The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. In order to determine whether green tea tannin ameliorates abnormal arginine metabolism as the result of excessive arginine, we have assessed the effects of the administration of green tea tannin mixture in rats treated 30 days with 2% arginine. In the arginine-treated group, the level of guanidino compounds such as arginine (Arg), guanidinoacetic acid (GAA), creatinine (Cr), methylguanidine (MG) and guanidinosuccinic acid (GSA), nitric oxide, urea, protein and glucose increased significantly in the serum, urine and kidney, whereas the oxygen species-scavenging enzymes of kidney were reduced as compared with the non-arginine-treated group. By way of contrast, the administration of green tea tannin reduced blood urea nitrogen and serum creatinine, and reduced the urinary excretion of guanidinoacetic acid, creatinine, and $NO_2^-+NO_3^-$. The increased levels of urinary urea, protein and glucose in the arginine-treated group were also lowered by the administration of green tea tannin. In these groups, the activities of superoxide dismutase and catalase in the kidney were increased, thereby suggesting the involvement of radicals in the normalizing of kidney function. These results show that the abnormal renal function induced by the adminstration of excessive arginine in rats may be restored by treatment with green tea tannin.
This study was done to investigate recovery effects of YJT, which has been used clinically in diabetes therapy. Mice were administerd Alloxan to induce diabetes. There body weight and kidney weight changes, BUN, creatinine, glucose, total protein and ALP activity in serum, urinalysis, excretion volume of Na+, K+ in urine were measured. The results were as follows: 1. Increase in body weight and kidney weight and the ratio of kidney to body weight of YJT treated group showed similar results with those of normal group. 2 The BUN, creatinine, ALP activity levels in serum of YJT treated group were similar with those of normal group. 3. The glucose level in serum of YJT treated group was better than that of control group 4. The result of the urinalysis in the group of YJT was almost same as that of control group The above results suggest that YJT partially improves the function of the kidney.
Urinary steroid levels were investigated in the treatment of CKD-501, a new anti-diabetic drug candidate. CKD-501 was administered orally at the dosage of 1, 2, 4 mg/day for 7 days to normal men (n=18). Urine was collected before, during and after stopping the drug administration and the urinary level of androgen, estrogen, progestin and corticoids were quantified using GC-MS (gas chromatography-mass spectrometry). Only urinary corticosteroid and an androgen, DHEA levels among all the analyzed steroids, have been found to increase progressively, reaching significant levels on the last day of drug treatment and later declined after the drug treatment is withdrawn. Therefore, it was thought that an increase in the urinary corticoid and DHEA levels could be a characteristic of CKD-501, since it prominently acts on the glucose sensitivity and suppresses the triglyceride levels. In conclusion, it was found that CKD-501, an anti-diabetic drug candidate, affects the glucocorticoid and DHEA levels and it plays a crucial role in glucose homeostasis.
This study was undertaken to determine if Plantaginis Semen Herbal Acupuncture(PSA) has a protective effect against glycerol-induced acute renal failure in rats. Rats were dehydrated for 24hr and then injected with 8 ml/kg of $50\%$ glycerol, one-half of dose in each hindlimb muscle. In experiments for PSA effect, rats received 0.1 ml of PSA extraction in both sides of corresponding Shenso($BL_{23}$) of human body for 3 days after injection of glycerol. The experimental group were di vided into the Normal group, the Control group, the PSA group. Glycerol injection decreased glomerular filtration rate and increased urine volume, serum creatinine, BUN level and fractional excretion of glucose, $Na^+$, $K^+$ and $CI^-$. These result show that glycerol injection result in acute renal failure. PSA significantly increased glomerular filtration rate and significantly decreased serum creatinine, BUN level and fractional excretion of glucose, $Na^+$ and $CI^-$ as compared Control group. This suggests that PSA could be used in prevention and treatment of acute renalfailure. However, the precise mechanisms of PSA protection remain to be determined.
Patients with chronic kidney disease (CKD) bear a significant financial burden and face numerous complications and higher mortality rates. The progression of CKD is associated with glomerular injury caused by glomerular hyperfiltration and oxidative stress. Factors such as uncontrolled hypertension, elevated urine protein levels, anemia, and underlying glomerular disease, contribute to CKD progression. In addition to conservative treatment, several medications are available to combat the progression of CKD to end-stage kidney disease. Renin-angiotensin-aldosterone system blockers could slow the progression of CKD by reducing glomerular hyperfiltration, lowering blood pressure, and decreasing inflammation. Mineralocorticoid receptor antagonists inhibit the mineralocorticoid receptor signaling pathway, thereby attenuating inflammation and fibrosis. Sodium-glucose cotransporter 2 inhibitors exhibit protective effects on the kidneys and against cardiovascular events. Tolvaptan, a selective vasopressin V2-receptor antagonist, decelerates the rate of increase in total kidney volume and deterioration of kidney function in patients with rapidly progressive autosomal dominant polycystic kidney disease. The protective effects of AST-120 remain controversial. Due to a lack of evidence regarding the efficacy and safety of these medications in children, it is imperative to weigh the benefits and adverse effects carefully. Further research is essential to establish the efficacy and safety profiles in pediatric populations.
