Objective : This study aimed to evaluate the effects of Dianthi Semen Herba herbal-acupuncture (DS-HA) at KI10 (Umgok) on nephritis induced by LPS in rat. Methods : Rats were injected with LPS and treated with DS-HA at KI10 3 times for a week(DS-HA), N.P. group was treated 26 gauge needle at KI10, saline group was treated with normal saline at KI10. To estimate the effects of DS-HA at KI10 on nephritis in rats, WBC, neutrophils in blood, BUN, creatinine TNF-${\alpha}$, CINC-1 in serum, urinal volume and creatinean and total protein in urine, reanl TNF-${\alpha}$, renal MPO were measured and histological analysis of renal tissue was performed. Results : DS-HA at KI10 significantly decreased WBC and neutrophil in blood and BUN and creatinin in serum, MPO in kidney, and significantly increased urinary volume in LPS-stimulated rats. DS-HA at KI10 reduced accumulation of neutrophil in renal tissue of LPS-stimulated rats. Conclusions : Taken together, DS-HA at KI10 has a protective or therapeutic effect for nephritis in LPS-stimulated rat. Therefore, it is suggested that DS-HA at KI10 may be an useful therapeutics in clinical field after further researches.
Objective : Oriental medicines have been applied to Membranous nephropathy(MN) for the purpose on increasing renal blood flow and modulating immune activity in according to Oriental medicine theories. Magsungsinyeom-bang (MSSYB) is one of the prescription which is known to show positive results in clinic with lack of laboratorial evidence. Thus, this study was aimed to evaluate the effects of MSSYB and partially investigate the mechanisms of it. Methods : The effect of MSSYB was evaluated by the morphology for the GBM thickening, protein excretion in urine and biochemical parameters in serum using cBSA-induced MN mice model. Mice were administered with MSSYB(250 or 500 mg/kg) or PBS for control group from experimental week 3 for 4 weeks. Results : 24 hrs proteinuria and the concentrations BUN was significantly decreased in the MS groups compared to the control group while the concentrations of serum albumin was higher in the MS groups than control group. MSSYB didn't affect the ratio of CD3e+/CD or 19CD4+/CD8 in the spleen and kidney, but inhibit the expression of IL-1${\beta}$, TNF-${\alpha}$, IL-6, and production of IgG and IgM. In histological analysis of kidney tissue, thickening of GBM was significantly decreased in the MS group compared to control group. Conclusions : MSSYB showed the positive results on the cBSA-induced membranous nephropathy in mice, thus, it could be a useful candidate for oriental drug for treating the membranous nephropathy in clinic.
To investigate the exposure effect of benzene, we measured airborne benzene as external doses, uninary tt-muconic acid as an internal dose of benzene exposure and analyzed the relationship between tt-muconic acid concentration and benzene exposure. The study population of eight businesses included 157 workers(87 workers in field; exposure group, 70 workers in board; control group) who produce or use benzene in petrochemical industry. The concentrations of airborne benzene were evaluated by personal samples and urine was sampled at the pre and end shift. Urinary t,t-muconic acid as internal dose was to analyze the relationship with airborne benzene. The geometric means(GM) of airborne benzene was 0.0231 ppm(range ND-1.0471 ppm) in exposure group and 0.0147 ppm(range ND-0.3162 ppm) in control group. The geometric means(GM) of urinary t,t-muconic acid at end-shift was $196.8{\pm}2.23{\mu}g/g$ creatinine in exposure group and $149.2{\pm}2.08{\mu}g/g$ creatinine in control group. There was significant correlation between the airborne concentration of benzene and the urinary concentration of t,t-muconic acid( r=0.711, p<0.01). From the results of stepwise multiple regression analysis about t,t-muconic acid at end-shift, significant independents was airborne benzene. In this study, there were significant correlation between the urinary concentration of t,t-muconic acid and the airborne concentration of benzene. More extensive studies ruling out healthy worker effect is needed.
The pharmacokinetics and tissue distribution of DWP20373, a novel fluoroquinolone, were examined in rats and beagle dogs after a single intravenous and oral administration. Analysis of DWP20373 in plasma, tissue, and urine was performed by both HPLC and microbiological assay. The plasma drug concentration declined biexponentially both rats and beagle dogs. In the rats, the terminal drug elimination half-life (t$_{1}$2$\beta$/) was 64 min (IV) and 57 min (PO) by bioassay, and 76 min (IV) and 77 min (PO) by HPLC. Whereas in beagle dogs, t$_{1}$2$\beta$/ was 196 min (IV) and 350 min (PO). The volume of distribution at steady-state (Vd$_{ss}$ ) was 811 ml/kg (bioassay) and 2061 ml/kg (HPLC) in rats, and 2738 ml/kg (bioassay) in beagle dogs. The total body clearance (Cl$_{t}$) of DWP20373 was 10 ml/min/kg (bioassay) and 7 ml/min/kg (HPLC) in rats, and 11 m1/min/kg (bioassay) in beagle dogs. The extent of bioavailability after oral administration was 49% (bioassay) and 67% (HPLC) in rats, and 84% (bioassay) in beagle dogs. The 24-h urinary recovery, measured by bioassay, was 2.7% after oral dosing and 5.5% after intravenous dosing in rats. Serum protein binding ratio determined at 27g/ml was 78%. This drug was also distributed in tissues in the decreasing order of liver, kidney, spleen, lung, heart, and muscle determined at 30 min after oral administration.on.
