Effect of Fluoride and Laser on Artificial Caries-like Lesion Formation in Bovine Enamel (불소 및 레이저가 우치법랑질의 인공우식병소에 미치는 영향)

  • The purpose of this study was to examine the morphological characteristics and combined effects of fluoride application and laser irradiation on artificial caries-like lesion formation in bovine enamel. Enamel specimens were divided into five experiment group and placed in no-treatment(group C), APF application alone(group F), laser irradiation alone(group L), APF application before laser irradiation (group FL), and APF application after laser irradiation(group LF) on artificial caries-like lesion. Sound enamel was used as a control group. The ultrastructural changes and physical effects of the fluorided and lased enamel has been investigated by using SEM, enamel solubility and microhardness test as well as distributions of calcium, phosphorus and fluoride in internal enamel by using EPMA. The following results were obtained. 1. In the all experiment groups, the amounts of dissolved calcium of enamel surfaces significantly decreased according to increasing exposure time of acid solution than control group(P<0.001). Group L showed higher than that group FL and LF in 30 and 60min(P<0.05). 2. The microhardness values of enamel surface in the control group was highest than that in the other experiment groups. Group F, L, FL and LF were significantly increased than group C(P<0.001). The enamel surface treated with APF produced deposites of numerous small globules and lased enamel showed a cracker-like appearance with microcrack and small pore. Numerous deposits were infiltrated in the fissured portion of enamel treated with APF after laser irradiation. 4. In the case of APF application alone, the elevation of the fluoride profile can be seen within $5{\mu}m$ of the outermost layer and a similar profile observed in the specimen treated with APF before laser irradiation. However, the specimen treated with APF after laser Irradiation showed a large elevation within $10{\mu}m$ of the outermost layer of the enamel. 5. The higher Ca/P ratios were observed in $10{\mu}m$ depth of lased and fluorided enamel when compared to the sound and carious enamel. The fluoride content decreased rapidly with distance from enamel surface, in the group F, fluoride concentration was significantly higher than that in the group C, L, FL, LF and control group according to increasing enamel depth (P<0.05).

  • Ionizing radiation is enough energy to interact with matter to remove orbital electrons, neutrons, and protons in the atom. Ionizing radiation like this leads to oxidizing metabolism that alter molecular structure through direct and indirect interactions of radiation with the deoxyribonucleic acid in the nucleus and cytoplasmic organelles or via products of cytoplasm radiolysis. These ionization can result in tissue damage and disruption of cellular function at the molecular level. Consequently, ionizing radiation-induced modifications of ion channels and transporters have been reported. When the harmful effects exceed those of homeostatic biochemical processes, induced biological changes persist and may be propagated to progeny cells. Also, Reactive oxygen species formed on the effect of ionizing radiation can get across into neighboring cells through the cell junctions that are responsible for intercellular chemical communication, and may there bring about changes characteristic to radiation damage. Depending on radiation dose, dose-rate and quality, these protective mechanisms may or may not be sufficient to cope with the stress. This paper briefly reviewed reports on ionization radiation effects on cellular level that support the concept of radiation biology. A better understanding of the biological effects of ionizing radiation will lead to better use of and better protection from radiation.

A Study in Bridging Sciatic Nerve Defects with Combined Skeletal Muscle and Vein Conduit in Rats (백서의 좌골신경에서 정맥 및 골격근을 이용한 결손신경 봉합술에 대한 연구)

