공단지역주민의 요중 비소농도와 혈압과의 관련성에 관한 연구 (Association Between Blood Pressure and Urinary Arsenic Concentration in Industrial Areas)

  • Objectives: The study examines the relation between urinary arsenic concentration and blood pressure, which is a risk factor of cardiovascular disease. Materials: In this study, the urinary arsenic concentration, history of diagnosed disease, and blood pressure of 782 local residents in Gwangyang, Yeosu, and Hadong regions from May 2007 to July 2007. Results: The urinary arsenic concentration of total participants was $9.06{\mu}g/g-ct$. The logistic regression analysis of medical diagnosed history and urinary arsenic concentration, showed statistically significance (p<0.05) of high urinary arsenic concentration in participants with diagnosed hypertension. In addition, diagnosed hypertension it was observed that the high blood pressure was related with the pulse pressure. Conclusions: The arsenic concentration level was low in this study, but the exposure to low levels of arsenic has an effect on hypertension. Also, hypertension is related to pulse pressure and mean arterial blood pressure as well as being risk factor of cardiovascular disease. Therefore, close supervision of low -level arsenic exposure is needed.

폐금속광산 지역 주민들의 요 중 비소종별 농도와 관련요인 평가 (Urinary Arsenic Species Concentrations and Related Factors among Residents Living near Abandoned Metal Mines)

  • Objectives: The purpose of this study was to evaluate urinary arsenic concentrations by arsenic species and to identify related factors among local residents near abandoned metal mines in Korea. Methods: Among the subjects of the Health Survey of Residents Near Abandoned Metal Mines for 2013-2017, 664 people were enrolled in this study. Urinary arsenic species analysis was performed using ICP/MS. Result: The geometric means (95% Confidence Interval) by urinary arsenic species were 0.15 (0.13-0.17) ㎍/L for AsIII, 0.64 (0.55-0.75) ㎍/L for AsV, and 1.21 (1.05-1.40) ㎍/L for inorganic arsenic. The geometric means of urinary MMA and DMA were 1.58 (1.35-1.86) ㎍/L and 77.93 (72.61-83.63) ㎍/L, respectively, and that of organic arsenic was 83.15 (77.80-88.88) ㎍/L. The concentration of inorganic arsenic in the group using groundwater as drinking water was 1.36 (1.13-1.64) ㎍/L, which was statistically significantly higher than the 1.00 (0.80-1.25) ㎍/L in the other drinking water groups. Regarding rice consumption, the concentration of inorganic arsenic in urine in the group whose consumption was more than half rice produced in the residential area was 1.32 ㎍/L, which was statistically significantly higher than that of the 1.12 ㎍/L for the group whose consumption was less than half. Conclusion: In the analysis of the factors affecting the urinary inorganic arsenic concentration of the residents of the abandoned metal mine area, the use of groundwater as drinking water and consumption of rice produced in the residential area were considered related factors.

HG-AAS법에 의한 요중 비소의 최적 분석법에 관한 연구 (A Study on the Optimal Analytical Method for the Determination of Urinary Arsenic by Hydride Generation-Atomic Absorption Spectrometry)

  • This study was carried out to examine the optimal analytical method for determination of urinary toxic arsenic (inorganic arsenic and its metabolites) by HG-AAS (hydride generation-atomic absorption spectrometry). In the analysis of SRMs (standard reference materials), method E (addition of 0.4% L-cysteine to pre-reductant and use 0.04M HCl as carrier acid) showed the most accurate results compared with the reference values. In the analysis of 30 urinary samples, analytical results were significantly different depend on the component of pre-reductant and the concentration of carrier acid. When the concentration of carrier acid was higher, the analytical result was lower. The recovery rates of MMA (monomethylarsonic acid) and DMA (dimethylarsenic acid) were varied by the concentration of pre-treatment acid and carrier acid and hydride generation reagents. When the concentration of carrier acid was 1.62 M (5% HCl), the recovery rates of DMA was 1%. The recovery rates of MMA and DMA in method E (=V) were 102% and 100%, respectively. The results of this study suggest that the component and concentration of pre-reductant and carrier acid must be carefully adjusted in the analysis of urinary arsenic, and method E is recommendable as the most precise analytical method for determination of urinary toxic arsenic.

