Physicochemical, Microbial, and Sensory Properties of Yogurt with Ulmus davidiana var. japonica During Storage (느릅 분말을 첨가한 요구르트의 이화학적 및 관능적 품질 특성)

  • This study evaluates changes in yogurt with Ulmus davidiana var. japonica (U. davidiana) in terms of its physicochemical, microbial, and sensory properties during storage. The pH value and mean microbial counts of this yogurt with 0.2 to 0.8% of U. davidiana added ranged from 4.1 to 4.3 and from $1.4{\times}10^8$ to $2.3{\times}10^8cfu/mL$, respectively, during storage at $7^{\circ}C$ for 16 d. In terms of its color, the L value was significantly higher in the control and U. davidiana yogurt 0.2 to 0.4% up to 4 d, than other yogurt samples, and the b value was influenced by the addition of U. davidiana over 0.6% throughout the storage. According to a sensory test, there was a significant difference in the yellowness score between U. davidiana yogurt and the control, but earthy, and bitterness, and viscosity scores were similar. Overall acceptability was not significantly influenced after 8 d of storage in all yogurt samples, including the control. According to these results, concentrations (0.2 and 0.4%) of U. davidiana yogurt had no significantly adverse effects on its physicochemical, microbial, and sensory properties.

  • Ulmus genus has excellent various physiological activities, including anti-ulcer, antioxidant, antibacterial, anti-cancer, immunity, and homeostasis maintenance effects, and it is known to have many additional drug effects And one of reasons for these excellentbiological activities is a flavan-3-ol chemical group in Ulmus genus. In this study a new flavan-3-ol compound was identified in Ulmus davidiana var. japonica. A flavan-3-ol,(2R,3S)-7-[(2S,3R,4R)-3,4-dihydroxy-4-(hydroxymethyl)oxolan-2-yl]oxy-2-(3,4-dihydroxyphenyl)-3,4-dihydro-2H-chromene-3,5-diol, named as catechin 7-O-beta-D apiofuranoside, was isolated from the stems and barks of Ulmus davidiana var. japonica for. suberosa, which is a species belonging to the genus Ulmus, growing throughout the Korea peninsula. The structure was elucidated by various spectroscopic methods including high-resolution TOF mass spectrometry, 1H-NMR and 13C-NMR spectrometry and comparison with chemical structures of defined compounds.

Anti-Inflammatory Activity of Constituents Isolated from Ulmus davidiana var. japonica

  • Twenty six compounds (1-26) were isolated from the root barks of Ulmus davidiana var. japonica. The anti-inflammatory activity of the isolated compounds were evaluated agai nst the generation of inflammatory chemical mediators in bone marrow-derived mast cells. Among them, compounds 10, 11, 13, 15 and 19 inhibited not only cyclooxygenase-2 dependent prostaglandin $D_2$ generation but also 5-lipoxygenase dependent leukotrien $C_4$ generation in a concentration-dependent manner. In addition, compounds 11, 12, 13, 15 and 19 also inhibited $\beta$-hexosaminidase release, a marker of mast cell degranulation reaction, from bone marrow-derived mast cell. These results suggest that the anti-inflammatory activity of U. davidiana might in part occur by both the inhibition of eicosanoid generations and the degranulation reaction of mast cells.

  • The extract of Ulmus davidiana var. japonica cortex has known as natural anti-inflammatory substance in East Asia. For the identification of antimicrobial substance, it was extracted by using methanol and fractionated by using different organic solvents. The fraction of butanol was represented the highest antimicrobial activities. Therefore, the butanol fraction was purified and identified the chemical structure by $^1$H and $^{13}$ C-NMR spectra, FT-IR and EI/MS spectroscopies. The isolated antimicrobial substance was identified as cis-2-[3,4-dihydroxy phenyl]-3,4-dihydro-2H-1-benzopyran-3,5,7-triol, which has commonly known as (-)epicatechin. Its minimum inhibitory concentrations (MICs) against Staphylococcus aureus and Listeria monocytogenes were shown as 100 $\mu\textrm{g}$/mL, respectively.