Objectives : This study was carried out to understand the prevalence and risk factors of green tobacco sickness (GTS) among Korean tobacco harvesters. Methods : The authors conducted a questionnaire among the tobacco harvesters (1,064 persons from 555 out of 723 tobacco harvesting households) in Cheongsong-gun for 4 days from May 7 to 10, 2002. Results : The study subjects were 550 males and 514 females. The recognition and experience of GTS up until 2001 were 96.4% and 61.9%, respectively. The prevalence of GTS in 2001 was 42.5%, and was significantly higher in females than in males (59.0% vs. 26.6%, p<0.01). The incidence density of GTS according to the number of workdays in 2001 was 12.3 spells/100 person..days. The GTS symptoms reported by the tobacco harvesters in 2001 were dizziness in 441 cases (97.6%), nausea in 414 (91.6%), headache in 349 (77.2%) and vomiting in 343 (75.9%). The use of gloves, hat and wristlets, sweating at work and the number of working hours significantly increased the prevalence of GTS (p<0.05). Multiple logistic re- gression analysis was used to determine the factors significantly associated with GTS. Odds ratios for smoking, working over 10 hours and sweating at work were 0.26 (95% CI:0.19-0.35), 1.64 (95% CI:1.26-2.14) and 1.60 (95% CI:1.14-2.25), respectively. Of those who reported GTS in 2001, 311 cases (68.8%) underwent treatment from their local medical facilities. Conclusions : In Korea, there are many tobacco-harvesting households, and most may be stricken with GTS. More extensive epidemiological studies, including the incidence and associated risk factors, are expected and a surveillance system including measurements of cotinine in urine should be conducted.
This study was performed to evaluate repeated-dose oral toxicities of Flos lonicerae extract in Fischer 344/n rats. Flos lonicerae was administered orally to rats at dose levels of 0, 37, 111, 333, 1,000 and 2,000 mg/kg/day. Each group consisted of 10 rats of each gender. The Flos lonicerae extract was given once a day, 5 times a week, for 90 day repeatedly. This study was conducted in accordance with the Protocol of Korea National Toxicology Program and The Standards of Toxicity Study for Medicinal Products. In the present study, there were no toxicologically significant changes in mortality, clinical signs, body weight gains, ophthalmoscopy, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histopathology, estrus cycle and sperm examination of all animals treated with Flos lonicerae extract. These results suggest that the oral no observed adverse-effect level of the test item, Flos lonicerae extract, in rats is higher than 2,000 mg/kg/day in both genders. The target organs were not established.
The purpose of this study was to assess calcium and sodium intakes and urinary excretion of adults in Busan and to evaluate the relationship between urinary calcium excretion (UCa) and the status of anthropometric, blood pressure, urine analysis, and nutrient intake of subjects. Nutrient intake by 24 hr recall, 24 hr urinary calcium and sodium excretion (UNa) were measured with 87 adults aged 20-59 yrs (42 men and 45 women). The mean calcium intake was 88.0% for men and 103.0% for women of Recommended Intake. The mean sodium intake was 283.4% for men and 250.5% for women of Adequate Intake (AI). The mean 24hr UCa was 127.4 mg in men and 107.3 mg in women. The mean 24 hr UNa was 3650.6mg in men and 3276.4mg in women. The intake and urinary excretion of calcium and sodium were not significantly different by gender. UCa showed significantly positive correlations with sodium intake and UNa in men (p < 0.001, p < 0.05) and women (p < 0.001, p < 0.001) and with age, systolic blood pressure (SBP) and sodium density in women (p < 0.05, p < 0.05, p<0.01). The UCa/creatinine showed significantly positive correlations with age, sodium intake, sodium density, and UNa in women (p < 0.05, p < 0.01, p < 0.01, p < 0.01). When UCa was stratified into quartile (Q1-Q4), age, SBP, UCa, UNa, sodium intake, and AI percentage of sodium (p < 0.01, p < 0.05, p < 0.001, p < 0.001, p < 0.001, p < 0.001) were significantly higher in Q4. The mean intake and AI percentage of sodium in Q4 were 4768.8mg and 329.0. Based on the results, UCa was related to age, SBP, UNa, and sodium intake. Therefore, nutritional education of decreasing sodium intake for decreasing UCa is needed.
