Objective - To evaluate the usefulness of chromium in erythrocytes as a biological marker of exposure to hexavalent chromium in chromate producers and chrome platers Methods - Blood and urine samples were ramdomly obtained from chromate producers (n=34) and chrome platers (n=35), and non-exposed workers (n=75), chromium level in erythrocytes and plasma, and urine were measured. Different chromium exposure workers were assessed through measurements of airborne hexavalent chromium concentrations using a personal air sampler. Linear associations between variables were evaluated with correlation analysis. Results - The chromate producers had mean chromium levels in erythrocytes five fold as higher than the chrome platers, and fifteen fold higher than non-exposed group. Among the chromium exposed workers, airborne hexavalent chromium was positively and strongly correlated with in erythrocytes (r=0.689, p<0.01), and erythrocytes chromium was inversely correlated with hematocrit (r=-0.441, p<0.01), hemoglobin (r=-0.465, p<0.01) and the number of red blood cells (r=-0.28, p<0.05). Conclusions - In conclusion, this study suggests that chromium in erythrocytes is a good indicator of the chromium body burden caused by exposure to hexavalent chromium.
Bioluminescence single-site immunometric assay for methamphetamine (MA) using the native aequorin, a photoprotein, as a signal generator was developed for the first time. MA is a potent sympathomimetic amine with stimulant effects on the central nervous system. MA abuse induces hallucinations and, thus, may cause a serious social problem. The single-site immunometric MA assay was optimized and its dose-response behavior was examined. The dose-response curve shows that the detection limit is 1.1 ${\times}$$10^{-10}$ M and a dynamic range is four orders of magnitude with 15 $\mu$g/mL BSA-MA conjugate and 1.0 ${\times}$$10^{-8}$ M anti-MA antibody-biotin conjugate. In order to evaluate this assay, the structurally similar compounds, amphetamine, ephedrine, norephedrine, benzphetamine and N-4-(aminobutyl)methamphetamine were examined for their crossreactivity. None of these five compounds showed any cross-reactivity. Additionally, an artificial urine solution spiked with MA was analyzed by the MA assay, and the result of the analysis demonstrated the usefulness of the present assay for the determination of MA in urine.
Normal range of mercury contents in blood and its relationship with urinary mercury excretion were studies with 68 healthy male adults living in Seoul city, who had no obvious evidence of .either occupational exposure to mercury or therapeutic use of mercurial agents. Mercury analysis was made by means of dithizone colorimetric method with coefficient of variation of 10.9% in .an average ranging from 5.1% to 18.0%. 1. Mercury contents in normal human blood were both normally and log-normally distributed, and better fitted to the latter. 2. Geometric mean and standard deviation of the mercury contents were $24.0(log^{-1}1.38){\pm}1.66{\mu}g/100ml(log^{-1}0.22{\mu}g/100ml)$ ranging from 7.2 to 79.7 ${\mu}g/100ml$ with 95% confidence interval. 3. Mercury contents in normal human blood differed from person to person (p<0.01), and the variability of the measurements was negligible (p>0.05). 4. Mercury in the blood was contained much higher in erythrocytes than in plasma (p<0.01), showing the geometric means of $21.0{\pm}1.25{\mu}g/100ml$ in red blood cells and $14.3{\pm}1.62{\mu}g/100ml$ in plasma, respectively. 5. Mercury contents in normal human blood had a relationship of power function with mercury excretion in urine corrected with a gram of creatinine excretion per liter of urine (p<0.10).
Objectives: The purpose of this study was to examine the relationship between dietary habits and concentrations of urinary phthalate metabolite in elementary school children. Methods: This study was conducted in Seoul, South Korea. We collected urine samples from 156 children from a single school in September 2016. Information on dietary habits was obtained from the parents of the children. Five types of urinary phthalate metabolites were analyzed using a high-performance liquid chromatography tandem mass spectrometer. Multiple linear regression analysis was used to determine the factors affecting the concentrations of urinary phthalate metabolite. Results: Girls had a significantly lower concentration of urinary mono-n-butyl phthalate (MnBP) (p=0.006) than did boys. When the consumption of bean processed food and wrapped delivery food were more than once a week, urinary MnBP (p=0.021) and monobenzyl phthalate (MBzP) (p=0.032) concentrations were the highest, respectively. Conclusion: Several urinary phthalate metabolite concentrations were associated with demographic and dietary habits. Particularly, urinary MnBP and MBzP were associated with several processed foods. The findings of this study might be useful as basic data for establishing and educating on environmental health policy.
Antineoplastic agents may exhibit effects not only in patients therapeutically exposed, but also in health workers who prepare and administer these drugs. This study was done to clarify whether nurses who handle anticancer drugs show signs of drug absorption. The experimental group was 14 nurses handling anticancer drugs at three medical wards of a hospital in J city ; the control group was 12 psychiatric nurses at the same hospital. The test materials were the nurses' 24hr urine specimens, which were concentrated by XAD-2 column chromatograpy. Tester strains were TA98(±S9mix), TA100(±S9 mix), TA1535(±S9 mix) and TA1537(±S9 mix) : the salmonella mammalian microsomal test (Ames test) was used for the urinary mutagenicity assay. The results are summarized as follow : 1. In qualitative analysis of the results, both experimental group and control group showed 15.4% urine toxicity. 2. The experimental group revealed significantly higher urinary mutagenicity both in the activation method test and non-activation method test of the tester strains TA98, TA100 and TA1535. In the case of TA1537, the two groups showed no difference in the non-activation method test, but the activation method revealed a difference. 3. In urinary mutagenicity of the experimental group by ward career, there was a significant difference between the group with more than 20 months experience and the group with less than 20 months on the tester strains TA98, TA100, and TA1537. No Significant difference was found between two groups by the tester strain TA1535.
