Growth of Ulva pertusa Kjellman (Chlorophyta, Ulvophyceae) by a Light-Emitting Diode (LED) Light Source (발광다이오드(LED) 조명을 이용한 녹조 구멍갈파래(Ulva pertusa)의 생장)

  • To determine an efficient growth system for algal cultivation and to develop adequate culture system utilizing LED light, we investigated the effects of fluorescent and light emitting diode (LED) light source on the growth of Ulva pertusa. U. pertusa was cultured at $17^{\circ}C$ under a light intensity of 35 ${\mu}mol$ photons $m^{-2}s^{-1}$ and a 10L:14D photoperiod using either fluorescent or LED lamps. After 1 week of incubation under the same environmental condition, the length and width of Ulva pertusa grown under LED light were 1.5- and 1.9-fold greater, respectively, than the length and width of algae grown under fluorescent light. After two weeks, length and width were 2.6- and 2.0-fold greater, respectively, in algae grown under LED light. Total length and width of Ulva pertusa after three weeks of incubation were 1.7- and 1.2-fold greater in algae grown under LED light than those grown under fluorescent light. Therefore, the LED light induced significantly higher growth of Ulva pertusa than fluorescent light.

Optimal Extracting Conditions Dimethyl-$\beta$-propiothetin from Ulva pertusa (구멍갈파래(Ulva pertusa)로부터 Dimethyl-$\beta$-propiothetin 최적추출조건)

  • This study was focused on the optimal extracting conditions of dimethyl-$\beta$-propiothetin as bioactive substance from green seaweed. Identification and quantification of dimethyl-$\beta$-propiothetin were measured by headspace gas chromatography after conversion to dimethyl sulfide by treatment with saturated NaOH solution. Dimethyl-$\beta$-propiothetin was extracted through various processes (solvent extraction, ultrasonication, boiling and autoclaving) from Ulva pertusa. The content of dimethyl-$\beta$-propiothetin extracted by autoclaving treatment showed higher than that of various extraction methods. Dimethyl-$\beta$-propiothetin content in extract of Ulva pertusa was 325,800 ng/g after autoclaving 121$^{\circ}C$ for 45 min. Dimethyl-$\beta$-propiothetin in exract of Ulva pertusa was comparative stable under low temperature. The retentions of dimethyl-$\beta$-propiothetin content in extract of Ulva pertusa were 76.6~99.8% by incubation at 10~6$0^{\circ}C$ for 2 hours. Chemical decomposition of dimethyl-$\beta$-propiothetin was observed under laboratory conditions at pH values higher than 9.5.

Marine Algal Vegetation of Samchonpo, South Coast of Korea (경남 삼천포 주변 해조류 식생)

  • Marine algal flora and intertidal vegetation around Samchonpo power plant, south coast of Korea, were investigated at three sites, specially referred to the seasonal variation of the species from April to October, 1982. A total of 84 species were identified; 8 green, 19 brown and 57 red algae. Among three sampling sites investigated, 65 species were collected at the first site located in front of the power plant, 66 at the second site, Namildae beach, and 61 at the third, Sinsudo, respectively. The algal vegetation at the first site was dominated in cover by Sargassum thunbergii and Ulva pertusa in April, Corallina spp. Ulva pertusa in July, and Chondria crassicaulis and Ulva pertusa in October. At the second site, it was dominated in cover by Sargassum thunbergii, Gigartina tenella and Ulva pertusa in April, Sargassum thunbergii and Ulva pertusa in July, and Chondria crassicaulis and Ulva pertusa in October, whereas at the third site Sargassum thunbergii and Ulva pertusa in April, and Chondria crassicaulis in October, respectively. The biomass among three investigated sites showed 2,360 g-fresh wt/$m^2$ on an average; 283 g-dry wt/$m^2$ at the first, 277 g-dry wt/$m^2$ at the second, and 256 g-dry wt/$m^2$ at the third site, respectively.

