Effects of Charcoal Powder on the Growth and Development of Red Pepper and the Changes of Soil Microflora (목탄분말 시용이 고추의 생육 및 토양미생물상 변화에 미치는 영향)

  • It was investigated the possibility to use charcoal powder as beneficial soil conditioner, which used frequently in environmentally friendly agricultural farming system. For this purpose, the effects of charcoal powder on the growth of red pepper and chemical and microbiological properties of soil were also determined. The application of charcoal powder resulted in no significant differences of pH and EC in the soil compared to those of control. However, small particle size of charcoal powder increased yield of red pepper while large charcoal powder resulted in decrease of root growth of red pepper. Furthermore, the application of charcoal powder resulted in changes of soil microflora relating to plant growth stage. The number of the nitrogen fixing bacteria and fungi increased at the early growth stage, while phosphate releasing fungi in the soil increased at the late stage of growth by charcoal powder application. These beneficial effect of charcoal powder on the soil microbial properties was larger by the use of smaller particle size of charcoal powder. Therefore, it indicated that the small size of charcoal powder might be more influential on the red pepper yield and soil microbial properties may be due to large capacity of nutrients uptake for the plant and microorganisms. Additionally the optimal application amount of charcoal powder for the red pepper could be suggested as much as 300kg 10a$^{-1}$ for the both purposes of improvement of crop yield and retardation of the nutrients accumulation by excess charcoal application.

Preparation and Characteristics of Partially Fluorinated-Sulfonated Poly(biphenylene-co-sulfone)ether Membranes for Polymer Electrolyte Membrane Fuel Cell (고분자전해질 연료전지용 부분 불소계 설폰화 Poly(biphenylene-co-sulfone)ether 막의 제조와 특성)

  • A series of partially fluorinated, sulfonated poly(biphenylene-co-sulfone)ether containing perfluorocyclobutane(PFCB) groups were prepared for fuel cell applications through three synthetic steps: synthesis of trifluorovinylether-terminated monomers, thermal cycloaddition and post-sulfonation. Two kinds of trifluorovinylether-terminated monomers were synthesized and statistically copolymerized via thermal cycloaddition to obtain a series of polymers containing 20-60 mol% of biphenyl units(PBS-X). The post-sulfonation of PBS-X was carried out using chlorosulfonic acid(CSA) to obtain copolymers with various sulfonation levels(SPBS-X). All the synthesized compounds, monomers and polymers were characterized by $^1H$-NMR, $^{19}F$-NMR and FT-IR. It was confirmed that the ion exchange capacity(IEC), water uptake and ion conductivity of SPBS-X increased with the increment of sulfonated biphenyl units. Particularly, SPBS-60 showed higher ion conductivity compared to Nafion$^{(R)}$-115 at 25~80 $^{\circ}C$.

Efficacy of Aquacell® Dressing in Partial Thickness Burn Patients (부분층 화상 환자에서 Aquacell®을 이용한 드레싱의 유용성)

  • In partial thickness burn injuries, silver sulfadiazine cream 1%(SSD, $Silvadene^{(R)}$) is the most commonly used topical agent worldwide. But silver sulfadiazine cream 1% has no exudate absorption property. Usually after escar is removed from wound surface, $Silvadene^{(R)}$ is changed to saline wet gauze dressing to promote epithelization. $Aquacel^{(R)}$(ConvaTec, UK) is a 100% sodium carboxymethylcellulose Hydrofiber material. It absorbs exudates directly into the hydrofibers by vertical wicking which allows rapid uptake of liquid into the fibers. The absorbed exudate fluid can be distributed to the entire dressing rather than just over the wound surface, which results in larger fluid absorption capacity. From April, 2003 to July, 2004 a study was done with 40 patients who had variable partial thickness burns. $Aquacel^{(R)}$ dressing was compared in 21 cases to silver sulfadiazine cream 1% and saline wet gauze dressings in 19 cases. In the $Aquacel^{(R)}$ cases, the average healing time on the face was $5.36{\pm}1.69$ a day; on the hands was $8.46{\pm}2.15$ a day; and, on the neck was $6.0{\pm}2.0$ a day. With the $Silvadene^{(R)}$ and Saline wet gauze dressing, the average healing time on the face was $6.44{\pm}1.74$ a day; on the hands was $13.79{\pm}5.35$ a day; and, on the neck was $11.17{\pm}3.31$ a day. As a result, the $Aquacel^{(R)}$ group showed a shorter healing time compared to the $Silvadene^{(R)}$ and saline wet gauze dressing group and patients were satisfied because of less pain and improved comfort. In conclusion, $Aquacel^{(R)}$ is a better choice for partial thickness burn injuries because of shorter healing time, less pain and more confortable dressing.

