Teicoplanin 생산성이 우수한 Actinoplanes teichomyceticus ATCC31121 변이주 선별 및 배양학적 특성 (Screening and Characteristics of a Mutant of Actinoplanes teichomyceticus ATCC31121 Highly Producing Teicoplanin)

  • Teicoplanin은 Actinoplanes teichomyceticus가 생산하는 밴코마이신과 유사한 글리코펩티드계 항생제의 일종으로 메티실린 내성 황색포도상구균에 대해서도 효과가 우수한 치료제이다. 공시균주인 Actinoplanes teichomyceticus ATCC31121에 자외선을 조사하여 teicoplanin 생산성이 우수한 균주를 얻었다. 본 연구에서 우수 변이주 획득을 위한 UV 조사에 대한 균주의 치사곡선을 구하고 변이 유도 최적 조건을 확립하였다. 또한 한천 확산법을 사용하여 모균주와 변이주들의 teicoplanin 생산성 분포를 비교하였다. 변이주 가운데 가장 생산성이 높은 T1014-1를 최종적으로 획득하였으며 효소 활성도, 항생제 내성, 자가내성 및 teicoplanin 생산성을 조사하였으며 발효조에서 발효특성을 조사하였다.

The Site-Directed A184S Mutation in the HTH Domain of the Global Regulator IrrE Enhances Deinococcus radiodurans R1 Tolerance to UV Radiation and MMC Shock

  • IrrE is a highly conserved global regulator in the Deinococcus genus and contributes to survival from high doses of UV radiation, ionizing radiation, and desiccation. Drad-IrrE and Dgob-IrrE from Deinococcus radiodurans and Deinococcus gobiensis I-0 each share 66% sequence identity. However, Dgob-IrrE showed a stronger protection phenotype against UV radiation than Drad-IrrE in the D. radiodurans irrE-deletion mutant (ΔirrE), which may be due to amino acid residues differences around the DNA-binding HTH domain. Site-directed mutagenesis was used to generate a Drad-IrrE A184S single mutant, which has been characterized and compared with the ΔirrE mutant complemented strain with Drad-irrE, designated ΔirrE-E. The effects of the A184S mutation following UV radiation and mitomycin C (MMC) shock were determined. The A184S mutant displayed significantly increased resistance to UV radiation and MMC shock. The corresponding A184 site in Dgob-IrrE was inversely mutated, generating the S131A mutant, which exhibited a loss of resistance against UV radiation, MMC shock, and desiccation. qPCR analysis revealed that critical genes in the DNA repair system, such as recA, pprA, uvrA, and ddrB, were remarkably induced after UV radiation and MMC shock in the ΔirrE-IE and A184S mutants. These data suggested that A184S improves the ability against UV radiation and MMC shock, providing new insights into the modification of IrrE. We speculated that the serine residue may determine the efficiency of DNA binding, leading to the increased expression of IrrE-dependent genes important for protection against DNA damage.

CAPS marker에 의한 Arabidopsis의 자외선 B 감수성 유전자 지도작성 (Mapping of UV-B sensitive gene in Arabidopsis by CAPS markers)

  • Arabidopsis thaliana columbia의 종자에 EMS를 처리하여 돌연변이체들을 만들었고 이중 UV-B에 감수성이 높은 돌연변이체를 골랐다. 이 UV-B 감수성 돌연변이체의 원인 유전자를 밝히기 위하여 교배 실험을 한 결과 이는 Mendel 유전법칙을 따르고 단일 유전자의 돌연변이에 의하여 나타나며 열성 유전을 하는 것으로 밝혀져 이 유전자를 uvs라 하였다. 염색체상의 uvs의 위치를 밝히기 위하여 CAPS maker를 이용한 연관분석을 하고자 하였고 이를 위하여 각각 maker의 primer 10종류를 제작하였다. 이를 이용, 각 PCR 산물에 대하여 uvs mutant와는 다른 제한효소 pattern를 갖는 Lansberg와 uvs mutant를 교배시켜서 얻은 것들로부터 DNA를 추출하여 PCR을 수행하였다. 이들과 자외선과의 감수성을 연관시켜 교차율을 계산한 결과 5번 염색체의 LFY3과 가장 가까웁게 연관되어 있었다.

