팔정산(八正散) 약침(藥鍼)이 글리세롤로 유발된 흰쥐의 급성 신부전에 미치는 영향 (The Effect of Paljeong-San Pharmacopuncture Treatment on Glycerol-Induced Acute Renal Failure in Rats)

  • Objective: The primary objective this study was to evaluate the effect of Paljeong-san (PJS) pharmacopuncture treatment on against the glycerol-induced acute renal failure in rats. Methods: Glycerol injection decreased GFR (glomerular filtration rate) and increased urine volume, serum creatinine, BUN level, urine albumine secretion and fractional excretion of Na+, K+. PJS was selected in the basis of invigorating kidney which can eliminate pathogens. Rats were treated with PJS pharmacopuncture on Shin-shu (BL23) and Chon-chu (ST25) point for 3 days, followed by 50% concentration of glycerol injection ($5m{\ell}/kg$ body weight). Results: After the 3 days treatment period, Paljeong-san (PJS) pharmacopuncture treatment improved renal function. In addition, Glycerol injection increased lipid peroxidation, and decreased Na-K-ATPase in renal cortex and which were prevented by PJS treatment. Conclusion: This study suggests that Paljeong-san (PJS) pharmacopuncture treatment show favorable effect on glycerol-induced acute renal failure in rats.

Removal and inactivation of bovine herpes virus and murine encephalomycarditis virus by a chromatography, pasteurization, and lyophilization during the manufacture of urokinase from human urine

  • The purpose of present study was to examine the efficacy of PAB (para-amino benzamidine) affinity column chromatography, pasteurization ($60^{\circ}C$ heat treatment for 10 h), and lyophilization steps, employed in the manufacture of urokinase from human urine, in the removal and/or inactivation of urine-born viruses. Bovine herpes virus (BHV) and Murine encephalomyocarditis virus (EMCV) were selected for this study. Samples from the relevant stages of the production process were spiked with the viruses and the amount of virus in each fraction was quantified by 50% tissue culture infectious dose ($TCID_{50}$). BHV and EMCV were effectively partitioned from urokinase during PAB chromatography with the log reduction factors of 6.71 and 5.27, respectively. Pasteurization was a robust and effective step in inactivating BHV and EMCV, of which titers were reduced from initial titers of $8.65\;log_{10}\;TCID_{50}$ and $7.81\;log_{10}\;TCID_{50}$, respectively, to undetectable levels within 1 hour of treatment. The log reduction factors achieved during lyophilization were 2.06 for BHV and 4.54 for EMCV. These results indicate that the production process for urokinase has sufficient virus reducing capacity to achieve a high margin of virus safety.

Successful EDTA-Tris treatment of Pseudomonas aeruginosa infection of urinary bladder secondary to urolithiasis in a dog

  • About 8 year-old castrated male Yorkshire terrier was presented for evaluation of dysuria, stranguria, hemtauria, and pollakiuria. On history taking, dysuria first was observed three months ago and these signs were waxed and waned. Physical examination revealed mild left perineal swelling. On routine laboratory examination, no significant findings were identified. Positive contrast urogram identified peritoneal herniation of urinary bladder. Urinalysis showed proteinuria and hematuria. Urine sediment revealed epithelial cells, white blood cells and rod-shaped bacteria. Pseudomonas aeroginosa was isolated from urine obtained through cystocentesis, and had resistance against fourteen antibiotics. Cystitis caused by P. aeruginosa concurrent with cystolithiasis and perineal hernia was diagnosed. Cystotomy, herniorrhaphy and EDTA-Tris solution lavage of bladder were performed. The patient was recovered to normal condition 2 days after treatment. Two weeks later, bacterial culture of urine was negative and any abnormality in ultrasonogram and urinalysis was not observed except calcium oxalate dihydrate crystals.

Toxic Effects of Polygalae Radix on Rat Kidney

  • The renal toxicity of the extract of Polygalae Radix was investigated in rats. Rats were treated with 3.5 mg/Kg of the extract, i.p., for 7 days. Changes in consumatory behavior, 24 hour-urine and the activities of urinary enzymes were determined during the administration of the extract. Significant decrease in body weight and food consumption and increase in 24 hour-urine volume were observed during the administration. However, the quantity of total creatinine in urine was decreased significantly. Those indicate that subacute treatment with the extract might induce diuresis and the ditiresis might be due to the decrease in water reabsorption. In the activities of urinary enzymes, the activities of alanine aminopejotidase (AAP) and gamma-glutamyl transpeptidase (GGT) were increased 4.3 and 3.5 times and then returned to the control. The activity of N-acetyl-${\beta}$-D-glucosaminidase (NAG) was increased 7.2 times and then decreased slowly. But, it was significantly higher than that of the control evea after the last administration. The activity of factate dehydrogenase (LDH) was increased continuozlsly during the treatment. It showed 32 times higher than the control. These results suggested that the extract of Polygalae Radix had toxic effect on kidney. Furthermore, the result suggested that the subacute administration of the extract induced resistance against the toxicity of Polygalae Radix.

