Spirulina maxima의 초음파 및 열수 추출 공정에 따른 항염증 효과 비교 탐색 (Comparison of Anti-Inflammatory Activity of Spirulina maxima Extract by Ultrasonication and Water Extraction Process)

  • 실험은 초음파 전처리를 통한 Spirulina maxima 추출물이 기존의 열수 추출 공정과 비교하여 항산화, 항염증 효과가 증진되는지에 대한 연구를 하였다. 낮은 세포 독성을 통해 안정성을 확보하였으며, 항염증성 효과를 증진시키기 위한 항산화 효능에서는 DPPH의 경우 약 50% 정도 이상의 효능을 보였다. Flavonoid 함량에서는 열수 추출물보다 초음파 공정을 거친 추출물은 10% 정도의 함량 증가를 보였으며, total phenol에서도 함량의 증가를 확인하였다. 또한 이 결과로 일반 천연물보다 항산화능이 좋을 뿐 아니라 독성도 낮은 결과를 얻어 S. maxima의 뛰어난 효능을 입증하였다. 항염증 효과의 경우 NO, TNF-${\alpha}$ 및 IL-6 생성량을 확인한 결과 3가지 생성량 모두 열수 추출물보다 초음파 공정을 통하여 항염증 효능의 증대를 보이는 것을 확인하였다. 이러한 결과를 바탕으로 초음파 공정을 이용해 낮은 온도에서 활성물질들의 파괴가 없을뿐더러 초음파의 높은 에너지로 인하여 더 많은 활성물질들을 추출하여 효능들의 증대로 보이며, 초음파 공정 전처리를 통하여 S. maxima의 두꺼운 세포벽을 효과적으로 파쇄하여 생리활성 물질의 추출이 효과적으로 진행되어 항산화능이 증가하게 되고, 항산화능의 증가로 항염증 효능에 영향을 미치는 것으로 사료된다. 이 실험 결과들을 바탕으로 S. maxima의 활성 증진 및 기능성 식품으로의 개발에 적용될 것이라고 사료된다.

미선나무의 저온 추출 공정에 의한 항주름 활성 증진 (Enhancement of Anti-wrinkle Activities of Abeliophyllum distichum Nakai through Low Temperature Extraction Process)

  • Skin anti-wrinkle activities of the stems and leaves of Abeliophyllum distichum Nakai were evaluated by the extracts obtained from various extraction processes such as using hot water at $100^{\circ}C$, 70% ethanol at $85^{\circ}C$, and 70% ethanol with ultrasonication at $60^{\circ}C$ The ultrasonicated extract showed 95.62% of the highest cell viability in addition of $0.3mg/m{\ell}$ of the extracts into the normal human fibroblast cell, CCD-986sk. For antioxidant activities, the extracts using ultrasonicated extract showed the highest DPPH free radical scavenging as 80.27%, followed by 75.88% and 62.44% for the extracts using ethanol extract and water extract. The ultrasonicated extract also showed the highest elastase inhibition activity as 25.32%, compared to ethanol extract and water extract based method at 22.01% and 12.88%, respectively. MMP-1 production was most effectively decreased down to $2908.1pg/m{\ell}$ with ultrasonicated extract while $6640.8pg/m{\ell}$ with water extract and $3609.3pg/m{\ell}$ with ethanol extract, in addition of $0.3mg/m{\ell}$. Collagen production was increased up to $154.7ng/m{\ell}$ in addition of ultrasonicated extract, and followed by $121.4ng/m{\ell}$ and $31.2ng/m{\ell}$ for ethanol extract and water extract, respectively. These results indicate that the ethanol extract should have skin anti-wrinkling activities and can be improved by the ultrasonication process that high energy input elute more amounts of bioactive substances eluting more amounts of bioactive substances from the high energy input of ultrasonication.

Hepatoprotective effect of ultrasonicated ginseng berry extract on a rat mild bile duct ligation model