DNA손상 및 돌연변이에 대한 명지버섯의 방어효능 (The Protective Effects of Ganoderma lucidum on the DNA Damage and Mutagenesis)

  • Ganoderma lucidum is commonly known as medically potent mushroom, which has been widely used in China and other oriental countries for the treatment of various diseases, including cancer. In this report, we investigated the anti-oxidant and protective effect of Ganodema lucidum extract (GLE) against the DNA damage induced by free radical and U.V. In the assay of cell growth inhibition, the inhibitory cell growth rate induced by hydroxyl radical was dose-dependently decreased by GLE. This results support that GLE has a detoxifying activity against cytotoxicity of hydroxyl radical in E. coli cell. GLE also protected ColE1 plasmid DNA damage in the concentration of 200$\mu\textrm{g}$ per reaction on the DNA fragmentation assay. The nuclear tailing by hydrogen peroxide in single cell gel electrophoresis(SCGE) was decreased by GLE in the concentration of 50$\mu\textrm{g}$/ml. These data indicate that Ganoderma lucidum has an anti-oxidative activity to hydrogen peroxide. The mutation rate after irradiation of U.V. was reduced by 50$\mu\textrm{g}$/ml GLE and total number of Rif (Rifampicin) resistant mutants was decreased in a concentration dependent manner when added the GLE exogenously in a culture media. According to the results, it is likely that GLE has not only an anti-oxidative activity to hydroxyl radical but also an anti-mutagenic activity to U.V. mutagenesis.

Sisomicin 생산균의 돌연변이와 고생산 균주의 선별방법 (Mutagenesis of Sisomicin-producing Strains and Selection Method of High Producers)

  • Sisomicin 생산균인 Micromonospora inyoensis 균주를 변이유기체로 처리하였을 때 활성이 높은 균주들을 얻기 위한 최적조건으로는 각각 자외선(UV light) 처리에서 90%, nitrosoguanidine(NTG)에 대해 99%, nitrous acid에 대해 99.3%의 사멸율을 나타내는 경우였다. 이 때 분리된 colony들의 sisomicin생성능의 측정은 액체배양 대신 쉽고 간편한 agar plug법을 사용하였다. 한편, 변이유기제로 처리한 후 우량균주를 선택적으로 분리하기 위한 방법으로 aminoglycoside계통의 항생물질 혹은 중금속염을 함유한 농도구배 agar plate에서 생성된 균주들의 화성을 살펴본 결과, 항생물질로서는 gentamicin, kanamycin 이, 중금속염으로서는 Co, Hg가 효과적이었다.

  • Actinoplanes teichomyceticus ATCC 31121 was mutated with UV to obtain a superior mutant strain with increasing level of teicoplanin production. In this investigation lethal curve was obtained and the optimal condition to induce mutagenesis was determined and to isolate the desirable mutant strain. It was also confirmed that teicoplanin activities by agar diffusion method to be compared to the mother strain. One mutant strain, T991014-1 that had the highest teicoplanin productivity was finally selected for further investigation including fermentation pattern. The mutant was characterized by the various tests such as amylase activity, protease activity, antibiotic resistance, autotoxicity, and productivity.

Activity of Human Dihydrolipoamide Dehydrogenase Is Reduced by Mutation at Threonine-44 of FAD-binding Region to Valine

  • Dihydrolipoamide dehydrogenase (E3) is a member of the pyridine nucleotide-disulfide oxidoreductase family. Thr residues are highly conserved. They are at the active site disulfide-bond regions of most E3s and other oxidoreductases,. The crystal structure of Azotobacter vinelandii E3 suggests that the hydroxyl group of Thr that are involved in the FAD binding interact with the adenosine phosphate of FAD. However, several prokaryotic E3s have Val instead of Thr. To investigate the meaning and importance of the Thr conservation in many E3s, the corresponding residue, Thr-44, in human E3 was substituted to Val by site-directed mutagenesis. The mutant’s E3 activity showed about a 2.2-fold decrease. Its UV-visible and fluorescence spectra indicated that the mutant might have a slightly different microenvironment at the FAD-binding region.