Objective : The clinical efficacy of Jowiseungchungtang(oriental herbal medication) over 6-month on the cognitive function in patients with mild Dementia of Alzheimer type(DAT) was investigated in this study. Method : The subjects for this study consisted of dementia patients who visited to the outpatient dementia department of Oriental Neuropsychiatry of the Kyunghee Oriental hospital. Patients were required to have at entry: a dignosis of mild DAT; Biochemical test, CBC, Urine analysis, Chest X -ray, EKG, Brain- MRI, Mini-Mental States-Korea(MMSE-K), Korean-Dementia Rating Scale(K-DRS), Samsung Dementia Questionaire(SDQ;to their caregiver). Patients(n=31) were assigned to treatment with Jowiseungchungtang (fourth/day for 6-month). The primary outcome measure was K-DRS. Result : 1. Patients' mean age was $71.4{\pm}6.6$ years(range 59 to 86 years). 2. Patients' mean education was $6.1{\pm}4.9$ years. K-DRS scores at baseline was $110.5{\pm}12.2$. K-DRS scores at 6-month was $109.0{\pm}17.0$. K-DRS scores at 6-month was not significantly different with K-DRS scores at baseline(t=.791 r=.435 p<.05). Conclusion : Treatment with Jowiseungchungtang for 6-month protected the cognitive function decline in patients with mild DAT.
This study was designed to investigate the effects of dietary calcium. serum estrogen level and physical activity on the bone status of 116 healthy elderly women living in urban area. Current calcium intake was assessed by convenient method(refered to as Ca intake) and calcium containing food frequency method(refered to as Ca index) Daily activity record was used for the estimation of physical activity level, and serum estrogen level was measured from fasting blood of subjects. The rate of bone resorption was evaluated by the determination of hydroxyproline(Hpr) in fasting urine with correction for creatinine excretion. The results of this study are summarized as follows : 1) Average daily Ca intake of subjects was 621.4$\pm$155.8mg, which is above the Korean recommended dietary allowances. However 44.8% of the subjects consumed Ca below RDA level. Ca index score was significantly correlated with the bone status(P<0.05), Ca intake did not show significant correlation with the bone status although a positive trend of influence was evident. 2) Average serum estrogen level of subjects was 18.7$\pm$9.8pg Contrary to our anticipation. estrogen level did not show any significant relation to age and bone status. 3) Daily physical activity was classified into four categories by activity intensity : sedentary. moderate, active and severe. The average physical activity of subjects belong to moderate level. and the bone status was significantly related to the physical activity(P<0.01) 4) Among other influential factors such as age, pocket-money. family type. drinking, smoking and BMI, there was a significant difference between bone status and BMI(P<0.05). 5) Multiple regression analysis of variables showed that physical activity has greater effect than other variables when the entire subjects were taken into account. However. eliminating the subjects whose bone status rated as excellent(Hpr/cr<0.009), Ca index showed higher correlation than physical activity. These results have demonstrated that dietary calcium intake is the primary important factor for keeping good bone health and that bone status of subjects with a sufficient calcium intake is affected by various factors such as physical activity, age, smoking. BMI and others.
The purpose of this study was to estimate the urinary excretion of sodium cho\ulcornerloride of adolescent in Jeon Bug. Korea and to evaluate the correlation of sodium chloride excretion by sex and area. The subjects were 261 healthy persons of 16 years old ( male 131. fermale 130) which were randomly choosen from different areas (city, farming area and fishing area). The urinary sodium chloride excretion in 24hrs were evaluated. which were cal\ulcornerculated from the analysis of urine sample gathered for 12hrs. The results were as follows; 1) The urinary excretion of sodium chloride by the group of male and female in 24hrs were 388.0$\pm$191.4mEq(22.68$\pm$l1.19gm) and 303.6\ulcorner138.0mEq(l7.75$\pm$8.07gm) reo spectively. 2) The urinary excretion of sodium chloride by the group of male was higher than by the group of female. 3) The urinary excretion of sodium chloride by the group of city, farming area and fishing area in 24hrs. were 256.2$\pm$96.8mEq(14.97$\pm$5.66 gm), 433.2$\pm$188.6 mEq (25.32$\pm$l1.62gm) and 356.0$\pm$169.8mEq (20.81$\pm$.92gm) respectively. 4) The urinary excretion of sodium chloride by the farming area group and the fishing area group were higher than the city group. Hence the daily urinary excretion of sodium chloride by adolescent of 16years old was estimated to be 20.21$\pm$9.63gm.
