Volatile organic compounds (VOCs) are an important public health issue in Korea and many important questions remain to be addressed with respect to assessing exposure to these compounds. Because they are ubiquitous and highly volatile, special techniques must be applied in their analytic determination Valid Personal exposure assessment methods are needed to evaluate exposure frequency, duration and intensity, as well as their relationship to personal exposure characteristics. Biological monitoring is also important since it may contribute significantly in risk assessment by allowing the estimation of effective absorbed doses. This study was on ducted to establish the environmental measurement, personal dosimetry and biological monitoring methods for VOCs. These methods are needed to compare blood, urinary and exhalation breath VOC levels and to provide tools for risk assessment of VOC exposure. Passive monitors (badge type) and a active samplers (trap) for the VOCs collection were used for air sampling. Methods development included determining the minimum detectable amounts of VOCs in each media, as well as evaluating collection methods and developing analytical procedures. Method reliability was assessed by determining breakthrough volumes and comparing results between laboratories and with other methods. A total capacity of trap used in this study was 60ι. Although variable by compound, the average breakthrough was 20%. Also, there was no loss of compounds in trap even if keep for 45 day in -7$0^{\circ}C$. The recovery of active and passive methods was 69% ~ 126% and method detection limit was 0.24 $\mu\textrm{g}$/trap and 0.07 $\mu\textrm{g}$/badge. There was no statistical difference (P > 0.05) between active and passive methods.
A gas chromatographic method was established for the simultaneous determination of urinary nicotine and cotinine. The analytes in basified urine containing a sufficient amount of Na$_2$S0$_4$were extracted into dichloromethane by vigorous shaking. Into the transferred organic phase was added a small amount of acidified methanol (0.5 N HCI in methanol), followed by concentrating the mixture to dryness using a mild stream of nitrogen gas. The concentrate was reconstituted with methanol and the final solution analyzed using the gas chromatograph equipped with the nitrogen-phosphorus detector. The reproducibility tests showed coefficients of variation less than 11% for both compounds. The percent recovery for both analytes ranged from 88 to 103%. The estimated method detection limits for nicotine and cotinine were 0.60 and 5.1 ng/mL, respectively. Extraction efficiencies for both nicotine and cotinine apparently declined without the addition of Na$_2$S0$_4$into the urine. Moreover, the absence of methanolic HCI in the extract resulted in almost complete evaporation of nicotine and partial loss of cotinine during the concentration process, indicating that the formation of nicotine-HCI and cotinine-HCI species is prerequisite to the suppression of the loss of both compounds.
Objectives: This study was conducted to examine the relationship between crude oil exposure and physical symptoms among residents participating in clean-up work associated with the Hebei Spirit oil spill, 2007 in Korea. Methods: A total of 288 residents responded to a questionnaire regarding subjective physical symptoms, sociodemographic characteristics and clean-up activities that occurred between two and eight weeks after the accident. Additionally, the urine of 154 of the respondents was analyzed for metabolites of volatile organic compounds (VOCs) and polycyclic aromatic hydrocarbons (PAHs) and heavy metals. To compare the urinary levels of exposure biomarkers, the urine of 39 inland residents who were not directly exposed to the oil spill were analyzed. Results: Residents exposed to oil remnants through clean-up work showed associations between physical symptoms and the exposure levels defined in various ways, including days of work, degree of skin contamination, and levels of some urinary exposure biomarkers of VOCs, metabolites and metals, although no major abnormalities in urinary exposure biomarkers were observed. Conclusions: This study provides evidence of a relationship between crude oil exposure and acute human health effects and suggests the need for follow-up to evaluate the exposure status and long-term health effects of clean-up participants.
