Ultrastructural Changes of the Bruch's Membrane and the Pigment Epithelial Cells of the Mouse Retina with Age (노화에 따른 마우스 망막의 바닥복합층과 색소상피세포의 미세구조 변화)

  • To study the age-related morphological differences of the retinal pigment cells and Bruch's membrane of mouse, retinae of one week-old, five weeks-old, eight weeks-old, six months-old, twelve months-old, eighteen months-old, twenty-four months-old and thirty months-old ICR mice were dissected out under anesthesia. Pieces of the tissue taken from the posterior region of the retina were fixed in 2.5% glutaraldehyde-1.5% paraformaldehyde (0.1 M Millonig's phosphate buffer, pH 7.3), and 1% osmium tetroxide (0.1 M Millonig's phosphate buffer, pH 7.3), and embedded in araldite mixture. The ultrathin sections were stained with uranyl acetate and lead citrate, and were observed under a JEM 100 CX-II electron microscope. Observed results were as follows: 1. Retinae of one week old mouse exhibit that some parts of the pigment cell provided with basal foldings, whereas other parts of the one contain without basal foldings. After (ive weeks-old, all retinal pigment cells have the basal infoldings. 2. In the one week-old, stage 1 and stage 2 melanosomes were observed in the retinal pigments cells, but after five weeks-old, most of the retinal pigment cells contain some matured stage melanosomes (stage III and stage IV). 3. The phagosomes in the retinal pigment cells were increased during aging. 4. After eighteen months-old, electron dense materials are observed within the basal infoldings. 5. After eighteen months-old, the thickness of the Bruch's membrane is prominently increased. The thickness of the basal laminae of the pigment cell and the choriocapillary endothelium is more prominently increased as compared with that of the other components of the Bruch's membrane. 6. The thickness of the basal lamina of the pigment cell is more prominently increased as compared with that of the choriocapillary endothelium on aging. From the above results, it was suggested that the pigment cell and Bruch's membrane matures structurally In five weeks, and the function of the pigment cell is prominently suppressed around eighteen months-old, and thereafter the functional suppression is continued on aging.

A Quantitative Ultrastructural Study on the Effects of Ischemia and Reperfusion on the Rat and Cat Hearts (허혈 및 재관류가 흰쥐 및 고양이 심장에 미치는 영향에 관한 형태계측학적 연구)

  • To understand the structural changes of the myocardial myocytes and endothelial cells in ischemic and reperfused heart, and to elucidate their roles in those conditions, the authors observed cat and rat myocardium ultrastructurally and evaluated them with morphometric techniques. In cat, mild ischemia and moderate degree reperfusion injury was induced by ligation of the anterior interventricular branch of left coronary artery and reperfusion. In rat, severe ischemia and irreversible reperfusion iniury was made using in vitro Langendorff techniques. In normal cat myocytes, the volume densities of cytoplasm, myofibrils, mitochondria, sarcoplasmic reticulum and T tubules were $0.11{\pm}0.013,\;0.51{\pm}0.096,\;0.25{\pm}0.082,\;0.09{\pm}0.008,\;0.02{\pm}0.010$ (Mean${\pm}$S.D.) respectively, and the myofibril/mitochondria ratio was $2.33{\pm}1.379$. The numerical density and average volume of mitochondria were $0.76{\pm}0.210/{\mu}m^3$ and $0.33{\pm}0.057{\mu}m^3$ respectively. In normal cat endothelial cells, the volume densities of cytoplasm, cytoplasmic vesicles, tubular systems (including endoplasmic reticulum and Golgi apparatus) and mitochondria were $0.43{\pm}0.023,\;0.28{\pm}0.007,\;0.22{\pm}0.021,\;0.03{\pm}0.014$ respectively. The mean thickness of endothelial cells was $230{\pm}45.2{\mu}m$. The numerical density and average volume of cytoplasmic vesicles were $508{\pm}55.0/{\mu}m^3,\;578{\pm}104.8nm^3$ respectively. In cat myocytes which received mild ischemic injury, the volume densities of organelles were not changed significantly in ischemic and reperfusion states. In reperfusion group myocytes, the numerical density of mitochondria was decreased significantly and the average volume was increased significantly. In endothelial cells, the volume density of tubular system in ischemic group and the average volume of cytoplasmic vesicles in reperfusion group were increased significantly. In rat myocytes which received severe ischemic injury, the volume density and average volume of mitochondria were increased significantly, and the volume density of sarcoplasmic reticulum and numerical density of mitochondria were decreased significantly in both ischemic and reperfusion groups. In ischemic and reperfused endothelial cells, the volume density and numerical density of cytoplasmic vesicles, the volume density of cytoplasm were decreased significantly. The volume densities of tubular system were increased significantly in both ischemic and reperfused groups. The volume density of mitochondria in ischemic group and the average volume of cytoplasmic vesicles in reperfusion group showed significant increase. The authors, based on the above observations, conclude that the mitochondria of myocytes and the cytoplasmic vesicles of endothelia are the first group of targets in ischemic and reperfusion injury and in this respect, the degree of ischemic insult is not significant. The role of myocyte mitochondria in reperfusion injury may be insignificant, but endothelial cells may contribute actively to reperfusion injury.

