참느릅나무의 성분에 관한 연구 (Studies on the Constituents of Ulmus parvifolia)

  • The bark of Ulmus parvifolia Jacq. (Ulmaceae) has been used for the treatment of gonorhea, edema, scabies and eczema marginatum. Previous investigations conducted with the heartwood and leaves have demonstrated it to contain sesquiterpenes as well as fat acids from the heartwood and flavonol glycosides from leaves. However, no phytochemical work has been done on the bark parts of this plant. Investigation of the phytochemical constituents in the barks of U. parvifolia has resulted in the isolation of sterols, sterol glucoside and a catechin glycoside, $(+)-catechin\;7-O-{\alpha}-{_L}-rhamnopyranoside$, all of which were isolated for the first time from this plant. Sterols were consisted of the three components, ${\beta}-sitosterol$, stigmasterol and campesterol in a ratio of 92.1:4.1:3.8, and sterol glucoside was identified as ${\beta}-sitosterol\;3-O-{\beta}-{_D}-glucoside$. The structure of the catechin $7-O-{\alpha}-{_L}-rhamnoside$ was established primarily by analysis of $^1H-and$ COSY-45 NMR, HMQC and HMBC and EI mass spectra of the heptaacetate. Especially, HMBC spectrum provides effective way for the determination of the point of attachment of the rhamnosyl group to catechin moiety.

Antioxidant and Anti-Nociceptive Activities of Ulmus davidiana var. japonica

  • Some pharmacological activities of Ulmus davidiana var. japonica were evaluated using its methanol extract (UDE). An acute anti-inflammatory activity of UDE was assessed using carrageenan-induced hind paw edema in rats. UDE exhibited an antioxidant activity when assayed by a stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH). Dose-dependent anti-nociceptive activity of UDE was assessed using the acetic acid-induced writhing test in mice. UDE was able to diminish the reactive oxygen species (ROS) level in the lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. UDE also suppressed production of nitric oxide and induction of inducible nitric oxide synthase and cyclooxygenase-2 in the stimulated macrophages cells. collectively, the results imply that U. davidiana var. japonica has antioxidant and anti-nociceptive activities in addition to anti-inflammatory activity.

Maslinic Acid, a Triterpenoid from the Root Barks of Ulmus davidiana var. japonica, Affects the Viability of HSC-T6 Hepatic Stellate Cells

  • Activation of hepatic stellate cells (HSCs) characterized by increased proliferation and extracellular matrix deposition is identified as the major pathological feature of hepatic cirrhosis. Therefore, suppression of HSC activation has been proposed as an important antifibrotic therapeutic strategy. In the present study, we investigated the antiproliferative activity of root barks of Ulmus davidiana var. japonica (Ulmaceae) by employing HSC-T6 hepatic stellate cells as an in vitro assay system. Further investigation of the n-hexane and $CHCl_3$ fractions of root barks of U. davidiana var japonica led to the isolation of six triterpenoids: friedelin (1), epifridelanol (2), oleanolic acid (3), maslinic acid (4), ${\beta}$-amyrin (5) and ${\alpha}$-amyrin (6), together with ${\beta}$-sitosterol (7) and daucosterol (8). Among these compounds, 2, 3 and 4 significantly inhibited HSC proliferation. In addition, 4 inhibited HSC proliferation in time- and concentration-related manners, via a partially direct toxic effect, as assessed by morphological changes and release of lactate dehydrogenase.

Antioxidant and Anti-inflammatory Activity of Stem Bark Extracts from Ulmus davidiana var. japonica

  • Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.

Ethanol Extract of Ulmus pumila Ameliorates Heat Stress through the Induction of Heat Shock Proteins Expression in RAW264.7 Macrophage Cells

  • Heat stress is a significant burden to animal production in most areas of the world. Improving our knowledge of physiological and metabolic mechanisms of acclimation may contribute to the development of procedures that may help to maintain health and production efficiency under hot temperature. The effect of Ulmus pumila (UP) extract in inducing Heat Shock Proteins (HSPs) expression in heat-stressed RAW264.7 macrophage cells was investigated. Cell viability assay showed a dose dependent increase in cells after treatment with UP for 24 hours. RT-PCR and western blot analysis showed that increasing concentrations of UP induce the expression of Heat Shock Factor 1 (HSF1) and dose dependently upregulated the expression of Heat shock protein 70 (Hsp70) and Hsp90. LPS-induced nitric oxide was dose-dependently reduced while phagocytic activity greatly recovered with UP treatment. These data demonstrated that UP can be a potential candidate in the development of cytoprotective agent against heat stress.

Investigations into the immunomodulatory activity of Ulmus davidiana Planch extracts

  • Objective: Although the effect of Ulmus davidiana Planch (UD) extracts on collagen-induced-arthritis (CIA) and bone metabolism has been studies, research on its effect on human immunomodulatory activity is further a due. The objective of the present study was to investigate the immunomodulatory activity of UD on cellular and humoral immunity. Methods : Oral administration of the ethanolic and water extracts of UD, at doses of 20, 100 and 200 mg/kg in mice, dose dependently potentiated the delayed type hypersensitivity reaction induced both by sheep red blood cells (SRBC) and oxazolone. Results : It significantly enhanced the production of circulating antibody titre in response to SRBC in mice. Extracts of UD failed to show any effect on macrophage phagocytosis. Chronic administration of UD extracts significantly ameliorated the total white blood cell count and also restored the myelosuppressive effects induced by cyclophosphamide. Conclusion : The present investigation reveals that UD extracts possesses immunomodulatory activity.