Objective: Cow urine possesses several bioactive properties but the responsible components behind these bioactivities are still far from identified. In our study, we tried to identify the possible components behind the antimicrobial activity of cow urine by exploring the peptidome and metabolome. Methods: We extracted peptides from the urine of Sahiwal cows belonging to three different physiological states viz heifer, lactation, and pregnant, each group consisting of 10 different animals. The peptides were extracted using the solid phase extraction technique followed by further extraction using ethyl acetate. The antimicrobial activity of the aqueous extract was evaluated against different pathogenic strains like Staphylococcus aureus, Escherichia coli, and Streptococcus agalactiae. The safety of urinary aqueous extract was evaluated by hemolysis and cytotoxicity assay on the BuMEC cell line. The urinary peptides were further fractionated using high-performance liquid chromatography (HPLC) to identify the fraction(s) containing the antimicrobial activity. The HPLC fractions and ethyl acetate extract were analyzed using nLC-MS/MS for the identification of the peptides and metabolites. Results: A total of three fractions were identified with antimicrobial activity, and nLC-MS/MS analysis of fractions resulted in the identification of 511 sequences. While 46 compounds were identified in the metabolite profiling of organic extract. The urinary aqueous extract showed significant activity against E. coli as compared to S. aureus and S. agalactiae and was relatively safe against mammalian cells. Conclusion: The antimicrobial activity of cow urine is a consequence of the feeding habit. The metabolites of plant origin with several bioactivities are eliminated through urine and are responsible for their antimicrobial nature. Secondly, the plethora of peptides generated from the activity of endogenous proteases on protein shed from different parts of tissues also find their way to urine. Some of these sequences possess antimicrobial activity due to their amino acid composition.

This study was conducted to analyze and determine the formation potential of chlorination DBPs from seven urinary compounds with or without Br$^-$. Three of seven components were kynurenine, indole and uracil that were relatively shown high the formation potential of chlorination DBPs concentrations. The reported results of THMs/DOC with or without Br$^-$ in kynurenine showed that THMs/DOC was detected 86.9 $\mu$g/mg when Br$^-$ was not added, and THMs/DOC was detected 100.8 $\mu$g/mg when Br$^-$ was presented. In indole, THMs/DOC was increased from 6.58 $\mu$g/mg to 31.4 $\mu$g/mg when Br$^-$ was added. Moreover, among them, the highest, second-highest and third-highest HAAs/DOC were shown in kynurenine, uracil and indole respectively. Specially, HAAs/DOC was significantly deceased in kynurenine and indole when Br$^-$ was presented. This was a totally different phenomenon for THMs/DOC. TCAA was dominated in HAAs for kynurenine and indole, and DCAA was also dominated in HAAs for uracil. The highest formation of HANs/DOC was shown in kynurenine whether or not Br$^-$ presented, and DCAN was predominant in HANs. HANs was not formed by chlorination in uracil. In addition, the formation of CH/DOC was relatively low in kynurenine and indole. The formation of CH/DOC was specially high(1,270 $\mu$g/mg) in uracil when Br$^-$ was not added. The formation of CH/DOC was 1,027 $\mu$g/mg in uracil when Br$^-$ was added. The formations of THMs and HAAs were also investigated in kynurenine, indole and uracil when Br$^-$ was presented or not. The formation of THMs/DOC was higher in kynurenine and indole when Br$^-$ was presented. The formation of HAAs/DOC was reduced in kynurenine when Br$^-$ was added. The result could be attributed to higher formation of THMs/DOC in kynurenine when Br$^-$ was added. The formation of HAAs/DOC was also reduced in indole when Br$^-$ was added. To the contrary, this result was not attributed to higher formation of THMs/DOC in indole when Br$^-$ was added.

Objectives: The purpose of this study is to investigate the atmospheric concentration of VOCs and the urinary concentration of t,t-MA, HA, MA in the industrial complex of Yeosu, South Jeolla Province. Methods: In order to study seasonal patterns of air concentration of VOCs, measurements were taken at five sampling sites around Yeosu from June 2013 to June 2014. Urinary metabolite excretionsfrom 671 subjects, exposure and comparison area were analyzed. Results: The average concentration of VOCs in the air was 1.53ppb for benzene, 0.73ppb for toluene, 0.22ppb for ethylbenzene, 0.52ppb for xylene and 0.12ppb for styrene. The concentration of benzene was somewhat higher than the year-average standard ($5{\mu}g/m^3$, about 1.5ppb) of the domestic air-environment criteria newly established in 2010.The metabolic concentration of VOCs in the urine of the entire sample was analyzed at $47.76{\mu}g/g\;cr.$, 213.07mg/g cr., and $290.09{\mu}g/g\;cr.$ for t,t-MA, HA, and MA, respectively. Compared with the average values for Korea as presented in the first basic survey of national environmental conservation ( $49.8{\mu}g/g\;cr.$ for t,t-MA, 0.17g/g cr. for HA, and 0.26mg/g cr. for MA), the metabolic concentrations of HA and MA in urine were higher than the average values. Conclusions: The concentration of VOCs in the air and urinary metabolites of the exposed and control areas showed that the concentrations of all substances were higher in the exposed area than in the control area.