First Record of Ulva pertusa Kjellman (Ulvales, Chlorophyta) in the Pacific Coast of Mexico

  • Based on samplings during 2006-2007 on the intertidal zone of Playa Tampico, Eréndira and Punta Baja, Baja California, Ulva pertusa Kjellman (Ulvales, Chlorophyta) was recorded for the first time for the Pacific coast of Mexico. An alga native to Asia, U. pertusa has just recently been recorded in the southern coast of California (USA). The identification of U. pertusa was based on the observation of the morphology and reproductive characters of the collected specimens. Furthermore, the species identity was confirmed by molecular comparison between nuclearencoded ITS2 sequences of the Mexican samples and those of other Ulva species in GenBnak data. Habitat and distribution of U. pertusa along the Pacific coast of North America are also described. Considering our new report of its occurrence in Mexico, we conclude that U. pertusa is expanding its geographical distribution by trans-ocean introductions.

Antioxidant and Immunomodulatory Effects of Ulva pertusa kjellman on Broiler Chickens (육계에서 구멍갈파래의 항산화 및 면역조절 효과)

  • This study was conducted to test the efficacy of Ulva pertusa kjellman as immunomodulators under lipopolysaccharide (LPS) challenge in broilers by investigating their effects on serum superoxide dismutase (SOD) like ability, immunoglobulin concentration, and splenic cytokine mRNA expression. A total of ninety six1-d-old male broiler chicks (Ross 308) were divided into 4 treatment groups with 8 replicates of 3 birds in each group. NC (negative control, no immune substances), PC (positive control, ${\beta}$-glucan 25 ppm), Ulva P (Ulva pertusa kjellman Powder 3%), and Ulva E (Extract form Ulva pertusa kjellman 0.3%) were added in feed with respective substance. Heparinized venous blood and spleens were collected from five birds per dietary treatment at 5 wk of age. The SOD like activities in Ulva P and Ulva E were significantly increased in comparison with other groups (P<0.05). The immunoglobulin M content was lower in the Ulva E than other groups (P<0.05). Expression patterns of splenic cytokine mRNA in the IL-$1{\beeta}$, IL-2 and IL-6 were similar. Expression rate of IL-$1{\beta}$, IL-2 and IL-6 in Ulva pertusa kjellman (Ulva P, Ulva E) were decreased (P<0.05) in comparison to other groups. Expression rate of IL-$1{\beta}$, IL-2 and IL-6 were significantly lower in groups treated with Ulva E than Ulva P. In conclusion, dietary supplementation of Ulva pertusa kjellman improved SOD like activity and affect immune system by inhibiting inflammatory response in broiler chicks. In addition, it is more effective to use Ulva pertusa kjellman extract than powder for immunomodulatory function. These results suggest the possibility that Ulva pertusa kjellman could be used as the immunomodulator for inflammatory response in broiler chicks.

Bioethanol Production from Seaweed Ulva pertusa for Environmental Application (해조류 구멍갈파래 (Ulva pertusa)의 친환경적 이용을 위한 바이오에탄올의 생산)

  • Ulva pertusa is one of the worst pollutant like a waste vinyl after agriculture and caused bad smell at seashore in Jejudo and south area of korean peninsular. For favorable environmental utilization of Ulva pertusa, it could be applied for ethanol production with its acid hydrolysate. The components of hydrolysate included fermentable sugar of glucose, xylose, mannose, galactose, and higher amounts of unfermentable rhamnose. Fermentable sugars were converted to ethanol with S. cerevisiae, also xylose to ethanol with P. stipitis, their maximun ethanol production at optimum conditions were 462 ${\mu}g$/mL and 475 ${\mu}g$/mL, respectively. While, rhamnose cannot be changed to ethanol with S. cerevisiae or P. stipitis, alone. Combination of S. cerevisiae and P. stipitis can convert rhamnose to ethanol, because P.stipitis degradaded rhamnose to pyruvate, and then S. cerevisiae convert to ethanol, at optimum conditions, ethanol reached to 782 ${\mu}g$/mL (30.24%) that is higher than that of 2 strain alone from 500 mg of dried Ulva pertusa contained 2586.45 ${\mu}g$/mL of reduced sugars. Ulva pertusa can be utilized for renewal energy insted of environmenatal enemy.