Anaerobic digestion is one of the well-known methods for biological treatment handling of concentrated organic matter such as swine $wastewater.^{1)} The anaerobic digestion can reduce organic loading but also hydrolyze non-biodegradable organic $matter.^{2)}$ The feces from the scrapper-type barn are usually collected to make compost and the urine is discarded with swine-slurry wastewater by ocean-dumping or treated by biological methods. The lagoon, aerobic digestion, anaerobic digestion, SBR, $A^{2}/O$, and UCT have been applied for treating swine $wastewater.^{3)} In this study, as a result of the analysis of swine wastewater, the total and soluble chemical oxygen demand was 130g/L and 60g/L, respectively. And the volatile fatty acid as chemical oxygen demand equivalent was 45g/L, which was 75% of soluble chemical oxygen demand. Before everything else, ammonia nitrogen concentration was 6.5 g/L. From biochemical acidogenic potential test, it was concluded that the enhanced acidification process to manage swine waste should be operated in the ammonia nitrogen concentration of less than 1.2 g/L. In the result of seeding ratio experiments with artificial $wastewater^{4)}, the lag period of acidogens was taken the long time because of the inhibition by the $ammonia^{5)}$, however no difference of period by the seeding ratio was not shown. The Haldane-based biokinetics were also evaluated using a method of fourth order Runge-Kutta $approximation.^{6,7)}$ The nonlinear least squares (NLLS) method with a 95% confidence interval was also used. The ranges of maximum microbial growth rate, ${/mu_{max}}$, and half saturation coefficient, $K_{s}$, for acidogenesis of various seeding ratio with artificial wastewater were 6.1 ~ 12.6 $d^{-1}$ and 45,000 ~ 53,500 mg glucose/L, respectively. Also, the methanogenic microbial yield coefficient, Y, and microbial decay rate coefficient, $k_{d}$, and inhibition substrate concentration, $K_{si}$, for the reactors were determined to be 0.32 ~ 0.465 ${/mu}g$/mg glucose; 0.42 ~ 1.01 $d^{-1}$ and 51,500 ~ 55,600 mg glucose/L, respectively.
The effect of dietary restriction was obserbed in Albino rats. Dietary restriction was at 2 levels: 10%, 20%, 30 males and same number of females of Albino rats, aged $30{\sim}40$ days were devided into following 6 groups, 5 rats each. 1 group; rice standard group 2 group; 10% restriction of rice standard 3 group; 20% restriction of rice standard 4 group; 65% rice+35% chinese cabbage group 5 group; 10% restriction of 65% rice+35% chinese cabbage 6 group; 20% restriction of 65% rice+35% chinese cabbage The rats were kept in individual cage and given 6 different diet for 7 weeks. The results of this study were elucidated as follow. 1. Final body weight was not obserbed any significant differences between non-restriction group and 10% restriction group, but 20% restriction group revealed the decrease of body weight. 2. According to dietary restriction, final organ weight was almost decreased in the restriction group of rice+chinese cabbage, and in rice standard group, there were no significants differences between none-restriction group and 10% restriction group, but organ weight was sharply decreased such as Liver, Heart, Kidney, Adrenal, Spleen in 20% restriction group. Sex organ weight of female was increased but weight of male was decreased in 20% restriction group of rice standard group. 3. The result of biochemical analysis such as nitrogen retention, serum cholesterol, glucose of urine, feces, and serum, and lipid of feces and liver, was not revealed any significant diffences, but in 20% restriction group, urinary glucose and serum glucose levels were somewhat decreased, and liver nitrogen level was increased. 4. Therefore there is no effect on albino rats by 10% restricted diet. It can be concluded that it is possible to restrict the diet within 10%.
A 2-year-old 16-kg, intact female lindo was presented with weight loss and poor hair coat. Abnormal serum biochemical values included mild hypokalemia (3.9 mmol/L, reference range 4.37 to 5.35 mmol/L) and mild hyperglycemia (124 mg/dl, reference range 65 to 118 mg/dl). in the complete blood count and diagnostic imaging examination, abnormal changes wer not seen. The analysis of urine sample obtained from cystocentesis revealed glucosuria (> 100 mg/dl) and mild proteinuria. Repeated analysis after admission showed persistent glucosuria and hypokalemia. But blood glucose values did not exceed the renal threshold fur glucose reabsorption. To differentiate cause of the glucosuria, the glucose tolerance test and the low-dosage dexamethasone suppression test were indicated. Results of both tests were normal. In addition, the serum total thyroxine $(T_4)$ value was within normal range. The arterial blood gas analysis showed no remarkable changes. The fractional reabsorption rates of amino acids and phosphorus were calculated above $97\%$. Based on these findings, the dog was diagnosed as renal glucosuria due to proximal renal tubular dysfunction. But this persistent renal glucosuria with hypokalemia may be the initial sign of Fanconi's syndrome or proximal renal tubular acidosis.
Objectives: This study was designed to evaluate the anti-diabetic effects of Spargaium stoloniferum Buchanan-Hamilton (Sparganii Rhizoma, SR) extract on diabetic rats. Methods: Diabetes was induced with Sprague-Dawley rats by high fat/high sucrose (HF/HS) diet for 4 weeks and injection of a single low dose of streptozotocin (STZ; 35 mg/kg). SR water extract at 500 mg/kg was orally administrated once a day for 4 weeks. Body weights, food and water intakes and urine volumes were measured. The levels of glucose, insulin, total cholesterol, glutamic oxaloacetic transaminase and glutamic-pyruvic transaminase (GPT) were measured in the sera of rats. Histological changes were observed in pancreas, liver, and kidney tissues by H&E staining. Results: The administration of Sparganii Rhizoma extract at 500 mg/kg in diabetic rats did not shown a significant difference in body weight changes and GPT levels, but showed meaningful changes in an increase of urination volume, and decrease of serum glucose and insulin levels. Total cholesterol and GPT levels were also significantly decreased after SR extract administration in diabetic rats. Furthermore, the abnormal changes of pancreas, liver and kidney were also improved by Sparganii Rhizoma extract administration. Conclusions: These results indicate that SR extract can improve HF/HS-diet and STZ-induced diabetic damages in rats through inhibition of the blood glucose and insulin increase.