충청남도 내 산업단지 주변에 거주하는 주민들의 요중 비소 농도 (Urinary Arsenic Concentrations among Residents in the Vicinity of a Chungcheongnam-do Province Industrial Complex Area)

  • Objectives: The purpose of this study was to evaluate the relationship between residential surroundings, such as a power plant, steel mill and petrochemical facilities, and urinary arsenic concentrations in Chungcheongnam-do Province, Korea. Methods: Stratified by fish consumption and residential district, median and maximum block sampling was applied. A total of 346 spot urine samples were speciated for $As^{5+}$, $As^{3+}$, monomethylarsonic acid(MMA), dimethylarsonic acid (DMA) and arsenobetaine (AsB). Exposure assessment was based on questionnaires including data on sex, age, current tobacco use, fish consumption, type of water consumed, and occupational category. Results: Urinary $As^{5+}+As^{3+}+MMA+DMA$ concentrations of people living in the vicinity of a power plant ($GM=50.39{\mu}g/g$) were 61% higher than those of people living in the inland area according to median block sampling. Urinary $As^{5+}+As^{3+}+MMA+DMA+AsB$ concentrations of people living in the vicinity of industrial complex area were higher than those of people living in the inland area according to block sampling by median and maximum. Conclusion: Urinary arsenic concentrations of people living in vulnerable areas such as around industrial complexes, especially power plants, were higher than those of people living in an inland area.

일부 폐금속광산 지역 거주 여성의 요중 비소와 8-hydroxydeoxyguanosine 농도 사이의 관련성 (The Relationship between the Urinary Arsenic and 8-Hydroxydeoxyguanosine Levels in Women of Abandoned Mine Area)

  • 본 연구는 비소에 대한 만성노출이 의심되는 일부 폐금속광산 주변 지역 주민 중 여성들을 대상으로 요중 비소 농도와 산화적 유전자 손상지표인 요중 8-hydroxydeoxyguanosine (8-OHdG) 농도와의 관련성을 평가하기 위해 시행되었다. 충청북도에 위치한 두 폐금속광산 지역 주민 중 여성 165명을 대상으로 요중 비소농도와 요중 8-OHdG의 농도를 측정하고 SPSS 12.0 통계프로그램을 이용하여 분석하였다. 조사 대상자들의 요중 비소농도는 기하평균이 $5.65\;{\mu}g/g$ creatinine으로 나타났으며 요중 비소와 8-OHdG 농도와의 관련성 분석에서는 상관계수 0.399 (p<0.001)의 유의한 상관성을 보였다. 이러한 관련성은 비소노출 농도가 낮은 경우가 노출농도가 높은 경우에 비해 상대적으로 높은 것으로 나타났다. 본 연구의 결과는 요중 8-OHdG 농도가 저농도의 비소에 만성적으로 노출된 여성에서 산화적 유전자 손상을 평가하는 좋은 지표가 될 수 있음을 시사한다.

울산 공단지역 초등학생들의 요중 비소, 카드뮴, 아연 농도에 관한 연구 (Urinary Levels of Arsenic, Cadmium, and Zinc of Children in Ulsan Industrial Area)

