Effects of Bisphenol A on Vitellogenin Synthesis in the Hepatocyte Primary Culture in the Olive Flounder, Paralichthys olivaceus (넙치 Paralichthys olivaceus 초대 배양 간세포의 Vitellogenin합성에 미치는 Bisphenol A의 영향)

  • The effects of bisphenol-A(BPA), a monomer of plastics used in many consumer products, on vitellogenin(VTG) synthesis were examined in primary hepatocyte culture of olive flounder, Paralichthys olivaceus. The hepatocytes were precultured for 2 days, and then estradiol-$17\beta(10^{-6}M)$ and BPA were simultaneously added to the incubation medium. The hepatocytes were cultured for 6 more days and then spent medium was analyzed by SDS-PAGE. BPA increased the rate of VTG to total protein concentrations in a concentration-dependent way, and a significant difference was obtained at concentrations of $10^{-6}M$ and $10^{-5}M$ (P<0.05). In particular, the rate of VTG to total protein concentrations was 26.36% at $10^{-5}M$ of BPA, and its level did not differ from the control level with $E_2$ alone. $E_2$ and/or BPA-primed VTG synthesis was markedly inhibited to about 80% of the control(with $E_2$) by the addition of tamoxifen($10^{-6}M$) to the incubation medium. Furthermore, In vivo $E_2$-primed VTG synthesis was significantly inhibited by in vitro $E_2$-free incubation of hepatocyte to about 22% of the control (with $E_2$) on Day 6. The effect of reducing was delayed in a BPA concentration-dependent way. These results suggest that BPA induce VTG synthesis by estrogenic activity through estrogen receptor mediated response and prolong VTG synthesis on vitellogenesis in olive flounder.

  • Bisphenol A (BPA), nonylphenol (NP), and 4-tert-octylphenol (OP) are known endocrine disrupting chemicals (EDCs) with estrogenic activity in fish. This study compared the effects of BPA, NP and OP on in vitro vitellogenin (VTG) synthesis in primary cultures of hepatocytes of the Chinese minnow Phoxinus oxycephalus. The VTG secreted into the culture medium was measured using enzyme-linked immunosorbent assay (ELISA), which we developed in this study using an antibody prepared from homogenates of Chinese minnow egg. VTG synthesis was induced by estradiol-17$\beta$ ($E_2$) and phenols (BPA, NP and OP) treatment. $E_2$ at concentrations of 10$^{-6}$ M or higher increased VTG levels significantly (P < 0.05). Exposure to 10^5\;M\;BPA\;or\;10^-4$M NP and OPinduced in vitro VTG synthesis (P < 0.01). However, $10^-3$ M BPA, NP or OP did not induce VTG synthesis. These results suggest that SPA has the highest estrogenic potential in Chinese minnow hepatocytes. Tamoxifen, an anti-estrogen, drastically blocked the production of VTG by phenols (BPA, NP and OP) suggesting that phenols (BPA, NP and OP) may act via binding to estrogen receptor (ER) in Chinese minnow hepatocytes.

  • Effects of bisphenol-A (BPA) on vitellogenin (VTG) synthesis and estrogen-estrogen receptor $(E_2- ER)$ binding activity were examined in primary hepatocyte cultures of rainbow trout, Oncorhynchus mykiss. Hepatocytes were precultured for 2 days and then $estradiol-17\beta\;(E_2,\;2\times10^{-6}M)\;BPA\;(10^{-5}-10^{-8}M)$ and/or 4-hydroxy-tamoxifen $(4-OHT,\;10^{-6} M)$ were simultaneously added to the incubation medium. Hepatocytes were cultured for 5 more days and then spent medium was analyzed by SDS-PAGE for VTG production. The addition of BPA to the incubation medium had no effect on the viability of hepatocytes in the culture. On the other hand, BPA increased VTG production in a concentration-dependent way and a significant increment occurred at BPA concentrations greater than $10^{-6}$M. Although VTG was increased by the addition of $E_2\;(2\times10^{-6}\;M)\;or\;BPA\;(10^{-5}M)$, its were reduced by a simultaneous 4-OHT $(10^{-6}\;M)$ addition. BPA inhibited $E_2-human$ ER binding activity by $72\%$ at $10^{-5}$ M of BPA. These results suggested that BPA induced VTG synthesis by BPA-ER binding activity in the hepatocyte of rainbow trout.

  • In teleost, vitellogenin (Vtg) is synthesised in the liver in response to estrogens and transported by the blood to the growing oocytes where it is incorporated by micropincytosis (Selman and Wallace, 1982).   • Effects of Estradiol-$17\beta(E_2)$ and 2, 4-dichlorophenoxy acetic acid (2, 4-D) on vitellogenin(VTG) production were investigated in primary hepatocyte culture of olive flounder, Paralichthys olivaceus. Highest survival rate of hepatocyte were observed at $27^{\circ}C$, which markedly declined equal to 50% of those of $15^{\circ}C$. Vitellogenin production peaked at the concentration of $10^{-6}M\;E_2$. No effect was observed on VTG production at various concentrations of 2, 4-D. However, a low concentration of 2, 4-D (ie, $10^{-8}M$) only appeared increased VTG production. $E_2$ or $10^{-8}M$ 2, 4-D-primed VTG production was markedly inhibited by the addition of $10^{-6}M$ tamoxifen to the culture medium(P<0.01). Inhibition was not affected by combinational treatment with $10^{-6}M$ $E_2$ and $10^{-6}M$ 2, 4-D. These results from the current investigation suggest that 2, 4-D mimics $E_2$, but the mechanism of reaction in inducing the $E_2$ receptor are different in VTG production in oliver flounder hepatocytes.

