Published data have shown that the levels of vascular endothelial growth factor (VEGF) and soluble VEGF receptor-1 (sVEGFR-1) in plasma and pleural effusion might be usefulness for lung cancer diagnosis. Here, we performed a prospective study to investigate the utility of VEGF and sVEGFR-1 in bronchoalveolar lavage fluid (BALF) for differential diagnosis of primary lung cancer. A total of 56 patients with solitary pulmonary massed by chest radiograph or CT screening were enrolled in this study. BALF and plasma samples were obtained from all patients and analyzed for VEGF and sVEGFR-1 using a commercially available sandwich ELISA kit. The results showed that the levels of VEGF in BALF were significantly higher in patients with a malignant pulmonary mass compared with patients with a benign mass (P < 0.001). However, no significant difference of sVEGFR-1 in BALF was found between malignant and non-malignant groups (P = 0.43). With a cut-off value of 214 pg/ml, VEGF showed a sensitivity and specificity of 81.8% and 84.2%, respectively, in predicting the malignant nature of a solitary pulmonary mass. Our study suggests that VEGF is significantly increased in BALF among patients with lung cancer than in benign diseases. Measurement of VEGF in BALF might be helpful for differential diagnosis of primary lung cancer.
Purpose: Nodal metastasis is the main prognostic factor in the patients with oral squamous cell carcinoma (OSCC). We investigated the association between tumor-associated lymphatics and OSCC characteristics. Methods: Thirty-four specimens were used for the immunohistochemical staining with the antibody for vascular endothelial growth factor (VEGF)-C, VEGF-D, VEGF receptor (VEGFR)-3, phosphorylated VEGFR-3, D2-40, and matrix metallproteinases (MMPs). We observed the distribution of the lymphangiogenic factors and quantified the degree of expression. We determined lymphatic vessel density (LVD) and lymphatic vessel dilatation with D2-40 immunostaining. We assessed the association of LVD or lymphatic vessel dilatation with tumor progression or tumor differentiation. Results: OSCC cells expressed lymphangiogenic ligands. Lymphangiogenic receptor, VEGFR-3, was expressed and activated in some tumor cells as well as in tumor-associated endothelial cells. LVD was not associated with tumor size or nodal status, but lymphatic vessel dilatation was higher in tumors with nodal metastasis, and also higher in poorly differentiated tumors. In stromal area of OSCC, MMP-1 and MMP-10 were up-regulated and the basement membrane of tumor-associated endothelial cells was destroyed by these collagenases. Conclusion: In the primary tumors with nodal metastasis, especially in poorly differentiated OSCC, tumor cells invaded the dilated lymphatic vessels via ruptured sites. MMP-1 and MMP-10 are important in the lysis of the glycocalyx inside the tumor-associated lymphatic endothelial cells.
위암은 최근 50년간 5년 생존율에 있어서 미약하게만 상승되었다. 이에 따라 위암에 대한 분자생물학적 치료에 대한 여러 연구가 진행되고 있다. 이에 진행성, 전이성 위암에서의 분자생물학적 치료에 대해 살펴보고자 한다. 위암의 분자생물학적 치료의 타켓으로는 EGFR (Epidermal growth factor receptor), HER(Human epidermal growth factor receptor), VEGF (Vascular endothelial growth factor receptor)를 들 수 있다. 대표적으로 분자생물학적 치료 중 trastuzumab은 HER2과발현 환자에게 유용하게 사용될 수 있어, 기존 항암화학요법에 복합투여해 볼 수 있다. Trastuzumab와 그 밖의 여러 분자생물학적 제재에 대해 본 연구에서 살펴보고자 한다.
