A PCR-aided monitoring of verotoxin-producing Escherichia coli (VTEC) was performed over the period of 12 months by using fresh feces collected monthly from 5 dairy cows that had been identified as VTEC carriers. The PCR products were confirmed to be verotoxin genes by Southern hybridization using a gene fragment of verotoxin 2 as a probe. Although seasonal variation of VTEC shedding seemed to depend on each cow, several factors may have influenced the frequency of detection. Shedding of VTEC tended to be reduced during grazing from the middle of May up to the beginning of October. Only one cow was positive for VTEC in August. Dry-off was also suggested to have a depressive effect on VTEC shedding, i.e. 3 of 4 dry cows showed no shedding of VTEC. Contrary to these factors, winter or indoor rearing tended to increase VTEC with only 5/24 samples being negative during the period from November to April. Total VFA concentration was higher (p<0.05) in VTEC-positive feces than in VTEC-negative feces, while fecal pH and VFA proportions were not different. Partial sequences of verotoxin genes from feces of 4 VTEC-positive cows were nearly identical (99-100%), suggesting that gut bacteria sharing the same gene were distributed among the cows. The present results indicate that grazing and dry-off could be factors which reduce VTEC shedding, while winter/indoor rearing may be a factor which increases the shedding, possibly through on-farm interactions.

Seventy-six animals including cattle, sheep, horses, 6 species of zoo animals were employed for collection of fresh feces in order to detect verotoxigenic Esherichia coli (VTEC) by safe, quick and sensitive PCR-based molecular methods. Bacterial cell disruption with bead-beating followed by bacterial DNA purification with hydroxyapatide chromatography and gel filtration allowed DNA preparation from animal feces with high recovery and purity. A mountain goat was firstly shown by PCR and sequencing to shed verotoxin 2 gene (vt2) that was used to generate vt2 probe and second primer set for nested PCR to attempt more sensitive detection. Most sensitive nested PCR revealed that 45% of tested cattle and 47% of tested zoo animals were VTEC-positive, while least sensitive normal PCR detected VTEC from none of these animals except a mountain goat. Moderately sensitive detection by PCR in combination with hybridization suggested that the VTEC density varied between the VTEC-positive cattle.
The models used for ionosphere error correction in positioning using Global Navigation Satellite System (GNSS) are representatively Klobuchar model and NeQuick model. Although these models can correct the ionosphere error in real time, the disadvantage is that the accuracy is only 50-60%. In this study, a method for polynomial modeling of Global Ionosphere Map (GIM) which provides Vertical Total Electron Content (VTEC) in grid type was studied. In consideration of Ionosphere Pierce Points (IPP) of satellites with a receivable elevation angle of 15 degrees or higher on the Korean Peninsula, the target area for model generation and provision was selected, and the VTEC at 88 GIM grid points was modeled as a polynomial. The developed VTEC polynomial model shows a data reduction rate of 72.7% compared to GIM regardless of the number of visible satellites, and a data reduction rate of more than 90% compared to the Slant Total Electron Content (STEC) polynomial model when there are more than 10 visible satellites. This VTEC polynomial model has a maximum absolute error of 2.4 Total Electron Content Unit (TECU) and a maximum relative error of 9.9% with the actual GIM. Therefore, it is expected that the amount of data can be drastically reduced by providing the predicted GIM or real-time grid type VTEC model as the parameters of the polynomial model.
Verotoxin non-producing E coli O157 strains have been isolated from cattle feces and compared in particular regard to biochemical properties and genotypes with verotoxin-producing E coli (VTEC). E coli O157 : nonH7 strains had different phenotypes in sorbitol fermentation and ${\beta}-glucuronidase$ activity from E coli O157 : H7. Regardless of verotoxin production ability of E coli O157 : H7, uidA gene was uniquely detected from sorbitol and ${\beta}-glucuronidase$ negative E coli O157 : H7. Forty five fecal samples from 6 dairy farms were obtained and VTEC was detected as 15.6% (7 strains) of the samples. Most VTEC isolates were positive for sorbitol fermentation and ${\beta}-glucuronidase$ activity but negative for eaeA gene. This study suggested that cattle could be a reservior for VTEC. However, absence of eaeA gene in VTEC isolates from most of healthy cattle suggested that they might be less virulent than eaeA-positive E coli against human health.

