A new concept of "photo" -antisense method has been evaluated, where the inhibition of gene expression by the conventional antisense method is enhanced by photochemical binding between antisense oligonucleotides conjugated with photo-reactive compound and target mRNA or DNA. Fluorescein labeled oligodeoxyribonucleotides (F-DNA) was delivered to cell nuclei in the encapsulated form in multilamellar lecithin liposomes with neutral charge. F-DNA was previously shown to photo-bind to the complementary stranded DNA, and the delivery system using neutral liposome to be effective in normal human keratinocytes. In the present study, we used human kidney cancer G401.2/6TG.1 cell line to be advantageous in reproducible experiments. p53 was adopted as a target gene since antisense sequence information has been accumulated. The nuclear localization ofF-DNA was identified by comparing the fluorescence ofF-DNA with that of Hoechst 33258 under fluorescence microscope. After 7hr incubation to accumulate p53 protein induced by UV -B, p53 protein was quantified by Western blot. After 2hrs from F-DNA application, about 30% of cell population incorporated F-DNA in their nuclei with some morphological change possibly due to liposomal toxicity. Irradiation of visible light longer than 400nm from solar simulator at this time enhanced the inhibitory action of antisense F-DNA. The present results suggest that photo-antisense method is promising to control gene expression in time and space dependent manner. Further improvement of F-DNA delivery to cancer cells in the stability and toxicity is in progress. progress.
SOS response is a conserved response to DNA damage in prokaryotes and is negatively regulated by LexA protein, which recognizes specifically an "SOS-box" motif present in the promoter region of SOS genes. Myxococcus xanthus DK1622 possesses a lexA gene, and while the deletion of lexA had no significant effect on either bacterial morphology, UV-C resistance, or sporulation, it did delay growth. UV-C radiation resulted in 651 upregulated genes in M. xanthus, including the typical SOS genes lexA, recA, uvrA, recN and so on, mostly enriched in the pathways of DNA replication and repair, secondary metabolism, and signal transduction. The UV-irradiated lexA mutant also showed the induced expression of SOS genes and these SOS genes enriched into a similar pathway profile to that of wild-type strain. Without irradiation treatment, the absence of LexA enhanced the expression of 122 genes that were not enriched in any pathway. Further analysis of the promoter sequence revealed that in the 122 genes, only the promoters of recA2, lexA and an operon composed of three genes (pafB, pafC and cyaA) had SOS box sequence to which the LexA protein is bound directly. These results update our current understanding of SOS response in M. xanthus and show that UV induces more genes involved in secondary metabolism and signal transduction in addition to DNA replication and repair; and while the canonical LexA-dependent regulation on SOS response has shrunk, only 5 SOS genes are directly repressed by LexA.
Ultraviolet B (UVB) radiation induces the production of reactive oxygen species (ROS) that promote apoptotic cell death. We showed that cytosolic $NADP^+$-dependent isocitrate dehydrogenase (IDPc) plays an essential role in the control of cellular redox balance and defense against oxidative damage, by supplying NADPH for antioxidant systems. In this study, we demonstrated that knockdown of IDPc expression by RNA interference enhances UVB-induced apoptosis of immortalized human HaCaT keratinocytes. This effect manifested as DNA fragmentation, changes in cellular redox status, mitochondrial dysfunction, and modulation of apoptotic marker expression. Based on our findings, we suggest that attenuation of IDPc expression may protect skin from UVB-mediated damage, by inducing the apoptosis of UV-damaged cells.
In order to study on the color change of silk dyed with natural colorant due to light fading, and find out the effect of combination dyeing, colorant extracts of safflower red, safflower yellow and amur cork tree were used, either singly or in combination. In combination dyeing, safflower yellow or amur cork tree dyeing process was added on the top of the silk fabric was dyed with safflower red. Color change and light fastness were investigated by $L^*,\; a^*,\; b^*$ H, V/C, and Color difference. Brightness of silk fabric dyed with safflower red and safflower yellow increased gradually with increasing the radiation time of UV light, but amur cork tree was decreased and turned to dull. Color difference of dyed with Amur cork tree showed higher than the others. Combination dyeing of safflower red and amur cork tree provided better light fastness than the one of safflower red and safflower yellow.