  • 저자들은 울산 공단지역 인근 초등학교 학생들의 요중 비소, 카드뮴, 아연 등 3 개 중금속 농도를 측정하여 초등학생들 의 건강관리를 위한 기초자료를 얻고 또 한 우리나라 소아들의 요중 중금속 농도에 대한 참고치를 알아보기 위해 공단지역 초등학교 학생 348명(남자 182명, 여자 166명) 교외지역 초등학교 학생 100명(남자 50망 여자 50명)을 대상으로 본 연구를 시도하였다. 그 결과는 다음과같다. 1. 조사대상 초등학생들의 요중 비소, 카드뮴, 아연의 기하평균 농도는 각각 3.69, 0.99, $282.49{\mu}g/l$ 이었다. 2. 조사대상 초등학생들의 요중 크레아티닌량으로 보정한 요중 비소, 카드뮴, 아연의 기하평균 농도는 각각 3.92, 1.05, $299.92{\mu}g/g$ creatinine이었다. 3. 공단지역의 초등학생들의 요중 비소 및 카드뮴 농도는 교외지역 초등학교 학생들보다 통계적으로 유의하게 높았다 (p<0.0l). 이상의 결과로 볼 때 공단지역 초등학생은 교외지역 초등학생에 비해 비소, 카 드뮴 등 중금속 폭로량이 많은 것으로 사료되며 향후 공단지역 공장들에 대한 유해인자의 방출 억제 및 개선대책과 함께 공단지역 초등학교 주변의 중금속 등 환경 유해요인의 주기적 측정 등이 필요한 것으로 사료된다.

울산지역 어린이들의 혈중 연, 요중 비소 및 카드뮴 농도 변화추이 - 3년 추적조사 (Trend of the Changes in the Level of Blood Lead, Urinary Arsenic and Urinary Cadmium of children in Ulsan ; 3-year Follow-up Study)

  • Objectives : To obtain basic data on blood lead level and urinary level of arsenic and cadmium of children living near a petrochemical estate and a suburban area in Ulsan, Korea and to observe the trend of the changes. In the level of these metals in these children. Methods : The study subjects comprised 626 children living near a petrochemical estate and 299 children living in a suburban area of Ulsan. We analyzed the level of lead, arsenic and cadmium using atomic absorption spectrometer. Results : The mean levels of blood lead in children living near the petrochemical estate were $5.25{\mu}g/dl,\;5.24{\mu}g/dl,\;and\;7.24{\mu}g/dl$, and in the years 1997, 1999, and 2000, respectively, whereas those of children living in the suburban area were $3.81{\mu}g/dl,\;4.75{\mu}g/dl,\;and\;7.19{\mu}g/dl$ respectively. The mean levels of urinary arsenic in children living near the petrochemical estate were $4.57{\mu}g/g$ creatinine, $4.78{\mu}g/g$ creatinine, and $6.02{\mu}g/g$ creatinine in the year 1997, 1999, and 2000 respectively, whereas those of children living in suburban area were $2.35{\mu}g/g$ creatinine, $4.75{\mu}g/g$ creatinine, and $7.07{\mu}g/g$ creatinine, respectively. The mean levels of urinary cadmium in children living near the petrochemical estate were $1.15{\mu}g/g$ creatinine, $1.05{\mu}g/g$ creatinine, and $1.71{\mu}g/g$ creatinine in the year 1997, 1999, and 2000, respectively, whereas those of the children living in the suburban area were $0.74{\mu}g/g$ creatinine, $1.29{\mu}g/g$ creatinine, and $1.48{\mu}g/g$ creatinine, respectively. There were increasing trends in the level of blood lead, urinary arsenic and cadmium of children in Ulsan, and the differences in the level of these metals were disappearing between the children living in other areas year by year. Conclusions : These results suggest that the amount of exposure to lead, arsenic, and cadmium is increasing from year to year, and there is a need for periodic biological and atmospheric monitoring of these metals in Ulsan.

FI-HG-AAS를 이용한 전처리 과정에서 사용되는 예비환원제의 종류에 따른 요중 비소 분석결과 비교 (Comparisons of Urinary Arsenic Analysis by Pre-reductant for Preconditioning via the FI-HG-AAS Method)

