Extract of Ettlia sp. YC001 Exerts Photoprotective Effects against UVB Irradiation in Normal Human Dermal Fibroblasts

  • The identification of novel reagents that exert a biological ultraviolet (UV)-protective effect in skin cells represents an important strategy for preventing UV-induced skin aging. To this end, we investigated the potential protective effects of Ettlia sp. YC001 extracts against UV-induced cellular damage in normal human dermal fibroblasts (NHDFs). We generated four different extracts from Ettlia sp. YC001, and found that they exhibit low cytotoxicity in NHDFs. The ethyl acetate extract of Ettlia sp. YC001 markedly decreased UVB-induced cytotoxicity. Additionally, the ethyl acetate extract significantly inhibited the production of hydrogen peroxide-induced reactive oxygen species. Moreover, it inhibited UVB-induced thymine dimers, as confirmed by luciferase assay and thymine dimer dot-blot assay. Thus, the study findings suggest Ettlia sp. YC001 extract as a novel photoprotective reagent on UVB-induced cell dysfunctions in NHDFs.

Photoprotection by Topical DNA Repair Enzymes

  • Many of the adverse effects of solar UV exposure appear to be directly attributable to damage to epidermal DNA. In particular, cyclobutane pyrimidine dimers (CPD) may initiate mutagenic changes as well as induce signal transduction responses that lead to a loss of skin immune surveillance and micro-destruction of skin structure. Our approach is to reverse the DNA damage using prokaryotic DNA repair enzymes delivered into skin using specially engineered liposomes. T4 endonuclease V encapsulated in liposomes (T4N5 liposome lotion) enhanced DNA repair by shifting repair of CPD from the nucleotide excision to the base excision repair pathway. Following topical application to humans, increased repair limited UV-induction of cytokines, many of which are immunosuppressive. In a recent clinical study, topical treatment of UV-irradiated human skin with T4N5 liposome lotion reduced the suppression of the nickel sulfate contact hypersensitivity response. Similarly, the photoreactivating enzyme enhances repair by directly reversing CPDs after absorbing activating light. Here also treatment of UV-irradiated human skin with photoreactivating enzyme in liposomes and photoreactivating light restored the response to the contact allergen nickel sulfate. These findings confirm in humans the observation in mice that UV induced suppression of contact hypersensitivity is caused in part by CPDs. We have tested the ability of T4N5 liposome lotion to prevent UV-induced skin cancer in patients with xeroderma pigmentosum (XP), who have an elevated incidence of skin cancer resulting from a genetic defect in DNA repair. Daily use of the lotion for one year in a group of 20 XP patients reduced the average number of actinic keratoses by 68% and basal cell cancers by 30% compared to 9 patients in the placebo control group. Delivery of DNA repair enzymes to skin is a promising new approach to photoprotection.

백부자산(白附子散)이 자외선 조사된 피부 손상과 색소침착에 미치는 영향 (The Effects of Baickbujasan Extract on the Skin Damage and Pigmendation Induced by Ultraviolet Irradiation)

  • Objective : The purpose of this study is to examine the effects of Baickbujasan(BB) on the skin damage and depigmentation. Method : The inhibition of tyrosinase activity, melanogenesis and cell viability in cultured B16 melanoma cells were measured. In order to test effects of reduction of melanogenesis, B16 F-10 mouse melanoma stem line was employed to extract melanin from cultured cell, where BB was added or not, and was dissolved in alkali for colorimetric analysis. Also, in order to test skin alteration in C57BL/6 after UV irradiation, the animals were grouped into a UV urradiation group and UV irradiation after BB application group. Dopa oxidase tissue staining was excuted to invesitage the change in the distribution of active melanin cell. The distribution of active melanin cell in inner skin of iNOS after damage from UVB irradiation and the manifestation condition of P53 which takes part in natural death of keratinocyte were examined. Result : The results indicate that BB has significant effects on tyrosinase activity, and melanogenesis in vivo test. BB seems to reduce C57BL/6, external dermatological damage, for instance, erythematous papule, eczema, loss of keratinocyte, reduction in pus, and relieves dermatological damages. Conclusion : BB can be applied externally for UV protection and depigmentation.

자외선 조사 마우스에서 만성 피부손상에 대한 분죽(Phyllostachys nigra var. henenis Strapf)잎 추출물의 효과 (Photoprotective Effect of Bamboo (Phyllostachys nigra var. henenis Strapf) Leaf Extract against Ultraviolet Radiation-induced Chronic Skin Damage in the Hairless Mouse)

  • To evaluate the ability of Bamboo (Phyllostachys nigra var. henenis Strapf) leaf extract (BL) to protect the skin from photodamage, the gross and microscopic changes in the skin of hairless mice and BL-treated mice exposed chronically to ultraviolet (UV) were examined. The skin of the UV-irradiated mice showed characteristic signs of photoaging, such as deep wrinkles across the back, increased epidermal thickeness, numerous cell infiltration, and many enlarged keratinizing cysts. BL-treated mice showed a significantly decreased wrinkling score and lack of proliferation of cysts. By the 22nd week, 88.9% (i.p. with saline) or 60.0% (topical administration with cream base) of the UV-irradiated mice developed at least one tumor. BL delayed tumor onset significantly. BL (i.p.) was also effective in reducing the occurrence of UV radiation-induced skin tumors and reduced the number of tumors per mouse. After 22 weeks of treatment, 37.5% (i.p.) of the mice treated with BL were tumor-free. Tumor multiplicity was reduced by 81.2% (i.p.) in the BL treated groups. It is noted that skin that is chronically exposed to UV is subject to photoaging and photocarcinogenesis and regular use of BL would prevent these photodamaging effects of UV.