Anti-cancer Potentials of Rhus verniciflua Stokes, Ulmus davidiana var. japonica Nakai and Arsenium Sublimatum in Human Gastric Cancer AGS Cells (AGS 인체위암세포에서 건칠, 유근피 및 신석 추출물의 항암 활성 비교 연구)

  • The anti-cancer activities of Rhus verniciflua Stokes (GC), Ulmus davidiana var. japonica Nakai (UGP) and arsenium sublimatum (SS) extracts, which have been used Oriental medicine therapy for various diseases, were investigated. The treatment of GC, UGP and SS alone, and combined treatment with GC, UGP and SS did not affect the cell viability in the mouse normal cell lines (RAW 264.7 macrophages and C2C12 myoblasts). However, co-treatment with GC, UGP and SS markedly induces apoptosis in human gastric cancer AGS cells, but not in other various cancer cell lines (human lung cancer A549, colon cancer HCT116, liver cancer Hep3B and bladder T24 cells) as evidenced by formation of apoptotic bodies, chromatin condensation, and accumulation of annexin-V positive cells. Co-treatment with GC, UGP and SS effectively induced the expression levels of Fas and Fas ligand, and inhibited the levels IAP family proteins such as XIAP, cIAP-1 and survivin, and anti-apoptotic Bcl-xL proteins compared with treatment with either agent alone. Combined treatment also significantly induced the loss of mitochondrial membrane potential, which was associated with the activation of caspases (-3, -8, and -9) and degradation of poly (ADP-ribose) polymerase. However, the cytotoxic effects induced by co-treatment with GC, UGP and SS were significantly attenuated by pan-caspases inhibitor, z-VAD-fmk, indicating an important role for caspases. These results indicated that the caspases were key regulators of apoptosis in response to co-treatment of GC, UGP and SS in human gastric cancer AGS cells and further studies will be needed to identify the active compounds.

Effects of Root Bark Components from the Elm (Ulmus davidiana var japonica) on Mastitis Pathogens (느릅나무 뿌리껍질 성분의 유방염균에 대한 항균효과)

  • The methanolic extracts from the elm (Ulmus davidiana var japonica) root barks which have been known as an oriental medicine Eubackpie for the treatment of edema, ulcer and inflammation were examined for their bacteriocidal effects on mastitis pathogens of milk cows. The growth of the pathogenic bacteria such as Staphylococcus aureus and Streptococcus sp. No. 12 was markedly inhibited by the chloroform fraction among the extracts, and these antibacterial activities were also supported by MIC test. Water-soluble mucilage was obtained from the root barks with a yield of 36.8~40.2% by dry weight basis. This substance appeared to be a kind of polysaccharide with $61.0{\pm}1.0%$ carbohydrate content, and to have hygroscopic and astringent properties due to its high viscosity and water absorption.

Antioxidant Activity of Ulmus davidiana var. japonica N. and Hemipteleae davidii P. (느릅나무와 시무나무의 항산화 활성)

  • Cortex of Ulmus davidiana var. japonica N. and Hemipteleae davidii P. have been used without classification according to countries as oriental medicine for treatment of various disorders. This study was conducted to elucidate the difference in antioxidant activity and total phenol content of these medicinal materials. Root cortex and trunk cortex of two medicinal plants were extracted with 80% ethanol $(at\;85^{\circ}C)$ and water (at room temperature), respectively. All of the extracts at $200{\sim}5\;{\mu}g/ml$, showed more effective scavenging activity on superoxide radical than ascorbic acid, and 80% ethanol and water extracts of Ulmus root cortex (URC) was more effective. Scavenging activities on DPPH radical of 80% ethanol extracts from URC and Ulmus trunk cortex (UTC) at 10 ${\mu}g/ml$, (41.4, 35.6%) were higher than those of the other six extracts and ${\alpha}-tocopherol\;(2.3{\sim}24.0%)$. Inhibitory activities on human low density lipoprotein (LDL) oxidation of 80% ethanol extracts from UTC and URC (84.3, 81.7%) at $10\;{\mu}g/ml\;(31.3{\sim}78.2%)$ were higher than those of the other six extracts and ${\alpha}-tocopherol$. However, antioxidant activities on linoleic acid peroxidation of all of the extracts were relatively lower than that ${\alpha}-tocopherol$. Total phenol content of 80% ethanol extract from URC was the highest value as 50.8% and that of water extract from HRC was the lowest as 5.6% among the extracts. From these results, it is suggested that Ulmus root cortex was the most effective in antioxidant activity.