The immunological mechanism of the responses to ultraviolet (UV) B radiation in mouse models were investigated by the suppression of contact hypersensitivity (CHS) and delayed type hypersensitivity (DTH), and susceptibility to infection. However, there are some differences in immune suppression according to the different models as well as the irradiation protocols. Therefore, this review focused on the differences in the suppressive effects on CHS and DTH, and susceptibility to infection in relation to the different in vivo models. Recent advances in cytokine knockout mice experiments have the reexamination of the role of the critical cytokines in UVB-induced immune suppression, which was investigated previously by blocking antibodies. The characteristics of the suppressor cells responsible for UVB-induced tolerance were determined. The subcellular mechanism of UVB-induced immune suppression was also explained by the induction of apoptotic cells through the Fas and Fas-ligand interaction. The phagocytosis of the apoptotic cells is believed to induce the production of the immune suppressive cytokine like interleukin-10 by macrophages. Therefore, the therapeutic UVB response to a skin disease, such as psoriasis, by the depletion of infiltrating T cells could be considered in the extension line of apoptosis and immune suppression.
자외선(UV)은 인체 피부 손상의 주된 요인이고 UV에 대한 피부의 초기 반응들은 홍반과 부종을 포함한다. 화장품에 사용할 효과적인 자외선 방어제를 발굴하기 위하여 세포수준의 효능평가법으로 다양한 식물 추출물들을 검색하였다. 시험된 총 38종의 식물 추출물들 중 제주조릿대(Sasa quelpaertensis), 접시꽃(Althaea rosea), 관중(Dryopteris crassirhizoma)에서 유래한 3종의 식물 추출물이 배양된 인체 표피 멜라닌세포에서 멜라닌 생성뿐만 아니라 UVB의 세포독성을 경감시켰다. 이 식물 추출물들의 항염증 작용은 동물 실험에서도 평가하였다. 대조 크림 혹은 식물 추출물을 1 % 함유한 시험 크림을 C57BL/6 쥐의 귀에 혹은 SKH?1 무모쥐의 등에 UVB 조사 전과 후에 도포하였다. UVB 노출에 의한 영향으로서 부종 생성은 귀의 두께 변화를, 홍반 생성은 등 피부의 붉기의 변화를 측정하여 추정하였다. 3종류의 시험 크림 모두 두 실험 모두에서 항염증효과를 보였다. 제주조릿대, 접시꽃, 또는 관중 추출물을 함유한 크림은 자외선 조사 4일째 부종 반응을 각각 53.8 %, 56.4 %, 31.1 % 경감시켰다. 또한 이 크림들은 자외선 조사 2일째 홍반 생성을 각각 45.7 %, 34.1 %, 20.5 % 억제시켰다. 본 연구는 화장품에 함유된 특정 식물 추출물들이 자외선 과다노출로 야기되는 피부 염증을 완화시켜 줄 수 있음을 시사하였다.
PEP-1 peptide has been used for transduction of native protein into mammalian cells. This work describes the findings that the fusion of PEP-1 to target proteins led to protein truncation likely in a non-protein-specific manner. Approximately 75% of PEP-1-MsrA fusion protein was truncated in the N-terminal region of MsrA between Lys-27 and Val-28 during expression in Escherichia coli and purification. This large protein truncation was also observed in another PEP-1 fused protein, PEP-1-MsrB2, in the N-terminal region of MsrB2. The full-length PEP-1-MsrA protein was rapidly transduced into keratinocyte cells within 15 min. The transduced PEP-1-MsrA was functionally active and could protect skin cells against oxidative stress- and ultraviolet radiation-induced cell death. Collectively, our data demonstrated the protective roles of MsrA in skin cells and, moreover, may raise a concern of protein truncation caused by fusion of PEP-1 about the general use of this peptide for protein transduction.