  • Objectives: The method of analyzing urinary arsenic by flow injection hydride generation atomic absorption spectrometry (FI-HG-AAS) is generally used because it shows relatively greater sensitivity, low detection limits, low blocking action, and is simple to operate. In this study, the results of analysis according to three pre-reductants commonly used in the FI-HG-AAS method were compared with each other. Methods: To analyze urinary arsenic, nineteen urine samples were collected from adults aged 43-79 years old without occupational arsenic exposure. Analysis equipment was FI-HG-AAS (AAnalyst 800/FIAS 400, Perkin- Elmer Inc., USA). The three pre-reductants were potassium iodide (KI/AA), C3H7NO2S (L-cysteine), and a mixture of KI/AA and L-cysteine (KI/AA&L-cysteine). Results: In the results of the analysis, the recovery rate of the method using KI/AA was 82.3%, 95.7% for Lcysteine, and 123.5% for KI/AA and L-cysteine combined. When compared with the results by use of high performance liquid chromatography inductively-coupled plasma mass spectrometry (HPLC-ICP-MS), the method using L-cysteine was the closest to those using HPLC-ICP-MS ($98.57{\mu}g/L$ for HPLC-ICP-MS; $74.96{\mu}g/L$ for L-cysteine; $69.23{\mu}g/L$ for KI/AA and L-cysteine; $13.06{\mu}g/L$ for KI/AA) and were significantly correlated (R2=0.882). In addition, they showed the lowest coefficient of variation in the results between two laboratories that applied the same method. Conclusion: The efficiency of hydride generation is considered highly important to the analysis of urinary arsenic via FI-HG-AAS. This study suggests that using L-cysteine as a pre-reductant may be suitable and the most rational among the FI-Hg-AAS methods using pre-reductants.

Substantial Evidences Indicate That Inorganic Arsenic Is a Genotoxic Carcinogen: a Review

  • Arsenic is one of the most toxic environmental toxicants. More than 150 million people worldwide are exposed to arsenic through ground water contamination. It is an exclusive human carcinogen. Although the hallmarks of arsenic toxicity are skin lesions and skin cancers, arsenic can also induce cancers in the lung, liver, kidney, urinary bladder, and other internal organs. Arsenic is a non-mutagenic compound but can induce significant cytogenetic damage as measured by chromosomal aberrations, sister chromatid exchanges, and micronuclei formation in human systems. These genotoxic end points are extensively used to predict genotoxic potentials of different environmental chemicals, drugs, pesticides, and insecticides. These cytogenetic end points are also used for evaluating cancer risk. Here, by critically reviewing and analyzing the existing literature, we conclude that inorganic arsenic is a genotoxic carcinogen.

폐금속광산 지역 주민의 비소 대사에 영향을 미치는 MTHFR, As3MT, GSTO1 유전자 다형성 (MTHFR, As3MT and GSTO1 Polymorphisms Influencing Arsenic Metabolism in Residents Near Abandoned Metal Mines in South Korea)

  • Background: In South Korea, areas around abandoned metal mines are designated as regions with high arsenic (As) contamination. However, studies assessing urinary As exposure, As metabolism, and relevant genetic polymorphisms in residents of these metal mine areas are lacking. Objectives: To identify factors associated with As exposure and evaluate the effects of MTHFR, As3MT, and GSTO1 genetic polymorphisms on As metabolism in residents of abandoned metal mine areas by measuring urinary As species. Methods: Urinary As species (arsenite [As3+], arsenate [As5+], monomethyl arsonic acid, and dimethylarsinic acid) were isolated using high-performance liquid chromatography in combination with inductively coupled plasma mass spectrometry (HPLC-ICP-MS). Four genetic polymorphisms (MTHFR A222V, MTHFR E429A, GSTO1 A140D, As3MT M287T) were analyzed in 144 residents of four areas around abandoned metal mines. Results: The study sample was comprised of 34.7% men and 65.3% women, with a mean age of 70.7±10.9 years. The urinary inorganic As concentration was higher among those consuming more than half locally produced rice (0.31 ㎍/L) than those consuming less than half such rice (0.18 ㎍/L). The urinary dimethylarsinic acid concentration was higher in the group that had consumed seafood in the past day (31.68 ㎍/L) than in those who had not (22.37 ㎍/L). Furthermore, individuals heterozygous in the MTHFR A222V and GSTO1 A140D polymorphism had higher urinary arsenic species concentrations than did individuals with a wild type or homozygous for the variant allele. Conclusions: Consumption of locally produced rice was associated with inorganic As exposure, whereas seafood consumption was associated with organic As exposure among residents of abandoned metal mine areas. There was no clear association between MTHFR A222V and GSTO1 A140D polymorphisms and As metabolism.