Comparison of Steel Slag and Activated Carbon for Phosphate Removal from Aqueous Solution by Adsorption (폐수 내 인 흡착 제거를 위한 제강슬래그와 활성탄 비교)

  • This study investigated the potential use of steel slag (SS) (0.5~2.0 g/200 mL) for the removal of phosphate from wastewater compared with activated carbon (AC) (3.0~6.0 g/200 mL). The adsorption equilibrium data were best represented by Langmuir isotherm and its calculated maximum adsorption capacity was 91 mg/g for SS, 27 mg/g for AC. The adsorption kinetics was found to follow the pseudo-second order kinetics model and its rate constant was $0.0232{\sim}0.1357g/mg{\cdot}min$ for SS, $0.0247{\sim}0.1221g/mg{\cdot}min$ for AC. The overall uptake for the SS and AC was maximum at pH 2. Therefore, it can be concluded that steel slag could play an effective role in reducing phosphate concentration compared with activated carbon.

  • Prolific females require better nutrition and feeding practice because of larger litter size and the substantial decline in body fat. Life-time pig output will be compromised if body protein and fat are not properly managed. First litter females are especially vulnerable because they can loose ${\geq}15%$ of whole-body protein. Conservation of body protein mass during first lactation minimizes wean to estrus interval and increases second litter size (up to 1.2 pigs). The ability to influence litter-size by amino acid nutrition is a new dimension in our understanding. A P2 fat depth below 12 mm at farrow and below 10 mm at wean compromised wean to estrus interval (>2 d) and next litter size (0.5 to 1.5 pigs) in sows. It is now clear that a 'modest' excess of feed during the first 72 h of pregnancy decreases embryo viability so that the potential for an increased litter size at birth is not realized. The capacity for milk production by prolific young sows is 25% higher than the standard used previously (NRC, 1988). First litter females averaged 9.82 kg milk/d for a 21 d lactation. Second and third litter counterparts averaged 10.35 kg/d. Milk production was 95% of peak by 10 d of lactation and sows were in greatest negative energy and lysine balance during the first 6 d. Nearly 45% of the total loss in body protein occurred within the first 6 d, but this could reduced to 30-35% by using a more aggressive feeding strategy after parturition. There appear to be 2 phases in lactation for lysine need (d 2-12 vs 12-21). Feeding to the higher level alleviates the second litter size decline. The lysine requirement for lactation can be predicted with accuracy, but we are not able to predict the second limiting amino acid. Mammary uptake of valine relative to lysine and recent work with practical diets suggest that the recent NRC (1998) pattern is realistic and that threonine and valine could be co-limiting for corn-soy diets for prolific sows nursing 10-11 pigs. Empirical studies are needed to refine the ideal pattern so that synthetic lysine can be used with more confidence. Milk fat output for the elite sow is extraordinary and poses an unnecessarily high energetic cost. Methods that reduce mammary fat synthesis will benefit the sow and may enhance piglet growth.

Cytotoxicity Assessments of Portulaca oleracea and Petroselinum sativum Seed Extracts on Human Hepatocellular Carcinoma Cells (HepG2)

  • The Pharmacological potential, such as antioxidant, anti-inflammatory, and antibacterial activities of Portulaca oleracea (PO) and Petroselinum sativum (PS) extracts are well known. However, the preventive properties against hepatocellular carcinoma cells have not been explored so far. Therefore, the present investigation was designed to study the anticancer activity of seed extracts of PO and PS on the human hepatocellular carcinoma cells (HepG2). The HepG2 cells were exposed with $5-500{\mu}g/ml$ of PO and PS for 24 h. After the exposure, cell viability by 3-(4,5-dimethylthiazol-2yl)-2,5-biphenyl tetrazolium bromide (MTT) assay, neutral red uptake (NRU) assay, and cellular morphology by phase contrast inverted microscope were studied. The results showed that PO and PS extracts significantly reduced the cell viability of HepG2 in a concentration dependent manner. The cell viability was recorded to be 67%, 31%, 21%, and 17% at 50, 100, 250, and $500{\mu}g/ml$ of PO, respectively by MTT assay and 91%, 62%, 27%, and 18% at 50, 100, 250, and $500{\mu}g/ml$ of PO, respectively by NRU assay. PS exposed HepG2 cells with $100{\mu}g/ml$ and higher concentrations were also found to be cytotoxic. The decrease in the cell viability at 100, 250, and $500{\mu}g/ml$ of PS was recorded as 70%, 33%, and 15% by MTT assay and 63%, 29%, and 17%, respectively by NRU assay. Results also showed that PO and PS exposed cells reduced the normal morphology and adhesion capacity of HepG2 cells. HepG2 cells exposed with $50{\mu}g/ml$ and higher concentrations of PO and PS lost their typical morphology, become smaller in size, and appeared in rounded bodies. Our results demonstrated preliminary screening of anticancer activity of Portulaca oleracea and Petroselinum sativum extracts against HepG2 cells, which can be further used for the development of a potential therapeutic anticancer agent.