Manifestation of metabolic Aberration in Rat urine Following Hypothalamic Lesions

  • The concentrations of metabolites of low molecular weights (90 to 310 amu) present in rat urine were determined by field ionization mass spectrometry. Three groups of rats were examined; intact controls, sham-operated rats and rats with selective lesions in their hypothalamus. The latter lesions are shown to induce characteristic aberrations in the metabolic profile, demonstrable five weeks after treatment, which are distinct from those induced by a sham operation.

  • This study was conducted to investigate on the changes of copper and zinc concentration in rat's tissues and urine after cadmium administration with atomic absorption spectroph otometric method. It is found that cadmium appeared to cause a change in the behavior of copper and zinc in vivo system even during 1 month after cadmium treatment.

  • Objectives: The purpose of this study was to report the clinical effects of pharmacopuncture, acupuncture, and ohrimsan on prostatitis that was not improved by antibiotics. Methods: The patient underwent So-Yeom pharmacopuncture injection, acupuncture, and ohrimsan. A numerical rating scale (NRS) was used to obtain a sense of residual urine and the National Institute of Health Chronic Prostatitis Symptom Index (NIH-CPSI) was used to evaluate the treatment effects. Results: NRS for the sense of residual urine decreased from 70 before treatment to 0 after treatment. The NIH-CPSI was 19 on the first day of admission and decreased to 8 on the day of discharge. No side effects were observed after treatment. Conclusions: The traditional Korean medicine of pharmacopuncture, acupuncture, and ohrimsan is effective for treating prostatitis that is not improved by antibiotics.

  • The word of stress crime from Latin language as stringere and it was used in medical fields from 1935. According to Selye, all the biological bodies reveal physilolgical changes when some stimulation exceed normal levels, and consequently the pituitary gland and adrenal systems are activated. Jacob expressed that stress is the loss of homeostasis by physical, chemical, and emotional stimulation. When biological organisms receive extreme stress the amount of catecholamine excretion are increase. Author investigated the catecholamine contents in rat urine after giving the low temperature stress, noise stress, and water immersion stress. The 24 hours rat urine was collected by adding 1 ml 6 N-HCl and the sample is passed through Bio-Rex 70 samples treatment column to extract catecholamine and detected the catecholamine with HPLC-fluorescence detetor. The highest epinephrine concentration was 67.14 ng in water immersion stress condition and the dopamine concentration of 221.37 ng was shown in the low temperature stress condition.

  • The effect of oral iron supplementation was assessed on blood iron levels and Pb and Cd levels in erythrocytes, hair and urine of 101 Puchon 5th grade school children with suboptimal iron status. Treatment with 25mg of elemental iron per day for 8weeks resulted in a significant increase in the intake of most nutrients in addition to iron. Iron supplementation resulted in significant improvements in hemoglobin, MU, MCH, MCHC, serum ferritin, serum iron, TIBC, and transferrin saturation of subjects(p<0.05 - p<0.01) and cocomitantly lowered Pb and Cd levels in erythrocytes, hair, and urine(p<0.01). Regression analysis showed that only iron intake contributed to significant increases in hemoglobin and serum ferritin. It seems that 25mg of iron supplementation is safe and adequate to improve iron status in school children with suboptimal iron status and it also has the benefit of alleviating Pb and Cd status.   • "The Act on Medication Treatment of Sexual Impulse of Sex Offenders" known as chemical castration has been effective since July 2011 in Korea. According to the law, monitoring of male sex hormone in urine is enforced to request National Forensic Service more than once a month after injection of medicine designed to reduce sex impulse. We established a rapid and sensitive method for the monitoring of testosterone (T) and epitestosterone (E) in human urine by liquid chromatography with tandem mass spectrometry (LC-MS/MS). Three mL of urine was pretreated by solid-phase extraction for purification and performed enzymatic hydrolysis. The pretreated samples were extracted twice with 2 ml of ethyl acetate and n-hexane (2 : 3). The separation was applied on Thermo Hypersil GOLD C18 column ($1.9{\mu}m$, $100{\times}2.1mm$). A gradient elution of methanol and water of 0.1% formic acid were used as mobile phase and the retention time was less than 10 min. LC-MS/MS system coupled with an electrospray ionization source was performed in multiple reaction monitoring mode. The transitions of the analytes executed as following: m/z $289{\rightarrow}97$, 109 for T and E, m/z $292{\rightarrow}109$ for $T-d_3$ and $E-d_3$ as internal standards. The validation results of the method were satisfactory. The limits of detection were 0.05 ng/ml and the limits of quantification were 0.1 ng/ml. This method was successfully applied to real human urine sample. The developed method will be useful for monitoring T/E ratio in urine of sex offenders.
