초음파처리가 홍화의 색소추출과 염색성에 미치는 영향 (Effect of Ultrasonic Treatment on Colorants Extraction and Dyeability of Safflower)

  • A new method for competent extraction of safflower (Carthamus tinctorius Linnaeus) colorants by using ultrasonic device was developed. When comparing to general extraction method, the ultrasonic method showed high extraction efficiency of safflower pigments. Ultrasonic method gave a higher extraction yield of red and yellow safflower pigments than using general method. It is supposed that the extraction efficiency is to be attributed to high vibration energies from ultrasound and finally induced physical changes of the pigments. Furthermore, this study explored the effects of ultrasonic treatment into the extracted safflower pigments on dyeing of cotton fabrics. Ultrasonic treatment into the extracted pigments exhibited significantly improved dyeing properties for the cotton fabrics.

Ultrasonic Extraction of Phenolic Compounds from Laminaria japonica Aresch Using Ionic Liquid as Extraction Solvent

  • An ionic liquid-based ultrasonic-assisted extraction method has been successfully applied to the effective extraction of phenolic compounds from Laminaria japonica Aresch. Three kinds of 1-alkyl-3-methyl-imidazolium with different cations and anions were evaluated for extraction efficiency. The results showed that both the characteristics of anions and cations have remarkable effects on the extraction efficiency. In addition, the ionic liquid-based ultrasonic-assisted extraction procedure was also optimized on some extraction parameters, such as ultrasonic power, extraction time and solid-liquid ratio. Compared with the conventional solvent, the optimum approach gained the highest extraction efficiency within the shortest extraction time. Average recoveries of phenolic compounds were from 75.5% to 88.3% at three concentration levels.

Preparation of High Quality Safflower (Carthamus tinctorius L.) Seed Extract by High-Pressure Extraction Process

  • Safflower seed extract was prepared by a high-pressure extraction technology and its quality characteristics were compared to that of other conventional extraction techniques, such ultrasonic and reflux extractions. Safflower seeds were extracted with 80% aqueous ethanol by three above extraction methods, and further fractionated with Diaion HP-20 column chromatography to obtain a partially purified safflower seed extract (PPSSE). Among the three extraction techniques examined, the reflux extraction showed the higher yields of EtOH extract and PPE than the ultrasonic and high-pressure extractions. Levels of most phenolic compounds in the EtOH extract of safflower seed are higher in reflux and ultrasonic extractions than the high pressure extraction, but levels of two serotonin aglycones, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS), in PPSSE were higher in the high pressure extraction than the reflux and ultrasonic extractions. In addition, color values (L and a) of the PPSSE were higher in the high-pressure extraction than the reflux and ultrasonic extractions, although there were no significant differences in pH and UV maxima absorption spectra among three extraction techniques. These results indicate that the high-pressure extraction technology is a simple and effective extraction for preparation of a high quality of safflower seed extract containing CS and FS with anti-wrinkle activity.

Sustainable water extraction of anthocyanins in aronia (Aronia melanocarpa L.) using conventional and ultrasonic-assisted method

  • The demand for sustainable extraction of bioactive compounds from food matrices has been increasing. Water extraction of anthocyanins in aronia was investigated using conventional and ultrasonic-assisted methods. The optimum extraction conditions for the conventional method included a sample-to-water ratio of 1:40 g/mL, extraction temperature 71℃, and extraction time of 39 min. The optimized conditions for ultrasonic-assisted extraction were a sample-to-water ratio of 1:40 g/mL, extraction temperature 80℃, extraction time of 20 min, and an amplitude of 87.2 ㎛. The anthocyanin contents of the two extracts were 155.32 and 158.02 mg/100 g fresh weight, respectively. The major anthocyanins were cyanidin 3-galactoside (65% of the total) and cyanidin 3-arabinoside (30% of the total). The contents of individual anthocyanins and phenolic acids were not significantly different between the two optimized extracts.

