연어(Oncorhynchus keta) 정자의 미세구조와 성분화 (Ultrastructures of Sperm, Gonadal Sex Differentiation in Chum Salmon(Oncorhynchus keta))

  • This study was carried out to investigate the ultrastructural changes of spermatozoa obtained from 20 of 3-year-old male chum salmon(Oncorhynchus keta) collected and analysed in middle October in 1995. The ultrastructural changes of gonad of fingerlings were examined to describe the sex differentiation of this species. The results obtained in this study were as follows : In spermatozoa, the nucleus is dense and homogeneous. Two spheroidal mitochondria(about 350nm long) are situated in parallel between the nucleus and the axoneme. Spermatozoa mitochondria are assembled into an organized sheath surrounding the outer dense fibres and axoneme of the flagellar midpiece. The sheath flagellum is situated beneath the base of the sperm head. The primordial germ cells of 6.8~7.2${\mu}{\textrm}{m}$ in size, which were buried under fibrous mesenchymal tissue between gut duct and notochord of larva with a total length of 2.4cm at 50 days after hatching. In juvenile of 10.5cm in total length at 70 days after hatching, the gonad was occupied by bundles of oogonia. The dense drumstick bodies(large arrows) are observed in the nuclei of the primordial gonad and surrounding tissue cells of fingerling at 70 days after hatching. The oval Barr bodies(asterisk) are observed in the nuclei of the primordial germ cells under the mitosis(2n). Note the large mitochondria, ribosomes and rough endoplasmic reticulum in the cytoplasm. Accordingly, the fingerlings at 70 days after hatching are identified as the female(xx). In result, the gonadal sex differentiation begins from the 70 days after hatching in chum salmon.

사람태아 성장기 모낭에서 결합조직-상피 경계부의 미세구조에 관한 연구 (Ultrastructural Study on Connective Tissue-Epithelial Junctions in Anagen Hair Follicle of Human Fetus)

  • The dermal papilla is known to playa major role in influencing the form and dynamics of the hair follicle, which probably involves regulatory substances crossing the basal lamina. But little is known about the junctions between the dermal papilla and the surrounding epithelial cells of the hair bulb, or between the connective tissue and the epithelial cells on the outside of the hair follicle. This study was performed to identify the ultrastructural differences between dermoepidermal junction of the skin and connective tissue-epithelial junctions on the outside of the hair follicle and around the dermal papilla of normal anagen hair follicles in the human fetal scalp skin. Electron microscopic findings of dermoepidermal junction in scalp skin showed that basal lamina was very irregular and undulated, and it contained many attachment plaques of hemidesmosomes with sub-basal dense plates, tonofilaments, and anchoring filaments. Also invaginations of plasma membrane of basal keratinocytes were seen. There were clear differences both on the outside of the follicle and around the dermal papilla as compared with similar junction in the skin. In particular, neither hemidesmosomes nor tonofilaments, as seen in dermoepidermal junction, were observed in the dermal papilla. Also attachment plaque, sub-basal dense plate and anchoring filaments were not observed at the junction on the outside of the follicle and the dermal papilla. There were some differences between connective tissue-epithelial junctions on the outside of the hair follicle and around the dermal papilla, ie, smoothness of basal lamina and orthogonal arrangement of collagen fibers were seen in the outside of hair follicle, but not in the dermal papilla. These results indicate that the mechanical connection between the hair follicle and the connective tissue component is much weaker than that between the corresponding components in skin, and it reflects the dynamic processes during the anagen phase of the hair follicle compared to the relatively permanent state of the epidermis.

수은이 계배 대뇌의 신경세포 분화에 미치는 영향(III) (Effects of Mercury on the Differentiation Cerebral Neuron of Chick Embry (III))