Site Characteristics, and Vegetation Structure, and Dynamics of Forest Communities growing Kalopanax septemlobus (Thunb.ex Murray) Koidz. in Gangwon-do (강원도 음나무 자생 임분의 입지환경, 식생구조 및 동태)

  • Site characteristics, vegetation structure and dynamics of Kalopanax septemlobus forests in Mt. Heungjeong, Mt. Balwang and Mt. Gariwang were studied to provide the information need for national resources conservation and restoration of K. septemlobus forest in Gangwon-do of Korea. The K. septemlobus forests were distributed at 780m to 1,300m in altitude, steep slope, ridges-slope areas with facing northeast to northwest in good nutrition area. The annual diameter growth of those trees ranged 1.60mm to 2.41mm. Forty-three plots(20$\times$20m) were set up at three natural population of Kalopanax septemlobus forest and TWINSPAN method was used for vegetation structure analysis. The result of TWINSPAN ordination show that geographical features played a role in determining community types in the study areas. Community type of the study areas classified into three groups by TWINSPAN; K. septemlobus-Quercux mongolica- Acer mono community(Mt. Balwang), K. septemlobus-Q. mongolica- Tilia amurensis community(Mt. Heungjeong), K. septemlobus-Betula costata-Q. mongolica community(Mt. Gariwang). In most of the K. septemlobus forests, major woody species appeared predominantly were Q. mongolica, T. amurensis, Acer pseudo-sieboldianum, Lindera obtusiloba, A. mono, Magnolia sieboldii, Ulmus davidiana var. japonica. And also, Those species were surveyed in all the areas. Species diversity(H') of investigated areas ranged 0.3124~0.13992 and this value relatively higher than that of other forests with similar site. The range of similarity indices between surveyed areas was 64.1~64.54%. The distribution pattern of Morista's index show that Kalopanax septemlobus distributed randomly in tree and concentrated a few sample plots in subtree layer and shurb layer. K. septemlobus occupied 37.1~58.8% of tree layer story, 8.4~17.6% of subtree layer, 0~0.38% of shurb layer each surveyed area. The association analysis showed that Kalopanax septemlobus positively associated with Betula ermani, Deutzia glabrata, Carpinus cordata, Fraxinus mandshurica and negatively associated with Symplocos chinensis var. pilosa, Styrax obassia, Acer mono, Euonymus macroptera, Ulmus davidiana var. japonica. According to the diameter distribution of Kalopanax septemlobus community, the community of Mt. Gariwang will gradually decline and replaced by Quercus community. The communities of Mt. Heungjeong and Mt. Balwang has its seedlings and saplings, so it may be sustained.

Bioactivities and Isolation of Functional Compounds from Decay-Resistant Hardwood Species (고내후성 활엽수종의 추출성분을 이용한 신기능성 물질의 분리 및 생리활성)

  • Wood of Robinia pseudoacacia and bark of Populus alba$\times$P. glandulosa, Fraxinus rhynchophylla and Ulmus davidiana var. japonica were collected and extracted with acetone-water(7:3, v/v) in glass jar to examine whether its bioactive compounds exist. The concentrated extracts were fractionated with hexane, chloroform, ethylacetate and water, and then freeze-dried for column chromatography and bioactive tests. The isolated compounds were sakuranetin-5-O-$\beta$-D-glucopyranoside from Populus alba $\times$Pl glandulosa, 4--ethyoxy-(+)-leucorobinetinidin frm R. pseudoacacia and fraxetion from F. rhynchophylla and were characterized by $^1H$ and$^{13}C $ NMR and positive FAB-MS. Decay-resistant activity was expressed by weight loss ratio and hyphae growth inhibition in the wood dust agar medium inoculated wood rot fungi. R. pseudoacacia showed best anti-decaying property in both test and its methanol untreated samples, indicating higher activity than methanol treated samples in hyphae grwoth test. In antioxidative test, $\alpha$-tocopherol, one of natural antioxidants, and BHT, one of synthetic antioxidants, were used as references to cmpare with the antioxidant activities of the extacted fractions. Ethylacetate fraction of F. rhynchophylla bark indicated the hightest activity in this test and all fractions of R. pseudiacacia extractives also indicated higher activities compared with the other fractions. In the isolated compounds, aesculetin isolated from F. rhynchophylla bark showed best activity and followed by robonetinidin from R. pseudoacaica.