초음파로 추출된 홍화색소의 특성 분석과 염색성 평가 (Physiochemical Properties and Dyeability of Safflower Colorants Extracted by Ultrasonic Treatment)

  • This study systematically investigated a method for extraction of safflower (Carthamus tinctorius Linnaeus) colorants by ultrasonic treatment. Compared to pigments productivity and cell wall structures of safflower after general and ultrasonic method, the ultrasonic method showed high extraction efficiency of safflower pigments due to destruction of safflower cell wall caused by high vibration energies. Microscopic analysis confirmed the hypothesis that the ultrasonic treatment of safflower caused its cell wall structure loosened and made efficient extraction of safflower pigments. And also, LC-MS/MS analysis revealed that productivities of the yellow and red safflower pigments by ultrasonic method were 21.9% and 14.6% higher, respectively, than those of pigments extracted by general method. The ultrasonic extracted yellow and red colorants could be used to dye not only natural fibers like cotton, silk and wool, but also synthetic fiber like nylon, and generally gave a better color tone than the general extracted colorants from safflower due to the affinities of red and yellow colorant on different fibers. As the yellow and red colorant were extracted by ultrasonic treatment in water, the K/S value on of 550/440nm of cotton and rayon was increased but in the case of silk and wool the change of this value was almost not detected. Finally, this technique might provide a solution to establish reproducibility and standardization for the extraction and dyeing methods on fabrics.

더덕 사포닌인 lancemasides의 최적 추출 방법 구명 (Determination the optimum extraction method for saponin lancemasides in Codonopsis lanceolata)

  • 본 연구는 더덕 사포닌의 최적 추출 방법을 구명하기 위해 수행되었다. 추출 방법에 따른 lancemasides 함량을 조사하기 위해 환류 추출(MeOH, BuOH), 열수 추출(Hot water), 초음파 추출(ultrasonic bath (40 kHz; continuous irradiation, interval irradiation), ultrasonicator (20 kHz))을 실시하였다. 그 결과, 총 lancemasides함량은 ultrasonic bath (MeOH; continuous irradiation) 추출에서 가장 높았고, ultrasonic bath (water; continuous irradiation) 추출>ultrasonic bath (MeOH; interval irradiation) 추출>ultrasonicator 추출>열수 추출>메탄올 환류 추출>부탄올 환류 추출 순으로 높았다. 따라서 더덕 사포닌의 최적 추출 방법은 MeOH을 이용한 초음파(ultrasonic bath, 40 kHz) 추출이었고, 추출 전 시료 건조 방법은 동결건조보다 음건으로 건조하는 것이 lancemasides 추출에 더 효과적이었다. 또한 초음파 추출 시 추출 시간은 2시간, 추출 온도는 $64^{\circ}C$에서 추출 하는 것이 효과적임을 확인할 수 있었다. 따라서 초음파(ultrasonic bath)추출은 기존에 주로 이용하던 환류 추출 및 분획을 실시했을 때 보다 많은 양의 사포닌(lancemasides)을 추출할 수 있었다. 그리고 추출 시간과 온도와 같은 추출 조건에 따라 aster saponin Hb가 lancemaside류로 성분 변화가 일어나는 것을 확인하였다. 특히 추출 온도가 증가함에 따라 aster saponin의 함량이 줄어들고, lancemaside A의 함량이 증가하는 것으로 보아 추출 온도가 사포닌 성분 변화에 영향을 주는 것으로 사료된다.

Taxus chinensis 유래 파클리탁셀 정제를 위한 초음파를 이용한 마이셀 추출 (Ultrasound-Assisted Micellar Extraction for Paclitaxel Purification from Taxus chinensis)

  • 본 연구에서는 식물세포 Taxus chinensis로부터 항암물질 파클리탁셀을 효율적으로 정제하기 위하여, 초음파 기반 마이셀 추출 공정을 개발하였다. 전통적 마이셀 공정(대조군)에서의 많은 추출 단계 및 긴 상 분리 시간 문제를 획기적으로 개선하였다. 초음파 파워 180 W, 초음파 조사 1.5 시간에서 가장 높은 파클리탁셀 수율(~96%, 2회 추출)을 얻었으며, 이는 대조군의 수율에 비해 24.7% 증가하였다. 또한 분배 계수(K)는 초음파 파워 180 W, 초음파 조사 1.5 시간에서 최대치(24.0)를 보였다. 파클리탁셀 순도에는 큰 차이가 없었으며, 초기 파클리탁셀의 순도(6.81%)가 정제 후 22%까지 증가하였다. 역 추출(back extraction)의 상 분리 시간은 대조군 대비 각각 40.7-56.2%(초음파 파워 80 W), 46.3-67.6%( 초음파 파워 180 W), 51.9-67.6%(초음파 파워 250 W) 감소하였다. 초음파 파워(80-250 W)와 초음파 조사 시간(0.5-2.5 시간)이 증가할수록 상 분리 시간이 감소하였다.