활성 RAW 264.7 세포에서 항염증 및 자외선 유도 마우스 피부손상의 개선에 대한 새송이 추출물의 효과 (Effects of Pleurotus eryngii extract against inflammation in activated RAW 264.7 cells and UV-induced skin damage in mice)

  • 본 연구는 먼저 새송이버섯 추출물인 PEE가 활성화된 면역세포에서 항염증 효과가 있는지 알아보았다. 그 결과 LPS로 자극된 RAW 264.7 세포에 PEE를 처리했을 때 5과 15 mg/mL 등 높은 농도에서 NO를 효과적으로 억제하였다. 그리고 LPS가 유도하는 $IL-1{\beta}$와 IL-6는 5 mg/mL 농도에서는 억제 없었으나, 15 mg/mL 농도에서 현저히 억제하는 효과가 있었다. 다음은 UVB가 유도하는 C57BL/6 마우스 피부손상에 대한 PEE의 개선 효과에 대해서 알아보았다. PEE를 7일간 경구투여한 후 7일, 10, 13일 등 3일 간격으로 하루에 1회씩 UVB ($250mJ/cm^2$)로 마우스 등 피부에 조사하여 피부손상을 유발하였다. UVB 조사 후부터는 PEE를 경구투여와 함께 피부에 도포하여 PEE가 피부 손상에 대한 개선 효과를 조사하였다. 그 결과 PEE 투여군은 UVB로 유도되는 표피두께, 홍반지수 및 멜라닌 지수가 참고약물로 사용한 비타민 C(AA) 투여군과 유사한 개선되는 효과가 있었다. 또한 PEE 투여군은 UVB가 유도하는 비만세포를 비롯한 염중세포 침윤이 현저히 억제되는 효과가 있었다. 이상의 결과를 종합해볼 때 PEE는 항염증 효과와 더불어 UVB가 유도하는 피부손상을 개선하는 우수한 효과가 있어 기능성 식품소재 및 화장품 소재로 활용할 수 있을 것으로 판단된다.

  • Fructose-1, 6-diphosphate (FOP), a glycolytic metabolite is reported to ameliorate inflammation and inhibit the nitric oxide production in murine macrophages stimulated with endotoxin. It is also reported that FOP has cytoprotective effects against hypoxia or ischemia/reperfusion injury in brain and heart. In this study, we examined whether FDP has protective effects on UV-induced oxidative damage in skin cell culture system and human skin in vivo. FDP had a protective role in UVB-induced LDH release and ROS accumulation in HaCaT although it did not show direct radical scavenging effect in the experiment using 1, 1-diphenyl-2-picrylhydrazyl (DPPH). FDP also preserved cellular GSH content after UV irradiation in HaCaT and normal human fibroblast culture system. Cellular oxidative stress induces multiple downstream signaling pathways that regulate expression of multiple gene including MMP-1 and collagen, we examined the effects of FDP on UV-induced alteration of these protein expression in fibroblast culture and human skin in vivo. The increased MMP-1 expression in fibroblast and human skin by UV irradiation was significantly decreased by FDP. FDP also prevented the UV-induced decrease of collagen expression in fibroblast and human skin. Moreover, the decreasing the intracellular levels of reducing equivalents in human fibroblast by glutathione (GSH) depletion lowered the UVA dose threshold for reduction of procollagen expression, indicating that the differences of glutathione contents define the susceptibility of fibroblasts towards UV-induced reduction of procollagen expression. FDP also preserved cellular GSH content after UV irradiation, indicating that FDP has protective effects on UV-induced reduction of procollagen expression, which are possibly through maintaining intracellular reducing equivalent. Based on these premises, we examined the effect of daily use of a moisturizer containing FDP on facial wrinkle in comparison with vehicle moisturizer lacking FDP. In the clinical study, FDP significantly decreased facial wrinkle compared with vehicle alone after 6 months of use. Our results suggest that FDP has anti-aging effects in skin by increasing cellular antioxidant system and preventing oxidative signal and inflammatory reaction. Therefore FDP may be useful anti-aging agent for cosmetic purpose.