  • Background: The Panax ginseng berry extract (GBE) is well known to have an antidiabetic effect. The aim of this study is to evaluate and investigate the protective effect of ultrasonication-processed P. ginseng berry extract (UGBE) compared with GBE on liver fibrosis induced by mild bile duct ligation (MBDL) model in rats. After ultrasonication process, the composition ratio of ginsenoside in GBE was changed. The component ratio of ginsenosides Rh1, Rh4, Rg2, Rg3, Rk1, Rk3, and F4 in the extract was elevated. Methods: In this study, the protective effect of the newly developed UGBE was evaluated on hepatotoxicity and neuronal damage in MBDL model. Silymarin (150 mg/kg) was used for positive control. UGBE (100 mg/kg, 250 mg/kg, 500 mg/kg), GBE (250 mg/kg), and silymarin (150 mg/kg) were orally administered for 6 weeks after MBDL surgery. Results: The MBDL surgery induced severe hepatotoxicity that leads to liver inflammation in rats. Also, the serum ammonia level was increased by MBDL surgery. However, the liver dysfunction of MBDL surgery-operated rats was attenuated by UGBE treatment via myeloid differentiation factor 88-dependent Toll-like receptor 4 signaling pathways. Conclusion: UGBE has a protective effect on liver fibrosis induced by MBDL in rats through inhibition of the TLR4 signaling pathway in liver.

Protective effect of ultrasonication-processed ginseng berry extract on the D-galactosamine/lipopolysaccharide-induced liver injury model in rats

  • Background: Acute hepatic failure is a life-threatening critical condition associated with rapid deterioration of liver function and liver transplantation. Several studies have shown that Panax ginseng Mayer has antidiabetic and hepatoprotective effects. However, the hepatoprotective effect of ginseng berry is still unveiled. In this study, we evaluated the hepatoprotective effects of ultrasonication-processed ginseng berry extract (UGBE) on acute hepatic failure model in rats. Methods: Ginseng berry extract (GBE) was ultrasonically processed. The GBE, silymarin, and UGBE were orally administered to male Sprague-Dawley rats for 4 wk. Twenty-four h after the last administration, rats were challenged with D-galactosamine (D-GalN)/lipopolysaccharide (LPS). Results: After ultrasonication, the component ratio of ginsenosides Rg2, Rg3, Rh1, Rh4, Rk1, Rk3, and F4 in GBE had been elevated. Administration of UGBE significantly increased the survival rate of D-GalN/LPS-challenged rats. Pretreatment with UGBE significantly decreased serum alanine aminotransferase, aspartate aminotransferase, and total bilirubin levels in D-GalN/LPS-challenged rats in a dose-dependent manner. The levels of enzymatic markers for oxidative stress (superoxide dismutase, glutathione peroxidase, catalase, and glutathione) were increased by UGBE treatment in a dose-dependent manner. Tumor necrosis factor alphalevel, inducible nitric oxide synthase activities, and nitric oxide productions were reduced by UGBE treatment. In addition, hemeoxygenase-1 levels in liver were also significantly increased in the UGBE-treated group. The protein expression of toll-like receptor 4 was decreased by UGBE administration. Hematoxylin and eosin staining results also supported the results of this study showing normal appearance of liver histopathology in the UGBE-treated group. Conclusion: UGBE showed a great hepatoprotective effect on D-GalN/LPS-challenged rats via the toll-like receptor 4 signaling pathway.

초음파 처리에 의한 인삼꽃대 엑스의 진세노사이드 성분 변화 (Changes in Ginsenosides Composition of Ginseng Flower Buds Extracts after an Ultrasonication Process)

  • The purpose of this study is to develop a new preparation process of ginseng (Panax ginseng) flower buds extracts featuring high concentration of ginsenosides Rg2, Rg3, Rg5, F4 and Rh1, red ginseng special components. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of ginseng flower buds were processed under several treatment conditions of ultrasonication (at $100^{\circ}C$). The results showed that the quantity of ginsenoside Rg6 increased by over 8.8% at the 16 hours of ultrasonication. Ginseng flower buds ethanol extract compared with other process times. The result of UGF-16 indicates that the ultrasonication processed ginseng flower buds extracts (at $100^{\circ}C$) treated for 16 hours produced the highest amount of ginsenoside F4 (8.833%), Rg3 (2.230%), Rg5 (2.339%) and Rg2 (1.002%).

초음파 처리가 채소 중의 sulforaphane 분석에 미치는 영향 (Effects of ultrasonication on the analysis of sulforaphane content in vegetables)

  • 초음파처리가 십자화과 채소의 sulforaphane 함량 분석에 미치는 영향을 GC/MS를 이용하여 검토하였다. 무, 알타리무, 순무, 양배추, 브로콜리를 각각 dichloromethane을 추출용매로 하여 초음파처리 후 용매 추출 및 질소 농축을 하고, m/z 72, 160, 55, 114을 선택이온측정법(selective ion monitoring, SIM)을 이용하여 분석하였다. 각 채소류의 sulforaphane 함량은 브로콜리가 149.0 ppm으로 가장 많았으며, 양배추(67.9 ppm), 무(35.4 ppm)의 순으로 높았다. Sulforaphane 추출율은 진탕교반(shaking)만 할 경우보다 초음파처리(sonication)후 진탕교반(shaking)을 할 경우 2.7배 높은 것으로 나타났다. 따라서 초음파추출공정이 sulforaphane의 추출효율 향상에 유효한 방법으로 판단된다.