  • A peripheral nerve when approximation of the ends imparts tension at the anastomosis and with a relatively long segment defect after excision of neuroma and neurofibroma cannnot be repaired by early primary suture. The one of the optimistic reconstruction method of severed peripheral nerves is to restore tension-free continuity at the repair site putting an autogenous nerve graft into the neural gap despite of ancipating motor or sensory deficit of the donor nerve area. To overcome the deficit of the autogenous nerve graft, several other conduits supplying a metabolically active environment which is able to support axon regeneration and progression, providing protection against scar invasion, and guiding the regrowing axons to the distal stump of the nerve have been studied. An author have used ipsilateral femoral vein, ipsilateral femoral vein filled with fresh thigh muscle, and autogenous sciatic nerve for the sciatic nerve defect of around 10 mm in length to observe the regeneration pattern in rat by light and electron microscopy. The results were as follows. 1. Light microscopically regeneration pattern of nerve fibers in the autogenous graft group was more abundant than vein graft and vein filled with muscle group. 2. On ultrastructural findings, the proxial end of the graft in various groups showed similar regenerating features of the axons, myelin sheaths, and Schwann cells. The fascicular arrangement of the myelinated and unmyelinated fibers was same regardless of the type of conduits. There were more or less increasing tendency in the number and the diameter of myelinated fibers correlated with the regeneration time. 3. In the middle of the graft, myelinated nerve fibers of vein filled with muscle group were more in number and myelin sheath was thinner than in the venous graft, but the number of regenerating axons in autogenous nerve graft was superior to that in both groups of the graft. The amount of collagen fibrils and amorphous materials in the endoneurial space was increased to elapsed time. 4. There was no difference in regenerating patterns of the nerve fibers of distal end of the graft. The size and shape of the myelinated nerve fbers were more different than that of proximal and middle portion of the graft. From the above results, the degree of myelination and regenerating activity in autogenous nerve is more effective and active in other types of the graft and there were no morphological differences in either ends of the graft regardless of regeneration time.

Ultrastructural Characteristics and Categorization of the Hemocytes in Spiny Top shell, Batillus cornutus (Lightfoot, 1786) (Gastropoda: Turbinidae) (소라, Batillus cornutus (Lightfoot, 1786) 혈구의 종류 및 미세구조적 특징)

  • Light and transmission electron microscopy of Batillus cornutus hemocytes revealed differences that the morphological distinctions between blast-like cell, granulocytes and hyalinocytes. Base on the morphological characteristics of the cells, we identified the eight types of hemocytes and present a categorization of the hyalinocytes into six sub-categories. The hemocytes of B. cornutus were observed basophilic cell under the light microscopy. Blast-like cells had a spherical profile with a central nucleus filling almost the whole cell. Granulocytes were characterized by presenting variable numbers of granules. This cell had spherical shape with diameter 7 ${\mu}m$ and smooth endoplasmic reticula, granules, mitochondria, glycogen granules in the cytoplasm. Hyalinocytes were the most abundant cell type. Especially, hyalinocyte VI had iirregular an amoebal shape and observed autophagosome and heterophagosome in the cytoplasm. From these results, it is concluded that there are eight types of cells in the hemolymph of B. cornutus. Further studies are now needed to identify the role of these hemocytes in the enzymological and immunological response.

Senescence as A Consequence of Ginsenoside Rg1 Response on K562 Human Leukemia Cell Line

  • Aims and Background: Traditional chemotherapy strategies for human leukemia commonly use drugs based on cytotoxicity to eradicate cancer cells. One predicament is that substantial damage to normal tissues is likely to occur in the course of standard treatments. Obviously, it is urgent to explore therapies that can effectively eliminate malignant cells without affecting normal cells. Our previous studies indicated that ginsenoside $Rg_1$ ($Rg_1$), a major active pharmacological ingredient of ginseng, could delay normal hematopoietic stem cell senescence. However, whether $Rg_1$ can induce cancer cell senescence is still unclear. Methods: In the current study, human leukemia K562 cells were subjected to $Rg_1$ exposure. The optimal drug concentration and duration with K562 cells was obtained by MTT colorimetric test. Effects of $Rg_1$ on cell cycle were analyzed using flow cytometry and by SA-${\beta}$-Gal staining. Colony-forming ability was measured by colony-assay. Telomere lengths were assessed by Southern blotting and expression of senescence-associated proteins P21, P16 and RB by Western blotting. Ultrastructural morphology changes were observed by transmission electron microscopy. Results: K562 cells demonstrated a maximum proliferation inhibition rate with an $Rg_1$ concentration of $20{\mu}\;mol{\cdot}L^{-1}$ for 48h, the cells exhibiting dramatic morphological alterations including an enlarged and flat cellular morphology, larger mitochondria and increased number of lysosomes. Senescence associated-${\beta}$-galactosidase (SA-${\beta}$-Gal) activity was increased. K562 cells also had decreased ability for colony formation, and shortened telomere length as well as reduction of proliferating potential and arrestin $G_2$/M phase after $Rg_1$ interaction. The senescence associated proteins P21, P16 and RB were significantly up-regulated. Conclusion: Ginsenoside $Rg_1$ can induce a state of senescence in human leukemia K562 cells, which is associated with p21-Rb and p16-Rb pathways.