목 적: Cytokine 중의 하나인 vascular endothelial growth factor(VEGF)와 VEGF 수용체들은 다양한 생체내 조절 및 질병 상태와 연관이 있음이 알려져 있다. 본 연구는 정신분열병 환자에서 항정신병약물 치료에 따른 혈청내 자유(free) VEGF와 가용성 VEGFR-1, 가용성 VEGFR-2의 변화를 보기 위한 것이었다. 방 법: 각 환자들은 DSM-IV 진단기준에 의해 정신분열병으로 진단을 받았고, 약물투여 시작일을 기준으로 4주째 및 8주째에 추적 관찰하였다. 모두 13명이 환자군에 포함되었으며 항정신병약물 투여전과 투여후 4주째, 8주째에 각각 PANSS에 의한 상태 평가와 함께 자유 VEGF, sVEGFR-1, sVEGFR-2의 농도를 측정하였다. 13명의 정상 대조군을 환자군의 나이와 성별에 맞춰 선정하였다. 결 과: 정신분열병 환자군의 혈청 자유 VEGF($295.2{\pm}43.7$pg/ml)와 sVEGFR-2($8259{\pm}336.7$)의 농도는 정상 대조군($199.0{\pm}28.8$ 및 $8481{\pm}371.9$)과 비교하였을 때 의미있는 차이를 보이지 않았다. 그러나 sVEGFR-1의 농도($86.2{\pm}10.3$, p<0.05)는 정신분열병 환자군에서 대조군($59.0{\pm}6.4$)에 비해 의미있게 상승하였다. 정신분열병 환자군에서 항정신병약물 투여 후 자유 VEGF 농도는 4주째($338.9{\pm}56.5$)와 8주째($309.5{\pm}58.7$) 모두 투여 전과 비교하여 차이가 없었다. 그러나 sVEGFR-1 농도는 약물 치료후 8주째($57.3{\pm}6.3$, p<0.05)에 측정한 결과에서 유의하게 감소하였다. sVEGFR-2의 농도도 치료전과 비교하였을때 약물 치료후 4주째($7761{\pm}403.0$, p<0.05)와 8주째($7435{\pm}333.5$, p<0.05) 모두 유의하게 감소하였다. 결 론: sVEGFR-1과 sVEGFR-2 농도의 감소는 항정신병약물이 작용하는 도파민 신경계와 관련된 것으로 추정된다.

Tumor angiogenesis is a process leading to formation of blood vessels within tumors and is crucial for maintaining a supply of oxygen and nutrients to support tumor growth and metastasis. Vascular endothelial growth factor(VEGF) plays a key role in tumor angiogenesis including induction of endothelial cell proliferation, migration, survival and capillary tube formation. VEGF binds to two distinct receptors on endothelial cells. VEGFR-2 is considered to be the dominant signaling receptor for endothelial cell permeability, proliferation, and differentiation. Bevacizumab(Avastin, Genetech, USA) is a monoclonal antibody against vascular endothelial growth factor. It is used in the treatment of cancer, where it inhibits tumor growth by blocking the formation of new blood vessels. The goal of this study is to identify the anti-tumor effect of Bevacizumab(Avastin) for oral squamous cell carcinoma cell lines. Human squamous cell carcinoma cell line(HN4) was used in this study. We examined the sensitivity of HN4 cell line to Bevacizumab(Avastin) by using in vitro proliferation assays. The results were as follows. 1. In the result of MTT assay according to concentration of Bevacizumab(Avastin), antiproliferative effect for oral squamous cell carcinoma cell lines was observed. 2. The growth curve of cell line showed the gradual growth inhibition of oral squamous cell carcinoma cell lines after exposure of Bevacizumab(Avastin). 3. In the apoptotic index, groups inoculated Bevacizumab(Avastin) were higher than control groups. 4. In condition of serum starvation, VEGFR-2 did not show any detectable autophosphorylation, whereas the addition of VEGF activated the receptor. Suppression of phosphorylated VEGFR-2 and phosphorylated MAPK was observed following treatment with Bevacizumab(Avastin) in a dose-dependent manner. 5. In TEM view, dispersed nuclear membrane, scattered many cytoplasmic vacuoles and localized chromosomal margination after Bevacizumab(Avastin) treatment were observed. These findings suggest that Bevacizumab(Avastin) has the potential to inhibit MAPK pathway in proliferation of oral squamous cell carcinoma cell lines via inhibition of VEGF-dependent tumor growth.
Angiogenesis must be precisely controlled because uncontrolled angiogenesis is involved in aggravation of disease symptoms. Vascular endothelial growth factor (VEGF)/VEGF receptor 2 (VEGFR-2) signaling is a key pathway leading to angiogenic responses in vascular endothelial cells (ECs). Therefore, targeting VEGF/VEGFR-2 signaling may be effective at modulating angiogenesis to alleviate various disease symptoms. Oleanolic acid was verified as a VEGFR-2 binding chemical from anticancer herbs with similar binding affinity as a reference drug in the Protein Data Bank (PDB) entry 3CJG of model A coordination. Oleanolic acid effectively inhibited VEGF-induced VEGFR-2 activation and angiogenesis in HUVECs without cytotoxicity. We also verified that oleanolic acid inhibits in vivo angiogenesis during the development and the course of the retinopathy of prematurity (ROP) model in the mouse retina. Taken together, our results suggest a potential therapeutic benefit of oleanolic acid for inhibiting angiogenesis in proangiogenic diseases, including retinopathy.