Micromonosporas purpurea로부터 효율적 gentamicin 생산을 위한 균주 개발 (Strain Improvement for High Gentamicin Production Using Micromonosporas purpurea)

  • Micromonosporas purpurea로부터 효율적 gentamicin 생산을 위해 protoplast fusion와 protoplast mutagenesis 방법이 검토 되었다. $CO^{60}\;irradiation\;(2.3{\times}10^5$ units, UV 3 min) 방법에 의해서 MP3-112, MP3-141, MP3-143을 분리 했다. 특히 MP3-143균주는 최대 gentamicin생산량이 얻어졌다. 개량된 MP3-143균주를 이용해서 탄소원 소비, 균체성장, 그리고 gentamicin 생산량이 batch culture에서 비교되었다. MP3-413와 parent 균주의 glucose 소비는 배양 2일과 3일 후에 각각 완전히 이루어졌다. 그러나 균체성장과 Soybean oil 소비는 비슷한 결과 얻어졌다. Gentamicin최대 생산량은 배양 5일 후 29756 U/ml였다. 이 결과는 parent 균주에 비해 생산량이 5.6배 증가했다.

Salmonella typhimurium내로의 pKM101 돌연변이체의 유도와 그 특성에 관한 연구 (Induction and Chatacterization of pKM101 Mutants in Salmonella typhimurium)

  • Mutator effect를 가진 plasmid pKMlO1을 MNNG처리와 tetrazolium-glactcse 배지에서 MMS에, 의한 Salmonella typhimuriun의 Gal-에서 Gal+로 전환시키는 능력에 따라 pKM101 변이체들을 선택하고 이들의 성질을 chemical과 UV를 사용하여 pKM101과 비교 하였다. pKM101변이체들은 Salmonella typhimurium내에서 모두 안정하였고 정상적으로 다른 균주로 전달되었으며 picillin에 대한 저항성은 원래의 plasmid보다 같거나 높아졌다. 이들 중 두 변이체는 chemical mutagen인 MMS와 4-NQO, 그리고 UV에 의한 Salmonella typhimurimlla의 돌연변이들을 모체 pKMIOI보다 증가시켰으며 다른 변이체들은 이에 대한 효과가 감소 되었거나 전혀 잃은 것이었다. Salmonella typhimurium uvr- 변이주에 있어서 이들 변이체들의 chemical과 UV에 의한 돌연변이율에 미치는 치사효과는 모체 pKMIOI과 비슷한 양상을 나타내었다. 이러한 결과는 plasmid pKMOI의 chemical 및 UV mutator effect가 한 유전인자에 의해 결정된다는 것을 시사한다.

Improved Biosurfactant Production by Bacillus subtilis SPB1 Mutant Obtained by Random Mutagenesis and Its Application in Enhanced Oil Recovery in a Sand System

  • Biosurfactants or microbial surfactants are surface-active biomolecules that are produced by a variety of microorganisms. Biodegradability and low toxicity have led to the intensification of scientific studies on a wide range of industrial applications for biosurfactants in the field of environmental bioremediation as well as the petroleum industry and enhanced oil recovery. However, the major issues in biosurfactant production are high production cost and low yield. Improving the bioindustrial production processes relies on many strategies, such as the use of cheap raw materials, the optimization of medium-culture conditions, and selecting hyperproducing strains. The present work aims to obtain a mutant with higher biosurfactant production through applying mutagenesis on Bacillus subtilis SPB1 using a combination of UV irradiation and nitrous acid treatment. Following mutagenesis and screening on blood agar and subsequent formation of halos, the mutated strains were examined for emulsifying activity of their culture broth. A mutant designated B. subtilis M2 was selected as it produced biosurfactant at twice higher concentration than the parent strain. The potential of this biosurfactant for industrial uses was shown by studying its stability to environmental stresses such as pH and temperature and its applicability in the oil recovery process. It was practically stable at high temperature and at a wide range of pH, and it recovered above 90% of motor oil adsorbed to a sand sample.
