Fixation of Cellular Ultrastructure by the Microwave Irradiation (마이크로파 조사에 의한 세포 미세구조의 고정 효과)

  • The microwave fixator has recently been introduced in morphological research. The present study was carried out to investigate the ultrastructural effects of microwave fixation of rat brain. kidney, liver and skeletal muscle tissues. The results are as follows: In the case of microwave fixed cerebrum. the cytoplasmic processes of neurons and the various membranous organelles such as nuclear envelope, mitochondria, rough endoplasmic reticulum and Golgi apparatus were well preserved, The myelin sheath wrapping neuronal axon was prominent. Microwave fixed hepatocytes showed the microvilli on the free surface of bile canaliculus, the evident nucleolar components, and typical organelles. In nephron, ultrastructures of glomerulus and Bowman's capsule were preserved, and also tubular wall were structurally observed. Among the skeletal muscle cells, plentiful collagen fibers were appeared, myofibrils and mitochondria were typically observed. In conclusion, the microwave fixation procedures result in an good preservation of the tissues and would be time- and reagent-saving.

  • Pulmonary alveolar proteinosis(PAP) is a rare disease in which the alveolar spaces are filled with an eosinophilic, PAS-positive material, whereas the interstitial architecture of the lung usually remains unaffected. Although a definitive diagnosis is usually made by an open lung biopsy, bronchoalveolar lavage(BAL) cytology may play a decisive role in the diagnosis and therapy of these patients and may spare a patient a more invasive diagnostic procedure. The author presents a patient in whom BAL cytology specimen contained the characteristic globules of amorphous proteinaceous PAS-positive material accompanied by background of rare macrophages and inflammatory cells. Ultrastructural study using BAL specimen can confirm the diagnosis of PAP.

  • A strain of Alcaligenes sp. Isolated from the sludge of industrial area was found to uptake 24.1 mg lead/g dried cell during incubation in medium containing 100g/ml of lead. Analyses of cellular subfractions reveal that fractions of cell wall contain 88.6 percent of lead found associated with the cells and the remainder is found associated with the cytoplasmic fraction. Ultrastructural examination of the cells cultured in media containing 500 and 1000g/ml of lead showed no major irregularities between cells of the treated and untreated cultures.

Accumulation of Heavy Metals in the Antarctic Clam Laternula elliptica

  • Immunohistochemical and ultrastructural experiments were conducted to find out heavy metal accumulation in some selected organs such as the kidney, the hepatopancreas, and the gills of the Antarctic Clam Laternula elliptica According to the immunohistochemical study the subject organs of the clam showed reactions indicating the presence of MT(metallothionein), a metal-binding protein involved in metal detoxifying process. Examination under the transmission electron microscope also revealed that other ligands(e.g. metal-rich granules in the kidney) may play a role in metal accumulating and detoxifying process in L. elliptica. In the artificial exposure of the clam to Cd, it showed immediate subcellular responses, suggesting that this species can be used as rapid and efficient bioindicators for Cd exposure in natural environment.

  • The radulae of six species of bithyniid snails, Bithynia striatula, B. tentaculate, B. siamensis, B. leachi, Gabbia misella and G. australis, were observed by SEM. The radular formula of all the bithyniids studied was 2 : 1 : 1 : 1 : 2. The all teeth were multicuspid and the central teeth had numerous basal denticles. The mesocones of the central and lateral teeth were spade or arrow-head shape. The mesocones of the central teeth and the lateral teeth of B. leachi, G. misella and G. australis were slightly larger than the other cusps in contrast of those of the other species. Number of cusp was varied by the specimen and the species.

  • The purpose of this study was to investigate the differentiation and degeneration of neurons in developing chick embryo. The activity of acid phosphatase(ACP) was measured and cytochemical study of ACP and ultrastructural changes were observed in prosencephalon, mesencephalon and rhombencephalon from day 4 to day 19 of incubation. As a result, the activity of ACP of all brain region was tend to increase from day 4 to day 19. On day 13, activities of ACP of mesencephalon and rhombencephalon were increased greatly and activity of ACP was decreased each region on day 17. On electron microscopic examination, the reaction product of ACP were localized at GERL complex, lysosome, Golgi body and vacuoles of neurons. Morphologically, disrupted nuclear envelope, mitochondrial destruction, vacuolization and ribosomal crystalization were observed.

