Vascular endothelial growth factor (VEGF) is a key regulator of angiogenesis under various physiological and pathological conditions. We found that the VEGF isoforms VEGF120, VEGF164, and VEGF188 were expressed in the bovine mammary gland and bovine mammary epithelial cells (bMECs). Expression of VEGF in the mammary gland was significantly higher during the lactation period than during the dry period. Although dexamethasone or prolactin alone had little effect on the expression of VEGF, that in dexamethasone-treated cells was significantly induced after additional treatment with prolactin. Furthermore, the VEGF expression induced by the combination of dexamethasone and prolactin was reduced by PD98059 in a dose-dependent manner. This combination also stimulated the phosphorylation of p44/p42 MAP kinase in these cells. These results strongly suggest that the combination of dexamethasone and prolactin stimulates VEGF expression in bMECs via p44/p42 MAP kinase.
혈관내피세포 성장 인자(Vascular endothelial growth factor, VEGF)는 혈관내피세포 특이하게 성장요인으로 작용하는 물질로 알려져 있다. 전리방사선에 대한 혈관내피세포의 효과는 정상조직에 대한 반응에 있어 아주 중요한 요소일 것으로 생각된다. 본 연구는 방사선 조사에 의해 배양시킨 혈관내피세포에서 apoptosis가 유도되는지, 유도가 된다면 VEGF에 의해 apoptosis가 억제되는지 그리고 apoptosis의 억제가 어떤 경로를 경유하는지를 실험하였다. 혈관내피세포에 방사선를 조사한 결과, 대조군에 비하여 선량이 증가함에 따라 apoptosis가 증가하였다. 같은 조건하에서 VEGF는 농도 의존적으로 apoptosis를 억제하였다. Antiapoptotic factor로서 VEGF가 어떤 신호 과정을 경유하는지를 밝히고자, 혈관내피세포에 방사선을 조사하여 apoptosis를 유도하면서 Flt-1과 Flk-1/KDR receptor를 처리하였다. 그결과 VEGF에 유도된 apoptosis 억제효과가 차단되었다. Phosphatidylinositol 3'-kinase(PI3-kinase) 특정 억제 물질인 Wortmanin과 LY294002를 방사선 조사한 혈관내피에 VEGF와 함께 처리했을 때 VEGF에 의해 유도된 apoptosis를 억제하였다. 이같은 결과는 VEGF가 방사선 조사로 일어나는 세포 치사를 억제하는 중요한 역할을 담당하며, In Vivo의 실험이 더 이루어져야 할 것으로 생각된다.
Vascular endothelial growth factor (VEGF)-A, a major regulator for angiogenesis, binds and activates two tyrosine kinase receptors, VEGFR1 (Flt-1) and VEGFR2 (KDR/Flk-1). These receptors regulate physiological as well as pathological angiogenesis. VEGFR2 has strong tyrosine kinase activity, and transduces the major signals for angiogenesis. However, unlike other representative tyrosine kinase receptors which use the Ras pathway, VEGFR2 mostly uses the Phospholipase-$C{\gamma}$-Protein kinase-C pathway to activate MAP-kinase and DNA synthesis. VEGFR2 is a direct signal transducer for pathological angiogenesis including cancer and diabetic retinopathy, thus, VEGFR2 itself and the signaling appear to be critical targets for the suppression of these diseases. VEGFR1 plays dual role, a negative role in angiogenesis in the embryo most likely by trapping VEGF-A, and a positive role in adulthood in a tyrosine kinase-dependent manner. VEGFR1 is expressed not only in endothelial cells but also in macrophage-lineage cells, and promotes tumor growth, metastasis, and inflammation. Furthermore, a soluble form of VEGFR1 was found to be present at abnormally high levels in the serum of preeclampsia patients, and induces proteinurea and renal dysfunction. Therefore, VEGFR1 is also an important target in the treatment of human diseases. Recently, the VEGFR2-specific ligand VEGF-E (Orf-VEGF) was extensively characterized. Interestingly, the activation of VEGFR2 via VEGF-E in vivo results in a strong angiogenic response in mice with minor side effects such as inflammation compared with VEGF-A, suggesting VEGF-E to be a novel material for pro-angiogenic therapy.
Vascular endothelial growth factor (VEGF) is essential for many angiogenic processes both in normal and pathological conditions. However, the signaling pathways involved in VEGF-induced angiogenesis are incompletely understood. The protein kinase D1 (PKD1), a newly described calcium/calmodulin-dependent serine/threonine kinase, has been implicated in cell migration, proliferation and membrane trafficking. Increasing evidence suggests critical roles for PKD1-mediated signaling pathways in endothelial cells, particularly in the regulation of VEGF-induced angiogenesis. Recent studies show that class IIa histone deacetylases (HDACs) are PKD1 substrates and VEGF signal-responsive repressors of myocyte enhancer factor-2 (MEF2) transcriptional activation in endothelial cells. This review provides a guide to PKD1 signaling pathways and the direct downstream targets of PKD1 in VEGF signaling, and suggests important functions of PKD1 in angiogenesis.