  • Effects of A23187 on estradiol$-17^{\beta}$-induced vitellogenin (VTG) induction were electrophore-tically examined in primary hepatocyte cultures in rainbow trout. hepatocytes were predultured for 2 days and then estradiol-$17^{\beta}$(E2, $2{\times}10^{-6}$M) and calcium ionophore (A23187, $10^{-7)$~$10^{-5}$ M) were added to the incubation medium. The hepatocytes were cultured for 7 more days. In addition, effects of A23187 on $E_2$-primed VTG production were investigated for 7 days. The addition of A23187 ($10^{-7)$~$10^{-5}$M) to the incubation medium specifically reduced VTG production by hepatocytes in a concentration-dependent way. The addition of A23187 significantly reduced the rate of $E_2$-primed VTG production to 18% of the control (E2 only) on Day 7. However, $E_2$-primed VTG production was reduced to 47% of the control by withdrawal of $E_2$ from the incubation medium. Therefore, these results suggest that intracellualr sequestered calcium could regulate VTG synthesis at the translational and/or post-translational stage.

Immunochemical Properties of Vitellogenins and Egg Yolk Proteins in Female Fusilier, Caesio diagramma (농어목 어류, Caesio diagramma의 vitellogenin과 난황단백의 면역화학적 특성)

  • This study was conducted to compare the immunochemical properties of female-specific serum proteins (vitellogenin, VTG) and egg yolk proteins in female fusilier, Caesio diagramma. VTG of fusilier was identified and characterized by using immunochemical analysis. Two types of VTG (VTG1 and VTG2) reacted clearly with antiserum against egg proteins, were confirmed in the serum of mature female. The results of sephacryl S-300 showed that the molecular weights of VTG1 and VTG2 were 560,000 and 410,000, respectively. Yolk proteins, E2 and E3, were isolated from egg extracts, and molecular weights of them were estimated 410,000 and 170,000, respectively. The treatment of $17\beta$-estradiol ($E_2$) to males has induced the synthesis of VTG of which immunological characteristics seems to be similar to the yolk proteins. The results suggest that VTG can be synthesized in the liver by the action of $E_2$ stimulation, and incorporated into the oocytes through the blood circulation. The level of serum $E_2$ was moderately high throughout the spawning period of June. The level of serum VTG was also sustained at high in May and June. The concentration changes of serum $E_2$ and VTG were corelated to the ovarian development in female fusilier. The results indicated that $E_2$ may have some important roles for the vitellogenesis in female fusilier. Also) the VTG can be a precursor protein of yolk not only because it could be synthesized in the liver then incorporated into the oocytes but also because an egg yolk protein had the similiar molecular weights and antigenecity with VTG.

  • Effect of 2,4-Dichlorophenoxy acetic acid (2,4-D) on vitellogenin (VTG) production and estrogen ($E_2$)-estrogen receptor (ER) binding affinity were examined in primary hepatocyte cultures of rainbow trout, Oncorhynchus mykiss. Hepatocytes were pre-cultured for 2 days; subsequently, $E_2$( 2$\times$$10^{-6}$/ M) and 2,4-D ($10^{-9}~10^{-6}/M$) were simultaneously added to the incubation medium. They were cultured for more than 5 days. VTG and $E_2$-ER binding affinities were analyzed by SDS-PAGE and ELISA, respectively. 2,4-D concentration used had no appreciable effect on the morphology, viability, and DNA content of hepatocytes in culture. It had also no effect on VTG production. However, it interfered with $E_2$-ER binding affinity, which was reduced with increasing concentration of 2,4-D. The affinity was inhibited by 25 and 30% at $10^{-7}$ M and $10^{-6}$ M of 2,4-D, respectively. This result suggested that although 2,4-D had no effect on VTG production, it acted as reno-estrogenic contaminant in ER.

Time-dependent Effects of Bisphenol Analogs on Ecdysteroid Pathway Related Genes in the Brackish Water Flea Diaphanosoma celebensis (Bisphenol 구조 유사체가 기수산 물벼룩 Ecdysteroid 경로에 미치는 영향)

  • Bisphenol A is a representative endocrine disruptor and continuously detected in aquatic environment due to wide use, resulting in adverse effects on growth, development, and reproduction in diverse organisms as well as human. Structural analogs have been developed to substitute BPA are also suspected to have endocrine disrupting effects. In the present study, the time-dependent expression patterns of ecdysteroid synthesis (nvd, cyp314a1), receptors (EcRA, EcRB, USP, ERR), and downstream signaling pathway - related genes (HR3, E75, Vtg, VtgR) were investigated using quantitative real time reverse transcription polymerase chain reaction (qRT-PCR) in the brackish water flea Diaphanosoma celebensis exposed to Bisphenol analogs (BPs; BPA, BPF, and BPS) for 6, 12, and 24 h. As results, the expression of nvd, cyp314a1, EcRs, USP, ERR and E75 mRNA was upregulated at 6 h exposure to BPF, which is earlier than BPA and BPS (12 h). On the other hand, HR3, E75 and VtgR mRNA levels were elevated at 6 h earlier at BPS and BPF than at BPA (12 h), but Vtg mRNA level was slightly changed within 24 h. These findings suggest that like BPA, BPF and BPS can also modulate the transcription of ecdysteroid pathway - related genes with different mechanisms, and have a potential as endocrine disruptors. This study will provide a better understanding the molecular mode of action of bisphenols on ecdysteroid pathway in the brackish water flea.