  • To investigate the effects of mercuric chloride $(HgCl_2)$ on the differentiation of the cerebral neuron of chick embryo 10 days, the ultrastructural changes in nerve cells injected with a various doses of mercuric chloride were observed with transmission electron microscope. The enzyme activity of the some dehydrogenases, cerebral proteins and adenosine triphosphate (ATP) were also analyzed. The results obtained are as follows; The ultrastructural changes in 1.0 mg-injected group, the nuclear membranes were irregular, outer of mitochondria membrances dispressioned, their cristae were destroyed. In 2.0 mg-injected group, the nuclear envelops were destroyed and divided, were not observed organelle except of few ribosome, the RER and mitochondria. The number of polypeptide bands were separated by SDS-PAGE in the normal group were 38 bands. According to the in creased dose of mercuric chloride, contends of the bands were increased in 4 bands, but were decreased in 1 band. The activities of dehydrogenases were declined by increasing the dose of mercuric chloride. Lactate dehydrogenase (LDH) activity fatted to 61% in 2.0 mg-injected group. Malate dehydrogenase (MDH) activity fatted to 90% in 1.0 mg-injected group, greatly to 76% in 2.0 mg-injected group. Succinate dehydrogenase (SDH) activity decreased to 79% in 1.0 mg-injected group and greatly to 62% in 2.0 mg-injected group. ATP content in 1.0 mg-injected group was almost near to the normal level, but it was increased greatly in 2.0 mg-injected group.

머리에 방사선 상해를 받은 흰쥐 흑색질의 미세구조 (Ultrastructural Study on the Substantia Nigra of the Head-Irradiated Rats)

  • An experimental study on the acute irradiation effects on the substantia nigra of head-irradiated rats were carried out. Rats anesthetized with sodium thiopental, were exposed only on their head areas with a single dose of 3,000 rads or 6,000 rads, respectively. Radiation was produced by Mitsubishi linear accelerator at the speed of 200 rads/min. Aminals were sacrificed on 6 hours, 2 days and 6 days following irradiations. By the perfusion fixation through the heart, rats were fixed with 1% glutaraldehyde-1% paraformaldehyde solution. Two hours later, brains were exposed and immersed in the same fixatives over night. Tissue blocks from subtantia nigra were punched out, and they were refixed in the 2% osmium tetroxide solution. Blocks were dehydrated through alcohol series, and embedded in the araldite mixture. Ultrathin sections were stained with uranyl acetate and lead citrate solutions, From the ultrastructural study, following results were made: 1. Six hours after irradiation, severe depletion of synaptic vesicles was occurred in the many axon terminals of the nigral neuropil. 2. Dramatical decrease of lysosomes and dense granules was observed. 3. Two days following irradiation, alterations of ribosomes, granular endoplasmic reticula, mitochondria, etc, were noticed. 4. Many of the malformations were seen to be repaired on the 6th day. 5. Above results were interpreted as follows. At the acute stage of heavy irradiation, neurotransmitters in the substantia nigra are released severely. But they are recovered within 6 days. It is concluded that acute head-irradiation may result severe disturbance of nigral motor control function during the first few days.

  • In an attempt to produce transgenic amphibia, bacteriophage ${\lambda}-DNA$ was microinjected into fertilized eggs of Xenopus laevis, and the effect on early embryogenesis and the ultrastructural behavior of exogenous DNA were investigated. The effect of microinjected gene on embryonic development showed differences according to the concentration of injected DNA and the incubation temperature. Various concentrations of ${\lambda}-DNA$ were tested. Among those, microinjection of 1-2 ng DNA dissolved in 20 nl TE buffer was not shown to disturb normal embryonic development and was recorded the highest survivability to the late tadpole stage (Stage 43); however, injection of increased concentrations of DNA than above provoked irregular cleavages or abnormal appearances, which resulted in reduced survivability. When the injected embryos were incubated at low temperatures (e.g., $12^{\circ}C$), 54.5% of the embryos developed to Stage 43, whereas 42.4% survived when incubated at room temperature. The survivability showed also differences according to the injection site. 58.0% of the embryos developed to Stage 43 when microinjected into the vegetal pole, whereas 44.9% survived when microinjected into the animal pole. To understand the structural fate or behavior of injected DNA a combined light and electron microscopical study was applied. The nucleus-like structure was observed in the ${\lambda}$ DNA-injected embryos, which was quite a similar to the interphase nuclei of normal Xenopus laevis. The nucleus-like structure showed the typical double-layered nuclear membrane and nuclear complexes; however, it consisted of unusual structures such as furrows of nuclear envelope into the nucleoplasm.