Effects of Scarification and Water Soaking Treatment on Germination of Hard-Seeded Legumes (두과 작물의 경실종자 발아촉진에 대한 종피연화처리의 효과)

  • The crops showing hard seed character have high seed viability after seed storage for long period. The low germination rate due to hard seed coat, however, cause a problem of low seedling establishment in field condition. Three legumes used in the experiment, lablab bean (Dolichos lablab L.), asparagus bean (Vigna sesquipedalis L. Fruwirth), and soybean (Glycine max L. Merr.), showed low germination rate (26, 17, and 5%, respectively) due to thick and hard seed coat. In this study artificial treatment for breaking dormancy was tested in hard seeds. The effect of proper treatment was various depending on species. The germination rate of lablab bean was highly improved up to 94% by soaking into water for 24 hours. In the case of asparagus bean, the rate was increased up to 90% by soaking for eight hours near boiling water until it cools. The germination rate of small hard seed soybean was increase to 96% by soaking into concentrated sulfuric acid for 10 minutes. Ultrastructural change revealed by scanning electron microscope (SEM) reflects that the structure of micropyle was changed and water uptake was facilitated with all treatments tested in the experiment. Especially, sulfuric acid treatment resulted in the degradation of micropylar tissue. These results demonstrate that the artificial treatment including sulfuric acid and (hot) water soaking treatment for promoting water uptake can be applied to improve seed germination in legume seed with thick and hard seed coat.

  • Spermatogenesis and reproductive cycle of the razor clam, solen grandis, were investigated monthly by histological and cytological observations. Samples were collected from natural intertidal population at Oshik-do, Kunsan, Korea, for one year, beginning from January to December, 1993. solen grandis is dioecious. Morphological structures of the spermatozoon of this species ar esimilar to those of other bivalve spermatozoa having a primitive type; i.e., a small head, a cap-shaped acrosome and a short mid-piece with four mitochondria surrounding axial filament. The head of spermatozoon is approximately 2 \mu m in length and sperm tail is about 20 \mu m long. The axoneme of tail flagellum consists of nine pairs of peripheral microtubules at the periphery and a pair of central microtubules at the center. Four spherical mitochondria form the paranucleus. Spawning occures once a year between early June and July, and the main spawning was observed in July when seawater temperature reaches above 20 \circ C. The reproductive cycle of male razor clam can be divieded into fivesuccessive stages; early active (December to january), late active (January to march), mature (March to early August), partially spawned (June to July), and spent/inactive stage (August to December).

  • The acinar cells and secretory granules of the parotid salivary gland were examined in the lesser white toothed shrew, Crocidura suaveolens and the big white-toothed shrew, C. lasiura. The parotid gland of both species were a serous gland having only one kind of serous acinar cells, and had conventional arrangement of acini and intercalated, granular and striated ducts. In case of C. suaveolens, serous acinar cells had well developed rER, prominent Golgi complex, several large mitochondria and abundant moderate dense secretory granules with various stages of the maturing or fusing process. Immature acinar secretory granules were only or mainly filled with fine strong dense specks and had an indistinct limiting membrane, and mature granules were filled with homogeneous pale large round center and had fine strong dense specks at the periphery of the homogeneous pale center and a distinct limiting membrane. In case of C. lasiula, serous acinar cells had well developed rER, prominent Golgi complex, several large mitochondria and abundant dense secretory granules with maturing or fusing process. Immature acinar secretory granules were only filled with pale rough specks and had an indistinct limiting membrane, and mature granules were only filled with rough dense specks and had a distinct limiting membrane. Eventually The acinar secretory granules of C. suaveolens were seen moderate at the light and ultrastuctural level, those of C. lasiura were strong dense at the light microscopic level and dense at the ultrastructural level.