Chitosan이 사과 겹무늬썩음병균 Botryosphaeria dothidea의 생육에 미치는 영향

  • To examine the potential utilization of chitosan, the biodegradable natural polymer, as a control agent of apple white rot caused by Botryosphaeria dothidea in a new control measure by coating it on the diseased branches, the various antifungal activities of chitosan was investigated. Chitosan showed significant inhibitory effect on the mycelial growth of B. dothidea, along with the morphological changes including hyphal swelling and ultrastructural changes on solid PDA medium. In liquid PD broth medium, the chitosan showed more significant effect on the growth of B. dothidea also forming cell clusters indicating affection on the hyphal extension. The growth of B. dothidea was inhibited more than 90% at the concentration of 1.0 mg/ml. Chitosan also detained the spore germination and induced the morphological change of germ tubes. Glucosamine, monomer of chitosan, did not affect on the growth of B. dothidea indicating the antifungal activity was caused by chitosan polymer.

  • The ultrastructural characteristics of shear fracture surfaces of laminated wood prepared from major four Korea wood specimens were examined. Commercial urea and urethan resin were used as adhesives for laminated woods of both homospecies and heterospecies. The morphology of fracture surface was observed using an optical microscopy and scanning electron microscopy. Three anatomical failure types were recognized : intercell failure, intrawall failure and transwall failure. In dry specimen, failure occurred mainly in woods. Laminated woods of softwoods showed mostly intrawall failure and transwall failure of tracheids, and them of hardwoods indicated mainly intrawall failure and interwall failure. Laminated woods prepared with urethan resin showed coarse fracture surface, on the other hand, those prepared with an urea formaldehyde resin had clean surface. In wet specimen, failure occurred dominantly in glue line. Intrawall failure and flags were characterized in laminated wood prepared with urethan resin. In heterospecies laminated woods, failure was occurred mainly in softwood. Consequently, fracture morphology of laminated wood may be influenced by adhesives, moisture content, species and anatomical characteristics.

  • The present study was designed in order to clarify ultrastructural characteristies of the spermatogenetic process and to examine survival state of the spermatozoa introduced into the female reproductive tract after autumnal coitus in the Korean greater horseshoe bats(Rhinolophus ferrumeguinum korai). The general morphological characteristics of spermatogenesis were principally similar to those of the other mammalian species; acrosomal formation, flagellar formation, middle piece formation and concentration of the spermatozoan nucleus. The spermatozoa introduced into the vagina were found to be dead forming a vaginal plug, the opaque central core of which consisted of trapped dead spermatozoa. Some spermatozoa introduced into the uterus were observed to be phagocytized by the polymorphonuclear leucocytes infiltrated into the uterine glandular lumen. The oviductal epithelium, consisted of ciliated and secretory cells; the luminal surface of secretory cells were covered by a number of microvilli with well developed glycocalyx, suggesting a close relationship to nutrient (e.g. glycogen) supply for the spermatozoa during hibernation.

  • The endosperm cells and the umbiliform layer of ginseng (Panax ginseng C.A. Meyer) seed are studied with light and electron microscope. Differentiated mitochondria, ER cisternae, proplastids and ribosomes are characteristically observed in the endosperm cells of matured seed. The cell inclusions contain the protein bodies and the spherosomes. Protein body contains, in proteinaceous matrix, globoids and crystalloids. Particularly the crystalloids have the lattice structure, and the formation of globoids is closely related with ER. Umbiliform layer has the positive reaction on alcian blue (pH 2.5) and the metachromasis on the toluidine blue. The umbiliform layer is formed by autolysis of endosperm cells, and composed of the deformated cell wall and the lipoprotein bodies. Particularly a part of the lipoprotein body and the fibrilar network structure have the positive reaction on acid phosphatase.
