Measurement of Glucose and Protein in Urine Using Absorption Spectroscopy Under the Influence of Other Substances (타 성분 영향을 고려한 요당과 요단백의 흡수분광학 진단)

  • Glucose and protein in urine are among the important substances for urine analysis and have generally been measured based on a reagent strip test. In this study, these two substances were measured using mid-infrared absorption spectroscopy. Samples were prepared from a commercial synthetic urine product. Glucose and albumin were added as well as red blood cells, which are expected to create the most spectroscopic interference of any substance. Concentrations of these substances were varied independently. Optimal wavelength regions were determined from a partial least squares regression analysis (glucose 980 - 1150/cm, albumin 1400 - 1570/cm). Interference by other substances increased the differences between measured and predicted values. Albumin measurement in particular weres heavily influenced by the presence of glucose and red blood cells. Depending on the inference by other substances, measurement errors were 29.85${\sim}$45.19 mg/dl for a glucose level between 0 and 1000 mg/dl and 14.0${\sim}$93.11 mg/dl for an albumin level of 0 ${\sim}$ 500 mg/dl. Our study proposes an alternative to the chemical test-strip analysis, which shows only discrete concentration levels.

Diagnostic ex vivo assay of glucose in live cell using voltammetry

  • The hand held voltammetry systems searched diabetic assay using glucose sensor of fluorine nafion doped carbon nanotube electrode (FCNE). An inexpensive graphite carbon pencil was used as an Ag/AgCl reference and Pt counter electrode. Upon combining and using three electrode systems, optimum square wave (SW) stripping results were attained to 1.0-9.0 ug/L with 8 points. Statistic RSD precision was of 6.02 % with n=15 in 0.1 mg/L glucose. After a total of 200 second accumulation times, analytical detection limit of 0.8 ug/L was obtained. This developed technique was applied to urine samples from diabetic patients urine for fluid analysis, it was determined that the sensor can be used with a diagnostics in the ex vivo of live cells and non treated biological fluid.

Hypoglycemic Effects of Fermented Chaga Mushroom (Inonotus obliquus) in the Diabetic Otsuka Long-Evans Tokushima Fatty (OLETF) Rat

  • Changes in the levels of analytes in the blood and urine of a rodent animal model were taken as a measure of the hypoglycemic effects of a diet containing fermented chaga mushroom. These studies were conducted using the genetically manipulated diabetic Otsuka Long-Evans Tokushima Fatty (OLETF) rat. The effects of 8-week long diets that included either fermented (FCM) or non-fermented (CM) chaga mushroom powder (5% in the diet) on the OLETF rat were compared to the normal diet fed OLETF rat and the non-diabetic Long-Evans Tokushima Otsuka (LETO) rat. Hypoglycemia was tracked by measuring serum and urine concentrations of glucose, insulin, fructosamine, and leptin. Serum and urine levels of glucose, fructosamine, and leptin in the OLETF rats were higher than in LETO rats when fed normal diets but insulin levels did not differ between the two animal groups. The FCM rats were characterized by dramatically low levels of serum glucose and leptin in the OLETF rats whereas the levels of fructosamine and urine glucose trended lower in response to FCM. The serum leptin level in the CM-fed OLETF rat was also lower than that in the normal diet fed OLETF control. Serum concentrations of insulin in the OLETF rats were higher following FCM or CM feeding compared to the normal diet. These observations imply that (a) a dietary supplement of fermented chaga mushroom may contribute to a hypoglycemic effect in the OLETF rat, and (b) the increased blood insulin concentration following 8 weeks of an FCM diet may be important to the noted improvement in hyperglycemia.

Effects of Okchunsan and Spinacia oleracea L.(Semen) on streptozotoicin-induced Diabetic Rats (실험적(實驗的) 당뇨병(糖尿病)에 대(對)한 옥천산(玉泉散) 및 파채자의 효과(效果))

  • In order to investigate the effects of Okchunsan (Sample A), Gamiokchunsan (Sample B) and Spinacia oleracea L. (semen: Sample C) on streptozotocin-induced diabetic rats, the experimal study was performed on the levels of glucose, insulin, total cholesterol, triglyceride, total protein, albumin, blood urea nitrogen (BUN), electrolytes $(Na^+,\;Ka^+,\;Cl^-)$, GOT and GPT in serum, hemoglobin, hematocrit, urine volume, glucose and protein in urine, body weight, liver and kidney weight. The results were obtained as follows: 1. Serum glucose levels were significantly decreased in Sample A. B and C group and serum insulin levels were increaseclin Sample A and B group. 2. Serum total cholesterol levels were recovered as the normal value in Sample A, B and C group and serum triglyceride levels were decreased in Sample A and C group. 3. Serum total protein levels were increased in Sample C group, BUN levels were decreased in Sample A and C group. 4. Serum $Na^+\;and\;Cl^-$ levels were increased, $K^+$ levels were decreased in Sample A, B and C group. 5. Serum GOT and GPT levels were remarkedly decreased in Sample A, B and C group. 6. Urine volume, urine glucose and protein levels were decreased in Sample A, B and C levles. 7. Body Weight showed a tendancy of increasing in Sample A, B and C group. 8. Levels of serum albumin, hemoglobin, nematocrit, liver and Kidney weight were not significantly changed. According to above results, Okchunsan, Gamiokchunsan and spinacia oleracea L. (Semen) were expected to be applied to the treatment of diabetes mellitus and its complications.