Ultrastructural Study on the Thymus following the Administration of 5-Fluoruracil or Mitomycin (5-Fluorouracil 및 Mitomycin이 가슴샘의 미세구조에 미치는 영향)

  • The experiment was performed to study the morphological responses of the thymus of the mice, to antitumour agents (5-Fluorouracil or mitomycin C). Healthy adult mice weighing 25 gm each were divided into normal and experimental groups. 5-Fluorouracil (60 mg/kg) or Mitomycin-C $(400{\mu}g/kg)$ were injected subcutaneously to the animals every other day. Animals were sacrificed at 4 days and 7 days following the first injection. Pieces of the tissue taken from the thymus were prefixed with 2.5% paraformaldehyde-l.5% glutaraldehyde, followed by post-fixation with 1% osmium tetroxide. Ultrathin sections stained with uranyl acetate and lead citrate were observed with a JEM 100 CX-II electron microscope. The observed results were as follow: 1. Apoptoses of T-lymphocytes were observed more frequently in the thymus of the experimental groups than in those of a normal group. 2. In the experimental group, the plasma cells with distended cisternae of the granular endoplasmic reticulum and the eosinophile leukocytes were observed frequently. 3. In the experimental group, newly forming Hassall's corpurscles were observed frequently. 4. In the mitomycin-treated group, the epithelial reticular cells containing distended perinuclear cisternae, distended the granular endoplasmic reticula and pyknotic nuclei were observed in the cortico-medullary junctional area. 5. In the mitomycin-treated group, nuclear bodies with medium electron dense materials were often observed in the T lymphocyte. 6. In the 5-fluorouracil-treated groups, fused and dissolved tonofilament bundles and apoptotic bodies were observed in the some epithelial reticular cells in the medullary area. 7. In the 5-fluorouracil-treated groups, some elongated and bar-shaped lysosomes with electron lucent gap were often observed in the macrophages. 8. In the 5-fluorouracil-treated group, membrane complex of the smooth endoplasmic reticulum were ofen observed in the macrophage. From the above results, it was suggested that 5-fluorouracil or mitomycin could induce rapid involution of the thymus, and disturb maturation and differentiation of T lymphocytes, and, in turn, supress immunity.

Protective Effects of Chitosanoligosaccharide on Gamma Ray-Induced Hepatic Damage in Mice (생쥐에서 키토산올리고당의 감마선 저항 효과)

  • The purpose of this study was to evaluate the effect the radiation-resistance of chitosan on the mice. A healthy male ICR mice were used for experiment. The SOD and MDA activity was measured from the liver of mice at 48 and 72 hours after the irradiation. The ultrastructural changes of the liver by irradiation was observed at 24, 48 and 72 hours after irradiation. The experimental groups were divided into three groups. Group 1 was the control group which was not treated with chitosanoligosaccharide before or after iradiation. Group 2 was the prefeeding group which chitosanoligosaccharide solution was supplied by feeding ad libitum for 30 days before irradiation. Group 3 was the postfeeding group which chitosanoligosaccharide solution was supplied by feeding after irradiation. In all groups 10 mice were used. The results were as follow: The SOD and MDA activity of the prefeeding group was decreased significantly (P<0.01). Control group - The nuclei were condensed. The mitochondria and rough endoplasmic reticulum (rER) were extended and the ribosome was dropped from the rER. Prefeeding group - The nuclei were rounded. The mitochondria was elongated. And the rER attached ribosomes. Postfeeding group - The nuclei were slightly condensed. The mitochondria and the rER were extended and the ribosome was dropped from the rER. It was concluded that the chitosanoligosaccharide was effective in the radiation-protection. So, chitosan would have the potential as the radiation-protection materials.