The objective of this study was to evaluate three verocytotoxin-producing Escherichia coli (VTEC) detection kits to detect the presence of VT genes: Doupath Verocytotoxin (GLISA) developed by MERCK, ProsPect Shiga Toxin E. coil (STEC) Microplate Assay (ELISA) developed by Remel, and a polymerase chain reaction method. Our laboratory verified artificially inoculated samples. All three methods could detect very low numbers of VTEC, but VT-PCR had the best sensitivity for VTEC detection. From April through September 2005, 257 ground-beefs from supermakets and traditional markets were examined for the presence of VTEC by polymerase chain reaction immediately after purchase and total viable counts (TVC) were determined. VTEC was isolated from 30 of 257 ground-beefs. A variety of serogroups was found, including 10 stains belonging to the virulence type EHEC, but major serogroups such as O157, O26 and O111 were nor found.
Ten isolates of Verotoxin-producing Escherichia coli (VTEC) were detected in slaughtered cattle and investigated their phenotypic characteristics and antimicrobial susceptibility. None of the isolates was positive for eae gene. Only one isolate was positive for uidA gene. Eight out of ten isolates of VTEC were originated from broker's cattle. Thus microbiological monitoring for broker farms should be performed to minimize VTEC contamination. In the antimicrobial susceptibility test, all the isolates were highly resistant to bacitracin and lincomycin whilst they are susceptible to apramycin and neomycin.

The ionospheric delay is currently one of the most significant error sources in precise GNSS surveys. The users of single-frequency receivers should apply some kind of ionospheric correction algorithms to remove or model the ionospheric delay. For real-time correction of the ionospheric delay, one can use Klobuchar or NeQuick model provided by navigation messages of GPS and Galileo, respectively. We evaluated the performance of those models by comparing their effectiveness at different seasons and latitudes. For the first test, we computed the vertical total electron content (VTEC) at the permanent GPS site SUWN for four different seasons. As the second test, we picked three sites in Korea (CHLW, SUWN, JEJU) with high, medium, and low latitudes and evaluated the dependency of VTEC on the site latitude. Computed VTEC values were compared with those from the IRI model and Global Ionosphere Maps (GIM). The root-mean-square (RMS) differences of Klobuchar and NeQuick with respect to IRI and GIM were analyzed. As a result, without regard to season and latitude, the RMS differences of NeQuick models were smaller than that of Klobuchar by about 0.01~3.50 TECU.
Surface swab samples from beef (188), pork (240) and chicken (95) carcasses were collected from slaughterhouse in Kangwon and Kyunggi areas from March through July 1996. The samples were examined on the level of E. coli biotype I relevant to fecal contamination due to unsanitary processing control and the existence of verocytotoxin-producing E. coli (VTEC). E. coli biotype I were confirmed from 38.8% of beef, 40.0% of pork, and 69.5% of chicken carcasses. Little variation was noted among three sampling points; rump, flank and neck of beef, ham, belly and jowls of pork. coli O157:H7 was only confirmed from 2 of 188 beef carcasses. E. coli biotype I. All the isolated E. coli O157 showed positive for vero cell cytotoxicity test. Isolation rate of E. coli O157 in summer was higher than in spring. In case of pork and chicken carcasses, E. coli O157 was isolated in summer only.

To quantitatively analyze the positioning error due to the ionosphere over the Korean peninsula, we created 2-dimensional ionosphere map using 44 permanent Global Positioning System(GPS) stations operated by Ministry of Land, Transport, and Maritime Affairs. We estimated Vertical Total Electron Content(VTEC) in a fine rectangular grids of $0.1^{\circ}{\times}0.1^{\circ}$ resolution. The observables we used were phase-leveled pseudoranges which are linear combinations of pseudoranges and carrier phases. VTECs were computed for five days during January 25-29, 2003 using the data from 45 permanent stations. In comparison with the Global Ionosphere Map of the Center for Orbit Determination in Europe, RMS differences were at the level of 8 TECU(TEC Unit).

Ionopheric deley is the largest error sources in GNSS positining. The single frequency receiver user needs an ionospheric model like the Klobuchar model or NeQuick model to eliminate the ionospheric error. In this study we estimated VTEC(Vertical Total Electron Content) over DAEJ station using the two models in each season. We compared the results with Global Ionosphere Maps and International Reference Ionosphere model predictions. As a result, the NeQuick model was more accurate than Klobuchar model.