배경: 혈관내피 성장인자(vascular endothelial growth factor, VEGF)는 혈관평활근세포(vascular smooth muscle cell)의 증식과 이동을 촉진함으로써 혈관신생에 중요한 역할을 한다. 당뇨병은 VEGF의 발현과 연관되어 정상 혈당상태에서 보다 세포의 증식을 더욱 촉진시킨다. 당뇨병쥐에서 VEGF 수용체의 선택적 차단이 손상된 혈관에서 신내막 형성과 혈관평활근세포의 이동에 미치는 영향에 대해 알아보고자 했다. 대상 뜻 방법: 당뇨병 쥐의 경동맥 풍선손상 모델에서 위약을 투여하거나, 혈관내피 성장 인자 수용체-1(VEGFR-1)에 선택적으로 작용하는 항-Flt-1 펩타이드(anti-Flt-1 peptide; Gly-Asn-Gln-Trp-Phe-Ile)를 풍선손상 2일 전부터 0.5mg/kg의 용량으로 2주간 매일 투여한 군으로 나누어 Hematoxylin-Eosin 염색을 하여 신내막의 형성정도와 혈관내강의 협착정도를 비교하였으며, proliferative cell nuclear antigen (PCNA)에 대한 면역조직화학염색법을 시행하여 세포의 증식정도를 관찰하였다. 혈관평활근세포를 고혈당환경에서 배양하고 transwell assay를 시행하여 혈관평활근세포의 이동 정도를 측정하였다. 고혈당 환경에서 자라고 있는 혈관평활근세포에 50ng/mL의 VEGF를 단독 또는 3ug/mL의 항-Flt-1 펩타이드와 함께 처리하고 일정시간이 지난 후 matrigel filter를 통과한 세포를 세어 아무런 처치를 받지 않은 세포가 이동한 정도와 비교하였다. 또한, 혈관평활근세포에 세포이동 정도 측정 시와 같은 처리를 한 후, RNA를 분리하고 reverse transcription-polymerase chain reaction (RT-PCR)을 시행하여 기질금속단백분해효소(matrix metalloprotenase, MMP)의 발현 정도를 관찰하였다. 결과: 신내막의 면적은 위약 투여 쥐는 $0.24{\pm}0.03 mm^2$이었으나, 항-Flt-1 펩타이드의 처리에 의해 $0.15{\pm}0.04 mm^2$로 유의하게 감소하였으며(p<0.01), 신내막 형성에 따른 내강의 협착 정도도 위약 투여 쥐는 $61.85{\pm}5.11%$, 항-Flt-1 펩타이드 투여 쥐는 $36.03{\pm}3.78%$로 항-Flt-1 펩타이드 투여에 의하여 유의하게 감소하였다(p<0.01). 신내막의 전체 세포수에 대한 PCNA(+)인 세포를 백분율로 구하였으며, 위약 투여 쥐와 항- Flt-1 펩타이드 투여 쥐에서 각각 $52.82{\pm}4.20%,\;38.11{\pm}6.89%$로 나타나 항-Flt-1 펩타이드를 투여한 쥐에서 PCNA(+)인 세포가 유의하게 적음을 보이고 있다(p<0.05). 혈관평활근세포의 이동 정도 측정에서는 항-Flt-1 펩타이드 처리에 의하여 VEGF에 의한 혈관평활근 세포의 이동이 유의하게 감소하였다(p<0.01). 또한, 항-Flt-1 펩타이드 처리에 의하여 VEGF에 의한 MMP-3와 MMP-9 mRNA의 발현 증가가 억제되었다. 결론: 항-Flt-1 펩타이드는 당뇨병쥐의 경동맥손상모델에서 신내막 형성을 억제하였으며, 고혈당 환경에서 배양된 혈관평활근세포의 이동과, MMP-3와 MMP-9의 활성을 억제하였다.