Cold Stress가 노화(老化) 흰쥐 심근(心筋)에 미치는 영향(影響) (Effect of Cold Stress on Myocardium of Aging Rat)

  • The present study was performed to determine the effect of cold stress on myocardium of aging rat. Control groups, which aged 6, 12 and 24 months, were compared with age-matched experimental groups that were exposed to moderate cold stress for a hours daily in a week at laboratory cold room $(4{\pm}1^{\circ}C)$. The histological, histochemical and ultrastructural changes of myocardium were observed. The results were summarized as follow: 1. Age-dependent histological change of control groups was observed the formation of contraction band in 24months aged group. The experimental groups submitted to cold stress showed a similar change pattern as seen in control groups. However, the degree of change in the experimental groups was significantly larger than that of control groups. In the 34 months aged group the formation of hypercontraction band was observed. 2. Regarding age-dependent histochemical changes of control groups, we observed the increase activities of PAS and Masson's trichrome. In experimental groups the activities of PAS and Masson's trichrome were also increased with age. Compare with control group, the activities of PAS was increased but the activities of Masson's trichrome was decreased. 3. Age-dependent ultrastructural changes on vacuolization, lysosome were observed. In control groups the structural changes occur at 12 months. The accumulation of lipofuscin, contraction band, hypercontraction band and a component of connective tissue were observed in 24 months. However, the degree of change in the experimental groups was significantly larger than that of control groups. In contract, the myelin body in intercalated discs was observed in 24 months of experimental groups.

난소제거 바퀴에서 알라타체의 이상비대화에 관한 연구 (Hypertrophical Changes of the Corpus Allatum Caused by Ovariectomy in Blattella germanica)

  • The present study is undertaken to investigate the hypertrophical changes of the corpus allatum (corpora allata, CA) after the ovariectomy in Blattella germanica. In particular, the ultrastructural aspects of the normal and ovariectomized conditions, and induced factors of the hypertrophic phenomenon are focused. Ultrastructure of the CA from an immediately emergent adult is similar to that of the last larval stage that has stopped secreting juvenile hormone. The CA is composed of undifferentiated cells, exhibiting an electron-lucent matrix, a few mitochondria and less smooth endoplasmic reticulum. The karyoplasm occupy most of the cytoplasm. Electron-dense materials are filled with the intercellular spaces and gap junctions are also found. Almost no ultrastructural changes have been noticed during seven days until the oviposition. However, considerable changes in structure have been detected soon after the oviposition. Mitochondria are increased dramatically in number and cristae, and changed to the filamentous form with a high electron density. In addition, Golgi complexes, microtubules, and polysomes are also increased. After an oviposition, the total volume of the CA are decreased again. The volume of the CA are increased continuously, hypertrophy, after the ovariectomy. Morphological aspects of the CA in an early stage after the removal are similar to the structure of the secondary egg maturation. Large and electron dense globules are observed in the ovariectomized CA cytoplasms and they are present in those cells for a long period of time. Yet such a hypertrophical phenomenon occur only in the specific cells. The hypertrophy are caused by hollowing the part of the CA cells and later filling such site with polysomes. In 42 days after the ovariectomy, the nuclear membranes disappear in the CA cells, thus, exhibiting the prokaryotic-like features. Some results of the current study will contribute to the establishment of the model that explain unusual changes accompanied by certain treatment in insects and/or further in animals.

$1\alpha$, Dihydroxycholecalciferol의 골다공증성 골절 치유효과 : 광학현미경 및 주사전자현미경적 관찰 (Effects of $1\alpha$, 25 Dihydroxycholecalciferol on Osteoporotic Fracture : Light Microscopic and Scanning Electron Microscopic Observation)

  • 12주령 랫트의 정상 비골골절의 치유과정을 이해하고, 난소적출 후 비골골절의 치유과정과 난소적출 후 비골골절의 치유에 미치는 $1\alpha$, 25 dihyoxycholecalciferol의 영향을 알아보고자 실험을 실시한 결과 다음과 같은 결론을 얻었다. 비골의 골절은 골절유발 후 5주에 성숙된 신생골 조직으로 충만되어 조직형태학적으로 완전한 치유가 이루어졌다. 난소적출 후 골절의 치유는 정상골절의 치유보다 지연되었으나, $1\alpha$, 25 dihydroxycholecalciferol 투여 결과 골절유발 후 5주에 성숙된 신생골 조직으로 골절단이 충만되어 정상상태의 골절과 비슷하게 치유가 되었다. 이상의 결과를 종합해보면 랫트의 난소를 적출하여 여성호르몬의 결핍을 유발하면 난소적출 후 7주에는 골다공증이 발생되고, 골다공증은 정상상태의 골절에 비하여 약 1주일 이상 골절의 치유를 지연시켰으며, $1\alpha$, 25 dihydroxycholecalciferol을 투여하였을 경우 정상상태의 골절치유와 비슷한 경과를 나타내어 $1\alpha$, 25 dihydroxycholecalciferol은 골다공증성 골절의 치료에 있어서 효과있는 치료제라는 결론을 내릴 수 있었다.