초음파 처리 인삼열매 엑스의 인삼사포닌 변화 (The Change of Ginsenoside Composition in Ginseng Berry Extract by the Ultrasonication Process)

  • The purpose of this study is to develop a new preparation process of ginseng berry extracts having high concentrations of ginsenoside Rh1, Rg2, Rg5, F4, a special component of red and black ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of ginseng (Panax ginseng) berry was processed under several treatment conditions including ultrasonication treatments. The content of total saponin reached their heights at 6 hr (UGB-6, 61.760%) of ultrasonication treatment, followed by 10 hr (UGB-10, 53.009%) and 9 hr (UGB-9, 50.652%) of ultrasonication treatment at $100^{\circ}C$. Results of those treatments showed that the quantity of ginsenoside Rh1 increased by over 15% at 10 hr of ultrasonication treatment at $100^{\circ}C$. The results of processing with UGB-10 indicate that the ultrasonication processed ginseng berry extracts that had gone through 10 hr treatments were found to contain the largest amount of ginsenoside Rh1 (15.358%), Rg2 (6.301%), Re (4.567%) and F4 (2.658%). In addition, UGB-6 contained ginsenoside Rg3 (13.632%) at high concentrations. It is thought that such results provide basic information in preparing ginseng berry extracts with functionality enhanced.

초음파 처리 인삼화뢰 추출물의 콜라겐 합성작용 및 MMP-1 발현저해 (Inhibition of MMP-1 Expression and Collagen Synthesis Activity of Ultrasonication Processed Ginseng Flower Buds Extract)

  • This study was to evaluate the effect of isopropyl alcohol fraction of ultrasonication processed ginseng flower buds(GFB-IF) on the collagen synthesis activity and inhibition of MMP-1 suppression in UV-irradiated human dermal fibroblasts. The higher contents of ginsenoside Rg2(8.234%), Rh1(5.749%), F4(3.881%) in isopropyl alcohol fraction of ginseng flower buds obtained by ultrasonication process at 600W(100℃) for 16 hours. GFB-IF had collagen synthesis effect. GFB-IF induced a significant dose-dependent decrease in the expression for MMP-1 protein. These results suggest that GFB-IF is a potential candidate for the prevention and treatment of wrinkle improving.

Zoogloea ramigera에 의한 생물고분자 생산에 관한 연구 (The Production of Biopolymer by Zoogloea ramigera)

  • 생물응집체 개발을 목적으로 Zoogloea ramigera115를 이용하여 균주배양 및 생물고분다 생성실험을 시시하였다. 다당류 최고 생성률을 얻을 수 있는 최적기질로 탄소원으로는 glucose, 질소원으로는 $NaNO_3$ 유기영양원으로는 yeast extract가 선택되었다. 초기 pH가 6일 때 가장 높은 세포성장과 점성을 나타내었다. 생성돈 생물고분자는 ultrasonication과 고속 원심분리를 거친 후 propanol 첨가에 의해 효율적으로 추출되었다. 생물고분자는 jae test에 의한 응집실험에서 높은 침강성을 보여 응집제 사용가능성을 확인하였다.

  • The purpose of this study was to develop a new ginseng (Panax ginseng) flower buds extract with the high concentration of ginsenoside Rg3, Rg5, Rk1, Rh1 and F4, the Red ginseng special component. Chemical transformation from the ginseng saponin glycosides to the prosapogenin was analyzed by the HPLC. The ginseng flower buds were processed at the several treatment conditions of the ultrasonication (Oscillator 600W, Vibrator 600W) and vinegar (about 14% acidity). The result of UVGFB-480 was the butanol fraction of ginseng flower buds that had been processed with ultrasonication and vinegar for 480 minutes gained the highest amount of ginsenoside Rg5 (3.548%), Rh1 (2.037%), Rk1 (1.821%), Rg3 (1.580%) and F4 (1.535%). The ginsenoside Rg5 of UVGFB-480 was found to contain 14.3 times as high as ginseng flower buds extracts (GFB, 0.249%).