Biosorption of Lead and Cobalt by Absidia coerulea and Thraustochitrium sp. (Absidia coerulea와 Thraustochitrium Sp. 에 의한 납과 코발트의 생물흡착)

  • Nonliving Absidia coerulea and Thraustochitrium sp. were used as biosorbents to remove lead and cobalt that are one of representative pollutant in wastewater and radioactive liquid waste. The optimum pH range for maximum lead and cobalt removal was increased 6.5~11.4 and 8.6~12.0 for Absidia coerulea and 4.2~10.5 and 8.9~11.6 for Thraustochitrium sp. to compared to biosorbent-free control, pH of 8.4~11.2 and 10.5~11.5, respectively. With 1 g biosorbent/L at initial solution pH 5.0. Absidia coerulea and Thraustochitrium sp. took up lead from aqueous solutions to the extent of 104 and 125 mg/g biomass, respectively, whereas Absidia coerulea and Thraustochitrium sp. at initial pH 6.0 took up only 2 and 20 mg/g biomass of cobalt, respectively. For initial 500 mg Pb/L at initial pH 5.0. optimum amount of biosorbent for maximum lead uptake was 0.2 g/L for Absidia coerulea and Thraustochitrium sp., whereas optimum 3.0 g biosorbent/L was needed for initial 200 mg Co/L at initial pH 6.0. Absidia coerulea and Thraustochitrium sp. had higher adsorption capacity for lead than that of cobalt.

  • From ^{15}N$ labelled nitrogen experiments on 13 fields use efficiency by difference method was higher than that by labelling method in 80% of fields tested indicating augmentation of soil nitrogen uptake by fertilizer nitrogen. Both methods showed very similar trend among fields. Sulfur coated urea(SCU) and point application increased fertilization efficiency(yield increment per fertilizer nitrogen applied, Fe) to 23 from 15 of split application through the increase of fertilizer use efficiency from 29(Eu) to 50 but tended to decrease efficiency of absorbed fertilizer nitrogen(yield increment per nitrogen derived from fertilizer, Ef) from 50. to 46 High yielding capacity of Tongil line appears to be attributed to the higher Ef, translocation efficiency and soil nitrogen preference index(soil nitrogen increment in plant per the increment of fertilizer nitrogen in plant, PI). This studies confirmed that yield under fertilizer application system depends on Fe which is the multiplication of Eu and Ef and that the improvement of fertilizer management(form, application method and time) increases principally Eu, the limit of which is controlled by Ef that is attributed mainly to varietal characteristics.

  • Fifteen naked barley cultivars including radiation breeding lines from three places were analized for crude protein, carbohydrates, P, K, Ca, Mg and tested protein by dye binding method and biuret method. Their content and simple correlation analyses among them were as follows. 1. Protein content was 7.67 for average (max. 10.3 in Baegdong, min. 6.0 in Bangju) that was lower than in milled barley and had significant (at p=0.01) correlation with dye binding capacity (r=0.769) and biuret absorbance (r=0.616). 2. Protein content also had significant correlation with $P_2O_5$(r=0.607, p=0.01) and with MgO(r=0.498, p=0.05). 3. There was great difference in protein content among radiation breeding lines(max. 8.40, min. 6.75%). 4. Naked barley appeared to be lower in carbohydrate content but higher in crude ash to compare with milled barley. 5. There was significant correlation(r=0.560, p=0.01) between Ca and K, indicating competition in uptake or translocation to grain. 6. Carbohydrate content showed the highest negative correlation with protein content but it was not significant. 7. The low protein variety (Bangju) showed higher yield than the high protein one (Baegdong) both with (16%) and without (48%) fertilizers.

Sulfonated Polystyrene/PTFE Composite Membranes for Direct Methanol Fuel Cell (직접 메탄올 연료전지를 위한 술폰화 폴리스티렌/테플론 복합막 제조 및 특성연구)

  • For the application of direct methanol fuel cell (DMFC), sulfonated polystyrene/teflon (PS/PTFE) composite membranes were developed by changing monomer ratio of styrene and DVB. The composite membranes were prepared as follows: first, the monomer mixtures consisting of styrene, divinyl benzene and AIBN were impregnated in porous PTFE film and then, polymerized under 8$0^{\circ}C$ to give PS/PTFE membranes. Finally, the membranes were reacted with chlorosulfonic acid in 1,2-dichloroethane to give the sulfonated composite membranes. The measurements of ATR-FTIR, SEM, solvent uptake test and ion exchange capacity (IEC) were done for the resulting membranes before or after sulfonation, respectively, which showed the composite membranes with proper crosslinking degree and sulfonic acid content were prepared well as a function of styrene/DVB ratio. ion conductivity and methanol permeability were studied for the sulfonated membranes. It was found that with decreasing the ratio of styrene/DVB, methanol permeability decreased from $6.6{\times}10^{-7}∼1.3{\timas}10^{-7}$ $\textrm{cm}^2$/s, which are much lower values than that of Nafion$^{(R)}$117($1.02{\times}10^{-6}$ $\textrm{cm}^2$/s). Under the same monomer condition, ion conductivity decreased from 0.11 S/cm ($25^{\circ}C$) to 0.08 S/cm ($25^{\circ}C$), which are similar or a little higher values compared with $Nafion^{(R)}117 (1.02{\times}10^{-6}$ $\textrm{cm}^2$/s, 0.0824 S/cm). These two results confirmed the composite membranes prepared could be applied successfully to DMFC.C.