Development of Urine Strip for Detection of Leukocytes in Urine using Peroxidase (과산화효소를 이용한 백혈구 측정용 뇨 검사지 제조에 관한 연구)

  • A new test strip to detect leukocytes using the myeloperoxidase in urine was developed. The reagent strip contains tetramethylbenzine, glucose and glucose oxidase. The detection limit was between 10 cells per 1$\mu$l urine(5 cells/hpf), showing greenish yellow color in the range of 10-25 cells/$\mu$l, green color in the range of 75-250 cells/$\mu$l, greenish blue color in the range of 500 cells/$\mu$l. The result can be obtained within two minute. The performance of the new method was evaluated by comparing the results of microscopic examination and other commercial products. Good correlations were shown between the values obtained by our urine strip and those by other commercial products with 172 urine samples. The results were proven that new methods were useful as primary screening reagents to detect leukocytes in urine.

  • This present study was carried out to investigate the effect of Saengjingamrotang and Saengjingamrotang plus Radix Trichosanthis in streptozotocin induced hyperglycemic rats. Body weight, serum levels of glucose insulin total cholesterol triglyceride and urine levels of volume glucose protein were measured in streptozotocin induced hyperglycemic rats orally receiving extracts of Saengjingamrotang and Saengjingamrotang plus Radix Trichosanthis for 4 days. According to above mentioned results, Saengjingamrotang and Saengjingamrotang plus Radix Trichosanthis have the effect of decreased blood sugar, serum lipid levels and urine volume, protein, glucose in streptozotocin induced hyperglycemic rats, and so they were expected to be appled to the treatment of diabetes mellitus.

  • This present study was carried out to investigate the effect of Chonhwasan and Okchunsan in streptozotocin induced hyperglycemic rats. Body weight, serum levels of glucose, insulin, total cholesterol triglyceride and urine levels of volume, glucose, protein were measured in streptozotocin induced hyperglycemic rats orally receiving extracts of Chonhwasan and Okchunsan plus Radix Trichosandis for 4 days. According to the result, Chonhwasan and Okchunsan have the effect of decreased body weight delate and decreased blood sugar, serum lipid metabolism and urine volume, glucose, protein in streptozotocine induced hyperglycemic rats, and they were expected to be applied to the treatment of Diabetes Mellitus.

  • For ex-vivo diabetic control, the voltammetric diagnosis of glucose (GU) was conducted with a modified carbon nanotube paste electrode, using handheld analytical circuits. The optimum analytical conditions were attained within the 0.5-4.0 ug/L working range and at the 0.06 ug/L detection limit, which system was interfaced to the feedback circuits and was applied to human urine for diabetic-patient diagnosis. It can be used for ex-vivo flow control analysis, vascular flow detection, and other medicinal assays.

Metabolism of $^{14}C$ Glycine-Glucose Melanoidin and Soybean Sauce ($^{14}C$ Glycine-Glucose Melanoidin 과 양조간장의 대사)

  • The metabolic transit of three samples( 14C glycine-glyucose melanoidin, glycine-glucose melanoidin and soybean sauce ) were studied on rats. The radioactivity of various organs and excreta intubated 14C glycine-glucose melanoidin on rats at various intervals(1, 3, 8, 12, 24, 48, 72 and 96 hrs were detected . And the brown pigment contents and hydrogen donating activities in the excreta which is obtained from three samples were detected during the 7 days after intubated. The total amount of 14C excreted in the fecese were 53% meaning that the rest of 47% melanoidin seemed to be retained in the body or metabolized . The radioactive compound showed a small retention in the liver and kidney. The brown pigment contents and hydrogen donating activities in the urine and feces increased proportionally to the activity of 14C. When the soybean sauce and glycine glucose melanoidin were intubated, the brown pigment contents excreted in the feces were found to be the highest after 1 st day of intubation. In the urine, the model melanoidin was excreted mostly after 3 days of intubation. The brown pigment contents and hydrogen donating activities of three samples of excreta agreed with each other. The soybean sauce retained longer than model melanoidin in the body is telling that it might have antioxidative activity in vivo.

Physiological Changes with Age by the Chronic Administration of Korean Red Ginseng in Spraque-Dawley Rats

  • The present study was designed to elucidate the physiological changes with age by chronic administration of red ginseng. All rats were reared in the conventional system. Ginseng treated rats were continously supplied with ginseng water extracts together water from 6 weeks of age to the age 24 months. Rats did not show any discernible signs or the rejection symptoms by red ginseng water extracts. A long-term administration of red ginseng extracts did not cause any physiological changes in the gain of body and organs weight, food intake and general properties of urine. However, red ginseng caused to decrease the level of serum cholesterol, glucose and TBARS, and it attenuated effectively the age-dependent decline of LDH activity. Other biochemical parameters measured from blood and general properties of urine were not significantly changed. These results suggest that long-term administration of red ginseng to rat does not cause any clear physiological changes in appearance and urine, and it retards age-related deteriorations in some biochemical parameters such as LDL-cholesterol, glucose and LDH in serum.