신장 상해물질이 생쥐 사구체곁세포의 미세구조에 미치는 영향 (Ultrastructural Influence of Nephrotoxic Agents on the Juxtaglomerular Cells of Mice)

  • This experiment was performed to study the ultrastructural changes of the juxtaglomerular cell of mice following subcutaneous injection of heavy metallic agents. Male mice were divided into normal and experimental groups. The mice were subcutaneouly injected with $HgCl_2$ (2mg, 5mg or 10 mg/Kg/BW) or with $K_{2}Cr_{2}O_7$(5 mg, 10 mg or 20 mg/Kg/BW). Mice were sacrificed on 6 hours, 3 days and 14 days after the injection. Kidneys were fixed in the 2.5% glutaraldehyde-1.5% paraformaldehyde solution, followed by refixation in the 1% osmium tetroxide solution. Dehydrated blocks were embedded in araldite mixture. The sections were cut on a LKB-V ultratome, and ultrathin sections stained with uranyl acetate and lead citrate were observed with JEM 100CX II electron microscope. The results were as follow: 1. Juxtaglomerular cell of the experimental groups showed some alterations, especially in the structures of protein synthesis including dilations and degradations of granular endoplasmic reticula, atrophy of Golgi complex, and numerous free ribosomes in the cytoplasm. 2. Juxtaglomerular cells treated groups showed a number of vacuoles, protogranules and some myelin figures in the cytoplasm, especially in the earlier groups. 3. Juxtaglomerular cells of treated groups, contained a large number of secretory granules showing variable electron densities and pleomorphism in later groups (2 weeks). From the above results, it was concluded that, the mercuric chloride or potassium bichromate induces acute renin release from juxtaglomerular cells of the mice, but many juxtaglomerular cells may secrete prematured secretory granules, or the synthetic system of the cell can not perform normal function.

Germ cell Differentiation During Spermatogenesis, and Ultrastructural Characteristics of Mature Sperm in Male Phacosoma japonicus (Bivalvia: Veneridae)

  • Some characteristics of germ cell differntiations during spermiogenesis and mature sperm ultrastructure in male Phacosoma japonicus were investigated by transmission electron microscope observations. The morphology of the spermatozoon of this species has a primitive type and is similar to those of other species in the subclass Heterodonta. Morphologies of the sperm nucleus and the acrosome of this species are the cylindrical type and cap shape, respectively. The spermatozoon is approximately 45-50 ${\mu}m$ in length, including a long curved sperm nucleus (about $3.70{\mu}m$ long with 45 $^{\circ}$ of the angle of the nucleus, an acrosome (about $0.55{\mu}m$ in length), and tail flagellum (about 42-$47{\mu}m$)The axoneme of the sperm tail shows a 9+2 structure. As some characteristics of the acrosomal vesicle structures, the basal and lateral parts of basal rings show electron opaque part (region), while the anterior apex part of the acrosomal vesicle shows electron lucent part (region). These characteristics of the acrosomal vesicle were found in the family Veneridae and other several families in the subclass Heterodonta. These common characteristics of the acrosomal vesicle in the subclass Heterodonta can be used for phylogenetic and systematic analysis as a taxonomic key or a significant tool. The number of mitochondria in the sperm midpiece of this species are four, as one of common characteristics appear in most species in the family Veneridae and other families in the subclass Heterodonta. However, exceptionally, only three species in Veneridae of the subclass Heterodonta contain 5 mitochondria. The number of mitochondria in the sperm midpiece can be used for the taxonomic analysis of the family or superfamily levels as a systematic key or tools.