치주인대에 일회성의 적절한 인장력을 가하였을 때 견인측 치근막에서 나타나는 혈관성장인자(VEGF)와 그 수용체(VEGFR)의 발현의 변화를 보기 위해 본 연구를 시행하였다. 8-10주된 Sprague-Dawley계 웅성 백서(rat)에서 상악좌측 제1대구치에 closed coil을 이용하여 근심 방향으로의 교정력을 가하였으며 1시간, 12시간, 1일, 3일, 1주, 2주 군으로 분리하여 각 군 당 5마리씩의 실험동물을 배정하였다. 우측 제1대구치는 치경부에 ligature wire만 결찰하고 동일 실험시간이 지난 후 대조군으로 이용하였다. 견인력이 가해진 치근막에서 VEGF와 VEGFR 및 이들의 mRNA의 발현 양상의 변화를 H&E 염색 및 면역조직화학적 염색과 in situ hybridization법으로 관찰하여 다음과 같은 결과를 얻었다. 1. 치주인대에 인장력을 가하면 치주인대의 신장으로 인한 혈관의 압박에 의해 울혈과 부분적인 출혈상이 초기에 나타났으나 3일 이내에 대조군과 같은 정도로 회복되었으며 신생골의 형성은 3일 이후 나타나서 2주간 지속되었다. 2. 치주인대에 인장력을 가하면 치주인대 세포와 조골세포, 백악아세포에서의 VEGF와 VEGF mRNA의 발현증가가 나타나며 이는 치주인대 혈관의 증가로 이어졌다. 3. 인장력을 가하고 3일 이후에는 VEGF와 VEGF mRNA의 발현은 주로 치조골 인접면의 치주인대세포와 조골세포에서만 관찰되었으며 2주후에는 VEGF와 VEGF mRNA, 치주인대혈관 모두 대조군과 유사한 정도로 감소하였다. 4. VEGF 수용체인 Flt-1과 Flk-1은 거의 동일한 발현 양상을 보였으며 주로 혈관 내피세포와 조골세포에서 관찰되었으나 치주인대에 인장력을 가하면 초기에 혈관내피세포에서 그 발현이 증가하였다. 조골세포에서의 발현증가는 내피세포에 비교해서 다소 늦게 나타났으나 발현의 증가는 더 뚜렷하였다. 결론적으로 교정력을 가했을 때 견인측 치주인대의 치주인대세포와 조골세포, 백악아세포에서 VEGF와 VEGF mRNA의 발현이 증가하며 이에 이어 혈관의 증가가 나타나고 신생골의 형성은 혈관의 증가 후에 나타나는 것을 관찰할 수 있었다. 백악아세포에서 발현이 증가된 VEGF mRNA는 치조골측으로 편재해 있던 혈관이 견인의 방향으로 치아를 향해 성장하도록 유도하는 것으로 사료되었다. VEGFR 및 VEGFR mRNA는 내피세포 뿐 아니라 조골세포와 골세포, 치주인대세포에서도 발현이 증가하여 VEGF가 paracrine한 방식 뿐 아니라 autocrine한 방식으로도 작용함을 알 수 있었다.
Objective: Taspine, isolated from Radix et Rhizoma Leonticis has demosntrated potential proctiective effects against cancer. Tas13D, a novel taspine derivative synthetized by structure-based drug design, have been shown to possess interesting biological and pharmacological activities. The current study was designed to evaluate its antiproliferative activity and underlying mechanisms. Methods: Antiproliferative activity of tas13D was evaluated by xenograft in athymic mice in vivo, and by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) and cell migration assays with human liver cancer (SMMC-7721) cell lines in vitro. Docking between tas13D and VEGFR and EGFR was studied by with a Sybyl/Surflex module. VEGF and EGF and their receptor expression was determined by ELISA and real-time PCR methods, respectively. Results: Our present study showed that tas13D inhibited SMMC-7721 xenograft tumor growth, bound tightly with the active site of kinase domains of EGFR and VEGFR, and reduced SMMC-7721 cell proliferation (IC=34.7 ${\mu}mol/L$) and migration compared to negative controls. VEGF and EGF mRNAs were significantly reduced by tas13D treatment in a dose-dependent manner, along with VEGF and EGF production. Conclusion: The obtained results suggest that tas13D inhibits tumor growth and cell proliferation by inhibiting cell migration, downregulating mRNA expression of VEGF and EGF, and decreasing angiogenic factor production. Tas13D deserves further consideration as a chemotherapeutic agent.
Vascular endothelial growth factor (VEGF) plays a key role in angiogenesis through binding to its specific receptors, which mainly occurs to VEGF receptor 2 (VEGFR-2), a kinase insert domain-containing receptor. Therefore, the disruption of VEGFR-2 signaling provides a promising therapeutic approach for the treatment of cancer by inhibiting abnormal or tumorinduced angiogenesis. To explore this potential, we expressed the catalytic domain of VEGFR-2 (VEGFR-2-CD) as a soluble active kinase in Escherichia coli. The recombinant protein was purified and the VEGFR-2-CD activity was investigated. The obtained VEGFR-2-CD showed autophosphorylation activity and phosphate transfer activity comparable to the commercial enzyme. Furthermore, the IC50 value of known VEGFR-2 inhibitor was determined using the purified VEGFR-2-CD. These results indicated a possibility for functional and economical VEGFR-2-CD expression in E. coli to use for inhibitor screening.

제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
