass productivity was highly sustained until day 22, chlorophyll a concentration peaked on day 22, pH value was > 12 on day 22, coil number was abruptly shortened on day 22, and floating activity was sustained at greater than 79% after day 22, indicating that day 22 is a criterion reflecting phase-transfer in cell physiology in a batch culture system. Many of these changes may have been caused by increased pH, suggesting that pH control is essential for mass production of S. platensis. Fluctuations in floating activity were likely induced by the number of cellular gas vacuoles. Consequently, coil number per trichome and floating activity of S. platensis could readily act as simple indicators for determination of culture status or harvesting time of cells.

Modification of Retinal Function by Hypothermia and Hyperthermia

  • Temperature-dependent electroretinogram responses were investigated in the dark adapted bullfrog eyes within the physiological temperature range 0-40$\^{C}$. In hypothermic process(25→0→25$\^{C}$), the amplitude of b-and c-wave decreased with lowering the temperature again. Both b-wave amplitude and threshold responses were maximal around 15$\^{C}$ during the temperature increment. Upon warming to room temperature again (25$\^{C}$), the b-wave amplitude was approximately doubled as compared to that of control without temperature changes. During the hyperthermic process (25→40→25$\^{C}$), however, the responses decreased with warming, and the wave amplitude failed to recover by cooling to 25$\^{C}$ again. As describe above, the recoveries of ERG in both processes show the striking difference. The hypothermia induces the amplification of the b-wave, that is, enhances the retinal function with the temperature recovery toward room temperature. While the hypertherima produces the decrease of the b-wave even though recovered to room temperature, which indicates an irreversible retina. The morphological alteration is shown both hypothermic and hyperthermic process, such as an appearance of large vacuoles and degenerating outer segments, more intense in hyperthermia, similar to light induced damage.

  • Serous otitis media is closely related with auditory tube function, but its etiology and pathogenesis are not clearly defined yet. So we tried to prove the theory of hydrops ex vacuo via the experimental study with cats by means of obstructing the pharyngeal orfice of the auditory tube and observe the serial changes in tympanic mucosa through light and electron microscopy. The results are as follows; 1. We confirmed the production of serous otitis media with auditory tube obstruction and have a new understanding of auditory tube function in middle ear aeration. 2. The effusion in serous otitis media was produced from the next day of experiment and increased till the fourteenth day, but decreased after the spontaneous perforation of ear drum. 3. Through the light microscopy, we observed the increasement of the sercretory cells including goblet cells, epithelial hyperplasia, capillary proliferation and invasion of inflammatory cells. 4. Through the elctron microscopy, we observed the protrusion of secretory cells, blobs in cilia, loss of cilia, increasement of vesicles, vacuoles and dense bodies in ciliated cells and Invasion of inflammatory cells. With above results, we concluded that aeration through auditory tube is the most important factor in serous otitis media and presumed the effusion was secreted by secretory cells.

  • Ultrastructures of the large and elongated cells (idioblasts) in the haustorium of a parasitic angiosperm, Cuscuta australis R. Brown growing on the host plant, Trifolium repens L. were investigated by the electron microscopy. The idioblasts were characterized by the presence of a large nucleus, small vacuoles, and dense cytoplasm including a number of various cell organelles such as ribosome, rough endoplasmic reticulum(r-ER), mitochondrion, dictyosome, proplastid, multilamellar structure(MLS), microfilament bundle(MFB), and cytosegresome. Therefore, it is suggested that the idioblasts are metabolical1y very active. Particularly, MLS, MFB, and cytosegresome observed in this study did not appear in the haustorial cells of the other parasitic angiosperms. MLS was transformed into vacuole and also incorporated with cell wall. MFB composed of microfilaments, about each 7.5 nm in diameter, was observed in nucleus and also cytoplasm. Some types of MFB were distinguished on the basis of arrangement of microfilaments. A part of cytoplasm sequestered by stacked cisternae of smooth ER(s-ER), cytosegresome, was altered into a vacuole which was formed by digestion of the sequestered cytoplasm and of cisternae of s-ER. Cell organelles such as MLS, MFB, and cytosegresome were discussed in relation to the metabolic control of the idioblasts.

Differential Diagnosis of Ovarian Mucinous, Serous, and Endometrioid Adenocarcinoma in Peritoneal Washing Cytology (복막세척액 세포검사에서 난소의 점액성, 장액성 및 자궁내막양 선암종의 감별진단)

  • This study presents the cytologic features of peritoneal washings, with particular emphasis on the cytologic discrimination among serous, mucinous, and endometrioid adenocarcinoma of the ovary. We selected histologically confirmed 27 cases of peritoneal washing : 8 cases of serous cystadenocarcinomas, 5 cases of mucinous cystadenocarcinomas, and 14 cases of endometrioid adenocarcinomas. The most frequent cytologic pattern of three tumors was clusters. Ball pattern was found in serous cystadenocarcinoma(36%) and acinar pattern in endometrioid adenocarcinoma(36%). Mucinous adenocarcinoma showed mucoid background(100%) and endometrioid adenocarcinoma revealed inflammatory background(43%). The cytoplasmic vacuoles were noted in 80%, 13%, and 43% of mucinous, serous, and endometriold adenocarcinoma, respectively. The endometrioid adenocarcinoma showed prominent nucleoli(64%). In conclusion, the cytologic findings of mucinous cystadenocarcinoma were different from that of serous and endometrioid carcinomas, such as mucoid background, abundant cyytoplasm with vacuolated cytoplasm, and peripherally located cytoplasm. Although endometriold carcinoma showed acinar pattern and prominent nucleoli, the differential diagnosis between serous cystadenocarcinoma and endometrioid adenocarcinoma in peritoneal washing cytology was was always possible.

Effects of copper sulfate poisoning on the fine structure of rat liver (유산동(硫酸銅) 중독(中毒)이 랫트 간장(肝臟)의 미세구조(微細構造)에 미치는 영향(影響))

  • This study was designed in order to investigate the effect of copper sulfate to the ultrastructural changes of the hepatocytes in Sprague Dawley rats. The animals were administered with copper sulfate (10mg/kg B.W.), which was dissolved in normal saline. The solution was injected into abdominal cavity every day. The animals were sacrificed at the 6th, 12th, and 24th day from the beginning of administration. The specimens obtained from the liver were observed with electron microscope and significant changes were as follows. 1. A prominent dilatation and disruption of the cisternae of rough endoplasmic reticulum were recognized. Also, the detachment of membrane bound ribosomes was shown. 2. The proliferation of smooth endoplasmic reticulum and the depletion of glycogen particles were noted. 3. The increase of primary lysosomes and autophagic vacuoles was obserbed. 4. The dilatation of mitochondrial cristae was obserbed. And it was irregulary scattered in the stroma of mitochondria. 5. The atrophy of microvilli in the bile canaliculi and space of Disse was prominent. 6. Membrane of hepatocytes was damaged and significant hydrophic degeneration was obserbed in the perisinusoidal regions. 7. The damage of Fat-storing cells was more significant than that of hepatocytes.