프로폴리스 농도가 항균활성에 미치는 영향 (The effect of propolis concentration on the antibacterial activity)

  • The objetives of this study are to set up optimum extraction temperature, time and organic solvent for propolis extraction, to investigate chemical properties, and to develop health foods from propolis preparation. In this study, ethanol and ultrasonic extracts method performed to optimum extraction temperature was at 60, $20^{\circ}C$, optimum extraction time was at 12, 4 hours and optimum extraction amount of solvent was at 20, 15 times of propolis weight. When various ethanol solutions were used, whereas flavonoid content was highest in 70, 80% aqueous ethanol, respectively. So the ultrasonic extracts method used gave better results than the ethanol extracts method in this work. Extraction of propolis with etanol and ultrasonic extracts method was performed by using the water and various concentrations of aqueous ethanol as solvent. Sensitivity of propolis samples to Staphylococcus aureus was investigated and the results were shown. Samples of water extract did not inhibit microbial growth, where as 50% aqueous ethanol extract the largest inhibitory zone for Staphylococcus aureus, then decreased inhibition with increasing ethanol concentrations.

카오스 특징 추출에 의한 용접 결함의 초음파 형상 인식 (Ultrasonic Pattern Recognition of Welding Defects Using the Chaotic Feature Extraction)

  • The ultrasonic test is recognized for its significance as a non-destructive testing method to detect volume defects such as porosity and incomplete penetration which reduce strength in the weld zone. This paper illustrates the defect detection in the weld zone of ferritic carbon steel using ultrasonic wave and the evaluation of pattern recognition by chaotic feature extraction using time series signal of detected defects as data. Shown in the time series data were that the time delay was 4 and the embedding dimension was 6 which indicate the geometric dimension of the subject system and the extent of information correlation. Based on fractal dimension and lyapunov exponent in quantitative chaotic feature extraction, feature value of 2.15, 0.47 is presented for porosity and 2.24, 0.51 for incomplete penetration The precision rate of the pattern recognition is enhanced with these values on the total waveform of defect signal in the weld zone. Therefore, we think that the ultrasonic pattern recognition method of weld zone defects of ferritic carbon steel by ultrasonic-chaotic feature extraction proposed in this paper can boost precision rate further than the existing method applying only partial waveform.

초음파 추출법에 의한 감태 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성 분석 (Phenolic Content, DPPH Radical Scavenging, and Tyrosinase Inhibitory Activities of Ecklonia cava Extracted with the Ultrasonic Wave Method)

  • 해조류 유래 유용성분 추출을 위한 초음파 추출법의 적용성을 검토하였다. 동결건조한 감태 분말로부터 열수($65^{\circ}C$), 물($24^{\circ}C$), 50% 에탄올 및 100% 에탄올 뿐만 아니라 초음파 추출을 수행하여, 추출물을 제조하였다. 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성을 분석하고, 초음파 추출과 전통적인 추출법의 차이를 비교하였다. 초음파 추출에 의한 감태 추출물의 페놀성 화합물의 함량(142.80 mg/g)은 열수 추출물의 페놀성 화합물 함량(10.03 mg/g)의 약 14배 이상 높았다. 열수 추출물에 비해 초음파 추출물의 DPPH 라디칼 소거 활성은 약 4배, tyrosinase 저해 활성은 약 14배 이상 높은 것으로 나타났다. 특히 초음파 추출법에 의한 추출물에서의 페놀성 화합물 함량과 DPPH 라디칼 소거활성 및 페놀성 화합물 함량과 tyrosinase 저해 활성간의 상관관계가 매우 높은 것(각각 $R^2$=99.47 및 99.99)으로 나타났다. 이상의 결과는 전통적인 용매 추출법보다 초음파 추출법이 감태의 유용성분 추출에 더 적합함을 의미한다.