VEGF는 tyrosine-kinase receptor인 VEGFR-1, VEGFR-2, VEGFR-3를 통해 그 역할을 이행하며, 이러한 VEGF-VEGFR 시스템은 암에서뿐만 아니라 비정상적인 혈관 및 림프관 형성으로 인해 야기되는 다른 질병들에 있어서도 핵심적인 요소로 각광받고 있다. 암의 측면에서 보았을 때, VEGF와 그 수용체는 종양관련 혈관 및 림프관을 형성하는 과정에서 필수적이라는 점에서 치료적인 타겟으로 이목을 끌고 있다. 때문에 암세포의 성장을 방해하기 위한 항VEGF 항체, 수용체 길항체, 수용체 기능 억제제 등과 같은 여러 가지 시도들이 있었지만, 아직까지 그 임상효과가 불확실하며 더 많은 연구들이 필요한 실정이다. 이 논문에서는 VEGF의 생리적 역할을 VEGF-A, VEGF-B, VEGF-C, VEGF-D, PLGF에 따라 나누어 설명하면서 VEGF/VEGFR 시스템의 중요성을 강조한다. VEGFR-1과 VEGFR-3은 각각 angiogenesis와 lymphangiogenesis에 핵심적인 인자이며, VEGFR-2의 경우 두 가지 모두를 일으킨다. 전반적으로 이 리뷰는 현재까지 밝혀진 암을 포함한 다양한 질병에서의 VEGF와 VEGFR의 역할에 대해 상세히 설명하고자 하였다. 이를 통해 치료 표적으로서 VEGF와 VEGFR의 활용이 더욱 촉진될 것으로 기대된다.
Vascular endothelial growth factor (VEGF)-VEGF receptor (VEGFR) system has been shown to play central roles not only in physiological angiogenesis, but also in pathological angiogenesis in diseases such as cancer. Based on these findings, a variety of anti-angiogenic drugs, including anti-VEGF antibodies and VEGFR/multi-receptor kinase inhibitors have been developed and approved for the clinical use. While the clinical efficacy of these drugs has been clearly demonstrated in cancer patients, they have not been shown to be effective in curing cancer, suggesting that further improvement in their design is necessary. Abnormal expression of an endogenous VEGF-inhibitor sFlt-1 has been shown to be involved in a variety of diseases, such as preeclampsia and aged macular degeneration. In addition, various factors modulating angiogenic processes have been recently isolated. Given this complexity then, extensive studies on the interrelationship between VEGF signals and other angiogenesis-regulatory systems will be important for developing future strategies to suppress diseases with an angiogenic component.
상용 복막투석액에 함유된 고농도의 포도당과 포도당 분해산물(glucose degradation products: CDP)이 복막의 비후, 복막 투과성의 증가 및 한외여과 부전과 같은 복막의 구조적, 기능적 변화를 초래하리라 추정되고 있다. 본 연구에서는 CDP성분이 사람 복막중피세포 활성화에 미치는 영향을 검색하였고 또 이때 ROS와 PKC가 관여하는지를 검색하였다. 혈청이 배제된 M199 배양액으로 성장을 동일화시킨 사람 복막중피세포를 GDP인 methylglyoxal(MGO), acetaldehyde, 그리고 3,4-dioxyglucosone-3-ene(3,4-DGE)으로 48시 간 동안 자극하였고, 복막의 투과성에 대한 지표로서 혈관내피성장인자(vascular endothelial growth factor VEGF)를, 섬유화의 지표로서 fibronectin과 heat shock protein 47(hsp47)의 단백을 정량하였다. 활성산소족(reactive oxygen species:ROS)과 protein kinase C(PKC)의 관여여부는 각각 항산화제 N-acetylcystein(NAC)과 PKC 억제제 calphostin C의 억제 효과로 검색하였다. MGO는 대조군과 비교하여 VEGF 분비를 1.9배, fibronectin분비를 1.5배 그리고 hsp47 표현을 1.3배로 유의하게 증가시켰다(p<0.05). MGO에 의한 VEGF 상향 조절은 calphostin C와 NAC에 의하여 유의하게 억제되었다. 사람 복막중피세포에서 VEGF 분비는 acetaldehyde에 의하여 증가하였으나 3,4-DGE에 의하여 억제되었고, fibronectin 분비와 hsp47 표현은 acetaldehyde나 3,4-DGE에 의하여 영향을 받지 않았다. 이상을 종합할 때, ROS생산과 PKC활성화가 상용투석액내 함유된 MGO에 의한 점진적인 복막의 투과성 증가, 세포외기질 축적 그리고 복막 섬유화를 유발하는 주된 신호체계로서 이를 선택적으로 억제함으로써 복막의 기능을 유지할 수 있을 것으로 생각된다.