  • Morphological difference of the renal glomerulus at different age groups have been studies in one week-old, five weeks-old, eight weeks-old, six months-old, twelve months-old, eighteen months-old, twenty-four months-old, and thirty months-old ICR mice. Pieces of the tissue taken from the renal corticies were fixed in 2.5% glutaraldehyde-1.5% paraformaldehyde solution (0.1 Millonig's phosphate buffer pH 7.3), and 1% osmium tetroxide solution (0.1 M Millonig's phosphate buffer, pH 7.3), and were embedded in araldite mixture. The ultrathin sections were stained with uranyl acetate and lead citrate solution, and were observed under a JEM 100CX-II electron microscope. The results were as follows: 1 In the one week-old mouse, thicknesses of the three layers of the glomeluar basal lamina (lamina densa, lamina rara interna and lamina rara externa) are similar, but in the five weeks-old mouse, thick lamina densa becomes a greater portion of the thickness of whole glomerular basal lamina. 2. No difference was noticed between thickness of the renal glomerular basal lamina of the five weeks-old mouse compare with that of the one week-old one, but basal lamina of the eight weeks-old one is thickened considerably and thicknesses were maintained through twelve months-old one. After eighteen months, the thickness of the glomerular basal lamina is increased remarkably. 3. After eighteen months, electron dense deposits within the basal lamina of the renal glomeruli are observed frequently. 4. Amount of the microfilaments in the mesangial cells and the mesangial matrices are increasing during aging. 5. The thicknesses of the basal laminae of the Bowman's capsule are increasing during aging. 6. After twenty four months, the proximal tubular cell-like parietal cells with well developed microvilli are observed frequently. From the above results, it was suggested that the renal glomerulus matures structurally in five weeks, and the function of the glomerulus is suppressed after eighteen months.

Occurrence of Nuclear Inclusions in Plant Cells (식물세포 내 핵 함유구조 발달 양상)

  • The occurrence of nuclear inclusions has been reported in various plant groups from primitive ferns to higher flowering plants. Their presence within a group seems to be randomly distributed without any phylogenetic relationships among species. According to the current survey, nuclear inclusions have been widely documented in more than several hundreds of species from various families of plants. The morphology and internal structures of nuclear inclusions are diverse and at least five types of inclusions develop within plant nuclei; amorphous, crystalline, fibrous, lamellar, and tubular form. Among these types, crystalline inclusions are the ones that are the most frequently reported. The inclusions are not bound by membranes and appear to be related to the nucleoli, either spatially by a close association or by an inverse relationship in size during development. The idea that nuclear inclusions are of a proteinaceous nature has been widely accepted. Further link to nucleolar activity as a protein storing site has also been suggested based on the association between the nucleolus and nuclear inclusions. Various investigations of nuclear inclusions have revealed more information about their structural features, but characterizing their precise function and subunit complexity employing molecular analysis and 3-D reconstruction remains to be elucidated. Tilting and tomography of serial sections with appropriate image processing can provide valuable information on their subunit(s). The present review summarizes discussion about different nuclear inclusions in plants from previous works, giving special attention to their fine, ultrastructural morphology, function, and origin.

Ultrastructure of the Fertilized Egg Envelope in Cichlasoma managuensis, Cichlidae, Teleost (경골어류 시클리드과 Cichlasoma managuensis의 수정란 난막 미세구조)

  • The ultrastructure of the fertilized egg envelope in Cichlasoma managuensis belonging to Cichlidae were investigated by routine light and electron microscopes. The fertilized eggs of Cichlasoma managuensis was of the light yellow, non-transparent, ellipsoidal, adhesive and nonfloted type. The size of fertilized egg was the major axis $1.92{\pm}0.08\;mm$, the minor axis $1.43{\pm}0.04\;mm$. The egg envelopes have a single micropyle, which is thought to the pathway of sperm in the area of the animal pole. An outer surface of fertilized egg envelope was covered by a adhesive reticular structures and the fertilized egg envelopes consisted of two distinct layers; an outer adhesive layer and an inner layer of $13{\sim}15$ horizontal lower electron density lamellae alternating with interlamellae of higher electron density. The external shape of fertilized egg is common trait of fishes belonging to Cichlidae and these ultrastructural characters of fertilized egg envelope can be utilized in taxonomy of teleost.