Antioxidant and Antidiabetic Activities of Ulmus davidiana Extracts

  • The antioxidant activities of water, ethanol, methanol, and chloroform extracts of Ulmus davidiana were evaluated using various antioxidant assays: DPPH (1,1-diphenyl-2-pricrylhydrazyl) free radical scavenging; hydroxyl radical scavenging; lipid peroxidation scavenging; and reducing power assays. All extracts, except the chloroform extract, demonstrated strong antioxidant activity in all assays. The chloroform extract had the highest hydroxyl radical scavenging activity, and its activity was equivalent to $\alpha$-tocopherol at a concentration of 0.5 mg/mL. Additionally, the antidiabetic activity of their extracts was also evaluated using a rat intestinal $\alpha$-glucosidase inhibition assay. Among all extracts investigated, the methanol extract had the highest $\alpha$-glucosidase inhibitory activity, although its activity was less than acarbose at $0.5\;{\mu}g/mL$. This result suggested that U. davidiana extracts may have antidiabetic activity. Total phenolic compounds and flavonoids were also measured. Phenolic compounds such as tannic acid, p-coumatric acid, and kaempferol were detected by high-performance liquid chromatography (HPLC). These results suggest that U. davidiana extracts may be useful as a potential source of antioxidant and antidiabetic materials.

환경 독성을 억제하는 느릅 당단백질이 마우스의 분뇨 악취저감 및 사료 효율에 미치는 영향 (Anti-ecotoxicological Glycoprotein Isolated from Ulmus davidiana Nakai Inhibits Fecal Malodor and Promotes Feed Efficiency in Mice)

  • Ulmus davidiana Nakai (UDN) has been traditionally used as a herbal medicine in Korea. In the present study, we investigated the anti-ecotoxic potential of a 116 kDa glycoprotein isolated from UDN (UDN glycoprot ein) in regulating fecal malodor and feed efficiency in mice. We found that UDN glycoprotein (200 μg/ml) has an inhibitory effect on the cell death induced by an ecotoxicological endocrine disrupting chemical, bisphenol A, in colon epithelial HT-29 cells. UDN glycoprotein did not show significant differences regarding the weight of ecotoxicity-related organs (liver, heart, kidneys, and spleen) and the levels of serum glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, and lactate dehydrogenase in mice for 2 weeks, compared to the control. Additionally, UDN glycoprotein reduced the levels of hydrogen sulfide and ammonia as markers of fecal malodor in mice. Interestingly, UDN glycoprotein can improve the mouse feed efficiency. In conclusion, our data indicate that anti-ecotoxicological UDN glycoprotein has the ability to increase the feed efficiency and reduce the fecal malodor by maintaining the viability of colonic epithelial cells in mice.

Inhibitory Effects of Ulmus parvifolia and Liriope platyphylla Wang et Tang on Histamine Release from Rat Peritoneal Mast Cells

  • Twenty-four different natural food materials extracted with 80% methanol were used to investigate the inhibition of cell-mediated immediate type allergic reactions induced by compound 48/80 in rat peritoneal mast cells (RPMCs). Nine 80% methanol extracts screened at a concentration of $10\;{\mu}g/mL$ inhibited histamine release from RPMCs induced by compound 48/80. Of these, two materials (Ulmus parvifolia and Liriope platyphylla Wang et Tang) were extracted and fractionated into four different solvent types (chloroform, ethylacetate, butanol, and water), and the fractions with major anti-allergic effects were assessed. The chloroform fraction of U. parvifolia (UP) at $5\;{\mu}g/mL$ and the ethylacetate fraction of L. platyphylla Wang et Tang (LPWT) at $1\;{\mu}g/mL$ showed the greatest inhibition of histamine release induced by compound 48/80. The chloroform fraction of UP and the ethylacetate fraction of LPWT in combination showed a greater inhibition of histamine release than either fraction alone. The cAMP levels in RPMCs treated with UP and LPWT were significantly greater than in cells treated with compound 48/80 alone. Our studies suggest that extracts from UP and LPWT may alleviate immediate type hypersensitivity reactions through the increase of cAMP levels in the mast cells.

Antimicrobial Activities of (-)Epicatechin from Ulmus davidiana var. japonica Cortex

  • The extract of Ulmus davidiana var. japonica cortex has known as natural anti-inflammatory substance in East Asia. For the identification of antimicrobial substance, it was extracted by using methanol and fractionated by using different organic solvents. The fraction of butanol was represented the highest antimicrobial activities. Therefore, the butanol fraction was purified and identified the chemical structure by $^1$H and $^{13}$ C-NMR spectra, FT-IR and EI/MS spectroscopies. The isolated antimicrobial substance was identified as cis-2-[3,4-dihydroxy phenyl]-3,4-dihydro-2H-1-benzopyran-3,5,7-triol, which has commonly known as (-)epicatechin. Its minimum inhibitory concentrations (MICs) against Staphylococcus aureus and Listeria monocytogenes were shown as 100 $\mu\textrm{g}$/mL, respectively.