The human eye, constantly exposed to solar radiation, can be damaged by UV radiation. In particular, ultraviolet B (UVB)-induced damage plays an important role in retinal degeneration and cell aging. In this study, we investigated the protective effects of the methanol extract of Oxya chinensis sinuosa (OCM), an edible insect known for its high protein content (64.2%), and various pharmacological effects, on human retinal pigment epithelial cells. ARPE-19 cells were treated with OCM and subsequently UVB irradiated. Our results showed that OCM effectively attenuates UVB-induced cell damage by reducing MAPK phosphorylation (JNK and p38 MAPK). Additionally, OCM increased the phosphorylation of Akt, and cell cycle regulators, including p21 and p27, in a dose-dependent manner. Moreover, OCM treatment increased ARPE-19 cell proliferation by activating the S6K1/S6 pathway. This study suggests that OCM prevents UVB-induced retinal cell damage by increasing cell proliferation via ROS reduction, suggesting its potential as a functional therapeutic superfood against retinal cell damage.
Background: Solar furnaces are used worldwide to conduct experiments to demonstrate the feasibility of solar-chemical processes with the aid of concentrated sunlight, or to qualify high temperature-resistant components. In recent years, high-flux solar simulators (HFSSs) based on short-arc xenon lamps are more frequently used. The emitted spectrum is very similar to natural sunlight but with dangerous portions of ultraviolet light as well. Due to special benefits of solar simulators the increase of construction activity for HFSS can be observed worldwide. Hence, it is quite important to protect employees against serious injuries caused by ultraviolet radiation (UVR) in a range of 100 nm to 400 nm. Methods: The UV measurements were made at the German Aerospace Center (DLR), Cologne and Paul-Scherrer-Institute (PSI), Switzerland, during normal operations of the HFSS, with a high-precision UV-A/B radiometer using different experiment setups at different power levels. Thus, the measurement results represent UV emissions which are typical when operating a HFSS. Therefore, the biological effects on people exposed to UVR was investigated systematically to identify the existing hazard potential. Results: It should be noted that the permissible workplace exposure limits for UV emissions significantly exceeded after a few seconds. One critical value was strongly exceeded by a factor of 770. Conclusion: The prevention of emissions must first and foremost be carried out by structural measures. Furthermore, unambiguous protocols have to be defined and compliance must be monitored. For short-term activities in the hazard area, measures for the protection of eyes and skin must be taken.
A decrease in stratospheric ozone probably caused by chloroflurocarbons (CFCs) emissions, has been observed large parts of-the globe. It is generally accepted that if ozone levels in the stratosphere are depleted, greater amounts of shortwave ultraviolet radiationB (UVB) will reach the earth's surface, resulting in increased incidence of nonmelanoma skin cancer. In this study, we evaluated several mathematical models, such as a power and an exponential model, and a geometric model considering the surface area of a human body part and ages for the prediction of Skin cancer incidence caused by exposure to the UVB radiation. These models basically estimated the risk of skin cancer based on those measurements of the local ozone in stratosphere and UVB. Both were measured at a part of Seoul with a Dobson ozone spectrometer and Robertson-Berger UV Biometer for 1995. As a result, we calculated the point estimation applying a biological amplification factor (BAF), UVB radiation and other factors. We used a Monte-Carlo simulation technique with assumption on the distribution of each considered factor. The sensitivity analysis of model by there components conducted using Gaussian sensitivity method. The annual integral of UVB radiation was 2275 MED (minimal erythema dose)/yr. Also, an estimate of the annual amount of UVB reaching the earth's surface at a korea's latitude and altitude was 3328 MED/yr. The values of the radiation amplification factor (RAF) were ranged from 0.9 to 1.5 in Seoul. To give the effective factors required to model the prediction of skin cancer incidence caused by exposure to the UVB radiation in Korea, we studied the pros and cons of above mentioned models with the application of those parameters measured in Seoul, Korea.