본 연구에서는 U-373-MG 세포를 이용하여 오디 추출물과 VEGF에 의한 MMPs 및 플라스민의 분비에 미치는 영향을 확인하고자 하였다. U-373-MG 세포에 오디 추출물을 $100{\sim}600\;{\mu}g/mL$의 농도로 처리한 결과, MMP-2의 분비는 거의 완벽하게 억제되었고 MMP-9의 분비는 약 $1.6{\sim}5.9$배 증가하였다. 이에 비해 플라스민의 분비는 오디 추출물 농도 $500{\sim}600\;{\mu}g/mL$에서 $36%{\sim}58%$ 감소하였다. 오디 추출물과 VEGF를 병용처리한 경우, MMP-2의 분비는 VEGF만을 처리한 것과 비교하여 약 85.5% 감소하였고, MMP-9의 분비는 약 89.8% 감소하였다. 또한, U-373-MG 세포에서 오디 추출물($100\;{\mu}g/mL$)에 의한 MMP-2와 MMP-9의 분비 증가와 플라스민 분비증가는 PI 3'-kinase 경로에 의존적이었다. 본 연구 결과를 통해 오디 추출물이 VEGF의 과발현에 따른 암의 성장을 억제 조절하는데 이용될 수 있는 가능성을 확인하였다.
Tumor angiogenesis is a critical step f3r the growth and metastasis of solid tumors. Vascular endothelial growth factor (VEGF) is the most important angiogenic molecule associated with tumor-induced neovascularization. VEGF exerts its activity through binding to its receptor tyrosine kinase, KDR/Flk-1, expressed on the surface of endothelial cells. This study was carried out to investigate inhibitory effect of extracts from root cortex of Paeonia suffruticosa Andrews on VEGF binding to VEGF receptor. The MeOH extract from P. suffrutiocosa Andr. inhibited the binding of KDR/Flk-1-Fc to immobilized VEGF$_{165}$ more than 45% at the concentration of 100 ${\mu}$g/mL. The MeOH extract was further fractionated into n-hexane, ethyl acetate, n-BuOH, and aqueous fractions. Among the four fractions, the ethyl acetate fraction from the root cortex of P. suffruticosa Andr. exhibited highly effective inhibition (${\approx}$ 79% inhibition) and then n-BuOH fraction (${\approx}$ 45% inhibition) on the binding of KDR/Flk-1-Fc to immobilized VEGF$_{165}$ at the concentration of 100 ${\mu}$g/mL. The ethyl acetate fraction from the root cortex of P. suffruticosa Andr. more efficiently blocked VEGF-induced human umbilical vein endothelial cell proliferation, than the growth of HT1080 human fibrosarcoma. Our results suggest that P. suffruticosa Andr. may be used as a candidate fur developing anti-angiogenic agent.
당뇨병의 주된 합병증 중 하나는 새로운 혈관 생성과 신경퇴화의 특징을 보이는 증식성 망막증이다. 당뇨병에서는 고혈당증, 저산소증과 부적절한 대사조절 능력이 혈관내피성장인자(vascular endothelial growth factor, VEGF)의 발현에 중요한 요인으로 제시되고 있다. 본 연구에서는 당뇨를 유발한 흰쥐에서 당뇨성 망막증에 대한 트레드밀 운동의 효과를 알아보고자 하였다. Sprague-Dawley계 흰쥐를 대조군, 운동군, 당뇨군, 당뇨운동군으로 분류하여 각 군당 8마리씩 배정하였다. 당뇨는 streptozotocin을 50 mg/kg의 용량으로 주사하여 유발하였다. 운동군은 분당 8 m의 속도로 하루 30분씩 주 5회, 총 12주 동안 트레드밀 운동을 실시하였다. 본 연구의 결과 당뇨쥐의 망막에서 phosphoinositide 3-kinase(PI3K), phospho-protein kinase B(pAkt), hypoxia inducible factor-1α(HIF-1α), 그리고 VEGF의 발현이 증가하였다. 트레드밀 운동은 PI3K/Akt 신호전달체계를 억제하여 HIF-1α의 발현과 VEGF의 발현을 감소시켰다. 본 실험 결과 트레드밀 운동은 망막의 새로운 혈관 생성을 억제함으로써 당뇨성 망막증의 진행을 억제하는데 효과적인 방안이 될 수 있을 것으로 생각된다.