An Ultrastructural Study on the Renal Glomeruli of the Aging Rat (연령에 따른 흰쥐 신사구체의 변화에 대한 전자현미경적 연구)

  • Morphological difference of the renal glomerulus at different age groups have been studied in young (three month-old), adult (twelve month-old) and old (thirty month-old) Fisher strain 344 rats. Pieces of the tissues were taken from renal corticies prefixed with 2.5% glutaraldehyde-1.5% paraformaldehyde (0.1 M Millonig's phosphate buffer, pH 7.3), following by post-fixation with 1% osmium tetroxide (0.1 M Millonig's phosphate buffer, pH 7.3) and embedded within Araldite. The ultrathin sections contrasted with uranyl acetate and lead citrate were observed under a JEM 100CX electron microscope. The mean thickness of glomerular basal lamina and Bowman's capsule were determined by measuring the thinnest portion of basal lamina, and by taking the average of 50 readings from electron micrographs at different ages. The numerical changes of the slit pores were compared based upon the numbers over the length of 10um of glomerular basal lamina. The results were as follow: 1. The thickness of glomerular basal lamina is increased during aging; 140.4 nm in young rats, 270.0 nm in adult ones, and 437.8 nm in old ones. 2. The thickness of basal lamina of parietal cells of Bowman's capsule is 187.5 nm in young rats, 914.0 nm in adult ones, and 2850.0 nm in old ones. 3. The numbers of the slit pores of basal lamina are reduced during aging, 30.3 slit pores/$10{\mu}m$ in adult ones, and 24.2 slit pores/$10{\mu}m$ in old ones. 4. Accumulation of dense intracytoplasmic filamentous material in the parietal cells of Bowman's capsule is increased in the vicinity of the basal lamina during aging. The proximal tubule-like epithelial cell in Bowman's capsule is observed at one glomerulus in a young rat. 5. The endothelial cells are edematous and form balloon-like structure protruding into capillary lumen in young and old rats. 6. Cytoplasm of the podocyte shows a variety of alteration during aging, such as swelling of mitochondria and of endoplasmic reticulum, and increase of microtubules, microfilaments, lysosomes and lamellated myelin structures, etc. Accumulation of dense intracytoplasmic material in the foot processes is increased in the vicinity of the basal lamina during aging. The podocytic membrane-like structures are seen in young and o]d rats. 7. The mesangial matrices and mesangial cells are increased during aging, and slight swelling of endoplasmic reticulum and Golgi cisternae in young and old rats.

  • Degranulation of the rat peritoneal mast cell induced by intraperitoneal injection of horseradish peroxidase(HRP) was studied using light and electron microscopes. 1. Rat peritoneal mast cells in the Tyrode's buffered salt solution injected control group did not show any particular morphological changes following the specified time course. 2. Under the light microscope, the majority of mast cells observed 10 minutes after HRP injection were nearly the same as those of the control group. However, after 30 minutes, granule densities or staining properties of certain cells began to decrease and these appearances increased gradually until 12 hours after injection, at which time small groups of granules being stained pale-red or pink with toluidine blue were easily identified in the cytoplasm of many cells, and numerous extruded granuleg were scattered around these cells. 3. In the mast cells representing the early stage of degranulation induced by HRP, the electron densities of certain granules decreased as the size enlarged, and perigranular cavities were formed by perigranular membrane expansion. As a result, a thin cytoplasmic septum was formed between the expanded perigranular membrane and the cytoplasmic membrane in the cell periphery, and fusion of the adjacent perigranular membranes was observed in the inner side of the cell. 4. In some mast cells, one or two changes in the peripheral cytoplasmic septum could be seen. One was a focal rupture of the peripheral septum and the other was the formation of a saccule containing one or more vesicles. This saccule was thought to be used for granule-extrusion site and/or material absorptive apparatus judging from the morphological characteristics. 5. As the degranulation proceeded, the granule was extruded from the cell after partial rupture of the peripheral cytoplasmic septum. This phenomenon proceeded to-ward the inner side of the cell through the fused perigranular cavities, and consequently several distinct cavities containing a few unextruded membrane-free granules were formed throughout the cytoplasm after 12 hours. As a rule, the granule-extrusion sites were relatively fewer while the cytoplasmic cavities resulting from degranulation were more numerously observed. Thus, it was thought that the granule-extrusion sites tended to be restricted in the HRP-induced degranulation.