Photoprotective effects of topical ginseng leaf extract using Ultraflo L against UVB-induced skin damage in hairless mice

  • Background: Abnormal activation of matrix metalloproteinases (MMPs) plays an important role in UV-induced wrinkle formation, which is a major dermatological problem. This formation occurs due to the degeneration of the extracellular matrix (ECM). In this study, we investigated the cutaneous photoprotective effects of Ultraflo L treated ginseng leaf (UTGL) in hairless mice. Methods: SKH-1 hairless mice (6 weeks of age) were randomly divided into four groups (8 mice/group). UTGL formulation was applied topically to the skin of the mice for 10 weeks. The normal control group received nonvehicle and was not irradiated with UVB. The UV control (UVB) group received nonvehicle and was exposed to gradient-UVB irradiation. The groups (GA) receiving topical application of UTGL formulation were subjected to gradient-UVB irradiation on $0.5mg/cm^2$ [GA-low (GA-L)] and $1.0mg/cm^2$ [(GA-high (GA-H)] of dorsal skin area, respectively. Results: We found that topical treatment with UTGL attenuated UVB-induced epidermal thickness and impairment of skin barrier function. Additionally, UTGL suppressed the expression of MMP-2, -3, and -13 induced by UVB irradiation. Our results show that topical application of UTGL protects the skin against UVB-induced damage in hairless mice and suggest that UTGL can act as a potential agent for preventing and/or treating UVB-induced photoaging. Conclusion: UTGL possesses sunscreen properties and may exhibit photochemoprotective activities inside the skin of mice. Therefore, UTGL could be used as a potential therapeutic agent to protect the skin against UVB-induced photoaging.

한방미용제제(자음미단)의 Hairless Mouse Model에서의 피부광노화 억제 및 섬유아세포의 UVB손상 저해효과 (Protective Effects of Jaummi-dan (Ciyinmei-dan) against Skin Photoaging in Hairless Mouse Model and UVB-induced Damage in Human Fibroblasts)

  • Objectives : To investigate the photoprotective effects and efficacy of Jaummi-dan (Ciyinmei-dan) on UV damage to animal skin/fibroblast cultures. Methods and Results : Hairless mice were orally administered Jaummi-dan (Ciyinmei-dan) extract and irradiated with UV for four weeks. In the Jaummi-dan (Ciyinmei-dan) treated group, a better skin appearance and less wrinkles were observed when compared to the control group. In addition, immunostaining for type 1 pN collagen showed that the amount of collagen deposition was higher in the Jaummi-dan (Ciyinmei-dan) treated group. The interstitial collagenase was measured in the cultured medium of fibroblasts after UVB irradiation using ELISA for MMP-l. Jaummi-dan (Ciyinmei-dan) treatment resulted in a significant decrease in MMP-1 secretion compared to the UVB-irradiated, untreated group. Conclusions : The results of our study indicate that orally administered Jaummi-dan (Ciyinmei-dan) seems to have photoprotective effects on UV damaged hairless mouse skin partly due to an inhibitory effect on collagen breakdown.

자외선으로 손상을 유도한 피부섬유아세포에서 스피룰리나 유래 피코시아닌의 보호 효과 (The Protective Effect of Spirulina-derived Phycocyanin on Dermal Fibroblasts Induced by UV Rays)

  • UV는 산화 스트레스를 유발하고 MMP(Matrix Metalloproteinase) 발현을 증가시켜 피부 노화를 발생시킨다. 따라서 자외선로 인한 피부 손상을 예방하면 피부 노화를 감소시킬 수 있다. 스피룰리나는 강력한 항산화제로 원핵생물로 구성되어 있다. 본 연구는 피부 섬유아세포를 사용하여 UVB 방사선에 대한 스피룰리나 유래 피코시아닌(PC)의 광보호 효과를 조사했다. 그 결과, PC는 섬유아세포 생존율 측면에서 5-40 ㎍/mL 농도에서 독성을 나타내지 않았다. UVB 조사된 섬유아세포의 생존율은 50.5%였으며 PC 처리로 73.5%로 증가했다. MMP-1 및 MMP-9 발현은 UVB 처리로 증가하는 반면 PC 처리한 군에서 감소했다. 이러한 결과를 종합해보면 PC는 UVB 조사로 인한 산화적 손상과 피부노화와 관련된 인자를 감소시켜 노화를 예방할 수 있을 것으로 사료된다.

천연물 도포가 UVB 파로 손상된 C57BL/6 mouse 피부의 색소침착과 염증생성에 미치는 영향 (Effects of Natural Extracts on UVB-induced Pigmentation and Inflammation in C57BL/6 Mouse Skin)

  • Ultraviolet(UV) radiation causes a variety of biological effects on the skin, including inflammation, pigmentation, photoaging and cancer. Free radicals are involved in inflammatory skin reactions induced by UVB radiation. In this study, we investigated the effects of antioxidants(Tea, Korean red ginseng, Ginkgo biloba extract) on UVB-induced skin damage. Tea, KRG and EGb 761 were topically treated to dorsal skin of ICR mouse. The mice were also treated soon after IMED ($1.4KJ/m^{2}$) of UVB irradiation. Skin pigmentation of irradiated mouse was observed by a chromameter after 2 weeks. Topical application of Tea, KRG and EGb 761 for 2 weeks decreased skin pigmentation compared to DVB control group(p<.05). Tea, KRG and EGb 761 also reduced UVB-induced infiltration of inflammatory cells. These results showed that Tea, KRG and EGb 761 as a topical application may have preventive effect against UVB-induced skin damage.
