Protective Activity against Ionizing Radiation of Antioxidative Plants Indigenous to Korea

  • We have screened the cytoprotective effect on ${\gamma}-ray$ radiation induced oxidative stress from forty one Korean plant extracts. Carpinus laxiflora (caulis), Quercus salicina (caulis), and Castanopsis cuspidata (caulis) were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of three plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by ${\gamma}-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Carpinus laxiflora, Quercus salicina, and Castanopsis cuspidata protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

Querus salicina, Clerodendron trichotomum, Lamium amplexicaule, Lozoste lancifolia and Malus baccata were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of these plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by $\gamma$-ray radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Quercus salicina, Clerodendron trichotomum, Lamium amplexicaule, Lozoste lancifolia and Malus baccata protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

천연약용식물의 활성산조종에 의한 산화적 손상의 보호 효과 (Protective Effect of Natural Medicinal Plants against Oxidative Damage Induced by Reactive Oxygen Species)

  • In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.

  • We screened the anti-oxidative effect on V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with fifteen herbal extracts. Uncariae rhynchophylla JACKS and Rheum coreanum NAKAI were found to show DPPH radical scavenge activity (25 and 29% compared to control). Rheum coreanum NAKAI and Siegesbeckia orientalis L. were shown to scavenge intracellular reactive oxygen species (57 and 55% compared to control) which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA). Rheum coreanum NAKAI which showed the most strong intracellular reactive oxygen species scavenging activity had low DPPH radical scavenging activity compared to Uncariae rhynchophylla JACKS.

Aspergillus niger 효소에 의한 길경 사포닌(플라티코딘)의 전환 및 항산화 활성 비교 (The Transformation of Saponin Platycodi Radix by Aspergillus niger and Anti-oxidation Evaluation of the Transformed Metabolites)

  • 본 연구에서는 길경을 추출, 농축, 정제하여 crude platycodin을 얻은 후, 식품 미생물을 이용하여 길경의 배당체인 platycodin의 당 사슬 부분을 일부 가수분해하여 전환할 수 있는 방법을 모색하였다. 그리고 platycodin의 전환 전과 전환 후의 세포주를 이용한 세포 독성, 항산화 활성 및 항산화 효소 활성에 대해 비교하여 보았다. Chinese Hamster lung fibroblast인 V79-4 세포 독성실험 결과, 전환 전에 비교하여 전환 후에 더 나은 세포 생존률을 보였다. 후에 진행된 DPPH 자유기 소거능을 측정실험과 lipid peroxidation 억제능을 알아보기 위하여 malondialdehyde(MDA) 양을 측정한 결과 전환 후에서 더 높은 항산화 활성이 나타나는 결과를 보였다. 따라서, 식품이나 생약 소재 배당체의 구조를 식품 미생물을 통해 안전하게 전환시키면 그에 따라 독성, 활성 등이 변화해 새로운 성질을 가진 유도체를 만들어 낼 수 있고, 이러한 전환체는 상대적으로 높은 생리활성을 가지는 것을 알 수 있었다. 따라서 이상의 결과를 종합하여 보면, 식품이나 생약 소재 배당체의 구조를 식품 미생물을 통해 안전하게 비당체로 전환시키면 그에 따라 독성, 활성 등이 변화해 새로운 성질을 가진 유도체를 만들어 낼 수 있다. 본 연구에서 살펴본 platycodin의 전환 전과 전환 후의 항산화 생리활성은 대부분이 전환 후의 platycodin 활성이 높게 나타났으며, 이는 전환체가 새로운 식품 소재로서의 가능성을 시사한다고 판단된다.

Antioxidant and Cytotoxicity Effects of Luteolin

  • Flavonoids are polyphenolic compounds that are ubiquitous in plants. They have been shown to possess a variety of biological activities, such as antioxidant and anticancer. Reactive oxygen species (ROS) lead to damages of cellular molecules and it is the one of various factors to induce cancer. The one of flavonoids, Luteolin, was found to scavenge 1,1-diphenyl-2-piculhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). Moreover luteolin showed protection on hamster lung fibroblast cells (V79-4 cell) damage induced by $H_2O_2$. And then it was investigated whether it may show cytotoxicity effect against various cancer cells by MTT, Luteolin at $10{\mu}g/ml$ showed the cell viability of 63.2%, 34.7%, 18.4% and 71.4% against NCl-H460, HeLa, U937 and MCF-7, respectively. As a result, luteolin shows more sensitive to U937 cells among the tested cancer cell lines. In summary, luteolin has antioxidant and cytotoxicity effect or various cancer cell lines.