Effects of Calcium Chelator on in vitro Fertilization of Rat Oocyte (흰쥐 난자의 체외수정에 미치는 Calcium Chelator의 영향)

  • In order to examine the effect of $Ca^{2+}$ -chelation on in vitro fertilization of rat zona-free oocyte, the formation of cortical granule envelope (CGE) and the rate of fertilization related to monospermy and polyspermy were determined. The ultrastructural characteristics of oocytes were observed with the scanning electron microscope and BAPTA/AM was used for calcium-chelation. The CGE formed by cortical reaction was observed in zona-free oocyte inseminated in vitro and it was also observed in the calcium chelator (1, 5, 10$\mu$M BAPTA/AM) treated zona-free oocytes inseminated in vitro. The CGE developed according to incubation time. The fertilization rate was decreased in the calcium chelator-treated group (59.8, 38.1, 37.0%) compared to the control group (60.6%) but monospermy rate was increased in the calcium chelator-treated group (45.0, 47.3, 50.9%) compared to control group (37.5%). The above results demonstrate that the CGE is formed during fertilization in rat and the extracellular calcium is used in cortical reaction. Also the results suggest that proper concentration of free calcium in oocyte acts as important factor in fertilization.n.

  • The androgenic gland secretes a hormone, androgenic gland hormone, which is believed to act on the differentiation of the primary, secondary, and behavioral sex characteristics in most malacostracan crustaceans. This report presents the ultrastructural morphology of the androgenic gland in the freshwater prawn Macrobrachium nipponense. This gland, located in the coxopodite of the last pair of walking legs, was attached to the subterminal region of the sperm duct. The gland was composed of simple cellular strands, encased by a fibrous sheath. Microvilli were situated in the fibrous sheath, especially at the edge of each cellular strand. The androgenic gland cells had the large and round nucleus and rough endoplasmic reticulum arranged either in spirals or in concentric circles throughout the cytoplasm of the cell. They also had the well-developed Golgi complex and long mitochondria with flat and transverse cristae. The Golgi complex was similar to microvesicular cluster, but usually in the shape of typical dictyosomes, These features of androgenic gland cells coincides well with the protein/peptide secretion in their function. However, despite the apparent ultastructual equipment for protein/peptide secretion, no accumulation of materials secreted were noticed in the cytoplasm. Therefore it is strongly suggested that the transient transportation of the materials into the hemocoel has occurred just after synthesis.

An Ultrastructural Study on the Effect of Exogenous $Ca^{2+}$ Stimulation to Ischemic Myocardium during Post-ischemic Reperfusion (재관류시 외인성 칼슘 자극이 허혈 심근에 미치는 영향에 대한 전자현미경적 연구)

  • The effects of exogenous $Ca^{2+}$ stimulation on the post-ischemic myocardial cells were studied using isolated Langendorff-perfused guinea pig hearts. At the starting point of reperfusion, Tyrode solutions, each containing 2.0mM, 4.0mM and 8.0mM $CaCl_2$ respectively, were administered for 2 minutes apart by descending, ascending, or by combined sequences followed by standard Tyrode solution containing 1.0mM $CaCl_2$. The minutes of global ischemia produced reversible but moderate to severe degree of myocardial ultrastructrual changes including focal destruction of sarcolemma, loss of nuclear matrix, clumping and margination of chromatins, mitochondrial swelling, destruction of mitochondrial cristae, shortening of sarcomeres, focal loss of myofibrils, and separation of cell junctions. In spite of reperfusion, the ultrastructure was more severely damaged and irreversible changes such as intracellular fluid accumulation, contracted sarcomeres, mitochondrial destruction, disruption of sarcolemma, loss of nuclear matrix, and separation of cell junction were observed in a large number of cells. In contrast, Tyrode-perfused $Ca^{2+}$-stimulated myocardial cells showed relatively well preserved ultrastucture, except slight changes including focal mitochondrial swelling, widening of T-tubule, and widening of cell junctions, especially at fasciae adherentes. The post-ischemic $Ca^{2+}$-stimulated reperfused myocardial cells produced focal changes such as mitochondrial destruction, disintegration of sarcolemma, widening of T-tubule, and intracellular fluid accumulation with slight variation in degree of changes by the method of $Ca^{2+}$ administration sequence. However, in a large number of the myocardial cells, chromatins were redistributed relatively evenly in the nuclear matrix, mitochondrial cristae were tightly packed, and a considerable number of intramitochondrial granules and glycogen granules reap-pealed. These results indicate that exogenous $Ca^{2+}$ stimulation in the initial period of reperfusion may be beneficial to salvage or to reduce the post-ischemic myocardium from further deleterious changes, and that the beneficial effects may be derived from the reserves of the function of the intracellular $Ca^{2+}$ regulating organelles and/or from the responsiveness of contractile apparatus to $Ca^{2+}$ stimulation.