Effect of salinity on growth and nutrient uptake of Ulva pertusa (Chlorophyta) from an eelgrass bed

  • The effects of salinity on various ecophysiological parameters of Ulva pertusa such as growth, nutrient uptake, photosynthetic performance and internal nutrient composition were tested. U. pertusa was collected from an eelgrass bed in a semi-protected embayment on the southwest coast of Korea. Under salinity regimes from 5 to 40 psu, the specific growth rates $(\mu)$ of U. pertusa ranged from 0.019 to $0.032\;d^{-1}$. Maximum growth rate was observed at 20 psu, and minimum at 40 psu. This species showed various uptake rates for nitrate and phosphate. Nutrient uptake was noticeably higher at intermediate salinity levels, and lower at both extremes. Salinity significantly influenced chlorophyll-$\alpha$ content and effective quantum yield. Tissue nitrogen content ranged from 1.5 to 2.9% N (dry weight), whereas tissue phosphorus ranged from 0.1 to 0.14% P (dry weight). The N : P ratio in the tissue of U. pertusa was considerably higher, ranging from 30 to 50. Increased growth at lower salinity suggests that the initial growth rate of U. pertusa is greater during the rainy season (i.e., late spring and early summer) than any other season during the year. The appearance of an Ulva bloom in eelgrass beds may be triggered by salinity more than by other environmental factors such as light and temperature.

Hydrothermal Pretreatment of Ulva pertusa Kjellman Using Microwave Irradiation for Enhanced Enzymatic Hydrolysis (구멍갈파래의 효소 가수분해 증진을 위한 마이크로파 이용 열수 전처리)

  • The green algae have cellulose as a main structural component of their cell wall and the cellulose content in green algae is much higher than other marine algae such as brown algae and red algae. Furthermore, green algae do not contain lignin in their cell wall and store starch as food in their plastids. Thus, it was investigated that the effect of hydrothermal pretreatment process utilizing microwave irradiation for Ulva pertusa Kjellman, a division of green algae, which is expected to be utilized for bioenergy production, on the enzymatic hydrolysis. The hydrothermal temperature have an effect on the pretreatment of Ulva pertusa Kjellman, but the effect of power of microwave irradiation is negligible. The rate of enzymatic hydrolysis was increased as the hydrothermal temperature increased until $140^{\circ}C$. The enzymatic hydrolysis of pretreated Ulva pertusa Kjellman under the optimum pretreatment conditions (50 W of microwave irradiation power and $150^{\circ}C$ of hydrothermal temperature) with cellulase, ${\alpha}$-amylase, and Novozyme 188 having ${\beta}$-glucosidase acitivity resulted in the saccharification of 96 wt% of total carbohydrate in Ulva pertusa Kjellman during 3 hrs, while it took 24 hrs for the enzymatic hydrolysis of untreated Ulva pertusa Kjellman. It confirmed that the hydrothermal pretreatment was effective on Ulva pertusa Kjellman for the enzymatic hydrolysis.

Adsorption Characteristics of the Heavy Metals, Cd(II) and Pb(II) Ions, on the Si-Immobilized Ulva pertusa (실리카고정 구멍갈파래에 대한 중금속 Cd(II), Pb(II)이온의 흡착 특성)

  • Adsorption characteristics of heavy metal ions, Cd(II) and Pb(II), on eastcoast-living algae, ulva pertusa, has been studied in our experiment. The Maximum adsorption amount of Cd(II) and Pb(II) ions on 1 g of the ulva pertusa were 2.3 mg, 3.1 mg in alkaline and 2.0 mg, 2.8 mg in acidic solution. However 3.4 mg, 7.3 mg in alkaline and 3.1 mg, 6.5 mg in acidic solution were shown on the Si-immobilized ulva pertusa in the same condition. Thus, Si-immobilized ulva pertusa adsorbs more amount of heavy metals, Cd(II), Pb(II), than the ulva pertusa, and more effective absorbent in alkaline. Furthermore, more amounts of Pb(II) ion were absorbed compare to Cd(II) ion in our work. Recovery ratio of Cd(II) and Pb(II) ions on the ulva pertusa were 55.0~61.0%, 59.7~66.8% respectively and 87.6~97.5%, 83.5~99.3% on the Si-immobilized ulva pertusa.