The abuse of zipeprol, an antitussive agent, is prevalent among young people. Ten fatal cases of zipeprol concentration in blood after its abuse had caused death are disscussed. GC equipped with TSD was used to quantify the drug in postmortem blood and GC/MS to identify the metabolites in urine after preliminary test. The calibration curve of zipeprol was linear (r = 0.998) and the recoveries of zipeprol 5, 10 and $20\;{\mu}g$ added to 1 ml postmortem blood were $97.2{\pm}3.9$, $92.5{\pm}3.9$ and $86.9{\pm}4.6%$ respectively. The blood zipeprol concentration varied 2.9 to $24.69\;{\mu}g/ml$ in 10 fatal cases. All the deaths were young adults with the age of 16-21. Three metabolites were identified in drug abuser's urine, 2-methoxy-2-phenyl ethylpiperazine, 2-hydroxy-3-methoxy-3-phenyl propylpiperazine and a demethylated zipeprol.
Urinary cytology has become an essential element in the diagnosis and management of transitional ceil carcinoma(TCC) of the urinary tract. It has the advantage of being noninvasive, inexpensive, and easily accessible. Besides that it can even detect malignancy when unsuspected at cystoscopy. We report a retrospective review of urine cytology un the diagnosis of 83 TCC cases that underwent 295 cytologic evaluation. All patients had biopsy-proven TCC of the bladder, ureter and renal pelvis, The overall incidence of the positive cytology cases was 66.2%. To define the cytologic features of tumor cells, we tried to use three cytologic gradings such as "grade 1", "grade 2", and "grade 3" according to the cytologic degree of anaplastic neoplastic cells. These cytologic gades of TCC were relatively well correlated with the histologic grade and tumor invasiveness. This result suggests that the recognition of characteristic cellular features of TCC can suspect the histologic grade and tumor stage. The false negative TCC cases were 78.9%. They showed severe inflammatory or bloody background and a few neoplastic cells. Therefore, a cautious approach for accurate interpretation, personal experience, and proper fixation and processing could expand the role of urinary cytology.
Purpose: The purpose of this study was to identify the risk factors for a nosocomial urinary tract infection in intensive care units with a foley catheterization which showed a positive urine culture. Method: Three-hundred eighty-seven patients were included in the study. A retrospective review of the electrical medical record system's databases and medical record sheets in hospitalized patients from January 2003 to December 2003 was used. The collected data was analyzed by descriptive statistics, t-test, chi-square test and logistic regression analysis. Result: The frequency of the participants' nosocomial urinary tract infection was 72.9%. Significant risk factors for a nosocomial urinary tract infection were 'age', 'place of catheter insertion', 'frequency of catheter change', and 'duration of catheterization'. These variables explained 18.4% of variance in the experience of nosocomial urinary tract infection in intensive care units with foley catheterization. Conclusion: Medical personnel can decrease the incidence of a nosocomial urinary tract infection by recognizing and paying attention to the duration of catheterization, frequency of catheter change, and place of catheter insertion. As a result, specific and scrupulous strategies should be developed to reflect these factors for decreasing nosocomial urinary tract infections.

The urine analysis system implemented to measuring the primary color reaction of urinalysis strip. Fuzzy classifier based on fuzzy theory implemented so as to classify of 9 items in the urinalysis strip and proposed the automatic turning algorithm of mambership function in the fuzzy classifier to progress the reproduction of classify. To evaluation of clinical capability, the fuzzy classifier and automatic turning algorithm apples to standard strip and standard reagent.
To investigate biomarker-based exposure to phthalates and related factors with demographics, 100 subjects who had participated in comprehensive health check-up were selected. We collected demographics through questionnaires and analyzed urine samples for 5 phthalates. Statistical likelihoods and regression methods were applied for data analysis using censored data. The highest levels of urine phthalates were 216$\mu\textrm{g}$/ml in di-isodecyl phthalate, 29.0$\mu\textrm{g}$/ml in di-butyl phthalate, 5.78$\mu\textrm{g}$/ml in di-(2-ethylhexyl) phthalate. The median values of di-(2-ethylhexyl) phthalate were 0.2340 $\mu\textrm{g}$/ml for male smokers, 0.0399 $\mu\textrm{g}$/ml for male non-smokers and 0.0085 $\mu\textrm{g}$/ml for female non-smokers, respectively. Di-(2-ethylhexyl) phthalate, benzyl butyl phthalate and di-isodecyl phthalate were higher in males than in females. In addition, mono-2-ethylhexyl phthalate was decreased with age. Our findings suggest that there might be significant demographic variations in exposure and/or metabolism of phthalates, and that health-risk assessment for phthalate exposure in humans should consider different potential risk groups.