Objectives: This study aims to investigate the atmospheric concentration of polycyclic aromatic hydrocarbons (PAHs) and volatile organic compounds (VOCs) and the urinary concentration of biomarkers in residents near the Shinpyeong·Jangrim Industrial Complex to compare them with those of residents in a control area. Methods: Hazardous air pollutants (PAHs and VOCs) were measured in an exposure area (two sites) and a control area (one site). Urine samples were collected from residents near the industrial complex (184 persons) and residents in the control area (181 persons). Multiple linear regression analysis was used to identify which factors affected the concentration of PAHs and VOCs metabolites. Results: The average atmospheric concentration of PAHs in Shinpyeong-dong and Jangrim-dong was 0.45 and 0.59 ppb for pyrene, 0.15 and 0.16 ppb for benzo[a]pyrene, and 0.29 and 0.35 ppb for dibenz[a,h]anthracene. The average atmospheric concentration of VOCs was 1.10 and 0.99 ppb for benzene, 8.22 and 11.30 ppb for toluene, and 1.91 and 3.05 ppb for ethylbenzene, respectively. The concentrations of PAHs and VOCs in residents near the Shinpyeong·Jangrim Industrial Complex were higher than those of residents in the control area. Geometric means of urinary 2-hydroxyfluorene, 1-hydroxypyrene, methylhippuric acid, and mandelic acid concentrations were 0.45, 0.22, 391.51, and 201.36 ㎍/g creatinine, respectively. Those levels were all significantly higher than those in the control area (p<0.05). In addition, as a result of multiple regression analysis, even after adjusting for potential confounding factors such as gender and smoking, the concentration of metabolites in urine was high in residents near the Shinpyeong·Jangrim Industrial Complex. Conclusion: The results of this study show the possibility of human exposure to VOCs in residents near the Shinpyeong·Jangrim Industrial Complex. Therefore, continuous monitoring of the local community is required for the management of environmental pollutant emissions.
Since ${\beta}-aminoethylphosphonic$ acid was discovered in the living organism, the biosynthesis and biological function of aminophosphonic acids have been extensively studied. The purpose of this project consists in the two parts: 1)the preparation of DL-1-amino-2-phenylethylphosphonic acid (Phenylalanine aminophosphonic acid) and DL-1-amino-3-methylbutyl-phosphonic acid (Isoleucine aminophosphonic acid) by the method of Chamber and Isbell. 2) the study of metabolism and biological functions of those synthetic materials by the animal experiment (white rats) The importance of this project proved to be the first experience fed by animals for the elucidation of biochemical and metabolic functions in the animal body. It was noteworthy that the higher glycogen content in the liver might indicate the significance in the incorporation of phenylalanine aminophosphonic acid into the intermediate of tricarboxylic acid cycle as activated state.
Objectives : To assess the protective effects of wearing protective devices among the residents and volunteers who participated in the cleanup of the Hebei Spirit oil spill. Methods : A total of 288 residents and 724 volunteers were surveyed about symptoms, whether they were wearing protective devices and potential confounding variables. The questionnaires were administered from the second to the sixth week following the accident. Spot urine samples were collected and analyzed for metabolites of 4 volatile organic compounds(VOCs), 2 polycyclic aromatic hydrocarbons(PAHs), and 6 heavy metals. The association between the wearing of protective devices and various symptoms was assessed using a multiple logistic regression adjusted for confounding variables. A multiple generalized linear regression model adjusted for the covariates was used to test for a difference in least-square mean concentration of urinary biomarkers between residents who wore protective devices and those who did not. Results : Thirty nine to 98% of the residents and 62-98% of volunteers wore protective devices. Levels of fatigue and fever were higher among residents not wearing masks than among those who did wear masks(odds ratio 4.5; 95% confidence interval 1.23-19.86). Urinary mercury levels were found to be significantly higher among residents not wearing work clothes or boots(p<0.05). Conclusions : Because the survey was not performed during the initial high-exposure period, no significant difference was found in metabolite levels between people who wore protective devices and those who did not, except for mercury, whose biological half-life is more than 6 weeks.
