Bulk fill 유동성 복합레진의 변연 누출에서 다른 중합시간의 영향에 대해 마이크로시티를 이용한 평가 (Evaluation of marginal leakage of bulk fill flowable composite resin filling with different curing time using micro-computed tomography technology)

  • 목적: 중합 시간이 bulk fill 유동성 복합레진의 미세누출에 미치는 영향을 마이크로시티를 이용하여 평가하고자 하였다. 연구 재료 및 방법: 30개의 발거된 인간 대구치에 변형된 2급 와동인 vertical slot cavity를 형성하였다. SonicFill과 SureFil SDR flow을 단일 충전하였으며, 다른 중합시간(20, 40, 60초)으로 광중합 하였다. 모든 표본은 5000번의 thermal cycle을 시행하였다. 치아는 silver nitrate 용액에 24시간 동안 침전 후 8시간 동안 현상액에 침전하였다. 마이크로시티를 사용하여 시편을 촬영 후 침투된 silver nitrate solution을 3차원적으로 재건하여 그 부피를 측정하였다. 변연 누출 결과값의 통계 처리는 Mann-Whitney test를 사용하였다. 결과: 광중합 시간의 증가는 두 bulk fill 유동성 복합레진에서 변연 누출을 증가시켰다. 두 복합레진 사이 유의한 차이는 없었다. 결론: 중합 시간은 bulk fill 유동성 복합레진의 변연 누출을 증가시킬 수 있는 요인이었다. 또한 마이크로시티를 통해 레진-치아 계면에서 변연 누출의 양을 비파괴적 방법으로 3차원적으로 재건하여 측정할 수 있었다.

Strengthened and flexible pile-to-pilecap connections for integral abutment bridges

  • Pile-to-pilecap connection performance is important as Integral abutment bridges (IABs) have no expansion joints and their flexible weak-axis oriented supporting piles take the role of the expansion joint. This connection may govern the bridge strength and the performance against various lateral loads. The intention of this study is to identify crack propagation patterns when the pile-to-pilecap connection is subjected to lateral loadings and to propose novel connections for improved performance under lateral loadings. In this study, eight different types of connections were developed and modeled, using Abaqus 6.12 to evaluate performances. Three types were developed by strengthening the connections using rebar or steel tube: (i) PennDOT specification; (ii) Spiral rebar; and (iii) HSS tube. Other types were developed by softening the connections using shape modifications: (i) cylindrical hole; (ii) reduced flange; (iii) removed flange; (iv) extended hole; and (v) slot hole connection types. The connections using the PennDOT specification, HSS tube, and cylindrical hole were shown to be ineffective in the prevention of cracks, resulting in lower structural capacities under the lateral load compared to other types. The other developed connections successfully delayed or arrested the concrete crack initiations and propagations. Among the successful connection types, the spiral rebar connection allowed a relatively larger reaction force, which can damage the superstructure of the IABs. Other softened connections performed better in terms of minimized reaction forces and crack prevention.

Expression of $\beta$-Galactosidase Gene of Lactococcus lactis ssp. lactis ATCC 7962 in Lactococcus lactis ssp. lactis MG1363

  • A 4.4 kb DNA fragment encompassing lacA (galactoside acetyltransferase) and lacZ($\beta$-galactosidase) genes from Lactococus lactis ssp. lactis ATCC 7962 (L. lactis 7962) was introduced ito a Lac strain, Lactococcus lactis ssp. lactis MG1363 (L. lactis MG1363) by using a lactococcal expression vector, pMG36e and expression level of lacZ was examined. Growth rates and $\beta$-galactosidase ($\beta$-gal) activities of MG1363 cells carrying recombinant plasmid, pMLZ3, on M17 broth containing different carbon sources (1%, w/v) were examined. Contrary to the expectations, MG1363 [pMLZ3] grown on lactose showed the lowest enzyme activity (17 units) and cells grown on galactose had the highest $\beta$-gal activity (41 units). Cells grown on glucose had intermediate activity (33 units). These activities are about one tenth of the values observed in L. lactis 7962 where lacZ is present as a single-copy gene in the chromosome. When the cellular concentrations of lacZ transcript were examined using slot blot hybridization, it was found that MG1363[pMLZ3] produced sufficient amounts of transcript. These results indicate that either proteolytic degradation of $\beta$-gal or other regulatory mechanism prevent the translation or accumulation of $\beta$-gal in L. lactis MG1363 cells. In regard to regulation, the presence of the ccpA gene in L. lactis MG1363 was confirmed by Southern blot.
